The study was designed to investigate the aspects of the goblet cell of the oviduct in laying Korean ring-necked pheasants by histochemical methods (PAS, alcian blue-PAS, alcian blue pH 2.5, alcian blue pH 1.0, and aldehyde fuchsin-alcian blue stain). Twelve laying Korean ring-necked pheasants were used. The results were summarized as follows. The goblet cell of the oviduct of the Korean ring-necked pheasant contained sulfated acid mucosubstances and neutral mucosubstances in the infundibulum. The compounds of sulfated acid mucosubstances, nonsulfated acid mucosubstances, and neutral mucosubstances are present in goblet cells of the magnum, vagina and openings of the tubular gland of the uterus of the oviduct in the Korean ring-necked pheasant. The goblet cell of the isthmus and uterus of the oviduct of the Korean ring-necked pheasant contained neutral mucosubstances except openings of the tubular gland of the uterus. The histochemical characteristics of secretory granules of goblet cells in oviduct of the Korean ring-necked pheasant were not related to the position of eggs in oviduct.
The morphological changes of the midgut epithelium during the metamorphosis of pine caterpillar are observed with light and electron microscope, being divided into 5 stages from the 8th instar larva to just after pupation. The midgut epithelium of 8th instar larva is composed of columnar cell, goblet cell, regenerative cell, and endocrine cell. The secretorials are arranged on the nuclear membrane in the columnar cell of the midgut epithelium in the 8th instar larva, and lysosomes are augmented in the apical portion. Cytoplasmic extrusions are observed in the apical surface of columnar cell but they have no cell organells. Nucleus, mitochondria, rER, Golgi complex, and free ribsomes are observed in the regenerative cell. Regenerative cells are differentiated into the form of goblet cell, and vacuoles are gradually increased in the cytoplasm. Just pupa stage, the materials, which appears to be mainly composed of Ca, are observed in the circular form and goblet cavity of regenerative cell are detached to lumen. As a result, it reflects the process of the degeneration of the midgut epithelium that lysosomes are gradually augmented in the columnar cell, that nuclear materials are removed to cytoplasm, and that cytoplasmic extrusions are observed in the apical surface. And though regenerative cells are differentiated into the form of the goblet cell, it is believed that goblet cavity is detached from regenerative cell to the lumen and midgut epithelium of pupa stage is formed.
Mast cells and goblet cells have the ability to protect against parasites by increasing mucus production that traps and excludes worms and prevents their intimate contact with the gut mucosa in the host. In this study, we investigated the function of mast cells and goblet cells for the rejection of Echinostoma hortense (E. hortense). In addition, we used both C3H/HeN and BALB/c mice in order to examine whether mast cells and goblet cells function differentially according to the strains of mice. After an oral infection with 30 E. hortense metacercariae, the number of mucosal mast cells and goblet cells, as well as worm recovery rate, were observed in experimentally infected mice between 1 week and 8 weeks post-infection (PI). Worm recovery rates in C3H/HeN and BALB/c mice were 65.7% and 23%, respectively, in week 1 P.I., indicating that worm expulsion in C3H/HeN mice was higher than in BALB/c mice. Our results demonstrate that the period (week 3 P.I.) in which worm recovery falls rapidly is the same period that the number of goblet cells and mast cells reaches a peak. These results indicate that worm recovery significantly correlates with the growth rate of goblet cells and mast cells (P=0.0482). However, worm expulsion is not associated with goblet cells or mast cells in BALB/c mice.

