Sparfloxacin is a new synthetic quinolone antimicrobial developed at the Research Laboratories of Dainippon Pharmaceutical Co, Ltd. To evaluate the efficacy and safety of sparfloxacin in acute pulmonary infection, we administered sparfloxacina(100mg) twice in a day to 30 patients who had signs and symptoms of acute pulmonary infectious diseases regardless of their underlying lung disease for 7 days. The results were as follows: 1) A total 30 patients were enrolled in the trial. Among them, 24 cases(80%) had underlying lung problems such as chronic obstructive pulmonary disease(36.4%), bronchiectasis(36.4%), bronchial asthma(3.3%), or lung cancer(3.3%). 2) In 26 cases(86.6%), we observed effective improvement, and 4 cases(13.4%) show mildly effective improvement of symptoms and signs of respiratory infection. 3) In 23 cases(73.4%), we observed bacteriological eradication in culture or decreased the number of bacteria by Gram stain which found dominantly in previous Gram stain. 4) The significant side effect was not noted. The above results suggested that sparfloxacin was effective as a first line therapy in patients with acute respiratory infection.
Helicobacter suis is a gram negative bacterium and colonizes in porcine stomach. It causes gastric diseases in the stomach and plays a significant role in daily weight gains in pigs. Recent studies about one of potential sources of human gastric diseases. Therefore, this study was conducted to compare histopathological lesions and molecular detection of Helicobacter suis in the pyloric mucosa of porcine stomachs transferred from slaughterhouses, based on gross and histological examinations and a PCR assay. A total 90 stomach samples were investigated to record gastric lesion scores by characteristic gastric lesions, followed by routine H & E and Warthin-Starry silver staining to detect Helicobacter-like organisms. For PCR assay, H. suis specific primers and conditions are used. Sixty-one samples (67.8%) showed gross gastric lesions, of which 38 samples (40.2%) presented grade 1, 12 samples (13.3%) presented grade 2, and 11 samples (12.2%) presented grade 3, respectively. In Warthin-Starry silver stain, Helicobacter-like organisms were detected from 11 samples (12.2%) with 4 samples (4.4%) for grade 0, 5 samples (5.6%) for grade 1, 1 sample (1.1%) for grade 2 and 1 sample (1.1%) for grade 3, respectively. The PCR resulted positive in 37 samples (41.1%) with 14 samples (15.6%) for grade 0, 14 samples (15.6%) for grade 1, 3 samples (3.3%) for grade 2 and 6 samples (6.7%) for grade 3, respectively. Positive samples for both examinations were 5 samples (5.6%). The result suggested that it should be considered as one of factors causing a gastric disease in pigs. Also, it could be acknowledged to research fundamental aspects of Helicobacter-induced gastritis in human as an animal model.

Cellular phones are the most used electronic device everyday in modern life and are always in contact with our hands, Although many studies have revealed microorganisms living on our hands, there are only a few reports on the research about products or places which are in contact with our hands. Therefore, the purpose of this study is to verify microorganisms living in cellular phones. Microorganisms were scraped from cellular phones of students and professors from the clinical laboratory science department in Daegu Health College, and cultured at Brain Heart Infusion agar and MacConkey agar following API kit to identify them. The average colony number was $1.5{\times}10^2$ on BHI agar and $40{\times}10$ on MacConkey agar. There was no difference according to gender. In Gram stain result, Gram(+) Cocci showed the highest frequency. Also in BHI agar plates, Micrococcus spp and Acinetobacter baumannii identified with high frequency. Moreover, S. aureus, which is very well known as strong food poisoning bacteria, was isolated. Klebsiella pneumonia ssp pneumonia was isolated with the highest frequency from the MacConkey agar or S-S agar plate. From these results show, there are as many different microorganisms from cellular phones as from our hands. This is the first report isolating strong food poisoning bacteria in cellular phones. Since infection in hospitals have been an important issue to be aware of, it is equally necessary to investigate cell phones and products which hospital workers touch with their hands.
