Effects of Glycyrrhizae Radix on Acetaminophen-induced Hepatotoxicity in Mice

  • In order to study if Glycyrrhizae Radix (GR) has protective effects on hepatotoxicity of acetaminophen in mouse, one of the species which are sensitive to acetaminophen-induced hepatotoxicity, effects of GR on liver weight to body weight ratio, serum alanine and aspartate transaminase (ALT and AST) activities, hepatic UDP-GT2 activity, and histopathologic changes were determined in acetaminophen-treated mice. Liver weight to body weight ratio and UDP-GT2 activity in mouse liver were not altered by GR. However, GR pretreatment lowered serum ALT and AST activities by 77% and 90% respectively, and diminished the degree of centrilobular necrosis caused by acetaminophen in liver as determined by histopathologic observation. These results suggest a possible protective effect of GR against the acetaminophen-induced hepatotoxicity in mice.

한약수치에 관한 연구(제5보) - 수치에 의한 감초의 성분변화 및 생리활성 - (Studies on the Processing of Crude Drugs (V) - On the Constituents and Biological Activities of Glycyrrhizae Raidx by Processing -)

  • In order to characterize the chemical change, of the constituents and pharmacological transformation of Glycyrrhizae Radix(GR) which may occur during its processing, processed GR has been investigated in comparison with unprocessed GR. It was found that glycyrrhizin(GL) and other several constituents contained in GR were remarkably changed by processing, and the decrease of contents of GL was identified by making use of thin layer chromatography-chromatoscanner. And also, it was found that glycyrrhetic acid (GA) was identified from processed GR. We recognized that commonly processed GR showed less hemolytic effect than unprocessed GR and processed GR at $190{\circ}c$. It was also found that commonly processed GR showed more effective analgesic effect and prolonged the duration of hypnosis induced by pentobarbital-Na in mice as compared with corresponding unprocessed GR and processed GR at $190{\circ}c$. And then, commonly processed GR significantly suppressed the increases of s-GOT, s-GPT and s-LDH activities in injured mice induced by $CCl_4$.

감초약침(藥鍼)이 PCA반응과 비만세포의 ${\beta}$-Hexosaminidase 및 Cytokine 분비에 미치는 영향 (Effect of Glycyrrhizae Radix Pharmacopuncture on PCA and Secretion of ${\beta}$-Hexosaminidase and Cytokines in RBL-2H3 Cells)

  • Objectives : The purpose of this study is to investigate anti-allergic effects of Glycyrrhizae Radix(GR) pharmacopuncture and GR extract. Methods : In vivo, animals were gotten GR pharmacopunctures at both sides of ST36s three times for 5 days. Then, we investigated anti-DNP IgE-induced passive cutaneous anaphylaxis of Sprague Dawley rats. In vitro, we measured cell viability, ${\beta}$-hexosaminidase release, and the secretion of interleukin-4(IL-4) and tumor necrosis factor-alpha(TNF-${\alpha}$) in RBL-2H3 cells after treatment of various concentrations of GR extract. Results : In vivo, we observed inhibition of passive cutaneous anaphylaxis after GR pharmacopuncture treatments at both sides of ST36s and optional points. In vitro, GR extract treatments did not affect cell viability, but inhibited ${\beta}$-hexosaminidase release and the secretion of IL-4 and TNF-${\alpha}$. Conclusions : These results suggest that GR pharmacopuncture and GR extract should be beneficial in the inhibition of allergic inflammatory response.

Neural Mechanisms Underlying Antidepressant-Like Effects of Glycyrrhizae Radix in Rats

  • Glycyrrhizae radix (GR) is an herbal medicine commonly used in East Asia for treating a variety of diseases, including stomach disorders. In this study, the antidepressant-like activity of GR was investigated using the forced swimming test (FST) in rats. After the FST, the expression of c-Fos and corticotrophin releasing factor (CRF) was assessed by immunochemistry of brain samples from the paraventricular nucleus of the hypothalamus (PVN). The results of the FST showed that a high dose (400 mg/kg) of extract was very effective in reducing immobility(P<0.01), and increased climbing. In addition, treatment with GR (400 mg/kg) significantly decreased the expression of c-Fos and CRF in the PVN, compared to controls. In conclusion, the findings of this study demonstrated that GR effectively reduced behavioral and physiological depression responses in an animal model of depression, suggesting that GR might be useful in the treatment of clinical depression.