The changing patterns of goblet cell hyperplasia, intestinal epithelial cell turnover, and intestinal motility were studied in ICR and C57BL/6 mice infected with Gymnophalloides seoi (Digenea: Gymnophallidae). Whereas ICR mice retained G. seoi worms until day 7 post-infection (PI), C57BL/6 mice showed a rapid worm expulsion within day 3 PI. Immunosuppression with Depo-Medrol significantly delayed the worm expulsion in C57BL/6 mice. Goblet cell counts were increased in both strains of mice, peaking at day 1 PI in C57BL/6 mice and slowly increasing until day 7 PI in ICR mice. In C57BL/6 mice infected with G. seoi, newly proliferating intestinal epithelial cells were remarkably increased in the crypt, and the increase was the highest at day 1 PI. However, in ICR mice, newly proliferating intestinal epithelial cells increased slowly from day 1 to day 7 PI. Intestinal motility was increased in G. seoi-infected mice, and its chronological pattern was highly correlated with the worm load in both strains of mice. Meanwhile, immunosuppression of C57BL/6 mice abrogated the goblet cell proliferation, reduced the epithelial cell proliferation, and suppressed the intestinal motility. Goblet cell hyperplasia, increased intestinal epithelial cell turnover, and increased intestinal motility should be important mucosal defense mechanisms in G. seoi-infected C57BL/6 mice.

Echinostoma hortense (E. hortense) is an intestinal trematode with the highest infection rate in South Korea. However, the immune response against E. hortense infection has not been explained well. In the present study, we investigated the effect of treatment with cyclosporin A (CsA) and histamine receptor antagonists on the cytokine expression and mucosal goblet cells in E. hortense-infected C3H/HeN mice. The alteration of cytokine mRNA expression ($TNF-{\alpha},\;IL-l{\beta},\;IL-4\;and\;IL-5$), intestinal worm recovery rate and goblet cell responses were measured weekly from 0 to 5 weeks post-infection (P.I.) in the control and the following three drug-treated groups: CsA, hydroxyzine and cimetidine. Compared with the control group, the expression of $TNF-{\alpha}$, IL-4 and IL-5 mRNAs decreased in the CsA- and hydroxyzine-treated groups, but only IL-4 mRNA expression did in the cimetidine-treated group. Worm recovery rate was significantly increased in the drug-treated groups. Mucosal goblet cells and their mucin response significantly decreased in the CsA-treated group (P<0.01), but significantly increased in the cimetidine- (P<0.05) and hydroxyzine- (P<0.01) treated groups. These data suggest that CsA treatment inhibits production of Th1- and Th2-type cytokines which are necessary for the worm expulsion. Histamine receptor increases goblet cells and their mucin activation, although it remains to be elucidated whether it directly affects the worm expulsion period of E. hortense in C3H/HeN mice.
In intestinal helminth infections, Th2 immune respones are generally associated with mucin secretion for worm expulsion from the host intestine. In particular, IL-4 and IL-13 are the important cytokines related with intestinal mucus production via STAT6 signalling in nematode infections. However, this perspective has never been studied in Gymnophalloides seoi infection. The present study aimed to observe the STAT6 signalling and cytokine responses in C57BL/6 mice, a mouse strain resistant to infection with this trematode. The results showed that worm expulsion occurred actively during days 1-2 post-infection (PI), when goblet cells began to proliferate in the small intestine. The STAT6 gene expression in the mouse spleen became remarkable from day 2 PI. Moreover, G. seoi infection induced a significant increase of IL-13 from day 4 PI in the spleen of infected mice. Our results suggested that goblet cell hyperplasia and worm expulsion in G. seoi-infected mice should be induced by STAT6 signalling, in which IL-13 may be involved as a dominant triggering cytokine.
Previous studies showed that Math1 homologous to human Hath1 can cause mouse goblet cells to differentiate. In this context it is important that the majority of colon cancers have few goblet cells. In the present study, the potential role of Hath1 in colon carcinogenesis was investigated. Sections of paraffin-embedded tissues were used to investigate the goblet cell population of normal colon mucosa, mucosa adjacent colon cancer and colon cancer samples from 48 patients. Hath1 and Muc2 expression in these samples were tested by immunohistochemistry, quantitative real-time reverse transcription -PCR and Western blotting. After the recombinant plasmid, pcDNA3.1(+)-Hath1 had been transfected into HT29 colon cancer cells, three clones were selected randomly to test the levels of Hath1 mRNA, Muc2 mRNA, Hath1, Muc2, cyclin D1 and p27 by quantitative real-time reverse transcription-PCR and Western blotting. Moreover, the proliferative ability of HT29 cells introduced with Hath1 was assessed by means of colony formation assay and xenografting. Expression of Hath1, Muc2, cyclin D1 and p27 in the xenograft tumors was also detected by Western blotting. No goblet cells were to be found in colon cancer and levels of Hath1 mRNA and Hath1, Muc2 mRNA and Muc2 were significantly down-regulated. Hath1 could decrease cyclin D1, increase p27 and Muc2 in HT29 cells and inhibit their proliferation. Hath1 may be an anti-oncogene in colon carcinogenesis.

