The Effects of Nicotine on the Mouse Oocyte Maturation In vitro (생쥐 난자의 체외 성숙에 미치는 Nicotine의 영향)

  • Objective: The present study was done to clarify the effects of nicotine and nicotine tartrate on the mouse oocyte maturation in vitro. Methods: GV (germinal vesicle) oocytes were isolated from Graafian follicle of ovaries with sharp needles under a stereomicroscope from female mouse of ICR strain (4 weeks old). Collected oocytes were cultured for 17 hours at $37^{\circ}C$, 5% $CO_2$ in air and 100% humidified condition in incubator. New MHBS was the basic medium used in which nicotine, nicotine tartrate, and mecamylamine (antagonist of nicotinic acetylcholine receptor) were added depending on the experimental group. GV oocytes were cultured in one of these media. Results: Nicotine ($300{\mu}M{\sim}5mM$) had no effects on GVBD (germinal vesicle breakdown) compared to the control, but increasing concentration of nicotine led to an decrease in the first polar body formation. However, nicotine ($10{\sim}500{\mu}M$) induced GVBD in a dose-dependent manner of GV oocytes in a medium containing dbcAMP. Nicotine tartrate ($50{\mu}M{\sim}5mM$) had no effects on GVBD compared to the control but, increasing concentration of nicotine tartrate led to an decrease in the first polar body formation. Mecamylamine $10{\mu}M$ added to the medium containing nicotine ($300{\mu}M{\sim}5mM$) showed higher percentage of the first polar body formation compared to the nicotine ($300{\mu}M{\sim}5mM$) treatment group. Mecamylamine $10{\mu}M$ added to the medium containing nicotine tartrate ($50{\mu}M{\sim}5mM$) showed higher percentage of the first polar body formation compared to the nicotine tartrate ($50{\mu}M{\sim}5mM$) treatment group. Conclusion: The present study suggest that nicotine and nicotine tartrate have the harmful effects on the meiotic maturation of the mouse oocytes in vitro. However, mecamylamine block harmful effects of nicotine and nictine tartrate.

Evaluation of the Midpalatal Suture Maturation in Young Koreans Using Cone-Beam Computed Tomography

  • Purpose: The aim of this study was to evaluate the ossification and maturation of the midpalatal suture in young Koreans using cone-beam computed tomography (CBCT). Materials and Methods: The study sample consisted of 40 patients with ages from 8.2 to 23.6 years who visited the Department of Orthodontics, Chung-Ang University Dental Hospital. CBCT images were taken for diagnosis. From the CBCT image, morphological stages and Hounsfield units (HU) of midpalatal suture were obtained to evaluate the midpalatal suture maturation. Spearman's correlation coefficients were calculated to analyze relationships between chronological age, morphological stage, and HU. Result: There was a wide variation in the morphology of the midpalatal suture in each age group. It showed a modest relationship between the chronological age and the degree of morphological stages. Also there was a modest relationship between the chronological age and HU. In contrast, there was a significant relationship between morphological stages and HU of midpalatal suture (P<0.01). Conclusion: CBCT images can be used for the evaluation of midpalatal suture maturation. For the estimation of the prognosis of rapid maxillary expansion, CBCT may be reliable for the assessment of the maturation of the midpalatal suture.

  • The sexual maturation occurred by the changes of steroid hormones was known to sex-dependently and/or age-dependently regulate the lipid metabolism in various animal species. Our current study demonstrates that lipid and its functional fatty acids can be changed depending on the status of sexual maturation. Of the functional fatty acids, ${\gamma}$-linolenic acid (GLA; 18:3n-6) is an important factor for maintaining human health. The purpose of our study was to investigate the level of GLA in mice with different stages of sexual maturation. To this end, the longissimus muscle (LM) of immature (3-week-old) and mature (7-week-old) female mice was analysed for the fatty acid composition by gas chromatography. Furthermore, both gene and protein level of ${\Delta}6$ desaturase (FADS2) which is involved in GLA metabolism by real time PCR and Western blotting, respectively. Mature females showed greater (P<0.05) serum $17{\beta}$-estradiol (E2) level and LM GLA contents than immature group. The mRNA and protein levels of FADS2, which converts precursor linoleic acid into GLA, were higher (P<0.05) in mature female mice than in immature mice. In conclusion, these results show that sexual maturation of female mice induces GLA and FADS2 contents in LM.

Reproduction and Maturation of Sea Bass, Lateolabrax japonicus, after Transportation from Net-Cages to Indoor Tanks

  • To determine whether the reproductive processes of sea bass, Lateolabrax japonicus, proceed normally after transportation from an outdoor net-cage into indoor tanks, we examined changes in the gonadosomatic index (GSI), histological gonadal tissue, and plasma levels of sex hormones (testosterone and estradiol-17ß) during their annual reproductive cycle. We also measured maturation and spawning across two sea water salinity levels (full and low salinity). Fecundity was estimated by the relationship between egg number and body size in female sea bass. Monthly changes in the GSI, histological gonadal tissues, and oocyte size showed both male and female sea bass reach final maturation in January and February, respectively, indicating that the spermiation of males occurs earlier than the spawning of females. The histological results indicated that the sea bass is a multiple spawner, similar to many marine teleosts, exhibiting group-synchronous oocyte development. Female maturation and spawning were enhanced in lower salinity seawater (29.6-31.0 psu) compared to that of normal salinity (34.5-35.1 psu). These results confirm that sea bass reproduction can occur successfully in captivity and imply that fertilized eggs can be collected from February to March. Additionally, our results show that lower salinity enhances oocyte maturation and spawning of female sea bass.

