  • Swainsonine is a toxic indolizidine alkaloid found in the plant, Swainsona species (Dorling, 1978; Colgate, 1979). It is a potent inhibitor of the glycoprotein processing pathway in the Golgi apparatus. Specifically, it inhibits mannosicase II resulting in abherrant high mannose glycans. Recent studies showed that swainsonine prevents metastasis of tumor cells and it inhibits solid growth tumor, at least partially (Goss et al., 1994; Baptista et al, 1994).

Electron Microscopic Studies on Effects of Korean Panax Ginseng on Rat Thyroid Follicular Cell (고려 인삼 투여가 흰쥐 갑상선 세포에 미치는 영향에 관한 전자현미경적 연구)

  • Changes in fine structure of thyroid follicular cells were studied in rats given oral administration of Korean Panax Ginseng for 60 days. The rough-surfaced endoplasmic reticulum was distended and formed large cisternae, the Golgi complex was hypertrophic, and enlarged colloid droplets and lysosomes were more numerous in the follicular cells of ginseng treated rats. Morphologic changes observed may represent stimulating effects on the thyroid gland in ginseng treated animals.

  • 細胞學의 發展은 電子顯微鏡의 分解能$(2.4\\mu 以上)$이 增加됨과 아울러 超薄切片法(Sjostrand 1953) 및 固定法의 改良(Palade 1952) 으로 細胞의 超微細構造를 追究하는 反面 物理化學的構成成分과 機能面에서 細胞生物學, 더 나아가 分子生物學으로 發展하였다. 細胞膜(原形質膜)의 分子的 構造는 二重의 脂質分子膜의 表面에 蛋白質分子가 結合하고 있는 單位膜(Robertson 1961, Danielli 1964)으로 되어있다. 이 單位膜構造는 모든 動植物 細胞의 表面膜만이 아니고 核膜, mitochondria, Golgi complex, 色素體等 細胞內 膜系가 同一하게 되어 있고(Robertson 1961), 膜의 무게는 位置(Sjostrand 1963) 및 小器官別(Yamamoto 1963)로 다르다. 또 各細胞는 그 機能에 따라 部分的으로 特殊하게 分化되어 있다.

Ultrastructural Observations of a Human Cutaneous Leishmaniasis (인체 피부에 기생한 Leishmania tropica의 전자현미경적 관찰)

  • A case of cutaneous leishmaniasis developed in a 48 year old Korean male who returned from middle east was studied by light and electron microscopic observations. Light microscopically, the lesion consisted of heavy chronic ill-defined granulomatous inflammation involving entire thickness of the dermis, composed of mainly histiocytic and small mononuclear cell infiltrations without evidence of necrosis or giant cell formation. Giemsa staining revealed numerous intracellular micro-organisms within histiocytes, showing dark stained central dot surrounded by light stained cytoplasm. Electron microscopically, the organisms were observed mostly ovoid in shape and frequently binary mitotic features within the host cells. follicle consisted of double unit membranes and microtubules, which are immediately below these membrnae. A long kinetoplast was noted within a very elongated mitochondrion at the center of the organisms and a flagella rose in front of the kineoplast but ended within the cytoplasm. Large numbers of free ribosomes, occasional Golgi complex and SER were also noted, but RER was seldom found. These ultrastructural features corresponded to promastigote stage of Leishmania tropica. In principle, leishmaniasis is a tropical disease and can not be found in temperate zone. However, travel to mideast by many Koreans may contract this disease while they are in endemic regions.

Ultrastructural Study on the Development of the Synovial Membrane in Human Fetuses (인태아(人胎兒) 활액막세포(滑液膜細胞)의 전자현미경적(電子顯微鏡的) 연구(硏究))

  • The development of synovial membrane from knee joint was studied by electron microscope in human fetuses ranging from 20mm to 260mm crown rump length (40days to 30weeks of gestational age). At 40mm fetus, developing synovial tissue was observed in homogenous interzone as a vascular mesenchyme around the periphery. The primitive joint space was appeared after the intermediate layer of the interzone in direct contact with chondrogenic layer at 60mm fetus. Differentiation of the synovial membrane coincided with clarification of the joint cauity. When dilatation of the synovial cavity occurred, the two types of synovial cells were well endowed with rough endoplasmic reticulum. At 100mm fetus, type A cells with a markedly attenuated cytoplasm were found as well as those cells which contained pinocytotic vesicles and vacuoles. By 150-200mm fetuses a majority of the intimal cells were type B. These cells were characterized by abundant rough endoplasmic reticulum and well developed Golgi complex. In contrast, A-type cell had numerous filopodia, pinocytotic vesicles lysosomes, and large vacuoles containing amorphous material. At 260mm fetus, the intimal cells were well developed and plentiful. The most marked difference between the synovial membrane of full-term fetus and adult was the large amount of collagen in the latter. During fetal period, the B-cells were most numerous cell type in the intimal cells. The B-cells were clearly distinguishable from the A-cells by their content of extensive rough endoplasmic reticulum and well developed Golgi complex.

