실험적 간섬유화에서 Fat-storing Cell의 미세구조의 변화에 대한 연구 (Ultrastructural Changes of Fat-storing Cells in Experimental Hepatic Fibrosis)

  • 간 섬유화에서 fat-storing cell의 변화를 미세 구조학적 측면에서 관찰하고자, 사염화탄소를 mineral oil에 녹여 1 : 1로 회석한 후 Kg당 0.5 ml의 용량을 주 2회씩 12주간 흰쥐에 복강 주입하였다. 실험기간 동안 1주 간격으로 도살하고, 적출된 간조직을 광학현미경과 연역조직화학적 검사 및 전자현미경으로 관찰하여 다음과 같은 결과를 얻었다. 광학현미경적 소견상 소엽 중심주의 염증 반응 및 세포 침윤 현상은 사염화탄소 투여 후 1주부터 나타나서 4주 째 가장 심하였으며, 이시기에 desmin염색 반응 양성세포의 수도 증가되었다. 괴사 및 섬유화는 2주부터 나타나기 시작하여 6주부터 결절이 형성되기 시작하였고 괴사성 병변은 8주까지 지속되었다. 그러나 세포 침윤 정도는 감소되면서 desmin에 대한 반응성도 낮아졌고, 경변성 변화는 10주 이후부터 나타났다. 전자현미경적 소견상 fat-storing cell의 숫적 증가는 1주부터 관찰되었으며 지방적이 감소하고 rER이 팽창된 이행세포는 2주째 출현하여 4주에 가장 현저히 증가되었다. 섬유아세포는 3주부터 나타나기 시작하였는데, rER이 확장되었으며 지방적은 없거나 1-2개의 소적이 세포돌기에서 관찰되었다. 섬유화가 진행됨에 따라 이행세포 및 fat-storing cell은 감소하고 섬유아세포는 증가된 상태를 유지하였다. 이상의 결과로 보아 fat-storing cell은 간섬유화 과정에서 활성화되고 증식하여 형태학적으로 이행세포 및 섬유아세포로 전환함으로써, 정지상태에 있는 섬유아세포의 전구체로서 간섬유화에 중요한 역할을 하는 것으로 사료된다.

토끼 뇌감염 모델의 CT 소견과 조영제 주입 후 동맥혈의 Hounsfield Unit의 변화 (CT Scan Findings of Rabbit Brain Infection Model and Changes in Hounsfield Unit of Arterial Blood after Injecting Contrast Medium)

  • 토끼 뇌에 대장균을 주입하여 CT 소견을 알아보고, 시간에 따른 동맥혈의 하운스필드 값의 변화를 알아보고자 하였다. 토끼 두개관에 천두공(burr hole)을 뚫고 2~3 mm 깊이에 대장균 $1{\times}10^7$ CFU/ml, 0.1 ml을 주입하여 뇌염증 모델을 제작하고, 조영 증강 CT와 동적 CT, 그리고 동맥혈의 CT영상을 얻었다.조영 증강 CT에서 뇌농양, 뇌실염 그리고 뇌막염등 다양한 뇌염증 소견이 보였다. 뇌농양은 중앙부가 거의 조영되지 않고 주변부가 강하게 조영되는 전형적인 양상을 보였고, 뇌실염은 측뇌실 벽을 따라 강하게 조영되는 소견을 보였으며, 뇌막염은 종뇌와 간뇌의 접히는 부위 뇌막이 강하게 조영되었다. 동적 CT영상에서 염증 중앙부의 조영제 주입 전 HU 값은 $31.01{\pm}3.55$였고, 주입 후 10분까지 $40.36{\pm}3.76$으로 서서히 증가하였다. 그리고 염증 가장자리구역에서 HU 값은 조영제 주입 전에 $47.23{\pm}3.12$였고, 조영제 주입 후 약 45초에 $63.59{\pm}3.31$로 가장 많이 증가 하였으나 이후 20분까지 약간 떨어졌다. 또한 균 주입 반대쪽 정상 뇌조직에서 측정한 HU 값은 조영제 주입 전에 $39.01{\pm}3.24$이었고, 조영제 주입 후 약 30초에 $49.01{\pm}4.29$로 가장 많이 조영되었고, 이후 서서히 낮아졌다. 동맥 혈액 CT에서 조영제 주입 전 HU 값은 $87.78{\pm}6.88$이었고, 조영제 주입 후 10초부터 30초까지 급격히 증가하여 $749.13{\pm}98.48$로 최대값을 보이고, 30초부터 45초까지 $467.85{\pm}62.98$로 급격히 감소하며, 45초에서 60초까지는 정체기(plateau)를 보였으며, 이후 20분까지 $188.28{\pm}25.03$으로 감소되었다. 결과적으로 대장균으로 뇌염증 모델을 만들 수 있고, 조영 증강 CT를 통하여 뇌염증의 특징적인 소견을 잘 알 수 있었으며, 동적 CT를 통해 염증 중앙부와 가장자리구역의 조영 양상을 알 수 있고, 동맥혈은 조영제 주입 후 10초부터 30초 까지 급격히 증가하다 정체기를 거쳐 서서히 감소하는 것으로 나타났다.

