Complete genome sequence of Lactococcus lactis strain K_LL005, a xylose-utilizing bacterium isolated from grasshopper (Oxya chinensis sinuosa)

  • Lactococcus lactis is a fermentative lactic acid bacterium that is used extensively in food fermentations. The L. lactis strain K_LL005 was isolated from the grasshopper (Oxya chinensis sinuosa) gut in Korea. In this study, we reported the complete genome sequence of Lactococcus lactis K_LL005. The final complete genome assembly consist of one circular chromosome (2,375,093 bp) with an overall guanine + cytosine (G + C) content of 35.0%. Annotation results revealed 2,281 protein-coding sequences (CDSs), 19 rRNAs, and 68 tRNA genes. Lactococcus lactis K_LL005 has a gene encoding xylose metabolism such as xylR, xylA, and xylB (xylRAB).

벼메뚜기(Oxya chinensis sinuosa Mistshenko) 분말 섭취와 유산소성 운동훈련에 의한 마우스의 에너지 대사 변화 (Effects of Grasshopper (Oxya chinensis sinuosa Mistshenko) Powder and Aerobic Exercise on Energy Metabolism in ICR Mice)

  • 본 연구의 목적은 벼메뚜기(Oxya chinensis sinuosa) 분말 섭취가 유산소성 운동훈련(트레드밀 달리기)의 병행 유무에 의해 ICR 생쥐의 에너지 대사를 증가시키는지를 알아보고자 하는 것이다. 이 목적을 달성하기 위하여, 28 마리의 ICR 생쥐를 보통식 대조군(CON), 보통식 대조군으로서 운동훈련 병행군(COEX), 벼메뚜기 분말이 혼합된 사료 섭취군(GH), 그리고 벼메뚜기 분말이 혼합된 사료를 섭취함과 동시에 운동훈련을 병행군(GHEX)으로 구분하였다. 벼메뚜기 분말 사료섭취 및 운동은 6주간 진행하였다. 체중증가율은 유의하지 않았다. 지방량은 GH와 GHEX에서 유의하게 감소하였다. 혈중 glutamic oxaloacetic transaminase와 glutamic pyruvic transaminase 수준은 처치 집단간 변화가 없었다. 제2형 당수송체 및 제4형 당수송체는 처치 집단간 유의한 차이가 없었다. GHEX의 fibronectin type III domain-containing protein 5 단백질 발현량이 가장 높았다. AMP-activated protein kinase 단백질 수준은 GHEX에서 유의하게 증가하였다. Glycogen synthase kinase 3 beta 단백질 발현량은 GHEX가 CON과 비교할 때 감소하였다. 이러한 결과들은 벼메뚜기 분말을 섭취하면서 지구성 운동훈련을 하는 경우에 에너지 대사에 영향을 준다는 것을 제시하고 있다.

  • 벼메뚜기 건제품의 가공저장중 품질의 열화에 가장 큰 영향을 미친 것으로 예상되는 지질의 조성을 분석하고 저장중 생성된 갈변반응물질이 지질산화에 미치는 영향을 검토하였다. 성숙한 벼메뚜기 암컷의 지질함량은 5.12%로서 인지질함량(27.35%)이 높았고 불포화지방산 함량이 68.18%나 되었다. 벼메뚜기 천일 및 열풍건조 과정중 COV는 크게 증가하였으나 POV는 거의 측정되지 않았다. 동결건조 과정중에는 과산화물과 카아보닐 화합물이 거의 생성되지 않았다. 저장중에 POV는 초기부터 급속히 증가하였으나 COV는 저장 98일까지 지속적인 증가를 보였고 저장 초기에 당-아미노반응에 의한 수용성갈변반응물질이 많이 생성됨과 동시에 환원당과 아미노질소 함량은 크게 감소하였다. 벼메뚜기 저장중 생성된 갈변반응물질이 지질화에 미치는 영향을 조사한 결과 수용성 구분에서 지질산화의 억제효과가 다소 있는 것처럼 보였으나 지용성 구분에서는 산화촉진 효과를 나타내었다.