Comparative Studies on the Ultrastructures of Non-Ciliated and Ciliated Epithelial Cells in the Ductus Epididymidis of Apodemus agrarius coreae (등줄쥐 (Apodemus agrarius coreae)의 부고환관의 무섬모상피세포와 섬모상피세포의 미세구조에 대한 비교 연구)

  • In order to the comparative morphological study of the non-ciliated and ciliated epithelial cells, and to elucidate the process of degeneration of non-ciliated epithelial cell of the ductus epididymidis, Korean striped field mouse, Apodemus agrarius coreae was examined with light and transmission electron microscopes. The morphological characteristics of non-ciliated epithelial cell, the cell types of the caput epididymidis (Cp), corpus epididymidis (Cr) and cauda epididymidis (Cu) were long-columnar, short-columnar and short-cuboudal, respectively. The mitochondria and rough endoplasmic reticulum tended to be broken as they immigrated from Cp to the Cu. The Golgi acted vigorously at the Cp, but the Golgi was inactive in Cr and Cu. The secretory vesicles and lysosomes were increased gradually from Cp to the Cu. The process of degeneration of the non-ciliated epithelial cells observed in the Cp, Cr and Cu epididymidis. The increase of the non-ciliated epithelial cells, and its degeneration were observed more often from Cp to the Cu. The morphological characteristics of the ciliated epithelial cells, the cell types of the Cp, Cr and Cu were long-columnar, short-columnar and short-cuboudal, respecptively like the non-ciliated epithelial cells. The stereocilia was long and slender at the Cp and Cr, while Cu was very short. The pinocytotic vesicles and absorptive vesicles were increased from the Cp to the Cu. Numerous disintergrated products was existed at the Cr including the Cp, but Cu were not observed. A significant amount of lysosomes existed at the Cp and Cr epithelial cells, but they were not observed in Cu epithelial cells.

Transition of Marker Enzymes of Rat Hepatocyte Organelles in Culture (배양중 흰쥐 간세포의 새포소기관 표지효소의 변천)

  • To investigate recovery, growth, and activity of hepatocyte in primary culture after cell separation, the authors followed up the marker enzyme activities of golgi complex, mitochondria and biologic membrane. Thiamine pyrophosphatase, the marker enzyme of golgi complex, activity approached the level of long term culture at 4th day. Succinate dehydrogenase, the marker enzyme of mitochondria, activity decreased with time, then it maintained constant level after 4th day. Alkaline phosphatase, the marker enzyme of biological membrane, activity increased from 3rd day, and after 5th day it showed strong reaction. These data suggested that hepatocytes were stabilized and recovered normal activity 4 day after cell separation, but the main secretory function was speculated to be reduced in culture.

Fine Structural Study of Pollen Wall Development at Late Stage of Microsporogenesis in Panax ginseng (인삼의 화분벽 발달에 관한 미세구조적 연구)

  • The ontogeny of pollen wall in Panax ginseng was studied with transmission and scanning electron microscopy from early tetrad stage until pollen maturity. Initial indication of exine development is undulation of plasma membrane for the preparation of bacular mound. The first recognizable structure of the pollen wall is the cellulosic primexine which is formed outside of the plasma membrane while microspore tetrads are still surrounded by callose wall. As development proceeds, foot-layer and baculum differentiation, callose dissolution and exine formation were progressed. During this process, sporopollenin is deposited into the exine, and then endexine development was followed. The intine, innermost pollen wall layer, is developing form hypertrophic Golgi vesicles. The thickness of exine is very even on all along the pollen wall, but intine thickness of apertural region is thicker than that of nonapertural region. Mature pollen of ginseng is $20{\mu}m$ in size, tricolpate and shows fine reticulate sculpturing.

Fine Structure of the Spermatogenic Cells during the Spermiogenesis of Paradoxornis webbiana (붉은머리 오목눈이 (Paradoxornis webbiana)의 정자변태 과정 중 정자형성세포의 미세구조)

  • The morphological characteristics of spermatogenic cells during the spermiogenesis of Paradoxornis webbiana were studied by transmission electron microscope. Spermiogenesis of P. webbiana was divided into ten phase. The chromatin granules became fibrous granules at the Golgi phase, gradually condensed at the cap phases, condensed as a stick at the acrosomal phase, and finally, a perfect nucleus was formed at the maturation phase. The formation of sperm tail began at the early Golgi phase, and completed at the late maturation phase. In particular, the dense materials existed in the sperm neck, which is wedged between the tip of segmented columns and the first mitochondria of the middle piece. The axone in the neck were surrounded by the dense materials. The axonema in spermatozoon contains a 9+2 arrangement of microtubules: 9 doublets, and 2 central single microtubules. Mitochondrial bundles of middle piece were composed of a pair of arms, which surrounded the axone of the middle piece by the $15^{\circ}$ angled-helical structure. The outer membrane of mitochondria were surrounded by microtubules in plasma membrane of the sperm. The undulating membrane had a helical structure, and the sperm plasma membrane was surrounded by undulating membrane.