청주 상대리 수막재배지의 지중 침투형 갤러리 설치와 예비 주입시험 (Setup of Infiltration Galleries and Preliminary Test for Estimating Its Effectiveness in Sangdae-ri Water Curtain Cultivation Area of Cheongju, Korea)

  • 우리나라 수막재배 지역은 성수기에 지속적인 지하수 사용으로 인해 수막재배 말기에 지하수위 하강과 더불어 지하수 고갈 문제로 항시 어려움을 겪고 있다. 최근에 국내에서는 이러한 지역에서의 지하수 부족 현상을 해결하기 위해 몇 가지 인공함양 방법들이 적용되고 있다. 이 연구에서는 인공함양 방식 중 하나인 지중 침투형 갤러리 설치를 소개하고, 이 중 3개 갤러리에서의 인공 주입시험 결과를 이용하여 인공함양의 효율성을 예비적으로 평가하였다. 연구지역은 청주시 상대리 수막재배지이며, 갤러리의 규모는 가로${\times}$세로${\times}$깊이 $50cm{\times}300cm{\times}50cm$이다. 갤러리는 비닐하우스 동 사이의 이격 공간을 활용하여 설치하였으며, 터파기 작업, 자갈석과 규사 메움, 피조미터와 상부 부직포 설치 순서로 진행하였다. 3개 갤러리에 대한 인공 주입시험은 1차 예비, 2차 단계, 3차 장기 주입시험으로 구성된다. 1차 예비 시험에서 B 갤러리는 주입율 $33.29{\sim}33.84m^3/d$, $45.60{\sim}46.99m^3/d$일 때 수위 상승이 각각 20 cm, 30 cm, C 갤러리는 주입율 $21.1m^3/d$, $33.98m^3/d$, $41.69m^3/d$일 때 수위 상승이 각각 0 cm, 16 cm, 33 cm, D 갤러리는 주입율 $48.10m^3/d$, $52.23m^3/d$일 때 수위 상승이 각각 29 cm, 42 cm 정도를 유지한 것으로 나타났다. 단계 및 장기 주입시험 결과를 이용하여 각 갤러리에 대한 주입율과 수위 관계식이 정량적으로 도출되었으며, 이와 같은 인공 주입시험 결과는 수막재배 기간 중에 사용 후 배출되는 지하수가 갤러리를 통해 자연적으로 주입되는 수량을 추정하는 데에 정량적으로 활용될 수 있을 것으로 기대된다.

Kinetics of Thermal Degradation of Polypropylene/Nanoclay/Wood Flour Nanocomposites

  • As a part of enhancing the performance of wood-plastic composites (WPC), polypropylene (PP)/ nanoclay (NC)/ wood flour (WF) nanocomposites were prepared using melt blending and injection molding process to evaluate their thermal stability. Thermogravimetric analysis (TGA) was employed to investigate thermal degradation kinetics of the nanocomposites both dynamic and isothermal conditions. Dynamic scans of the TGA showed an increased thermal stability of the nanocomposites at moderate wood flour concentrations (up to 20 phr, percentage based on hundred percent resin) while it decreased with the addition of 30 phr wood flour. The activation energy $(E_a)$ of thermal degradation of nanocomposites increased when nanoclay was added and the concentration of wood flour increased. Different equations were used to evaluate isothermal degradation kinetics using the rate of thermal degradation of the composites, expressed as weight loss (%) from their isothermal TGA curves. Degradation occurred at faster rate in the initial stages of about 60 min., and then proceeded in a gradual manner. However, nanocomposites with wood flour of 30 phr heated at $300^{\circ}C$ showed a drastic difference in their degradation behavior, and reached almost a complete decomposition after 40 min. of the isothermal heating. The degree of decomposition was greater at higher temperatures, and the residual weight of isothermal degradation of nanocomposites greatly varied from about 10 to 90%, depending on isothermal temperatures. The isothermal degradation of nanocomposites also increased their thermal stability with the addition of 1 phr nanoclay and of wood flour up to 20 phr. But, the degradation of PP100/NC1/MAPP3/WF30 nanocomposites with 30 phr wood flour occurs at a faster rate compared to those of the others, indicating a decrease in their thermal stability.

