Evaluation of Growth Inhibition Causes on Perennial Ryegrass(Lolium perennial L.) in Afforesting Area (인공배양토 식생지역에서의 페레니얼 라이그래스 생육저해 원인 평가)

  • To minimize the danger of soil erosion and settle habitats earlier, afforestation, which vegetates bare slopes, is selected as an environmental recovering technology. Large portions of these areas often are suffered by a bad germination and growth inhibition of sprayed seeds. Afforested materials collected in the normal and damaged sites were not any big difference in chemical characteristics and biological response to ryegrass. But background soil of the damaged site has very low pH (3.6) and high contents of iron and aluminum compared with them of the normal sites. Both germination and root growth of ryegrass were inhibited severely in the water extracts of damaged soils, but not in the water extracts of normal sites. Groundwater collected nearby the damaged sites was very strong acidic (pH 33) and exhibited a high value of electrical conductivity and high contents of iron and aluminum. In the ground water, germinated ryegrass was scarcely grown. In Al standard solution, the root growth of ryegrass was inhibited over 50% in 0.5 mM in pH 3.5-4.5 and in 1.4 mM in pH 5.5, which seems to be related to $Al^{3+}$ activity in solution. In the ferric Fe ($Fe^{3+}$) standard solution, ryegrass growth was inhibited over 50% in the concentration of 14-19 mM in root and 23-25 mM in shoot. This strong tolerance of ryegrass to $Fe^{3+}$ might be concerned with the very low activity of $Fe^{3+}$ at pH 3.5-5.5. In contrast, ryegrass responded very sensitively to ferrous Fe ion ($Fe^{2+}$), especially in root growth: $Fe^{2+}$ concentrations corresponding to 50% growth reduction were 0.3-0.4 mM at pH 3.5-5.5 in roots. This high growth inhibition should be related to the high ion activity of $Fe^{2+}$ irrespective of different pH conditions. In conclusion, low pH and high contents of $Fe^{2+}$ and aluminum seem to be caused by pyrite and be closely related to the growth inhibition of ryegrass seeded in afforested area.

The Organizational Citizenship Behavior and Organizational Effectiveness of Hospital Employees (병원근로자의 조직시민행동과 조직효과성 관계 연구)

  • Background: The organizational citizenship behavior is generally known as the important factor relevant to the organizational effectiveness. This research examined the mediating effect of the organizational citizenship behavior of hospital employees on the organizational effectiveness. Methods: Data were collected from 1,112 employees located in city of Seoul, Kyunggi and Chungnam province through self-administered questionnaires. Collected data were analyzed using IBM SPSS ver. 20.0, frequency analysis, t-test, analysis of variance, regression analysis, and path analysis. The main findings of the study are as follows. Results: First, it was found that many characteristics variables of personality, job, and relationship together affected organizational citizenship behavior of hospital employees. Especially, the following variables of negative affectivity, desire for growth, job value, job significance, and job security were found to have significant effect on the organizational citizenship behavior of hospital employees. Second, the results of path analysis showed that, through the mediating effect of organizational citizenship behavior, personality variables of positive and negative affectivity, and desire for growth, job characteristics variables of job value, job significance, and job security, and relationship variables of organizational support and task interdependence, had significant total effects on the level of job satisfaction of hospital employees. Conclusion: As a result, the organizational citizenship behavior seems to have both direct and indirect effects on the organizational effectiveness of hospital employees. Based on above findings, some theoretical and practical implications were discussed.

Dendritic Cell Based Cancer Immunotherapy: in vivo Study with Mouse Renal Cell Carcinoma Model (수지상세포를 이용한 항암 면역 치료: 생쥐 신장암 모델을 이용한 연구)

  • Background: As a potent antigen presenting cell and a powerful inducer of antigen specific immunity, dendritic cells (DCs) are being considered as a promising anti-tumor therapeutic module. The expected therapeutic effect of DCs in renal cell carcinoma was tested in the mouse model. Established late-stage tumor therapeutic (E-T) and minimal residual disease (MRD) model was considered in the in vivo experiments. Methods: Syngeneic renal cell carcinoma cells (RENCA) were inoculated either subcutaneously (E-T) or intravenously (MRD) into the Balb/c mouse. Tumor cell lysate pulsed-DCs were injected twice in two weeks. Intraperitoneal DC injection was started 3 week (E-T model) or one day (MRD model) after tumor cell inoculation. Two weeks after the final DC injection, the tumor growth and the systemic immunity were observed. Therapeutic DCs were cultured from the bone marrow myeloid lineage cells with GM-CSF and IL-4 for 7 days and pulsed with RENCA cell lysate for 18 hrs. Results: Compared to the saline treated group, tumor growth (E-T model) or formation (MRD model) was suppressed in pulsed-DC treated group. RENCA specific lymphocyte proliferation was observed in the RENCA tumor-bearing mice treated with pulsed-DCs. Primary cytotoxic T cell activity against RENCA cells was increased in pulsed-DC treated group. Conclusion: The data suggest the possible anti-tumor effect of cultured DCs in established or minimal residual disease/metastasis state of renal cell carcinoma. Systemic tumor specific immunity including cytotoxic T cell activity was modulated also in pulsed-DC treated group.