메뚜기 체내와 체외에서 혈구생성 경로 추적에 관한 연구 (Study on the Detection of In vivo and In vitro Hemocytic Pathway in Grasshopper, Euprepicnemis shirakii)

  • 메뚜기 체내와 체외에서 혈구 분화경로를 광학현미경과 전자현미경으로 관찰하였다. 조혈기관에서 혈구의 형성은 망상세포에 둘러싸인 줄기세포에서부터 유래되었으며, 줄기세포에서부터 원시혈구, 무정형혈구, I형 과립혈구, II형 과립혈구, 소구혈구 및 편도혈구가 각각 분화되는 것을 확인하였다. 곤충배지에 배양된 조혈조직에서 각각 다른 형태의 혈구들이 분화되어 방출되었다. 그러나, 이들 혈구들의 유사분열상은 관찰되지 않았다. 배지의 조혈기관에서 분화된 세포들의 형태학적 특징들은 메뚜기 체내의 조혈기관에서 분화된 세포들과 같았다. 이와 같은 결과는 줄기세포가 각각의 서로 다른 혈구들로 직접 분화하는 것을 의미한다. 본 연구 결과 조혈기관의 줄기세포는 각각의 혈구로 직접 분화할 수 있는 기능을 가지고 있었으며, 체내와 체외에서 한번 형성된 순환 혈구는 다른 혈구의 형태로 변형되지 않았다. 메뚜기에서 순환혈구의 유지는 복부 등쪽 첫 번째 마디에서 여덟 번째 마디 사이의 익상근 위에 광범위하게 존재하고 있는 조혈기관에 전적으로 의존하였다.

Characterization of Cooked Meat Models using Grasshopper (Sphenarium purpurascens) Soluble Protein Extracted by Alkalisation and Ultrasound as Meat-Extender

  • The most abundant Orthoptera in Mexico is a small grasshopper (Sphenarium purpurascens) which is considered a food source with increased nutritional value due to its high protein content. Insect proteins have gained relevance because of their high potential as gelling, texturing, and extender agents in the food industry. The objective of this study was to evaluate the effect of substituting meat with a soluble protein extract from grasshopper obtained by alkalisation or alkalisation-piezoelectric ultrasound, on the techno-functional, physicochemical, and sensory characteristics of cooked meat models (sausages). The soluble protein was extracted in NaHCO3 pH 8 and a piezoelectric ultrasound 5-mm sonotrode at 20 kHz with 99% amplitude. Different formulations with meat substitution: 0%, 5%, 10%, and 15% were prepared and characterised for their rheological behaviour, emulsion stability, weight loss by cooking, total protein content, colour, and texture. Sensory evaluation was conducted with consumers using a test involving check-all-that-apply and overall liking. The alkalisation-piezoelectric ultrasound method improved the solubility and the techno-functional properties of the soluble grasshopper protein when applied in sausages at maximum levels of 10% meat substitution. The sensory evaluation indicated that the formulation with 5% meat substitution exhibited the same acceptability as the control sample. Given these results, the soluble protein treated with alkalisation and piezoelectric ultrasound could be used as an extender in meat products.

Study on Integrated Workflow for Designing Sustainable Tall Building - With Parametric method using Rhino Grasshopper and DIVA for Daylight Optimization

  • Purpose: The Objective of this study is to explore the capabilities of an integrated modelling and simulation workflow when applied to an experiment-based research process, aimed at deriving daylight optimization strategies specific to tall buildings. Methods: Two methods were devised to apply this workflow with the help of DIVA and Rhino/Grasshopper. The first method is a multiple variant analysis by setting up an appropriate base case and analysing its daylight and energy performance, forming the basis of comparison for subsequent cases for design variants. The second method involved setting up the base case within a site context and conducting a solar irradiation study. An architectural variables such as overhang and shading device, were then defined as inputs in the parametric definition in Grasshopper to control the selected variable. Results: While the first method took advantage of the speed and efficiency of the integrated workflow, the second method was derived based on the ability to directly process simulation data within the integrated, single-software platform of the proposed workflow. Through these methods, different architectural strategies were explored, both to increase daylight penetration and to reduce radiant heat gain. The focus is on methods by which this workflow can be applied to facilitate the experimental derivation of daylight optimization strategies that are specific to tall building design.

