Clinical characterization of 3-month-old pigs infected with African swine fever virus from Vietnam

  • African swine fever (ASF) is a fatal viral disease in pigs, with a short incubation period and causing immediate death. Few studies exist on the Asian epidemic ASF virus (ASFV) challenge in older pigs, including growing and fattening pigs and sows. We aimed to investigate clinical outcomes, pathomorphological lesions, and viral distribution in organs of 3-month-old growing pigs that were inoculated with the ASFV isolated in Vietnam. The clinical outcomes were recorded daily, and the dead or euthanized pigs immediately underwent necropsy. Viral loads were determined in 10 major organs using quantitative polymerase chain reaction. The average incubation period in growing pigs was more delayed (5.2±0.9 dpi) than that in weaned pigs, and the clinical signs were milder in growing pigs than in weaned pigs. The digestive and respiratory clinical signs in growing pigs showed at the end period of life, but these were observed at an early stage of infection in weaned pigs. The pathomorphological features were severe and nonspecific with hemorrhagic lesions in various organs. The viral loads in organs from growing pigs were higher than those from piglets, and the number of viral copies was related to gross lesions in the tonsil and intestine. In the absence of vaccines against ASF, early clinical detection is important for preventing the spread of the virus. Our findings elucidated that the clinical signs and gross lesions in growing pigs differed from those in weaned pigs, which provide valuable information for diagnosis of pigs with suspected ASF infection.

Effects of particle size and lipid form of corn on energy and nutrient digestibility in diets for growing pigs

  • Objective: Two experiments were conducted to evaluate the effects of corn particle size and lipid form on the apparent total tract digestibility (ATTD) of energy and nutrients in diets for growing pigs. Methods: In Exp. 1, thirty barrows (initial body weight [BW], 53.1±3.9 kg) were allotted to 1 of 5 diets formulated with 96.9% corn ground to 441, 543, 618, 659, and 768 ㎛, respectively. In Exp. 2, thirty-six barrows (initial BW, 54.7±3.6 kg) were allotted to 1 of 6 diets formulated by including 2% or 15% corn germ (CG 2 or CG 15), 1% or 6% corn oil (CO 1 or CO 6), 1% CO+2% corn germ meal (CO 1+CGM 2), or 6% CO+15% corn germ meal (CO 6+CGM 15), respectively. Results: The ATTD of gross energy (GE) and the digestible energy (DE) in diet and corn grain linearly decreased as the corn particle size increased (p<0.05) from 441 to 768 ㎛. Particle size had a quadratic effect (p<0.05) on the ATTD of neutral detergent fiber and acid detergent fiber in diets, and which firstly increased and then decreased as the corn particle size increased from 441 to 618 ㎛ and 618 to 768 ㎛, respectively. The ATTD of GE, ether extract (EE), and the DE in CO 1 diet and CO 6 diet was greater (p<0.05) than that in CG 2 diet and CG 15 diet, respectively. The ATTD of EE in CO 6 diet and CO 6+CGM 15 diet was greater (p<0.05) than that in CO 1 diet and CO 1+CGM 2 diet. Conclusion: Less than 618 ㎛ was recommended for corn particle size in growing pig's diet and extracted lipid had greater digestibility than the intact lipid in corn. Higher concentration of extracted CO had greater digestibility of EE compared with lower concentrations of CO diet.

  • A total of 701 swine sera from 55 swine farms (Mar, 1998 through Feb, 2001) were nation-widely collected for the presence of antibody to porcine parvovirus (PPV) in sows and 30-, 60-, 90-day-old pigs. Sero-prevalence by haemagglutination inhibition assay with guinea pig red blood cells was investigated on the basis of year, region and season, respectively. In general, there was no significant difference with gradual decrease of passive immunity for the sero-prevalence to PPV in sows and 30-, 60-, 90-day-old pigs for the period of 1999 and 2000. However, regional variation was observed in the provinces of Kyonggi, Choongnam and Kyungnam, Natural infection of the virus in 90-day-old pigs was increased during the fall and the winter. Thus, it seems that the natural infection of PPV in growing pigs may be attributed to the increased outbreak of postweaning multisystemic wasting syndorme in co-infection with porcine circoviruses.

