Leuconostoc mesenteroides KCTC3505를 이용한 발효자몽 추출물 생산 조건의 최적화 (Optimum Conditions for Production of Fermented Grapefruit Extract using Leuconostoc mesenteroides KCTC3505)

  • This study was conducted in order to determine the optimum conditions for the production of fermented grapefruit extract showing high cell growth, antioxidant activity and total flavonoid content. Five lactic acid bacterial strains - Lactobacillus plantarum KCTC3104, Lactobacillus brevis KCTC3102, Weisella cibaria KCTC3746, Leuconostoc citreum KCTC3526 and Leuconostoc mesenteroides KCTC3505 - were evaluated first in order to determine the optimum strain able to grow with high efficiency on grapefruit as a substrate and possesses higher antioxidant activity and flavonoids content. Among these strains, L. mesenteroides KCTC3505 was selected as a starter culture. To estimate the available or effective content of grapefruit in basal medium, the effects of 30%, 50%, and 70% grapefruit contents on the performance of fermentation were tested, and it was found that grapefruit can be added at 70% levels to medium. In this study, three factors of fermentation conditions - incubation time, sucrose, and glucose contents - were evaluated for their effects on fermentation performance. Taguchi experiment design was employed and the responses of experiments were calculated using signal and noise ratio calculation with larger-the-best characteristics. Finally, the optimum conditions for the manufacture of fermented grapefruit extract were as follows: grapefruit 70%, sucrose 10 g/L, glucose 10 g/L, sodium acetate 1 g/L, NaCl 1 g/L, dipotassium phosphate 0.1 g/L, magnesium sulfate 0.01 g/L and 16 hr of incubation.

  • The result of the study for essential oil(Bergamot, Grapefruit essential oil)'s Anaiysis of Blood Serum Epidermis proteins by exposure of nature lays on treatments are as follows. 1. There is no significant difference between control group and the groups treated by essential oil in feed efficiency, but in water intake, there are significant differences between each group. Water intake of the groups treated by Bergamot and Grapefruit essential oil without treatment by sun cream is higher than that of any other group. It is presented that the effect of losing water of the surface of skin is related to the balance effect of water. 2. As a result of serum analysis without applied UV-rays, the total cholesterol in the groups treated by Bergamot essential oil is higher than that of the groups treated by Grapefruit essential oil. It is showed that the dissolution of cellulite of Grapefruit essential oil is stronger than that of Bergamot essential oil. After applied UV-rays, however, the change of total cholesterol of the groups treated by Bergamot essential oil is lower than that of the groups treated by Grapefruit essential oil, namely, the group treated by Bergamot essential oil drops is used to make sebum membrane on the surface of skin, so the quantity of influx into blood vessel was low. With the result that, atherosclerotic index(A.I) also was low in the group treated by Grapefruit essential oil. In the aspect of increasing cholesterol for obesity treatment, it was judged that the effect of treatment with Bergamot essential oil is somewhat slower than that with Grapefruit essential oil, but it is safer than the effect of treatment with Grapefruit essential oil in the aspect of health. 3. As a result of protein analysis, the groups applied UV-rays in the group of treated by Bergamot essential oil, the biggest change of protein's band was appeared under part of 20.0 1Kd in the change pattern of protein. As the trend of changing protein at the groups treated by SPF 15 and 30 was similar to that of the groups of the control, it was showed that the effect of sun cream can prevent the change of the composition of skin. In the groups treated by Grapefruit essential oil, after the groups treated by Grapefruit essential oil applied UV-rays, the biggest change of protein's bend was appeared the upper part of 43Kd in the change pattern of protein. It was also judged that it means the change of the composition of skin. However, the trend of changing protein at the groups treated by SPF 15 and 30 was also similar to that of the groups of the control. Eventually, it was also showed that it can prevent the change of the composition of skin by the effect of sun cream.

  • 광범위한 천연항균제로 그 활용이 고려되고 있는 grapefruit 종자추출의 안전성을 검토하기 위하여 급성 경구독성, 피부자극 및 급성 안점막 자극시험을 수행하였다. 실험 흰쥐에 대한 급성 $LD_{50}$값은 3.75g/kg이었으며, 피부에 대한 독성은 약한 편으로 부종 등의병변이 거의 생기지 않았으나 눈에 대한 자극성이 있는 것으로 나타났다.