Analysis of the Development of the Nasal Septum and Measurement of the Harvestable Septal Cartilage in Koreans Using Three-Dimensional Facial Bone Computed Tomography Scanning

  • Background The septal cartilage is the most useful donor site for autologous cartilage graft material in rhinoplasty. For successful nasal surgery, it is necessary to understand the developmental process of the nasal septum and to predict the amount of harvestable septal cartilage before surgery. Methods One hundred twenty-three Korean patients who underwent three-dimensional (3D) facial bone computed tomography (CT) were selected for evaluation of the midsagittal view of the nasal septum. Multiple parameters such as the area of each component of the nasal septum and the amount of harvestable septal cartilage were measured using Digimizer software. Results The area of the total nasal septum showed rapid growth until the teenage years, but thereafter no significant change throughout the lifetime. However, the development of the septal cartilage showed a gradual decline due to ossification changes with aging after puberty in spite of a lack of change in the total septal area. The area of harvestable septal cartilage in young adults was $549.84{\pm}151.26mm^2$ and decreased thereafter with age. Conclusions A 3D facial bone CT scan can provide valuable information on the septal cartilage graft before rhinoplasty. Considering the developmental process of the septal cartilage identified in this study, septal surgery should not be performed until puberty due to the risk of nasal growth impairment. Furthermore, in elderly patients who show a decreased cartilage area due to ossification changes, septal cartilage harvesting should be performed carefully due to the risk of saddle nose deformity.

Epidermal Growth Factor Receptor Tyrosine Kinase Inhibitor Versus Placebo as Maintenance Therapy for Advanced Non-small-cell Lung Cancer: A Meta-analysis of Randomized Controlled Trials

  • Background: Use of epidermal growth factor receptor inhibitors (EGFR-TKIs ) is now standard for non-small-cell lung cancer (NSCLC). However, the effects of EGFR-TKIs in maintenance therapy for advanced NSCLC patients are still unclear. The preent meta-analysis was performed to examine pooled data of randomized control trials (RCT) where EGFR-TKIs were compared against placebo in maintenance regimens for patients with advanced NCSLC to quantify potential benefits and determine safety. Methods: Several data bases were searched, including PubMed, EMBASE and CENTRAL, and we performed an internet search of conference literature. The endpoints were objective response rates (ORR), progression-free survival (PFS) and overall survival (OS). We performed a meta-analysis of the published data, using Comprehensive Meta Analysis software (Version 2.0). with a fixed effects model and an additional random effects model, when applicable. The results of the meta-analysis are expressed as hazard ratios (HRs) or risk ratios (RRs), with their corresponding 95% confidence intervals (95%CIs). Results: The final analysis included six trials, covering 3,758 patients. Compared with placebo, EGFR-TKIs maintenance therapy improved ORR and PFS for patients with advanced NSCLC, the difference being statistically significant (P<0.05), but proved unable to prolong patients' OS. The main adverse reactions were diarrhea and rashes. Conclusion: EGFR-TKIs demonstrated encouraging efficacy, safety and survival when delivered as maintenance therapy for patients with advanced NSCLC after first-line chemotherapy, especially for the patients who had adenocarcinomas, were female, non-smokers and patients with EGFR gene mutations.

Early outcomes of cleft and palatal width following anterior palate repair (vomerine flap) in infants with wide cleft lip and palate

  • Background Anterior palatal repair performed during cleft lip repair using a vomerine flap may assist in recruiting additional soft tissue for subsequent completion of palatoplasty, especially in patients with a wide cleft. We present our early results in the hope of triggering a re-evaluation of this technique regarding its advantages for maxillary growth through further studies of patients with a wide cleft. Methods A retrospective analysis of patients with complete unilateral and bilateral cleft lip and palate was performed, including cleft and palatal measurements taken during initial surgery (lip repair together with anterior palate repair) and upon completion of palatoplasty. Results In total, 14 patients were included in this study, of whom nine (63.3%) had unilateral cleft lip and palate and five (37.5%) had bilateral cleft. All patients had a wide cleft palate. Lip and anterior palate repair was done at a median age of 3 months, while completion of palatoplasty was done at a median age of 10.5 months. Measurements taken upon completion of palatoplasty showed significant cleft width reduction in the mid-palate and intertubercle regions; however, the palatal arch distances at nearby landmarks showed non-significant marginal changes. Conclusions Anterior palate repair using a vomerine flap significantly reduced the remaining cleft width, while the palatal width remained. Further research is warranted to explore the long-term effects of this technique in wide cleft patients in terms of facial growth.