Supplementation of protease and different nutrient density diets in growing-finishing pigs

  • This study was conducted to investigate the effects of protease supplementation and different nutrient density of diets in growing-finishing pigs. A total of one hundred-eight crossbred growing pigs ([Landrace × Yorkshire] × Duroc) with an initial body weight (BW; 18.74 ± 3.46 kg) were used for 15 weeks. Pigs were randomly assigned to six dietary treatments with 6 replicates of 3 pigs per pen in a 3 × 2 factorial through the following arrangement: Three groups of protease (1, Basal diets; 2, Protease A: 125 mg/kg protease derived from Streptomyces sps; 3, Protease B: 100 mg/kg protease derived from Bacillus licheniformis) at two different nutrient density diets (1, Basal requirement; 2, 0.94%-0.98% higher than requirement in dietary protein and 50 kcal/kg in energy). High nutrient (HN) diets showed higher average daily gain (ADG) (p < 0.05) and apparent total tract digestibility (ATTD) of crude protein (CP) (p < .0001) compared to basal nutrient (BN) diets during growing periods. Supplementation of protease showed higher BW (p < 0.05) and ADG (p < 0.05) compared to non-supplementation of protease during growing periods. Also, supplementation of protease showed higher ATTD of CP (p < 0.01), ATTD of gross energy (p < 0.05) and decreased blood urea nitrogen (BUN) level (p = 0.001) compared to non-supplementation of protease during finishing periods. Pigs which fed the protease showed decreased ammonia (NH3) emissions (p < 0.05) during experiment periods and decreased hydrogen sulfide (H2S) emissions (p < 0.01) during finishing periods. Interactions between nutrient density and protease were observed, which decreased the feed conversion ratio (p < 0.05) in HN diets without protease compared to BN diets without protease during weeks 4 to 6. Also, interaction between nutrient density and protease was observed, which resulted in improved ATTD of CP (p < 0.01) in response to PTA supplementation with HN diets during the finishing period. In conclusion, supplementation of protease reduces NH3 in feces and BUN in whole blood by increasing the digestibility of CP and improves growth performance. Also, diets with high nutrient density improved growth performance and nutrient digestibility in growing periods.

Cellulose Digestibility Increased with CelD Transgenic Pigs

  • This study was performed to test the cellulose digestibility using the transgenic pigs harboring cellulose degradation gene D (CelD). After delivered offsprings between normal pig and transgenic swine, DNA was isolated from piglets tail for PCR analysis. In first generation, five out of 65 piglets showed CelD positive. Unfortunately, four CelD-positive pigs were died during growing, but one survived pig was used as a transgenic founder to produce F$_1$ descendents. Among 3 F$_1$ transgenic pigs produced, one died and the remaining two pigs were used to test the fiber digest efficiency. An assorted feed was composite of 5% fiber with other ingredients. The feed of 3 kg per day was provided to the pigs including transgenic founders and littermate controls. The manure quantity was measured daily for a month, and all manures were dried for three days to analysis nitrogen, phosphate and fiber concentrations. The fiber digestion efficiencies of the transgenic F$_1$ pigs showed approximately 10% higher than those of control pigs. Fiber digestion was not greatly improved in transgenic pigs as it had been expected approximately 30%. Nitrogen concentration of transgenic pig's manure was slowly decreased compare to the control pigs. Because there were only two transgenic pigs tested, a large number of transgenic pigs may be necessary to obtain more reliable data. Breeding of animals to obtain sufficient transgenic pigs subjected for a further study is on progress. Taken together, this study demonstrated successful production of transgenic pigs with increase of cellulose digestibility in the porcine feed.