Effect of Cysteine and Glutathione on In Vitro Maturation of Porcine Follicular Oocytes (Cysteine 및 Glutathione 이 돼지난포란의 체외성숙에 미치는 영향)

  • This study was conducted to investigate the effect of cysteine (CySH) and glutathione (GSH) on in vitro maturation of porcine follicular oocytes. The results obtained were summarized as follows: 1. When the immature oocytes were cultured at 0, 0.04, 0.14, 0.6 and 1.2 mM of cysteine (CySH) for 36h, the germinal vesicle breakdown (GVBD) rates were 90.8, 89.9, 90.5, 92.0 and 91.3%, respectively, and the maturation rates of the oocytes with metaphase-II were 56.1, 50.7, 41.9, 49.0 and 61.5%, respectively. Especially, the maturation rates of 0.14 and 0.6 mM treated groups were significantly lower than those of control (non-treated) group (P<0.05). After 44h of culture in the same treatments of CySH, the GVBD rates of porcine immature oocytes were 90.0, 91.8, 89.8, 90.5 and 89.6%, respectively, and the maturation rates were 80.2, 76.3, 69.4, 66.7 and 72.6%, respectively. Especially, the maturation rates of 0.14 and 0.6 mM treated groups were significantly lower than those of control group (P<0.05). 2. When the immature oocytes were cultured at 0, 0.05, 0.1, 0.5 and 1.0 mM of glutathione (GSH) for 36h, the GVBD rates of porcine immature oocytes were 91.0, 90.9, 89.5, 92.0 and 91.1%, respectively, and the maturation rates were 59.0, 48.5, 47.8, 38.6 and 37.5%, respectively. All treated groups of GSH showed lower maturation rates than the control group (P<0.05). After 44h of culture in the same treatments of GSH, the GVBD rates of porcine immature oocytes were 91.8, 94.1, 89.1, 91.3 and 91.1%, respectively, and the maturation rates were 84.6, 57.1, 69.6, 71.3 and 64.3% respectively. All treated groups of GSH showed lower maturation rates than the control group (P<0.05).

Use of a Xanthine-Xanthine Oxidase System on In Vitro Maturation and Fertilization in Pig

  • This study was undertaken to evaluate the effects of catalase using xanthine (X) - xanthine oxidase (XO) system on in vitro maturation and fertilization in pig. When follicular oocytes were cultured in maturation medium with X and/or XO, the maturation rates were not significantly different between in medium with and without catalase despite of different culture periods. However, significantly (P<0.05) higher maturation rates were obrained in culture with X-XO system. The rates of degenerated oocytes were increased with culture periods prolonged, and were significantly (P<0.05) higher in medium without than with catalase at 120 h of culture. On the other hand, the parthenogenetic oocytes were observed with high proportions at 72 h of culture, hut were not different in medium with and without catalase at various times of culture. In another experiment, the frozen-thawed boar spermatozoa treated with X-XO system for in vitro fertilization. The penetration rates were higher in medium with that than without catalase during the in vitro fertilization with, none (P<0.05), XO and X+XO. On the other hand, when sperm were treated with none, X, XO and X+XO, lipid peroxidation were higher in medium without that than with catalase. However, the changes in sperm penetration and lipid peroxidation showed opposite patterns. The sperm suspensions were also treated with X and/or XO for assay of sulfhydryl (-SH) group content. Under the above all conditions, sperm-SH group were higher detected In medium with that than without catalase. The activity of sperm binding to zona pellucida was also evaluated through binding to salt-stored porcine oocytes. In control group, sperm binding to zona pellucida were higher than in medium with X, XO and X+XO groups. No significant differences, however, were observed between medium with and without catalase. In conclusion, the exposure of follicular oocytes and spermatozoa to X-XO system may be caused stimulating in vitro maturation and fertilization in pig. Effects of epigallocatechin-3-gallate on bovine oocytes matured in vitro