Effect of the Harvest Season on the Yield and Growth of Unripe Fruit and Biennial Flowering of 'Miyagawa' Satsuma Mandarin in Open Field Cultivation (노지재배 풋귤 수확시기가 수량과 과실생육 및 다음해 착화에 미치는 영향)

  • BACKGROUND: As consumption of unripe mandarin increases, its cultivation has increased in open field cultivation areas. Because unripe mandarin must be harvested before ripening and color change, the optimum harvest time must be determined. This study investigated the effect of the harvest season on the yield of unripe fruit and biennial flowering of 'Miyagawa' satsuma mandarin. METHODS AND RESULTS: Two areas of unripe mandarin orchard were selected, and the yield, fruit growth, working time, and flowering of trees the following year were investigated. Fruit was harvested at 40, 60, 80, 100, and 120 days after full bloom and at general ripening. Fruit yield of unripe mandarin increased with later harvest time from 100th to 120th day except normal ripening. The next year, biennial occurred with normal ripening and harvesting, but not at the 120th day after full bloom. At the 40th day (earliest harvest time), summer and autumn shoots were present, but not after the 100th day. The 40th day required the most harvesting time; because the time gradually decreased with later harvest, the harvest time was shortest on the 120th day, and general ripening occurred shortly after the 120th day. CONCLUSION: Harvesting of unripe mandarin 100-120 days after full bloom was ideal to reduce harvesting time, enhance yield, and enable flowering the following year.

Lack of Mutation in p53 and H-ras Genes in Phenytoin Induced Gingival Overgrowth Suggests its Non Cancerous Nature

  • Background: There have been case reports of oral squamous cell carcinoma arising from gingival overgrowth induced by phenytoin - an antiepileptic drug. However, a detailed analysis for the presence of mutations in p53 and ras genes, which are the two most frequently mutated genes in cancers, in phenytoin induced gingival overgrowth tissues has hitherto not been performed. Methods: Cellular DNA isolated from twenty gingival overgrowth tissues collected from patients undergoing phenytoin therapy were amplified using primers for p53 (exons 5-8) and H-ras (exons 1-2) genes. The PCR amplicons were then gel purified and subjected to direct sequencing analysis to screen for mutations. Results: Direct sequencing of twenty samples of phenytoin induced gingival growth did not identify mutations in any of the exons of p53 and H-ras genes that were analyzed. Conclusion: Our result indicates that mutational alteration of p53 and H-ras genes is infrequent in phenytoin induced gingival growth, which thus suggests a non malignant nature of this pathology. The findings in the present study are clinically significant as a large number of epileptic patients are treated with phenytoin.

Antifibrotic Effect of Curcumin in TGF-β1-Induced Myofibroblasts from Human Oral Mucosa

  • Background: Myofibroblasts play an important role in the development of oral submucous fibrosis (OSF). In the current study, we investigate the effect of curcumin on growth and apoptosis of myofibroblasts derived from human oral mucosa. Methods: Myofibroblasts were generated by incubating fibroblasts, obtained from human oral mucosa, with transforming growth factor-${\beta}1$ (TGF-${\beta}1$). MTT, PI staining, and FACS assays were used to investigate curcumin's effect on proliferation and cell cycle of fibroblasts and myofibroblasts. Annexin V/PI binding and FACS assays were used to examine apoptosis of myofibroblasts, Western blotting to determine the levels of Bcl-2 and Bax, and enzyme-linked immunosorbant assay was employed to examine the levels of collagen type I and III in the supernatants of myofibroblasts. Results: Curcumin inhibits proliferation of fibroblasts and myofibroblasts; it also disturbs the cell cycle, induces apoptosis and decreases the generation of collagen type I and III in myofibroblasts, which are more sensitive to its effects than fibroblasts. Curcumin induces apoptosis in myofibroblasts by down-regulating the Bcl-2/ Bax ratio. Conclusion: Our results demonstrate the antifibrotic effect of curcumin in vitro. It may therefore be a candidate for the treatment of OSF.

Comparison of Epidermal Growth Factor Receptor Mutations between Primary Tumors and Lymph Nodes in Non-small Cell Lung Cancer: a Review and Meta-analysis of Published Data

  • Background: Epidermal growth factor receptor (EGFR) mutations in non-small cell lung cancer (NSCLC) can predict the clinical response to tyrosine kinase inhibitor (TKI) therapy. However, EGFR mutations may be different in primary tumors (PT) and metastatic lymph nodes (MLN). The aim of this study was to compare EGFR mutations between PT and the corresponding MLN in NSCLC patients, and provide some guidelines for clinical treatment using TKI therapy. Materials and Methods: A systematic review and meta-analysis was performed with several research databases. Relative risk (RR) with the 95% confidence interval (CI) were used to investigate the EGFR mutation status between PT and the corresponding MLN. A random-effects model was used. Results: 9 publications involving 707 patients were included in the analysis. It was found that activation of EGFR mutations identified in PT and the corresponding MLN was 26.4% (187/707) and 19.9% (141/707), respectively. The overall discordance rate in our meta-analysis was 12.2% (86/707). The relative risk (RR) for EGFR mutation in PT relative to MLN was 1.33 (95%CI: 1.10-1.60; random-effects model). There was no significant heterogeneity between the studies ($I^2$=5%, p=0.003). Conclusions: There exists a considerable degree of EGFR mutation discrepancy in NSCLC between PT and corresponding MLN, suggesting that tumor heterogeneity might arise at the molecular level during the process of metastasis.