Morphology, histology, and histochemical characteristics of the digestive tract of the guirine goby (Rhinogobius giurinus: Gobiidae), collected from the coast of Jeju Island, Korea, were investigated. The digestive tract of R. giurinus, which is a brackish water species, has a short (relative length of gut=0.42), simple, and narrow gut. The gastric glands are well developed in the stomach, but pyloric caeca are absent. The mucosal folds are regularly branched, and the muscularis extern a is thickest in the esophagus, which also contained the most mucus-secreting goblet cells (P<0.05). In R. giurinus, digestive function occurs in the anterior portion of the digestive tract, where mucus protects the tract from the actions of digestive enzymes and functions to activate digestion.
Objectives: This study was undertaken to investigate the effects of Yijin-tang and GamiYijin-tang on the gastrointestinal functions of rats Methods: Sprague-Dawley rats were used as experimental animals, and were administered Yijin-tang (Sample I group, 47.5 mg/ml) and GamiYijin-tang(Sample II group, 38.37 mg/ml, Sample ill group, 85.3 mg/ml) water extract once a day. Changes of gastric juice volume and intestinal mobility index were measured. The effects on colitis induced by dextran sulfate sodium in the rats were also observed. Results: 1. Gastric juice volume was decreased significantly in the sample I group (P<0.05) compared to the control group; there was not significant effect in the sample II and sample III groups. 2. The moving distance of carbon bolus was increased significantly in the sample n (p<0.05) and sample II (p<0.05) groups compared to the control group; there was not significant effect in the sample I group. 3. The intestinal mobility index was increased significantly only in the sample II group (P<0.05) compared to the control group. 4. The feces consistency was increased significantly on the 3rd and 5th day of the sample I group (P<0.05), on 3rd, 4th, and 5th day of the sample II (p<0.05) and the sample III (p<0.05) groups compared to the control group. 5. The feces property index was increased significantly only on the 5th day of the sample III group (p<0.05) compared to the control group. 6. The number of WBC and RBC, levels of hemoglobin and hematocrit were not changed in all sample groups compared to the control group. 7. The number of the type B Goblet cells were increased significantly in the sample II (p<0.05) and the sample III (P<0.05) groups, but the number of the type C Goblet cells were decreased significantly only in the sample ill group (P<0.05) compared to the control group. Conclusions: According to the above results, GamiYijin-tang compared to the Yijin-tang were decreased hight significantly in gastrointestinal mucose and histological antidiarrheal function with protection of the goblet cell more excellently were observed.
Mucosal immune responses against Pygidiopsis summa (Trematoda: Heterophyidae) infection were studied in ICR mice. Experimental groups consisted of group 1 (uninfected controls), group 2 (infection with 200 metacercariae), and group 3 (immunosuppression with Depo-Medrol and infection with 200 metacercariae). Worms were recovered in the small intestine at days 1, 3, 5, and 7 post-infection (PI). Intestinal intraepithelial lymphocytes (IEL), mast cells, and goblet cells were counted in intestinal tissue sections stained with Giemsa, astra-blue, and periodic acid-Schiff, respectively. Mucosal IgA levels were measured by ELISA. Expulsion of P. summa from the mouse intestine began to occur from days 3-5 PI which sustained until day 7 PI. The worm expulsion was positively correlated with proliferation of IEL, mast cells, goblet cells, and increase of IgA, although in the case of mast cells significant increase was seen only at day 7 PI. Immunosuppression suppressed all these immune effectors and inhibited worm reduction in the intestine until day 7 PI. The results suggested that various immune effectors which include IEL, goblet cells, mast cells, and IgA play roles in regulating the intestinal mucosal immunity of ICR mice against P. summa infection.
