Effect of 11-ketotestosterone (11-KT) on Gonadal Sex Reversal and Spermatogenesis of Honeycomb Grouper Epinephelus merra

  • We investigated the androgenic effects of 11-ketotestosterone (11-KT) on gonadal sex reversal and spermatogenesis in honeycomb grouper Epinephelus merra by method of gonadal biopsy. 11-KT was injected intramuscularly at a concentration of 1 and $10{\mu}g$ body weight. The proportion of cross sectional area of the gonad occupied by each germ cell type was measured and compared pre- and post-injection group. During the sex change phase, the distribution ratio of oocytes was decreased in all fish of 11-KT treatment group while the distribution ratio of spermatocytes was increased than pre-injection group. In male phase, all fish of 11-KT treatment group shown the increased distribution ratio of spermatocytes, but the distribution ratio of spermatozoa was decreased than pre-injection group. The present results suggest that 11-KT can stimulate degeneration of oocytes, proliferation of spermatocytes and spermiation in honeycomb grouper.

참남여중간몸증으로 오인된 혼합생식샘발달장애 (Mixed Gonadal Dysgenesis Mimicking True Hermaphroditism)

  • A differential diagnosis between the true hermaphroditism (TH) and mixed gonadal dysgenesis (MGD) has important clinical implications for gender assignment and the decision for early gonadectomy; however, variable clinical and histological features frequently lead to the confusion of TH with MGD. A 17-month-old boy was presented with proximal hypospadias with chordee and right non-palpable testis in his scrotum. He also had right auricular anomaly including a separated tragus with skin tag. Left testis was well palpable in his left scrotum. Diagnostic right inguinal exploration showed Mullerian structures such as a gonad like an ovary and a fallopian tube with a uterus, which were removed. Repair of hypospadias and right auricular anomaly was also done. Following ultrasonography (USG) showed a normal looking testis in left scrotum. His chromosome was 45, XO/46, XY. We report a difficult case of mixed gonadal dysgenesis mimicking true hermaphroditism which combines ipsilateral congenital auricular anomaly.

담수순화 감성돔 Acanthopagrus schlegeli의 생식소발달, 성 스테로이드호르몬 및 채정시기 (Sperm Collection Time, Sex Steroid Hormones, and Gonadal Development of Black Porgy, Acanthopagrus schlegeli, Acclimated in Freshwater)

  • The sperm collection time, sex steroid hormones, and gonadal development of protandrous black porgy, Acanthopagrus schlegeli, acclimated in freshwater for more than 2 years were investigated to evaluate its reproductive capability. The gonadal development of black porgy reared in seawater and freshwater could each be classified into four successive stages. For black porgy reared in seawater (BSW) as the control, these were the growing (December to February), mature (February to March), spent (March to June), and degeneration and resting (July to December) stages; for black porgy reared in freshwater (BFW), these were the growing (November to January), mature (January to February), spent (February to May), degeneration and resting (June to November) stages. In both BSW and BFW, the plasma cortisol levels were the highest in March. The plasma testosterone (T) levels of BSW and BFW were the highest in March and February, respectively. The plasma estradiol-$17{\beta}$ ($E_2$) levels did not differ significantly between BSW and BFW. The 11-ketotestosterone (11-KT) levels in the plasma of BSW and BFW were the highest in April. Sperm was collectible from March to June in BSW and from February to May in BFW. The results indicated that the gonadal maturation of BFW was about 1 month faster than that of BSW.

원발성 무월경 환자에 이어서 세포유전학적 연구 (Cytogenetic Studies in 236 Patients with Primary Amenorrhea)

