Reproductive Cycle of Seabass, Lateolabrax japonicus (농어, Lateolabrax japonicus의 생식주기)

  • Annual reproductive cycle of seabass, Lateolabrax japonicus, was histologically investigated based on samples captured on the coast of the Tongyoung, South Korea. The gonadosomatic index (GSI) of females began to increase in October and reached its maximum in February. The GSI of males reached its maximum in December. The change of GSI and gonadal tissue showed that the annual reproductive cycle was classified into the following successive four stages: (1) the degenerative and resting stage from March to August, (2) the growth stage from September to November, (3) the mature stage from November to December, and (4) the ripe and spawning stage from December to March.

  • This study aims to find out the effect of heavy metals, as is the case of EDCs (endocrine disrupting chemicals), on reproductive endocrine disruption of aquatic animals. In the present experiment zinc, which is a heavy metal well known for its androgenous activity, was used. The experimental period was 24 weeks, starting in November during the inactive stage of the clam's reproductive cycle. The experimental groups were composed of one control condition and three zinc exposure conditions (0.64, 1.07, and 1.79 mg/L). The sex ratio (F:M) was 1:1.06 in the control group and 1:1.70 in all the exposed group, illustrating the tendency for higher proportion of males with increases in zinc concentration. Gonad maturity was higher in 1.07, and 1.79 mg/L groups compared to the control group, with higher maturity observed in males than females. Intersex individuals made up 24.7% of the exposed group, while females exhibited a higher ratio than the males with increasing zinc concentration. The results of this study indicate that zinc functions as an androgenic effector on the reproduction of Gomphina veneriformis.

Intersexuality of Scomberomorus niphonius from the Coastal Area around Jeju Island, Korea (Teleostei: Scombridae)

  • This study histologically describes the intersexuality of Scomberomorus niphonius collected from the coastal area near Jeju Island. A total of 126 S. niphonius, collected from March to July 2012 with a total length of 62.4 cm (${\pm}17.5$) and a total weight of 1,701.9 g (${\pm}1,528.9$) were used for analyses. From a histological perspective, two types of intersex were confirmed. One type had scattered germ cells from the opposite sex within the gonad. The second type developed germ cells from the opposite sex in the connective tissue of the outer gonadal membrane. The intersexuality was 14.3% (n=18/126), with females (21.3%; n=16/75) exhibiting a higher rate than males (3.9%; n=2/51). There was no displayed correlation between intersexuality and the total length and weight.

  • Intracranial germ cell tumors (iGCTs) are a heterogeneous group of tumors with peculiar characteristics clearly distinguished from other brain tumors of neuroepithelial origin. Diverse histology, similarity to gonadal GCT, predilection to one sex, and geographic difference in incidence all present enigmas and fascinating challenges. The treatment of iGCT has advanced for germinoma to date; thus, clinical attention has shifted from survival to long-term quality of life. However, for non-germinomatous GCT, current protocols provide only modest improvement and more innovative therapies are needed. Recently, next-generation sequencing studies have revealed the genomic landscape of iGCT. Novel mutations in the KIT-RAS-MAPK and AKT-MTOR pathways were identified. More importantly, methylation profiling revealed a new method to assess the pathogenesis of iGCT. Molecular research will unleash new knowledge on the origin of iGCT and solve the many mysteries that have lingered on this peculiar neoplasm for a long time.

Hemorrhage- and Restraint-induced Analgesia in Male and Female Conscious Rats

  • It is well known that stress induces analgesia. This study was designed to demonstrate the stress-induced analgesia by employing hemorrhage and restraint and to investigate its mechanism and sex difference. The degree of pain was assessed by measuring the magnitude of jaw opening reflex produced by a noxious electrical stimulation in the dental pulp and by measuring the latency to withdraw the tail from a heat ray. Restraint showed an antinociceptive response. A significant increase in pain threshold on bleeding was shown and the increase was larger in male group than in female group. The tail flick latency (TFL) on bleeding after AVP antagonist injection into the ventricle was decreased and the decrease was greater in male rats than in female rats. Castration resulted in a significant reduction of TFL. This effect was reversed by treatment with sex hormones. TFL was decreased during hemorrhage in castrated rats. This response was opposite to that in non-castrated rats. TFL was further decreased during hemorrhage after infusion of AVP antagonist, and there was a significant sex difference. These results suggest that both restraint and hemorrhage produce an antinociception and that, in hemorrhage-induced analgesia, AVP and sex hormones may play an important role and male rats show a greater analgesic response.

Maturity and Growth of Marbled Sole, Pseudopleuronectes yokohamae in the West Sea of Korea (한국 서해 문치가자미(Marbled Sole, Pseudopleuronectes yokohamae)의 성숙과 성장)

  • We determined the maturity and growth of the Marbled sole, Pseudopleuronectes yokohamae based on monthly sampling in the West Sea of Korea from February 2009 to December 2010. Determination of sex of P. yokohamae was by gonadal inspection, and age and growth were determined by analyzing the otolith. The biological minimum size of P. yokohamae female was 24.5 cm. The seasonal changes in the ratio of a translucent zone to an opaque zone revealed that the end of the translucent zone of the otolith were annuli formed in May once a year. We compared 8 type growth equations with several length-at-age data. By using the length-at-monthly age data of these, the calculated von Bertalanffy growth equations were $L_t=41.7(1-e^{-0.47(t+0.24)})$ for females and $L_t=32.7(1-e^{-0.75(t+0.01)})$ for males. We estimated that 24.5 cm was the total length of 2 year old P. yokohamae spawns in the cold water season from January to April, and grows to 8 years old for females and 6 years old for males in the West Sea of Korea.