연속수증기증류추출법에 의한 오렌지와 자몽의 휘발성 유기화합물 확인 (Determination of the volatile flavor components of orange and grapefruit by simultaneous distillation-extraction)

  • 오렌지와 자몽의 휘발성 향기성분을 분석하기 위하여 과육과 과피로 분리하여 각각 SDE 방법으로 휘발성 성분을 추출한 후 GC-MS로 확인하였다. 오렌지의 과육 및 과피의 휘발성 향기성분은 각각 120.55 및 4,510.81 mg/kg이며, 자몽의 과육과 과피는 195.60 및 4,223.68 mg/kg으로 확인되었다. 오렌지와 자몽의 휘발성 향기성분 중 가장 높은 함량으로 확인된 성분은 monoterpene류인 limonene으로 과육은 65.32 및 105.00 mg/kg으로 과피는 3,008.10 및 1,870.24 mg/kg으로 정량되었다. Limonene, sabinene, ${\alpha}$-pinene, ${\beta}$-myrcene, linalool, (Z)-limonene oxide, (E)-limonene oxide 등은 오렌지와 자몽의 공통적인 주요 향기로 확인되었다. 오렌지의 특징적인 향기성분은 valencene이며, 자몽은 (E)-caryophyllene 및 nootkatone으로 확인되었다. $\delta$-3-Carene, ${\alpha}$-terpinolene, borneol, citronellyl acetate, piperitone 및 ${\beta}$-copaene 등은 오렌지에서만 확인된 화합물이며, ${\beta}$-pinene, ${\alpha}$-terpinyl acetate, bicyclogermacrene, nootkatol, ${\beta}$-cubebene 및 sesquisabinene는 자몽에서만 확인되었다. Phenylacetal dehyde, camphor, limona ketone, (Z)-caryophyllene은 과육에서 확인되었으며, ${\alpha}$-thujene, citronellal, citronellol, ${\alpha}$-sinensal, ${\gamma}$-muurolene, germacrene D 등은 과피에서만 확인된 성분이다. 본 연구결과 오렌지 및 자몽의 과육과 과피의 휘발성 향기성분 함량과 조성의 차이를 확인할 수 있었다.

두유에서 자몽씨 추출물의 Bacillus 균에 대한 항균효과 (Antibacterial Effects on Bacillus Stearothermophilus by Adding Natural Grapefruit Seed Extracts in Soymilk)

  • 천연향균물질인 자몽씨 추출물을 0.01~0.03% 첨가하여 내열성 포자형성균인 Bacillus stearothermophilus를 비롯하여, 저온성 세균인 Bacillus subtilis, 식품 독성을 일으키는 Staptylococcus aureus 등에 대하여 생장억제 효과를 분석 하였다. 그리고 이 추출물을 실제 공정에 이용하기 위한 현장실험에서는 두유에 자몽씨 추출물을 0.2% 첨가 시 B. stearothermophilus의 성장은 억제 되었으나 완전사멸은 되지 않았고 B. subtilis와 S. aureus는 각각 48 h과 72 h내에 사멸되었다. 그러나 이러한 조건은 경제성, 유화안정성, 관능 상에 문제가 발생되며, 그 해결방안으로 두유에 자몽씨 추출물의 0.015% 첨가와 $121^{\circ}C$에서 10 min간 처리하는 열처리조건을 병행하므로서 균을 완전히 사멸하고 제품의 안정성과 관능 상의 문제점 해결 가능성을 높일 수 있었다.

자몽 종자 추출물의 항균성 (Antimicrobial Activity of Grapefruit Seed Extract)

  • 자몽 종자 추출물의 항균성을 조사하기 위하여 그람 양성균인 Bacillus cereus, Bacillus subtilis, Listeria monocytogenes와 그람 음성균인 Escherichia coli, Salmonella enteritidis, Serratia marcescem를 대상으로 하여 최소 저해 농도, 생육 저해 활성, 콜로니 형성 저해 활성을 실험하였다. 그람 양성균에 대한 자몽 종자 추출물의 최소 저해 농도는 12.5 ppm정도로 상대적으로 낮았으며, 그람음성균의 최소저해 농도는 50ppm 이상으로 상대적으로 높았다. 균주의 증식에 대한 생육 저해 활성을 조사한 결과 Bacillus cereus는 1ppm 이하, Bacillus subtilis는 6.25 ppm, Listeria monocytogenes는 1ppm 이하, Escherichia coli는 6.25 ppm, Salmonella enteritidis는 25 ppm Serratia marcescem는 25pp부터 생육이 저해되었다. 그람 양성균의 콜로니 형성 저해활성은 Bacillus cereus는 93.9%, Bacillus subtilis는 94.0%, Listeria monocytogenes는 99.9%, 그람 음성균의 콜로니 형성 저해 활성은 Escherichia coli는 4.4%, Salmonella enteritidis는 82.7%, Serratia marcesem는 86.4%로, 그람 양성균에 대해서는 높은 저해 활성을 보였으며, 그람 음성균에 대해서는 비교적 낮은 저해 활성을 보였다.