  • The high rate of chromosomal abnormalities in patients with primary amenorrhea implies the need for routine screening for chromosomal abnormalities among such patients. This study was designed for the cytogenetic analysis of 236 patients with primary amenorrhea, which was referred to Yonsei University Medical Center, from January, 1, 1974 to December, 31, 1985. The results were as follows: 1. Of the 236 patients, 145 cases (61.4%) showed normal karyotype, and 91 cases (38.6%) showed chromosomal abnormalities. 2. Gonadal dysgenesis was found in 56 cases, consisting of 42 cases, Turner's syndrome, 12 cases, pure gonadal dysgenesis, and 2 cases mixed gonadal dysgenesis. a) Turner's syndrome was found in 42 cases, consisting of 18 cases of 45, X and 24 cases of mosaicism. b) Pure gonadal dysgenesis was found in 12 cases, consisting of 10 cases of 46, XX and 2 cases of 46, XY. c) Mixed gonadal dysgenesis was found in 2 cases, consisting of 1 case of 46, XY and 1 case of 45, X/46, XY. 3. Intersex was found in 80 cases, consisting of 35 cases of 46, XX, and 45 cases of 46, XY. 4. Congenital anomalies of reproductive system was found in 82 cases and all cases were normal karyotype.

  • 연구는 양질의 피조개 수정란 확보와 안정적 종묘생산을 위하여 피조개 모패의 먹이생물로 미세조류 3종 (Isochrysis galbana, Tetraselmis tetrathele, Phaeodactylum tricornutum) 을 단독 또는 혼합 공급하여 먹이생물에 따른 피조개 모패의 비만도, 생식소발달, 성숙유도율 및 생존율을 조사하였다. 사육 45일 경과 후 피조개 모패의 완숙기 출현율, 성숙유도율 및 생존율은 3종의 미세조류를 혼합 공급하거나 T. tetrathele를 단독공급한 실험구에서 가장 높았다.

자연산 바지락, Ruditapes philippinarum의 생식주기와 수온-먹이섭이 및 절식조건에 의한 생식소발달의 인위적 제어 (Reproductive Cycle of Natural Population and Artificial Control of Gonadal Development of Ruditapes philippinarum by the Conditions of Water Temperature-Feeding and Starvation)

  • 자연산 바지락 (Ruditapes philippinarum) 개체군의 생식 주기와 수온-먹이섭이 및 절식조건에 따른 생식소 발달의 인위적 제어 결과를 조직학적 관찰에 의해 조사하였다. 곰소만 자연산 바지락 개체군의 생식소 발달에 따른 암수 개체의 생식주기는 초기활성기 (4-5월), 완숙기 (4-8월), 부분산란기 (5-10월), 퇴화 및 비활성기 (8-3월)의 연속적인 5단계로 구분할 수 있었다. 수온에 따른 생식소 발달의 인위적 제어 실험에서, 바지락은 저수온 (10$^{\circ}C$) 에 의해 억제되었다. 19$^{\circ}C$ 및 22$^{\circ}C$의 고수온에 노출시킨 실험구에서 생식소 발달은 고수온에 의해 촉진되었는데, 자연산 개체군의 것보다 약 1달 빠르게 나타났다. 고수온-먹이섭이 실험구에서, 작은개체 실험구의 생식소 발달 단계가 큰개체구 실험보다 더 빠르게 나타났으며, 저수온 (10$^{\circ}C$)의 것보다 더 빠르게 나타났다 (p=0.01). 암, 수 모든 개체들에서 고수온 (22-28$^{\circ}C$)-절식구 실험구의 생식소 발달관계가 고수온-섭이 실험구의 것들보다 더 빠른 것으로 나타났다. (paired sample t-test, p=0.004). 42일 경과 후 암컷과 수컷의 먹이섭이-고수온 실험구에서, 고수온-작은개체 실험구 조건이 큰개체의 생식소 발달보다 더 민감하게 나타났다. 그러나 저수온-수컷개체 실험구는 암컷 개체들의 실험구보다 반응이 좀 더 민감하게 나타났다. 전반적으로 고수온 실험구의 성적성숙이 저수온구보다 빠르게 나타났으며, 암수 개체 간에 유의한 차이를 보였다 (paired sample t-test, p=0.001). 42일 경과 후 절식 실험구에서, 고수온-수컷 큰개체 실험구의 생식소 발달이 고수온-암컷 큰개체 실험구의 것들보다 빠르게 나타났다. 그러나 작은 개체들의 생식소 발달 단계는 먹이섭이 실험구와 절식 실험구사이에 동일한 양상을 보였다. 산란 성기중 암수 개체들의 경우는 고수온-절식 실험구의 생식소 발달이 암컷과 수컷의 성 및 개체의 크기에 관계없이 고수온-먹이섭이 실험구의 것들보다 더 빠르게 나타났으며, 암수간 유의한 차를 보였다(paired sample t-test, p=0.004).

