Soil-Environmental Factors Involved in the Development of Root Rot/Vine on Cucurbits Caused by Monosporascus cannonballus

  • A root rot/vine decline disease occurred naturally on bottle gourd-stocked watermelon, melon, oriental melon and squash grown in greenhouses, but not on these plants grown in fields. Self-rooted watermelon, cucumber, pumpkin and luffa were also proven to be hosts of the pathogen by artificial inoculation in this experiment. The pathogen was identified as Monosporascus cannonballus by comparing microscopic characteristics of fungal structures with those of previously identified fungal strains. Our field investigations showed that the temperature and electric conductivity of soil in infected greenhouses were higher and the soil moisture content was lower than in noninfected greenhouses. To investigate soil-environmental factors affecting disease development, greenhouse trials and inoculation experiments were conducted. The host plants inoculated and grown under conditions of high soil temperature and electrical conductivity ($35\pm2^{\circ}$, 3.2-3.5 mS) and with low soil moisture content (pF 3.0-4.5) were most severely damaged by the fungal disease. Since plants growing in greenhouses ae usually exposed to such environmental conditions, this may be the reason why the monosporascus root rot/vine decline disease has occurred only on cucurbits cultivated in greenhouses but not in field conditions.

Resistance of Cucurbits to Podosphaera xanthii Race 1 (Podosphaera xanthii Race 1에 대한 박과 작물의 저항성)

  • This study was conducted to establish an efficient screening method to identify cucurbits resistant to powdery mildew. Powdery mildew fungus was obtained from a single lesion of infected cucumber leaf in 2010 at Daejeon. The fungus was identified as Podosphaera xanthii race 1 based on morphological characteristics and resistance responses of four melon differentials. Development of powdery mildew caused by the fungal isolate on 34 commercial cultivars of cucumber was investigated at three plant growth stages in a greenhouse. The degree of resistance of cotyledons of each cultivar to the fungus was not correlated with that of whole plant, but powdery mildew occurrence in the first true leaf was highly correlated with resistance at the level of the whole plant. Based on these results, the first true leaf of cucurbit cultivars can be used for screening of resistance to powdery mildew. In addition, variation of resistance of commercial 12 cucumber and 26 melon cultivars to the powdery mildew fungus due to different growing seasons was tested. In the case of cucumber, the resistance response in some cultivars was influenced by growing season. The resistant cultivars showed higher resistance in the warm season than in the cool season. By contrast, the resistant melon cultivars demonstrated strong resistance in all the tested growing seasons. Interestingly, the tested powdery mildew pathogen, a member of P. xanthii race 1, was not pathogenic on seven cultivars of watermelon (Citrullus lanatus). To follow up on this, diverse race 1 isolates of P. xanthii should be collected and tested.

Sequential Sampling Plan for Aphis gossypii (Hemiptera: Aphididae) based on Its Intra-plant Distribution Patterns in Greenhouse Cucumber at Different Growth Stages (온실재배 오이의 생육단계별 목화진딧물의 주내 분포 특성에 기초한 축차표본조사법)

  • This study describes the development of a method for monitoring Aphis gossypii in greenhouse cucumber fields that was used during 2013 and 2014. The dispersion pattern of A. gossypii was determined by commonly used methods: Taylor's power law (TPL) and Iwao's patchiness regression (IPR). The sample unit was determined by linear regression analysis between mean density of sample unit versus whole plant. The optimum sample unit for different plant growth stages was two leaves (median and the lowest + 1 leaf) when the total number of leaves was less than nine, and three leaves (4th, 7th from canopy, and the lowest +1 leaf) when the total number of leaves was greater than nine. A. gossypii showed an aggregated distribution pattern, as the slopes of both TPL and IPR lines were greater than 1. TPL provided a better description of the mean-variance relationship than did IPR. The slopes and intercepts of TPL and IPR from leaf samples did not differ between the surveyed years. Fixed precision levels (D) for a sequential sampling plan were developed using Green's and Kuno's equations based on the number of aphid in a leaf sample. Green's method was more efficient than Kuno's to stop sampling. The number of samples needed to estimate the density of A. gossypii increased at higher D levels and lower mean densities. The cumulative number of aphids needed to stop sampling increased at higher D levels and with fewer plants sampled. Thus to estimate 10 aphids per leaf, 13 plants needed to be sampled, and the cumulative number of aphids to stop sampling was 131.

