Fruit juices such as grape juice are associated with healthy products by consumers because of the many health benefits they provide. Farm produced (FPGJ) and commercially produced grape juice (CPGJ) in South Korea were compared and studied through the evaluation of their physicochemical, functional, and sensory properties. The results of this study show that FPGJ's physicochemical properties are more varied than CPGJ. The pH, titratable acidity, and total soluble solids of FPGJ were higher than CPGJ. FPGJ had a higher mean value for total phenolics ($2,558.20{\pm}50.06\;mg/L$ GAE), total flavonoid ($3,236.80{\pm}56.11\;mg/L$), total anthocyanin ($559.88{\pm}3.51\;mg/L$), and radical scavenging activity (86.48%) than CPGJ, although the differences were significant only with regard to total flavonoid and total anthocyanin. This study also demonstrates that CPGJ is preferred in terms of sensory evaluation. These properties may be used as a basis for the optimization of processing to produce a higher quality grape juice.
Grapes (Vitis vinifera) are one of the most important fruits worldwide and are eaten raw or after conversion to jelly, jam, juice and wine. Grape skins are a major source of resveratrol (3,5,4'-trihydroxystilbene), which has the ability to reduce blood sugar as well as anticancer, anti-inflammatory, and other beneficial cardiovascular effects. In this study, we investigated the increased accumulation of resveratrol in grape skin and leaves following ultrasonication treatment, which has been shown to induce resveratrol accumulation in several plants. Various ultrasonication treatment times and incubation periods were employed to identify the optimum conditions for the maximum accumulation of resveratrol. Treatment and further incubation led to increased resveratrol in both grape skins and leaves, with the highest increases of 7.7-fold and 1.9-fold occurring in response to 5 min ultrasonication treatment followed by 6 hour incubation and 15 min ultrasonication treatment followed by 3 hour incubation, respectively. The underlying mechanism for the increased amounts of resveratrol were studied by employing a semi-quantitative RT-PCR to monitor the expression levels of the resveratrol synthase (RS) gene in response to ultrasonication treatment. The RS gene increased the expression in response to ultrasonication treatment, suggesting that up-regulation of the RS gene by ultrasonication treatment triggers increased amounts of resveratrol. Taken together, these data indicate that this simple ultrasonication treatment of grapes can be an efficient post-harvest technology for increasing resveratrol in grape skins in addition to cleaning the fruits.
Many of the natural dyes used for natural dyeing are difficult to maintain colorfastness due to their complex structure and specific properties. Therefore, there is a need for developing of color sustainable ability for use as an advanced coloring agent for fabrics, which would eco benign or not. In this study, the natural dye extracted from the waste of grape fruits was used to dye cotton fabric. Thus, the present study aims at extraction of color from grape seeds, skin, and stem through fermentation and then employing the same in dyeing and mordanting of cotton. Dyeing experiments were done under different conditions of fermentation and protein type mordants which were treated before and after dyeing. Experimental fabrics were used with cotton after scouring. Color value of dyed fabrics and color fastness of cotton dyed fabrics to washing and light were measured. The fastness of dyed experimental fabrics was increased by mordanting of protein fermentation and the color of dyed cotton was light red purple. The color of dyed fabric found with the optimum mordant treatment when treated with pre milk-mordant at $40^{\circ}C$ for 30min and 3% grape seed extract. On the whole, reddish tone very slightly increased with the milk pre-mordant. The color fastness of dyed cotton fabrics to light and washing was increased after fermentation.
Exopolysaccharide produced by the yeast Papiliotrema flavescens, isolated from wine grape berries of Champagne vineyard, was investigated for both chemical and functional characterization. SECMALLS and colorimetric assay analyses showed that the EPS is a high MW heteropolymer (2.37 × 106 g/mol) majorily consisting of mannose, glucose, xylose and glucuronic acid as monosaccharide constituents, with two substituents (sulphate and phosphate groups), and a minor protein moiety. Structural enchainment of these carbohydrates based on methylation, GC-MS and NMR analyses revealed a linear main backbone built up of α-(1 → 3)-D-mannopyranosyl residues on which are branched side chains consisting of a single β-D-glucopyranosyluronic acid residue and β-(1 → 2)-xylopyranoses (2-5 residues). Suggestion of some xylopyranose side chains containing a mannose residue at the nonreducing terminal end was also proposed. This is first report on EPSs from the grape P. flavescens yeast with such structural characteristics. Furthermore, investigations for valuating the application performance of these EPS in relation with their structural features were carried out in 8% alcohol experiment solutions. Very exceptional viscosifying and foaming properties were reported by comparison with commercial biopolymers such as Arabic, gellan and xanthan gums. The intrinsic properties of the natural biopolymer from this wild grape-associated P. flavescens yeast make it a potential candidate for use in various biotechnology applications.