Crocodile farms are getting popular in Bangladesh in an economic point of view. In one of the farms, some crocodiles were found sick and three of them died between May and July in 2006. This investigation was performed to diagnose the cause of the death. Routine postmortem examination was conducted. Samples were collected in 10% neutral buffered formalin for histopathology and in falcon tube for microbiological study. Additional swabs were collected in nutrient broth. Histopathological and microbiological studies were conducted using routine procedures. In addition Giemsa, Gram and PAS stains were performed to detect the organism in tissues. Grossly, esophagus, trachea, lungs, liver, spleen, heart and kidney were congested. Intestine, rectum and colon were hemorrhagic. Clay colored material was found in colo-rectum. Purulent exudates in lungs and thick and cloudy pericardial fluid in pericardial sac were found. Histologically, multifocal granulomatous inflammation was evident in lung, liver, kidney, intestine and colon with bacterial colonies, fungal spores and hyphae. These bacteria were appeared as Gram negative. Fungal hyphae and spores were detected in liver, lungs and colon by using PAS stain. Bacteriologically, E. coli were isolated from lungs exudates, pericardial fluids and intestinal fluids. Therefore, it can be concluded that 3 crocodiles died due to E. coli septicemia concurrent with mycotic infection.
Dermatopathologic findings on cutaneous streptothricosis (bovine dermatophilosis) were conducted in Dinajpur of Bangladesh for one year. The animals were examined clinically at hospital during the physical visit of the affected animals and clinical manifestations and patients' history were recorded. The gross lesions were systematically examined, noted and categorized. An impression smear was made by the skin scraps, stained with Gram's stain and examined microscopically for the presence of causative bacteria. The suitable sizes of skins of 3 animals typically affected with dermatophilosis were collected surgically from the animals, and prepared for the histopathological study. The cardinal clinical features and gross lesions were characterized as paint-brush cutaneous surface with matted hairs, scab and crust formation, roughened coat; istopathologically as hyperkeratosis, parakeratosis, typical "palisading encrustations", extensive proliferation of coccoid shaped, bluish coloured bacteria (zoospores) in the cells of the outer rootsheath of the hair follicles, separation of keratinized layer from the epidermal epithelium, epidermal hyperplasia, densely cellular dermis with moderate destruction of glandular structures. Coccoid shaped gram positive organisms which form multidimensional arrangement were also observed by impression smear. The clinical signs, gross lesions, morphology of the organisms, staining properties, and histopathological features of the affected skin were diagnostic criteria for the bovine cutaneous streptothricosis.
The role of bacteria in root canals and periapical infections is well known and established. In these bacteria, black-pigmented bacteria(BPH) play important role in endodontic infection. BPB are Gram negative anaerobic rods which are closely related 50 clinical symptoms such as pain, percussion, tenderness, foul odor, etc. In America and Europe, many studies on BPB have been done and are continued. But, relatively few studies have been done in Korea, especially its prevalence in Korean population is not yet studied. The purpose of this study is to establish prevalence of BPB in infected root canals and periapical abscesses in Korean people. Microbial samples were collected from the root canals of 34 intact tooth with periapical rarefactions of endodontic origin and 3 periapical abscesses. All samples were incubated in an anaerobic chamber(Coy, Model No. 77. Ann Arbor, Michigan, USA.). Identification of In microorganism was based on its growth in the anaerobic chamber, colonial pigmentation, colonial morphology, Gram stain, and Rapid ID32A(BioMericux SA/69280 Marcy-l'Etoile/France) results. In addition, the polyme ase chain reaction using specific primers for 16S rRNA genes were used differentiate Prevotella nigrescens for Prevotella intermedia. The results were as follows : 1. In this study, thirteen (35%) of thirty seven samples were positive for the growth of BPB. In thirteen samples, sixteen strains of BPR were found. 2. The most frequently identified BPB in root canals and abscesses in Korean were P. nigrescens 5/37(14%) and P. intermedia 5/37(14%). Porphyromonas gingivalis 3/37(8%), Porphyromonas endodontalis 2/37(5%) and Prevotella loecheii 1/37(3%) were also found. 3. In this study, no significant differences were found between the prevalence of BPB in Korean and that of American and European.
A fourteen-month-old Holstein bull from a private cattle fattening farm at Ansung county in Kyunggi Province Korea was submitted on August 2nd 1997, for examination at the Pathology Division of the National Veterinary Research Institute. The bull died within 24 hours after developing clinical signs of abrupt inertia, complete anorexia high fever(40.3℃) diffuse severe emphysematous swelling of upper part of the right hind leg lateral recumbancy and paralysis. At necropsy diffuse severe subcutaneous redness and influx of serosanguinous exudate containing gas bubbles had accumulated under the thorax right hip and upper region of right hind leg. Muscles in upper right hind leg were blackish to dark red and yellowish brown in color. Muscle bundles were dry and separated by gas bubbles and serosanguinous fluids and muscles sections from affected regions floated in water. Histopathologically muscle fibers were partially or entirely degenerated fragmented and separated by exudate and gaseous substance mixed with polymorphonuclear cells. Blood vascular walls in affected regions showed severe acute fibrinoid necrosis. Typical large rod-shaped bacteria with or without oval central to subterminal spores were frequently observed in tissue sections stained with H-E and Gram stain. The large Gram-positive anaerobic endospore-producing rods were isolated from the suspension of muscle lesions. Isolated bacteria were identified as Clostridium(CL) chauvoei and CL. sordellii by biochemical tests. This case was diagnosed as blackleg based on the typical clinical signs gross finding histopathological observation and bacteriological results. This is the first case report on blackleg associated with Cl. chauvoei and Cl. sordellii in Holstein cattle in Korea.