Pituitary and Gonadal Response to GnRH in Prepubertal Buffaloes (Bubalus bubalis)

  • The objective of this study was to investigate the responsiveness of hypophysis and gonads to synthetic GnRH among prepubertal buffalo heifers at 12 months of age. Peripheral plasma FSH, LH, estradiol and progesterone level were measured in blood samples collected at 1 hr before and up to 18 days subsequent to the administration of $200{\mu}g$ GnRH (n=6) or saline (n=6) in Murrah buffalo heifers. The pretreatment peripheral plasma FSH, LH, estradiol and progesterone among GnRH treated heifers were $7.35{\pm}0.45ng/ml$, $1.08{\pm}0.3ng/ml$, $22.93{\pm}1.06pg/ml$ and $0.27{\pm}0.04ng/ml$ respectively. A quick elevation (p < 0.01) of FSH and LH within five min of GnRH administration was observed in all geifers. Although the peak FSH $(89.57{\pm}23.43ng/ml)$ and LH $(7.52{\pm}3.08ng/ml)$ reached by 10 min of GnRH administration, yet the animals differed both in terms of their amplitude response of FSH and LH release as well as in terms of time which animals took to exhiit maximum response to GnRH administration. The GnRH administration did not cause alteration in plasma estradiol and progesterone level. The present study suggests that the pituitary of 12 month buffalo heifers has capacity to synthesize and store of gonadotropin and have developed receptors for GnRH for a spike of gonadotropin release.

Gametogenic Cycle and Fine Structure of Ripe Germ Cells in the Pacific Oyster, Crassostrea gigas on the South Coast of Korea

  • The gonadal development and the gametogenic cycle and the fine structure of ripe germ cells of the cultured Pacific oyster, Crassostrea gigas were investigated using oysters monthly collected from the southern coast of Korea from October 2000 to September 2001. Monthly changes in the condition index were similar to that of meat weight rate and the highest value was observed in between April and May, and the lowest value in August. The external colors of the testis and the ovary were milky white and yellowish, respectively. The spawning period of the Pacific oyster was continued from May to September, with a peak in July. The gametogenic cycle could be classified into five successive stages: multiplicative stage (December to March), growing stage (March and April), mature stage (April to June), spawning stage (June to August) and resting stage (August to January). Variety of egg yolk granules, lipid granules, mitochondria, and endoplasmic reticula were observed in cytoplasm of ripe oocyte. The spermatozoon consisted of the head, middle piece and tail; including cap-shaped acrosome with domed structure, elliptical shaped nucleus, four mitochondria, two centrioles and flagellum.

Maternal caffeine consumption has irreversible effects on reproductive parameters and fertility in male offspring rats

  • Objective: Concerns are growing about the decrease in male reproductive health. Caffeine is one of the popular nutrients that has been implicated as a risk factor for infertility. In the present study, we examined whether in utero and lactational exposure to caffeine affects the reproductive function of the offspring of rats. Methods: Pregnant rats received caffeine via drinking water during gestation (26 and 45 mg/kg) and lactation (25 and 35 mg/kg). Body and reproductive organ weight, seminiferous tubule diameter, germinal epithelium height, sperm parameters, fertility rate, number of implantations, and testosterone level of the offspring were assessed from birth to adulthood. Results: Significant dose-related decreases were observed in the body and reproductive organ weight, seminiferous tubule diameter, and germinal epithelium height of the offspring. Sperm density had declined significantly in offspring of the low-dose and high-dose groups, by 8.81% and 19.97%, respectively, by postnatal day 150. The number of viable fetuses had decreased significantly in females mated with male offspring of the high-dose group at postnatal days 60, 90, 120, and 150. There were also significant reductions in testosterone levels of high-dose group offspring from birth to postnatal day 150. Conclusion: It is concluded that maternal caffeine consumption impairs gonadal development and has long-term adverse effects on the reproductive efficiency of male offspring rats.

Phosphoproteomic Analysis of the Brain of Ovariectomized Adult Rat

  • Aging in females is associated with a reduced metabolic function, increased incidence of neurodegenerative diseases, and cognitive dysfunction, as a result of loss in gonadal function. The change can alter the states of phosphorylation on the proteins, which cause dramatic changes in the cellular location or activity of the proteins. In this study, the differential phosphorylation of the proteins responsible for the functions related to cognition was studied using the ovariectomized adult rats. Phosphoproteomic analysis using the cerebral and hippocampal tissues could identify 51 differentially phosphorylated proteins including 12 proteins for energy metabolism, 8 cytoskeletal proteins, 6 signaling proteins, and other functional proteins in the ovariectomized rats. Further, anti-oxidative enzymes, superoxide dismutase and peroxiredoxin-2, which are known to be inactivated by phosphorylation, were found to be differentially phosphorylated in the cerebellum and hippocampus of the ovariectomized rats, respectively. Many of the deactivated proteins by differential phosphorylation identified in this study were overlapped to those of Alzheimer's disease cases. These results will provide information for neurodegenerative learning and memory impairments in women as brought about by menopause.