  • Objective: Epigallocatechin-3-gallate (EGCG) is a major ingredient of catechin polyphenols and is considered one of the most promising bioactive compounds in green tea because of its strong antioxidant properties. However, the protective role of EGCG in bovine oocyte in vitro maturation (IVM) has not been investigated. Therefore, we aimed to study the effects of EGCG on IVM of bovine oocytes. Methods: Bovine oocytes were treated with different concentrations of EGCG (0, 25, 50, 100, and $200{\mu}M$), and the nuclear and cytoplasmic maturation, cumulus cell expansion, intracellular reactive oxygen species (ROS) levels, total antioxidant capacity, the early apoptosis and the developmental competence of in vitro fertilized embryos were measured. The mRNA abundances of antioxidant genes (nuclear factor erythriod-2 related factor 2 [NRF2], superoxide dismutase 1 [SOD1], catalase [CAT], and glutathione peroxidase 4 [GPX4]) in matured bovine oocytes were also quantified. Results: Nuclear maturation which is characterized by first polar body extrusion, and cytoplasmic maturation characterized by peripheral and cortical distribution of cortical granules and homogeneous mitochondrial distribution were significantly improved in the $50{\mu}M$ EGCG-treated group compared with the control group. Adding $50{\mu}M$ EGCG to the maturation medium significantly increased the cumulus cell expansion index and upregulated the mRNA levels of cumulus cell expansion-related genes (hyaluronan synthase 2, tumor necrosis factor alpha induced protein 6, pentraxin 3, and prostaglandin 2). Both the intracellular ROS level and the early apoptotic rate of matured oocytes were significantly decreased in the $50{\mu}M$ EGCG group, and the total antioxidant ability was markedly enhanced. Additionally, both the cleavage and blastocyst rates were significantly higher in the $50{\mu}M$ EGCG-treated oocytes after in vitro fertilization than in the control oocytes. The mRNA abundance of NRF2, SOD1, CAT, and GPX4 were significantly increased in the $50{\mu}M$ EGCG-treated oocytes. Conclusion: In conclusion, $50{\mu}M$ EGCG can improve the bovine oocyte maturation, and the protective role of EGCG may be correlated with its antioxidative property.

Successful In Vitro Development of Preantral Follicles Isolated from Vitrified Mouse Whole Ovaries

  • The purpose of this study was to assess follicular viability and competence through in vitro culture of preantral follicles isolated from vitrified mouse whole ovaries. Mouse preantral follicles were enzymatically isolated from vitrified- warmed and fresh ovaries and cultured for 10 days followed by in vitro oocyte maturation. In vitro matured oocytes were fertilized and cultured to the blastocyst stage. Five minutes pre-exposure to vitrification solution of whole ovaries had significantly higher (p<0.05) oocyte survival and maturation rates than between 10 min exposure groups. Oocyte diameter was significantly smaller (p<0.05) in the 5 and 10 min exposure groups ($69.4{\pm}2.8$ and $67.8{\pm}3.1$) when compared to that of control group ($71.7{\pm}2.1$). There was no statistical significant difference in blastocyst development rates between vitrification group (8.6%) and the fresh control group (12.0%). The mean number of cells per blastocyst was significantly lower (p<0.05) in the vitrification group ($41.9{\pm}20.2$) than in the fresh control group ($55.1{\pm}22.5$). The results show that mouse oocytes within preantral follicles isolated from the vitrified whole ovaries can achieve full maturation, normal fertilization and embryo development.

Effects of IVM and IVF Duration on In Vitro Development and Cell Numbers of Embryos in Korean Native Cattle

  • The present study was performed to investigate the effects of in vitro maturation (IVM) and in vitro fertilization (IVF) duration on the development of Korean Native Cattle embryos. The time of blastocyst formation and the quality of blastocysts based on cell numbers were examined. The cleavage rate increased with the length of IVF duration in the groups of 18-hr IVM, but was constant in the groups of 24-hr IVM. The development rate to the 8-cell stage was significantly higher in the IVM 18: IVF 20 group than in the IVM 24: IVF 24 group. The development rate to the blastocyst stage was highest in the IVM 18: IVF 20 group, significantly different from that of the IVM 18: IVF 16, IVM 24: IVF 20 and IVM 24: IVF 24 group. The time of blastocysts formation tended to be shorter when IVM and IVF duration were decreased. The number of inner cell mass, trophoblast and the total cells were significantly higher in the IVM 18: IVF 16 group than in the IVM 24: IVF 24 group (P<0.05). These results demonstrated that the IVM and IVF duration should be adequate for the efficient production of bovine embryos, and it might particularly be essential to determine the proper combination of IVM and IVF duration.

The relationship between malocclusion and menarcheal age, and its secular trend for Korean women

  • Objective: This study aimed to evaluate the mean age of menarche, its secular trend in Korean women, and the relationship between malocclusion and the rate of skeletal maturation, as defined by menarcheal age. Methods: We retrospectively collected data on menarcheal age from 931 Korean women born during 1961 - 1997. Subjects were divided by the malocclusion type and birth-year decade into 3 and 4 groups, respectively. The mean menarcheal age for each group was determined, and one-way ANOVA was performed for intergroup comparison (p = 0.05). Two-way ANOVA was also performed to compare all the 12 subgroups (p = 0.05). Results: The mean age of menarche was 12.82 years for Korean women born during 1961 - 1997. A distinct downward secular trend of menarcheal age was noticed (p < 0.05). For the birth-year decade 1961 - 1970, the Class III malocclusion group showed earlier onset of menarche than the other malocclusion groups (p < 0.05), but the other birth-year groups did not show any significant difference in the type of malocclusion (p > 0.05). Conclusions: A positive secular trend towards earlier menarche exists among Korean women. Malocclusion does not show any significant relationship with the rate of skeletal maturation, as defined by menarcheal age.