Anthocyanins were isolated and identified from grape peels which were wasted much in Korea, and their characteristics were as follows .Isolated pigments from grape peels were 11 varieties such as 5 varieties of 3, 5-diglucoside (DG), 6 varieties of 3-monoglucoside (MG), and acylated pigment was 2 varieties of them. Malvidin was 4 varieties , petunidin , peonidin and delphinidin 2 varieties in each, and cyanidin 1 variety of 11 vareities. Malvidin -3, 5-diglucoside and peonidin -3, 5-diglucoside of anthocyanins were above 48% in total anthocyanins content of 114.99mg/g in dried skins. Breakdown of anthocyanins was higher become intimate neutral pH, but stable to stroage period for 7-days. Hyperchromic effects were showed when sugars were added in pigment extract of grape peels, the highest value was glucose and the next ordor was fructose and sucrose. Breakdown velosity of anthocyanins was higher when ascorbic acid was added, but its velocity was reduced in anaerobic state . Absorption degree by organic acid treatment was higher than control, and anthocyanins were stable to storage period for 7 days.
The aim of the present study was to assess the anti-obesity effects of grape seed extract (GSE) supplement in C57BL/6J mice. Thirty mice were divided into three groups; normal diet control group (ND), high fat diet control group (HD) and high fat diet plus grape seed extract supplemented group (HD+GSE). Results were as follows: 1. GSE supplement reduced the weight gain in mice fed high fat diets; epididymal and back fat weights, were lower compared to non-supplemented HD group. 2. Blood lipid concentrations were lower in the HD+GSE group than in the HD group. Serum HDL-C concentrations were higher in the HD+GSE group compared with the other groups. 3. The concentrations of acid-insoluble acylcarnitines, (AIAC) in serum and liver were higher in the HD+GSE group than in the HD group. 4. GSE supplementation increased mRNA levels of lipolytic genes such as carnitine palmitoyltransferase-l (CPT-1) and decreased mRNA levels of lipogenic genes such as acetyl CoA carboxylase (ACC). These findings suggest that grape seed extract supplements in high fat diet might normalize body weight, epididymal and back fat weights, lipid concentrations, and carnitine levels through controlling lipid metabolism.
현재 식품공전 및 식품첨가물 공전에서 포도씨유 및 추출물의 catechin 함량 측정방법으로 널리 사용되고 있는 바닐린비색법을 polyamide cartridge를 사용함으로써 개선하였고 보다 정확하며 재현성 있게 catechin 함량을 측정할 수 있는 HPLC 방법을 개발하였다. 포도씨유 및 용매(열수, 에탄올, 메탄올 및 아세톤)추출물의 catechin 함량을 바닐린비색법으로 측정한 결과 각각 $30{\sim}40\;mg%$ (g/포도씨유 100g) 및 $17{\sim}43%$(g/추출물 100g) 범위를 차지하였다. 반면, polyamide cartridge를 사용한 개량 바닐린비색법으로 위의 포도씨유 및 추출물의 catechin 함량을 측정한 결과 각각 미량($1{\sim}5ppm$) 및 $4.0{\sim}7.5%$ 차지하였다. 한편, HPLC 방법으로 포도씨유의 catechin 함량을 측정한 결과 포도씨의 주된 4가지 catechin 성분[(+)-catechin, procyanidin $B_2$, (-)-epicatechin 및 (-)-epicatechin gallate]은 거의 확인할 수 없었다. 그러나 포도씨 용매추출물의 catechin 함량을 측정한 결과 4가지 주된 catechin 성분을 확인할 수 있었으며, 그 중 (+)-catechin 및 (-)-epicatechin 함량이 각각 $1.35{\sim}2.60%$ 및 $2.35{\sim}4.59%$ 범위로서 상당히 높은 반면, procyanidin $B_2$ 및 (-)-epicatechin gallate 함량은 각각 $0.77{\sim}1.36%$ 및 0.06${\sim}0.30%$ 범위로서 상대적으로 낮았다 또한, HPLC에 의해 국내의 포도 품종별 및 산지별 포도씨의 catechin 조성 함량을 측정하였다. 이와같이 위에서 정립된 HPLC 방법에 의한 포도씨의 4가지 주된 catechin 성분의 회수율은 95% 이상으로 높았으며, 재현율은 모두 5% 미만의 편차를 나타내어 분리효율이 우수하였다. 그리고 4가지 catechin 성분의 검출한계는 $1{\sim}5\;ppm$ 범위를 나타내었다.