바이오 필터를 이용한 비육돈사 배기덕트 시설의 악취저감 효과 (Odor Removal Efficiency of Biofilter Ducting Systems in Indoor Pig House)

  • 무창 육성 비육돈사에 대한 악취 제어 효율 비교분석을 위하여 Biofilter 덕트에 의한 악취 제어 연구를 수행하였다. 본 실험은 서울대 부속목장 실험돈사에서 실시하였으며 실험결과는 다음과 같다. 1. 무창 육성비육돈사에서 여름철 최대 배기시 바이오필터 외부로 배출되는 공기속도는 볏짚(.0.77 m/s), ALC(0.56 m/s)으로 측정되었다. 2. 실험시작 5일 후 가스검지기(GV-100)에 의한 간이측정시 암모니아($NH_3$) 농도는 필터 통과 후 볏짚($2mg/{\ell}$), ALC(3ppm)로 측정되어 부자재 사이의 공극율이 높은 볏짚이 높게 나타났다. 부자재에 따른 필터외부에서의 먼지측정 결과 볏짚($93\;mg/m^3$), ALC($32\;mg/m^3$)으로 측정되어 볏짚은 볏짚 간의 넓은 공극에서 발생하는 먼지 및 돈사내부의 먼지가 혼합되어 필터 외부로 배출되어 돈사내부보다 높게 측정되었다. 3. 황화수소($H_{2}S$) 가스농도 변화는 돈사내부에서는 측정일 7일(6 ppm), 21일(5 ppm) 및 36일(7 ppm) 경과함에 따라 어느 일정 시점에서 황화수소 가스농도는 증가하지 않았으나 다른 필터여재에서는 일정한 경향은 보이지 않았다. 이상의 실험결과를 종합해 볼 때 무창 육성비육 돈사에서 Biofilter를 이용한 악취저감 효과를 확인할 수 있었다.

Effect of Vitamin Levels and Different Stocking Densities on Performance, Nutrient Digestibility, and Blood Characteristics of Growing Pigs

  • This study was conducted to evaluate the effects of dietary vitamin levels and stocking densities on growth performance, nutrient digestibility, and blood characteristics in growing pigs. A $2{\times}3$ factorial (two vitamin levels, three regimens of stocking densities) arrangement was utilized with 96 pigs ($23.10{\pm}0.95$ kg initial body weight and 63 d of age) for 36 d. The pigs were allocated to pens with different stocking density (0.64, 0.48, and 0.38 $m^2/pig$, respectively). The diets used in this study were a normal diet (based on NRC) and a high level of vitamin diet (2-fold higher than normal diet). The ADG and ADFI of pigs were decreased as the stocking density increased (p = 0.03 and p = 0.01, respectively). The G/F of pigs was 5% lower in the high vitamin treatment (p = 0.03) as compared with the control treatment. The apparent total tract digestibility (ATTD) of DM and N digestibility was negatively affected by the high level of vitamin in diets (p = 0.05 and p = 0.04, respectively). Moreover, a significant and negative effect on the ATTD of N was detected in the large groups (linear, p = 0.02). Blood cortisol concentration was increased with increasing stocking density (linear, p = 0.05), and was decreased by high level of vitamin (p = 0.04) at the end of this experiment. Stocking density also caused a linear reduction in WBC concentration (p = 0.05). Our data indicated that the principal effect of stocking density was not reliant on dietary vitamin levels. In conclusion, results indicated that doubling the vitamin supplementation did not improve the growth performance of pigs in high density. However, the blood cortisol concentration was decreased but the ATTD of N digestibility was impaired by high level of vitamin diet.