토끼의 관절병증에 미치는 알긴산올리고당의 치료효과 (Effect of Alginic Acid on Experimentally Induced Arthropathy in Rabbit Model)

  • For the induction of arthropathy, 5% hydrogen peroxide($H_2O_2$) was injected for 5 weeks into the intraarticular space of the New Zealand white rabbits to damage articular cartilage. Alginic acid of low molecular weight (2%) made from macromolecular alginate treated with enzyme was administered into articular space at the dose of 5 mg/kg twice a week for 3 and 6 weeks using 1 ml syringe and 26 G needle. Saline was injected for the control. Tissues surrounding the articulation were obtained for the measurements of superoxide dismutase(SOD) activity as a major antioxidant enzyme and malondialdehyde (MDA) as a lipid peroxidation level. Histopathologic examination on the surface of articular cartilage was carried out. Data showed that injection of hydrogen peroxide for 5 weeks had led to the induction of free radical damage and of articular cartilage change as confirmed by microscopic observation. The application of hydrogen peroxide caused a gradual increase in the SODs and MDA. These patterns were similar after 3 and 6 weeks of alginate treatment. Furthermore, microscopic examinations revealed that hydrogen peroxide caused flaking, fibrillation, fissuring, denudation, and hypocellularity in the articular surfaces. In conclusion, lipid peroxidation was demonstrated in the articular cartilage by the administration of hydrogen peroxide in the rabbit model. This lipid peroxidation could be caused by oxygen free radicals. The histologic and enzymatic correlations on lipid peroxidation in the articulation have provided a better understanding of arthropathy. It is possible to take advantage of these findings to evaluate effective alginate dosage more efficiently.

Pharmacokinetics of a sustained-release bovine somatotropin in lactating cows

  • Bovine somatotropin is known to improve the growth rate and lactation in cattle. In this study, we examined the concentration-time profiles of a sustained-release formulation of bovine somatotropin (BST) and insulin-like growth factor-1 (IGF-1) in plasma and milk in cows. In addition, the possible effect of co-administrated vitamin ADE complex on the pharmacokinetic parameters of BST and IGF-1 was evaluated. 1. Plasma BST and IGF-1 levels reached the peak at 12~24 and 48 hours after the administration of BST, and plasma half-lives ranged 100 to 137 and 201 to 310 hours, respectively. To 8th day after administration, BST and IGF-1 levels in milk were not significantly different from the control levels. 2. Plasma BST levels showed cyclic pattern with high concentrations in early stage after each injection and following gradual declining during repeated administrations at 2 week intervals, while plasma IGF-1 levels in treated animals did not show such a cyclic pattern, but remained higher than the control levels. 3. Milk BST and IGF-1 levels during repeated treatments were not significantly different from the control levels. 4. Co-administration of vitamin ADE complex yielded slightly increased AUC of plasma BST for high dose group, but such effect was not evident in the IGF-1 levels. Co-administration of ADE complex tended to increase plasma BST levels and decrease the elimination half-life of IGF-1. 5. These results suggest that the BST formulation tested is one of the ideal sustained-release formulation for long term use in dairy industry. As for the co-administration of vitamin ADE complex, the benefit of co-administration with BST is needed to be further evaluated.

RADIO EMISSION FROM WEAK SPHERICAL SHOCKS IN THE OUTSKIRTS OF GALAXY CLUSTERS

  • In Kang (2015) we calculated the acceleration of cosmic-ray electrons at weak spherical shocks that are expected to form in the cluster outskirts, and estimated the diffuse synchrotron radiation emitted by those electrons. There we demonstrated that, at decelerating spherical shocks, the volume integrated spectra of both electrons and radiation deviate significantly from the test-particle power-laws predicted for constant planar shocks, because the shock compression ratio and the flux of inject electrons decrease in time. In this study, we consider spherical blast waves propagating through a constant density core surrounded by an isothermal halo with ρ ∝ r−n in order to explore how the deceleration of the shock affects the radio emission from accelerated electrons. The surface brightness profile and the volumeintegrated radio spectrum of the model shocks are calculated by assuming a ribbon-like shock surface on a spherical shell and the associated downstream region of relativistic electrons. If the postshock magnetic field strength is about 0.7 or 7 µG, at the shock age of ∼ 50 Myr, the volume-integrated radio spectrum steepens gradually with the spectral index from αinj to αinj + 0.5 over 0.1–10 GHz, where αinj is the injection index at the shock position expected from the diffusive shock acceleration theory. Such gradual steepening could explain the curved radio spectrum of the radio relic in cluster A2266, which was interpreted as a broken power-law by Trasatti et al. (2015), if the relic shock is young enough so that the break frequency is around 1 GHz.