A New Disinfestation Approach Against Some Greenhouse Pests Using Ethyl Formate Fumigation (훈증제 에틸포메이트를 이용한 몇 가지 시설하우스 해충에 대한 새로운 방제 전략)

  • Ethyl formate (EF) is a rapid kill, environmentally safe, and low mammalian toxicity fumigant, registered to disinfest quarantine insect pests from imported agricultural products. A new concept for controlling insect pests of agricultural crops was tested in a fumigation chamber with EF. Control efficacy of and phyto-toxicity due to EF fumigation were evaluated against four pests (Thrips palmi, Bemisia tabaci, Myzus persicae, and Tetranychus urticae) and on seedlings of four fruit vegetables (FVs; yellow melon, cucumber, tomato, and pepper). Ethyl formate fumigation at a dose of 1.5 g m-3 for 12 h produced >93.3% mortality in T. palmi, B. tabaci, and M. persicae. However, T. urticae was tolerant to fumigation, showing only 20% mortality at 2.0 g m-3. In terms of concentration × time (CT) products, at least 8.9 g·h m-3 CT at 20 ± 1.5℃ was needed to achieve > 90% mortality against the three susceptible insect pests. Fumigation at 1.5 g m-3 for 12 h caused no phyto-toxicity to any of the four FV seedlings. Ethyl formate application, as a new disinfestation method in greenhouses, could be an alternative to reduce the use of conventional insecticides. However, further studies are needed to determine the efficacy of this method at different pest developmental stages and in different greenhouse environments. Additionally, research is needed to elucidate the phyto-toxicity of EF application at different growing stages of a wide variety of crops.

The Usage Status of Pesticides for Vegetables under Greenhouse Cultivation in the Southern Area of Korea (남부지역 시설채소 재배 농가의 농약 사용실태)

  • This study was undertaken to survey the pesticides used under the greenhouse cultivation of 9 vegetable crops including cucumber, tomato, strawberry, eggplant, hot pepper, paprika, lettuce, perilla leaves and green onion in the Southern area during the period of 2003-2004. The total number of farmers investigated was 202 which consisted of 124 conventional growers and 78 environment-friendly growers. The farmers were requested to record with respect to the crop's name, cultivation acreage, growing practices (conventional/environment-friendly), use purpose, pesticide kinds, usage quantity and application time of pesticides. The average usage quantity of pesticides for the 9 crops was 3.30 kg ai/ha in conventional growing and 0.47 kg ai/ha in environment-friendly growing. The average application time of pesticides was 11 times in conventional and 2 times in environment-friendly growing, with more than 90% cases as a foliar application. The number of active ingredients of pesticides was 34 in conventional and 10 in environment-friendly growing on the average per crop. The purpose of pesticide application was as a fungicide in 56% cases and as an insecticide in 43% cases.

The Growth of Cucumber and Variation of Soil Temperature Used by Warming Water Irrigation System (가온관수 시스템에 따른 지온변화와 오이의 생육)

  • A soil temperature was known as extremely important factor in terms of measuring the values of the growth and yield of vegetable in the greenhouse. A low temperature water irrigation was had much trouble in its growth. This study was performed to analyze the effect of the heating water irrigation on the soil temperature and the growth of a cucumber within a greenhouse environment. Soil temperature was 5-7$^{\circ}C$ below to 10cm in depth and 2-3$^{\circ}C$ to 20cm when the irrigation water temperature was 13$^{\circ}C$ (non-warme water irrigation). Soil temperature was similar to irrigation water temperature at 5cm in depth and was 1.5-2$^{\circ}C$ below at 10cm when the irrigation water temperatures were 2$0^{\circ}C$, $25^{\circ}C$. The early growth rates of heating water irrigation were 109-110% in plant height, 107-108% in leaf number, 103% in node number compared with those of unheated water irrigation for 30 days after planting it. The rates of total yield were 115% in 2$0^{\circ}C$ water irrigation plots and 121% in $25^{\circ}C$ water irrigation plots while those of unheated water irrigation plots were.

Cross-Protection Effectiveness of Cucumber mosaic virus (CMV) Isolates Associated with Satellite RNA for Prevention of CMV Disease in Pepper Plants (Satellite RNA 보유 Cucumber mosaic virus(CMV)의 고추 CMV병에 대한 교차방어 효과)