분만후 젖소의 자궁내 미생물상을 조사하고 미생물로부터 분리한 Lipopolysaccharide를 적용하여 소의 번식효율 증진에 기여하고자 분만후 젖소의 도축장 유래 자궁을 채취하여 혐기적 상태에서 균분리 동정을 실시하였다. 균분리 동정을 위하여, 시료를 1cm$\times$1cm로 채취하여 혐기상태에서 거품이 생길 때까지 vortexing한 후 균액 300$\mu l$를 뽑아 혐기배지에 도말하였고 도말한 plate는 $37{\circ}C$ 혐기chamber에서 24시간 배양하였다. 혐기배지에서 자란 균의 colony를 따서 Mac, BHI+B, BHI 배지에 배양한 후 Gram stain을 실시하였다. BHI 배지에서 자란 균의 colony를 따서 BUA+B 배지에 계대배양하였고 BUA+B 배지에서 자란 균중에 가장 마지막으로 자란 균을 따서 An-IF에 넣고 탁도를 63%T로 맞춘 후 An micro plate에 100$\mu l$씩 분주하였다. 분주한 plate를 $37{\circ}C$ 혐기 chamber에서 20~24시간 동안 배양한 후 Biolog를 실시하였다. 시료의 UV측정을 위하여 Sonic Processor로 세포를 분쇄하였고 분쇄한 세포를 $4{\circ}C$에서 10,000rpm으로 10분간 원심분리한 후 상층액을 분리하여 0.45$\mu m$ 필터로 여과한 다음 여과액을 취하여 UV로 standard(E.coli O26 B6 LPS)와 sample(10배 및 20배 희석액)을 측정하였다. The activated sludge from the aeration basin of the Su-yeong municipal wastewater treatment plant which has operated by a standard activated sludge process in Busan, Korea was investigated during April 2004 and January 2005 with several bio-indicators. The number of bacteria and fungi per gram of dry weight of MLSS were estimated to be $3.1\times10^6\sim1.5\times10^8\;and\;l.1\times10^3\sim1.1\times10^5$ colony forming units, respectively, by the plate agar method. By cultivation-independent methods, such as 4',6-diamidino-2-phenylindole stain and fluorescence in situ hybridization, the ratio of eubacteria to the entire biomass was evaluated by more than $80\%$ (v/v). The ratio of ammonia-oxidizing bacteria and nitrite-oxidizing bacteria to the total eubacteria was detennined to be $7.0\sim9.8\%\;and\;3.3\sim6.2\%$ without heavy variation in spite of a period of relatively low temperature in the basin. It would be expected that the nitrification would occur or at least co-exist throughout the year in the sludge of many municipal WWTP with influents that contain the sufficient nitrogen sources although the WWTP does not have any specialized processes for the removal of nitrogen.
Toll-like receptor 4 (TLR4) together with MD2, one of the key pattern recognition receptors for a pathogen-associated molecular pattern, activates innate immunity by recognizing lipopolysaccharide (LPS) of Gram-negative bacteria. Although LBP and CD14 catalyze LPS transfer to the TLR4/MD2 complex, the detail mechanisms underlying this dynamic LPS transfer remain elusive. Using negative-stain electron microscopy, we visualized the dynamic intermediate complexes during LPS transfer-LBP/LPS micelles and ternary CD14/LBP/LPS micelle complexes. We also reconstituted the entire cascade of LPS transfer to TLR4/MD2 in a total internal reflection fluorescence (TIRF) microscope for a single molecule fluorescence analysis. These analyses reveal longitudinal LBP binding to the surface of LPS micelles and multi-round binding/unbinding of CD14 to single LBP/LPS micelles via key charged residues on LBP and CD14. Finally, we reveal that a single LPS molecule bound to CD14 is transferred to TLR4/MD2 in a TLR4-dependent manner. These discoveries, which clarify the molecular mechanism of dynamic LPS transfer to TLR4/MD2 via LBP and CD14, provide novel insights into the initiation of innate immune responses.