  • Collection of large number of gonadal Primordial germ cells(gPGCs) is a prerequisite factor for improving germline transmission efficacy in the aves, In this study a magnetic-activated cell sorter(MACS) was applied for improving retrieval efficacy of quail gPGCs and the migration capacity of MACS-separated gPGCs was further examined after being transplanted to recipient embryos. We also induced germline chimeras by transfer of MACS-separated quail gPGCs at the efficiency of 17.4% on average.

붕어의 발육 및 생식에 미치는 비소의 영향 (Effects of Arsenic on the Gonadal Development in Crucian Carp)

  • To evaluate the arsenic effect of fish endocrine disruption, crucian carp (Carassius auratus) were treated with 0.5 mg/L and 3.0 mg/L of As$_2$O$_3$ for 14 days and gonadal development was examined by histological analysis. In ovaries from female crucial carps exposed to 3.0 mg/L, immature follicles and atretic follicles were appeared while various normal developmental stages of oocytes were shown in control group. Body weight and Gonadosomatic Index (GSI) of treated carp were also decreased compared to control group. However, no other significant histological changes in liver or kidney were shown in this exposure scheme. This means that reproductive organs are more sensitive than other organs and arsenic may exert endocrine disrupting effect through inhibiting the development of reproductive organ in fish.

  • Most vascular disorders tend to affect both the brain and heart, and among them, a clinical syndrome constituting cerebral aneurysm and aortic coarctation(AC) has been well recognized. Persistent hypertensive impact to the cerebral vasculature with developmental anomaly of the neural crest, precursor of ectomenchymal, would be closely associated with development of the cerebral aneurysm in AC. Gonadal steroid hormone, a guardian of the cardiovascular system, has been known for its protective effects on the vascular wall. Gonadal steroid hormone (androgen) insensitivity such as 46,XY female syndrome may increase the risk of hypertention and subsequent vascular anomalies. The authors report on a 46-year-old 46,XY female patient with AC who underwent surgical clipping of the ruptured cerebral aneurysm. Clinical implications and proposed pathogenetic mechanisms of aneurysm in this intersex syndrome are presented and discussed.

자귀나무 추출물이 송사리(Oryzias latipes) 생식소 성숙에 대한 억제효과 (Inhibitory Effects of Extracts from Albizzia julibrissin on Gonadal Maturation in a Medaka (Oryzias latipes))

  • 사료에 첨가된 saponin이 함유된 자귀나무(Albizzia julibrissin) 껍질 추출물이 송사리(Oryzias latipes)의 생식소 성숙과 산란에 미치는 영향을 조사하기 위하여 자귀나무(A. julibrissin) 껍질의 n-BuOH 추출물로부터 Diaion HP-20, Silica gel과 Sephadex LH-20 chromatography들을 이용하여 조 사포닌 분획물(HaBC)을 분리하였다. 실험 어류들은 순환여과 장치 시스템의 수조에서 사육하였으며, HaBC를 첨가한 사료를 급여하여 암컷의 생식소 성숙 억제 및 산란 억제 효과를 조사하였다. 미성숙 송사리들에 대한 실험에서 사료에 HaBC를 20 mg/g-feed 이상 첨가한 사료를 먹인 어류들은 생식소의 성숙과 산란을 개시하는 시기가 지연되었다. 또한 성숙한 암컷 송사리들도 HaBC를 20 mg/g-feed 이상 첨가한 사료를 먹었을 때 대조구에 비해 낮은 GSI 값을 보였다. 미성숙한 송사리에 대한 길이 성장이나 체중에 대한 변화는 HaBC의 첨가량에 관계가 없었으나, 비만도(CF)에서는 HaBC를 첨가한 사료를 먹인 어류들이 대조구에 비해 높은 값을 보였다. 이러한 결과로 자귀나무(A. julibrissin)의 껍질로부터 분리한 saponin 분획물은 송사리 암컷의 성숙을 억제할 수 있었지만, 성장 촉진에는 아무런 작용을 하지 않았다. 이들 작용 기전에 대해서는 더 많은 연구들이 있어야 할 것이다.