국내의 산간지역을 중심으로 재배면적이 확대되고 있는 개량머루의 소비촉진의 방안으로 안전재배를 도모하고 머루재배 농가의 소득증대에 이바지하고자 개량머루주의 고품질화 기술개발연구를 실시하고 다음과 같이 요약 정리한다. <시험 1> 개량머루의 술 가공적성 구명 가. 개량머루의 과립중은 3.7g으로 포도 과립중 12.2g의 1/4배 정도였다. 나. 개량머루의 pH는 3.34로 포도의 pH 3.62 보다 0.3정도 낮고 적정산도함량이 높아 신맛이 강하며, 당도는 개량머루가 17°Brix%로 포도 15°Brix 보다 높았다. 다, 개량머루의 색차는 포도에 비해 밝기, 적색도 및 황색도가 낮아 검붉은 색을 띤다. <시험 2> 개량머루주의 품질향상 가. 개량머루의 원형, 파쇄 및 즙형으로 술을 제조하였을 머루의 형태별 pH는 3.70~3.75, 적정산도는 0.91~0.96㎖, 환원당은 0.34~0.42%, 알콜함량은 16.2~16.3%로 처리 간에 큰 차이가 없었으며, 색차와 식미에서도 차이가 거의 없었지만 발효속도를 고려할때는 머루를 약간 파쇄하여 담그는 것이 좋을 것으로 생각한다. 나. 개량머루주에 참나무의 조제 형태별 및 첨가량별에 따른 1년동안 숙성된 머루주의 pH, 적정산도, 환원당 및 알콜함량은 거의 차이가 없었다. 그러나 참나무의 조제형태별 첨가량별 모든 처리구의 식미가 무첨가 3.0(보통이다)보다 높은 점수를 얻었으며, 특히 참나무톱밥을 1.0% 첨가로 맛, 냄새, 색에서 품질이 향상되었다. 다. 개량머루주 발효 후 숯을 첨가하여 숙성시킨 머루주는 숯 첨가량이 많을수록 pH는 3.82에서 3.86으로 증가하였으며, 적정산도는 0.91㎖에서 0.86㎖로 감소하였다. 그리고 알콜함량은 각 처리간에 차이가 없었다. 색차에서 명도는 약간 떨어지는 경향을 보였으며, 적색도는 증가하는 경향을 보였다. 숯 3% 첨가로 냄새와 색은 보통이상으로 향상되었다. 라. 개량머루주의 숙성시 참나무톱밥 1%나 참나무통에 숯 3% 첨가시 pH, 적정산도는 별차이가 없었으나 무첨가 머루주와 차가 있었다. 색차에서는 숯의 첨가량이 많을수록 명도와 적색도도 높아지는 경향이었다. 식미에서는 참나무톱밥 1%나 참나무통에 숯 3%를 넣고 숙성시켰을 때 식미가 "좋다"라는 평가를 받았다.
We analyzed and compared profiles of flavonols extracted from leaves and exocarps of the grape Kyoho by TLC, HPLC and UV spectrophotometry. In the exocarps, quercetin 3-O-glucoside was the main compound while isorhamnetin 3-O-glycoside (I) was present in minor amounts. In leaves, on the other hand, quercetin 3-O-glucoside and quercetin 3-O-glucoside-7-O-glucronide were the major compounds while isorhamnetin 3-O-glycoside (II) and kaempferol 3, 7-O-diglycoside were present in minor amounts.

Polyphenolic compounds have been reported to exhibit a wide range of pharmacological properties. In this study. we investigated the hepatoprotective effect of skin and seed of grape which contain abundant polyphenol compounds on dimethylnitrosamine(DMN)-induced liver damage in rats. Ingestion of skin and seed of grape (10% diet, daily for 4 weeks) into the DMN-treated rats remarkably prevented the elevation of serum alanine transaminase, aspartate transaminase and alkaline phosphatase, and bilirubin levels.