Organoleptic Evaluation of the High-Protein Yoghurt containing the Edible Insect Oxya chinensis sinuosa (Grasshopper): A Preliminary Study

  • This objectives of this study were to assess the pH and titratable acid (TA) and conduct sensory evaluation of the high-protein yogurt supplemented with Oxya chinensis sinuosa (grasshopper). High-protein yogurt containing Oxya chinensis sinuosa powder displayed TA of 0.93% to 1.1%, and a pH of 4.3 or 4.4. There were no significant differences between the control and treated groups in pH and TA. Organoleptic evaluations revealed that, except for color and texture, taste, flavor, and overall acceptability showed decreased trends in proportion to the amount of Oxya chinensis sinuosa powder. Further studies will explore the potential of Oxya chinensis sinuosa powder as a protein with health benefits for humans.

Modern computer simulation for the design of concrete catenary shell structures

  • The purpose of this study was to model and design a concrete catenary shell using a modern computer program without performing experiments. The modeling idea stems from the study by Pendergrast, but he listed supplementary items that should be improved in his paper. This study aims to resolve those issues and overcome the drawbacks of the study by Pendergrast. The process of experiment for the design of a catenary shell was reproduced by Grasshopper script. In order to ensure credibility, two models designed from the Grasshopper script were analyzed using a finite element program, SAP2000; one is a square-based catenary shell and the other is a special catenary shell called as the Naturtheater $Gr{\ddot{o}}tzingen$ shell, which was completed in 1977. First, the developed modeling approach was proved to be reasonable from the analysis of the square-based shell. The reliability was further confirmed by a comparison between the current and previous analysis results for the Naturtheater $Gr{\ddot{o}}tzingen$ shell.

OAPR-HOML'1: Optimal automated program repair approach based on hybrid improved grasshopper optimization and opposition learning based artificial neural network

  • Over the last decade, the scientific community has been actively developing technologies for automated software bug fixes called Automated Program Repair (APR). Several APR techniques have recently been proposed to effectively address multiple classroom programming errors. However, little attention has been paid to the advances in effective APR techniques for software bugs that are widely occurring during the software life cycle maintenance phase. To further enhance the concept of software testing and debugging, we recommend an optimized automated software repair approach based on hybrid technology (OAPR-HOML'1). The first contribution of the proposed OAPR-HOML'1 technique is to introduce an improved grasshopper optimization (IGO) algorithm for fault location identification in the given test projects. Then, we illustrate an opposition learning based artificial neural network (OL-ANN) technique to select AST node-level transformation schemas to create the sketches which provide automated program repair for those faulty projects. Finally, the OAPR-HOML'1 is evaluated using Defects4J benchmark and the performance is compared with the modern technologies number of bugs fixed, accuracy, precession, recall and F-measure.

  • The Lactococcus taiwanensis strain K_LL004 was isolated from the gut of a grasshopper (Oxya chinensis sinuosa) collected from local farm in Korea. L. taiwanensis strain K_LL004 is the functional probiotic candidate with an ability to hydrolyse plant polysaccharides. The complete genome of the L. taiwanensis strain K_LL004 contains one circular chromosome (1,995,099 bp) with a guanine + cytosine (GC) content of 38.8%. Moreover, 1,929 Protein-coding sequence, 19 rRNA genes, and 62 tRNA genes were identified based on results of annotation. L. taiwanensis strain K_LL004 has a gene, which encodes hydrolytic enzymes such as beta-glucosidase and beta-xylosidase, that hydrolyzes plant polysaccharides.