일차성 간림프종: 자기공명영상과 병리소견의 연관 (Primary Hepatic Lymphoma: MR Imaging and Pathologic Correlation)

  • 일차성 간림프종은 전체 림프절외장기를 침범하는 림프종의 1% 미만에서 나타나는 매우 드문 질환이다. 저자들은 수술로 확진된 일차성 간림프종의 자기공명영상과 병리 소견의 특징을 보고한다. 간세포 특이 조영제를 주입하여 얻은 역동적 조영증강 영상에서 점차적으로 조영증강되어 간내담관암과 유사하게 보였다. 그러나 20분 지연 후 얻은 간담관기 영상과 확산강조영상에서 서로 다른 신호강도를 보이는 세 층의 띠모양의 병변이 특징적으로 보였으며 이는 병리적으로 외층의 생존 종양, 중간층의 괴사된 종양, 및 종양에 둘러싸인 중앙부의 괴사된 간 실질과 일치되는 소견을 보였다.

Triptolide improves myocardial fibrosis in rats through inhibition of nuclear factor kappa B and NLR family pyrin domain containing 3 inflammasome pathway

  • Myocardial fibrosis (MF) is the result of persistent and repeated aggravation of myocardial ischemia and hypoxia, leading to the gradual development of heart failure of chronic ischemic heart disease. Triptolide (TPL) is identified to be involved in the treatment for MF. This study aims to explore the mechanism of TPL in the treatment of MF. The MF rat model was established, subcutaneously injected with isoproterenol and treated by subcutaneous injection of TPL. The cardiac function of each group was evaluated, including LVEF, LVFS, LVES, and LVED. The expressions of ANP, BNP, inflammatory related factors (IL-1β, IL-18, TNF-α, MCP-1, VCAM1), NLRP3 inflammasome factors (NLRP3, ASC) and fibrosis related factors (TGF-β1, COL1, and COL3) in rats were dete cted. H&E staining and Masson staining were used to observe myocardial cell inflammation and fibrosis of rats. Western blot was used to detect the p-P65 and t-P65 levels in nucleoprotein of rat myocardial tissues. LVED and LVES of MF group were significantly upregulated, LVEF and LVFS were significantly downregulated, while TPL treatment reversed these trends; TPL treatment downregulated the tissue injury and improved the pathological damage of MF rats. TPL treatment downregulated the levels of inflammatory factors and fibrosis factors, and inhibited the activation of NLRP3 inflammasome. Activation of NLRP3 inflammasome or NF-κB pathway reversed the effect of TPL on MF. Collectively, TPL inhibited the activation of NLRP3 inflammasome by inhibiting NF-κB pathway, and improved MF in MF rats.

정맥영양의 조제와 감염관리 (Infection Control in Parenteral Nutrition Preparation and Compounding)

  • Recently, in Korea, the importance of preparation and use of injectable drugs has been emphasized due to successive fatal accidents caused by injection infections. Parenteral nutrition (PN) has also been identified as a cause of infection. Cases of infection due to PN have been reported not only in Korea, but also abroad, and contamination occurs mainly during the preparation of PN. Because sterile preparation and compounding of injections are very important for infection control and patient safety, this article reviews the major guidelines outlined thus far. The Korea Ministry of Food and Drug Safety in 2006 published guidelines and the KSHP (Korean Society of Health-System Pharmacists) recently issued guidelines for the aseptic preparation of injections. In addition, as US guidelines, the ASHP (American Society of Health-System Pharmacists) guidelines and United States Pharmacopeia (USP) <797> are also reviewed. The recent guidelines published by the KSHP have significance in that they were adopted in accordance with the domestic reality, even though they conform to foreign guidelines, and are expected to be guidelines for hospital pharmacists performing aseptic preparation work. In addition, the Korea Ministry of Health and Welfare is considering appropriate guidelines for the safe management of medications, training staff for infection prevention and strengthening staff capacity. Furthermore, the gradual expansion of aseptic compounding facilities and human resources, as well as the provision of adequate medical costs are also considered. Based on the establishment and standardization of injectable drugs compounding guidelines for Korean hospitals, it is believed that if human resources and facilities are supported and medical charges are improved, it will be possible to expect the safer preparation and use of injections.