Changes in expression of insulin signaling pathway genes by dietary fat source in growing-finishing pigs

  • This study investigated changes in gene expression by dietary fat source, i.e., beef tallow, soybean oil, olive oil, and coconut oil (each 3% in feed), in both male and female growing-finishing pigs. Real-time PCR was conducted on seven genes (insulin receptor; INSR, insulin receptor substrate; IRS, phosphatidylinositol (3,4,5)-triphosphate; PIP3, 3-phosphoinositide-dependent protein kinase-1; PDK1, protein kinase B; Akt, forkhead box protein O1; FOXO1 and cGMP-inhibited 3', 5'-cyclic phosphodiesterase; PDE3) located upstream of the insulin signaling pathway in the longissimus dorsi muscle (LM) of pigs. The INSR, IRS, PIP3, and PDE3 genes showed significantly differential expression in barrow pigs. Expression of the PIP3 and FOXO1 genes was significantly different among the four dietary groups in gilt pigs. In particular, the PIP3 gene showed the opposite expression pattern between barrow and gilt pigs. These results show that dietary fat source affected patterns of gene expression according to animal gender. Further, the results indicate that the type of dietary fat affects insulin signaling-related gene expression in the LM of pigs. These results can be applied to livestock production by promoting the use of discriminatory feed supplies.

Effect of Nonstarch Polysaccharide-Rich By-Product Diets on Nitrogen Excretion and Nitrogen Losses from Slurry of Growing-Finishing Pigs

  • An experiment was conducted to investigate the effect of diet for growing-finishing pigs with high level of non-starch polysaccharides (NSP) from by-products on nitrogen excretion and nitrogen losses from slurry during storage. Sixteen commercial crossbred barrows of about 68 kg BW were randomly allotted to one of four diets. The control diet was formulated using tapioca and rice as basal energy sources. In the other diets, tapioca was replaced by either coconut expellar, rice bran or beer by-product. The diets differed mainly in the amount and compostition of NSP. After a 12-day adaptation period, urine and faeces were collected separately in metabolism cages for 9 days. Urine and faeces from the first four days were used to analyse the nitrogen partitioning. Urine and faeces from the last 5 days were mixed as slurry. The slurry was sampled at the end of the collection period and again after 30 days storage, to analyse for nitrogen to calculate the losses. Increasing dietary NSP reduced urinary nitrogen and nitrogen losses from the slurry during storage. The pigs fed the diet based on beer by-product excreted the most nitrogen via faeces and the least nitrogen via urine. Nitrogen losses from slurry of pigs fed the beer by-product were from 34 to 65% lower than from the other three diets. It is concluded that including NSP-rich by-products in the diet of growing-finishing pigs reduces urinary nitrogen excretion and nitrogen losses from slurry during storage.

Shrimp By-product Feeding and Growth Performance of Growing Pigs Kept on Small Holdings in Central Vietnam

  • The effect studied was that of the feeding of shrimp by-product meal, as a source of eicosapentaenoic and docosahexaenoic acid, on growth performance and fatty acid composition of adipose tissue in growing pigs kept on small holdings in Central Vietnam. Shrimp by-product meal was exchanged with ruminant meal so that the diets contained either 0, 10 or 20% shrimp byproduct meal in the dry matter. The diets were fed on 6 different small-holder farms. The farmers fed a base diet according to their personal choice, but were instructed as to the use of shrimp by-product and ruminant meal. The diets were fed to the pigs from 70 to 126 days of age. There were three animals per treatment group per farm. The diets without and with 20% shrimp by-product meal on average contained 0.01 and 0.14 g docosahexaenoic acid/MJ of metabolisable energy (ME). Due to the higher contents of ash and crude fiber, the shrimp by-product meal containing diets had lower energy densities than the control diets. Eicosapentaenoic acid was not detectable in adipose tissue; the content of docosahexaenoic acid was generally increased after consumption of shrimp by-product meal. In spite of the concurrent high intakes of ash and crude fiber, the feeding of shrimp by-product meal had a general stimulatory effect on growth performance of the growing pigs. The intake of docosahexaenoic acid or its content in adipose tissue was not related with average daily gain. It is suggested that shrimp by-product meal may contain an unknown growth enhancing factor.