Grapefruit 종자추출물을 이용한 Aspergillus parasiticus의 생육 및 Aflatoxin 생성억제 효과 (Inhibitory Effects of Grapefuit Seed Extract on Growth and Aflatoxin Production of Aspergillus parasiticus)

  • 농산물 및 그 가공식품을 수확·저장·가공하는 기간중에 Aspergillus parasiticus가 오염되어 aflatoxin을 생성하는 것을 방지할 목적으로 생육배지에 graefruit 종자추출물(GFSE)을 처리하여 곰팡이의 생육 및 aflatoxin 생합성을 저해하는 뚜렷한 효과를 관찰할 수 있었다. 4000 ppm의 GFSE농도 처리로 Aspergillus parasiticus의 생육을 92% 저해하였으며, 5000ppm 농도에서는 곰팡이 생육을 완전히 저해하였고 3000ppm 이상의 농도에서 aflatoxin 의생성을 100% 억제하였다. 한편, GFSE 처리에 의하여 aflatoxin 생합성 경로증, averufin, versiconal acetate 등의 중간대사산물이 aflatoxin으로 주입되는 것이 완전히 저해된 반면, versicolorin A, versicolorin A hemiacetal, sterigmatocystin 등은 aflatoxin 으로 전환되는 결과를 보여주어 GFSE는 versiconal acetate를 포함한 그 이전의 중간 대사물이 반응기질이 되는 효소 반응계를 저해하는 것으로 나타났다. 전자현미경을 이용한 곰팡이균체 및 포자의 형태변화는 GFSE처리에 의하여 세포막기능에 파괴되어 세포구성 내용물이 유실되고 곰팡이가 사멸하고 aflatoxin 생합성 기작이 중단되는 결과를 초래하였다.

  • To investigate the effect of several bactericides and fungicides against fruit & vegetable decay grapefruit seed extract(GFSE) was tested in a dilution solution. GFSE was shown to be effective against decay and rind breakdown and to extend the shelf-life of fruits and vegetables during storage and tiansport. Antimicrobial efficiency of GFSE on a wide spectra of gram + & - bacteria, moulds and yeasts was demonstrated by the measurement of minimal inhibitory concentrations performed (Bacteria 10-500ppm, Fungi:250-1,000ppm, Yeasts:100-250ppm). GFSE was separated and extracted into water-soluble fraction, water-insoluble and non-dialyzed fraction to isolate the antimicrobial substances. The water-soluble fraction showed the most active antimicrobial effect. The antimicrobial substances were isolated by gas chromatography. As the result of the isolation using GC, Peak-D was found to be the antimicrobial compound in GFSE. The identification of the most antimicrobial substance was carried out by using GC-MS.

  • The antibacterial and antifungal effect of grapefruit seed extract(GFSE) was investigated for its purpose of application to a diverse spectrum of field as sanitizers, disinfectants and preservatives. GFSE showed coparatively high content of such flavoniods as naringin and hesperidin and ascorbic acid. GFSE containing a low level of organic acids is a heavy viscous and water-soluble liquid. As a result of the antimicrobial test of GFSE, Bacillus subtilis and Aspergillus oryzae did not survive at detectable levels when treated with more than 100 ppm of GFSE. The minimum inhibitory concentrations of GFSE for a wide variety of pathogenic and putrefactive bacteria, fungi and yeasts were 100 ppm and 250 ppm, respectively. In the comparable electron micrograph of microbial cells treated with GFSE or not, we could conclude that GFSE destroy microorganisms by disrupting the functions of the cell wall membrane and microbial spores.

  • The antimicrobial effect of mixed solutions of grapefruit seed extract(250ppm) and ascorbic acid(1%) on the spoilage microorganisms such as Staphylococcus epidermis, Escherichia coli, Pseudomonas syringae, Candida albicans and Corynebacterium xerosis isolated from the spoiled soybean sprouts were investigated. Cell wall and membrane were partially destroyed and the contents of the destroyed cell were exuded after treatment. Packages with 30 m cast polypropylene(CPP), 16 m polyolefin(RD 106) and 10 m high density polyethylene(HDPE) were applied for soybean sprouts dipped in mixed solutions respectively. Oxygen and carbon dioxide concentration inside packages were dependent on the kind of films during storage at 5oC. The antimicrobial activity of mixed solutions was maintained for 5 days at CPP package. Package with HDPE showed a severe browning than the others after 5 days. Ascorbic acid content of mixed solution treatment was higher than that of control for each package.