  • Two attenuated Cucumber mosaic virus (CMV) isolates, Paf-CMV and Rs2-CMV that had been selected from CMV isolates associated with satellite RNA (satRNA) were tested for cross-protection effect in pepper plants. The viruses selected as attenuated strains appeared to be identical serologically and physically to the challenge virus (Mf-CMV), but they were lower in the dilution end-point of infectivity of crude sap than Mf-CMV When symptoms were observed in several indicator plants after inoculation, Paf-CMV and Rs2-CMV were symptomless or showed mild mosaic symptoms while another satRNA isolate Ap-CMV developed severe mosaic symptoms on the leaves as Mf-CMV. The nucleotide sequences of the satRNAs were determined by sequencing full-length cDNA clones. Paf-, Rs2- and Ap-satRNAs were 386, 335, and 347 nucleotides long, respectively, The sequences were then compared with the other known Y-satRNA, revealing that nucleotide sequences of the satRNAs consisted of 5'- and 3'-terminal conserved regions. However variations occurred on the middle regions of the sequences, especially those related to symptom interference, showing significant differences between Paf-satRNA and other isolates. Infectious transcripts of Paf-satRNA and Rs2-satRNA induced mild mosaic symptoms in pepper plants when supported by genomic RNAs of Mf-CMV. Under greenhouse conditions, Paf-CMV and Rs2-CMV were tested for cross-protection effect in pepper and tobacco (Nicotiana tabacum cv, Xanthi nc) plants against Mf-CMV. No symptoms were developed on the plants vaccinated with Paf-CMV until 3 weeks after inoculation with the virulent strain; however another attenuated isolate, Rs2-CMV, showed less effectiveness in cross-protection. Depending on the concentration of the challenged virus, symptoms sometimes appeared later in the upper leaves. However, in plants challenged with low concentrations (below 0.2 mg/ml) of the challenge inoculum, symptoms caused by the virulent strain did not develop on the plants vaccinated with Paf-CMV. In the field experiments, the number of pepper plants with severe mosaic symptoms in the control plots was progressively increased after transplanting and reached approximately 50% after 50 days. On the other hand, the incidence of mosaic disease appeared very low on the plants that had received the protective inoculation with Paf-CMV.

Residual evaluation of ethyl formate in soil and crops after fumigation in green house (에틸포메이트의 하우스 농작물 훈증처리 후 토양 및 작물 중 잔류양상)

  • Ethyl formate (EF) is a potent fumigant replacing methyl bromide. The use of EF is limited to a quarantine process. Appling EF to agricultural field as a safe insecticide in greenhouse give us valuable benefits including less residual concern. In this regard, residual pattern after EF fumigation in greenhouse should be undertaken. In the previous study, we have established agricultural control concentration of EF to control pests in a greenhouse. EF was fumigated at 5 g m-3 level for 2 h. The concentration of EF inside a greenhouse was analyzed to be 4.1-4.3 g m-3 at 30 min after fumigation. To prepare an analytical method for residues in cucumber crops and soil in the greenhouse, the limit of detection(LOD) of the method was 100ng g-1 and the limit of quantitation(LOQ) of this method was 300 ng g-1. R2 values of calibration curves for crops and soil were 0.991-0.997. In samples collected immediately after ventilation, EF concentration was determined to be below LOQ level. In addition, EF level was below LOQ in samples collected at 3 h after ventilation except that leaf samples of melon during the flowering period showed a level of 1,068.9 ng g-1. Taken together, these results indicate that EF used in quarantine can be applied to agricultural fields without residual issue as an effective fumigant for insect pest control.

Spectrophotometric microtitre assay for rapid screening of membrane-disrupting herbicides (분광분석을 이용한 막과산화작용 제초제의 신속한 검정법)

  • This study was conducted to establish an efficient screening method of membrane-disrupting herbicides, based on the result that membrane leakage leading to evolution of carbonyl compounds and aldehydes could be colorimetrically assessed by formation of stable chromophores with MBTH and ferric chloride. Assay conditions including plant material, light intensity, irradiation time, concentrations of reagents for colorization, and reaction time were optimized, and activities of different types of known herbicides were investigated with cucumber cotyledon discs. That result indicated that this assay was sensitive to membrane-disrupting herbicides, such as protoporphyrinogen oxidase inhibitors (acifluorfen, oxyfluorfen, oxadiazon) and an electron interceptor from photosystem I(paraquat). With seven compounds newly synthesized, highly positive correlation was observed between herbicidal activities measured by greenhouse bioassay and MBTH method. Because only a single disc(4 mm diameter) from cucumber cotyledon in 60 ${\mu}L$ of incubation solution was required to assay, a large number of herbicides could be tested simultaneously using 96-well microtitre plates. But the assay efficiency is not likely to significantly increase because of the boiling step during the reaction.

Biochemical Methane Potential of Agricultural Residues and Influence of Ensiling on Methane Production (시설농업부산물의 잠재메탄발생량 평가 및 사일로 저장에 따른 메탄 발생 변화)

  • In this study, the biochemical methane potentials of different agricultural residues produced from agricultural plastic greenhouse were determined. Additionally, ensiling storage practice was applied on agricultural residues for its effect on biogas production. Agricultural residues of cabbage, strawberry, tomato, cucumber, and oriental melon were selected as sample. The methane potential and biodegradability of agricultural residues ranged from 149~286 mL-$CH_4/g$-VS, 27~48% (by vol.), respectively and methane production was in order of cabbage > oriental melon > strawberry ${\approx}$ cucumber > tomato. Ensiling caused difference in methane production in a range of -11~36% (by vol.) per VS compared with raw material. An increase in methane potential was presumably linked to the organic acid accumulation, cellulose degradation and decrease in methane potential was due to chemical composition change, ammonia accumulation during the storage process.