감초를 이용한 눈꽃동충하초 균사체 배양 추출물의 항균 활성 및 항염증 활성 분석 (Antimicrobial and Anti-inflammatory Activities of Extracts from Glycyrrhizae radix cultured with Paecilomyces japonica)

  • This study was carried out to investigate the antimicrobial and anti-inflammatory activities of ethanol extract from Glycyrrhizae radix (GR) and ethanol extract from Glycyrrhizae radix cultured with Paecilomyces japonica mycelium (GRPM). Antimicrobial activity was measured by paper disc diffusion assay and minimum inhibitory concentration (MIC). Anti-inflammatory activity was evaluated by measurement of NO production in LPS-stimulated RAW264.7 cells. For the results of the paper disc diffusion assay, GRPM showed high antimicrobial activities against Bacillus cereus, Listeria monocytogenes, and Staphylococcus aureus. In addition, the MIC of GRPM (100 ppm) was lower than that of GR (200 ppm) against L. monocytogenes. When the morphology of L. monocytogenes treated with GRPM was observed using a FE-SEM, the surface of cells treated with GRPM were damaged, and some parts of the cell wall were destroyed. The inhibitory effect on NO production in LPS-stimulated RAW264.7 cells was significantly increased by GRPM treatment. In conclusion, GRPM is superior to GR in terms of antimicrobial and anti-inflammatory activities.

LPS로 유발된 우울증에 대한 감초(甘草)의 효과 (Effect of Glycyrrhizae Radix Water Extract on the depression induced by LPS in Rats)

  • Objectives : Investigation of the antidepressant effect of Glycyrrhizae Radix (GR) through the anti-inflammatory effect. Methods : Depression in rats was induced by LPS (i,p.3days). The rats were treated with GR100 mg/kg (GR 100) or GR400 mg/kg (GR 400). The depressive immobility was examined with Tail Suspension Test(TST) and Forced Swimming Test(FST). The expression of nuclear factor-${\kappa}B$(NF-${\kappa}B)$, $I{\kappa}B$ was measured with western blotting. The concentration of corticosterone, cytokine in plasma was measured with ELISA. The expression of c-Fos in the paraventricular nucleus(PVN) and tyrosine hydroxylase(TH) in the locus coeluleus(LC) were measured with immunostaining method. Results : In the TST, GR400 group significantly decreased immobility time compared with the LPS group. In the FST, GR100, GR400 group significantly decreased immobility time comparing with the LPS group. c-Fos expression in GR100 and GR400 group was decreased comparing with the lipoplysaccharide(LPS) group. The $I{\kappa}B$ expression of GR100 and GR400 group was increased comparing with the LPS group. The level of corticosterone of GR100 group was decreased comparing with the LPS group. The concentration of cytokine of GR100 and GR400 group was decreased comparing with the LPS group. TH expression in the LC was increased in LPS group, but in GR100 and GR400 group was not shown significant decrease. Conclusion : According to this results obtained, GR has antidepressant effects by the anti inflammatory action through the suppression of HPA axis activity, not through the action against the catecholaminergic system.

Effect of Glycyrrhizae Radix on the Glucuronidation in Rat Liver

  • Pretreatment of Glycyrrhizae Radix(GR) to male Sprague-Dawley rats was demonstrated to increase excretion of acetaminophen-glucuronide ocnjugate when bile nad urine were assayed after administration of acetaminophen. In order to study the effect of GR on the glucuronidation in rats, we examined enzymatic activities of hepatic UDP-glucuronosyl-transferases (UDP-GT1 and UDP-GT2) and intracellular concentrations of hepatic UDP-glucuronic acid (UDP-GA), upon the administration of GR (1 g/kg body weight, p.o.) or glycyrrhizin (23 mg/kg body weight, p.o.) a major component of GR, for 6 days. GR and glycyrrhizin caused increases in specific activities of UDP-GT2 111% and 96% respectively. Specific activity of UDP-GT1 was increased 25% by GR treatment whereas it was not significantly increased by glycyrrhizin. Concentrations of UDP-GA were increased 257% by GR and 484% by glycyrrhizin. These data indicate that GR activated glucuronidation and thus suggest the possibility that GR may influence detoxification of xenobiotics in rat liver.

한약수치에 관한 연구(제4보) -수치에 의한 감초중 Glycyrrhizin의 물리화학적 변화- (Studies on the Processing of Crude Drugs (IV) -Physico-chemical Transformation of Glycyrrhizin in Glycyrrhizae Radix by Processing-)

  • In order to investigate the chemical and pharmacological characterization of crude drug processing, the triterpenoidal constituents of Glycyrrhizae Radix(GR) were examined. Processed GR has been often used to reinforce and change the efficacy of GR. Glycyrrhizin(GL) is one of the main constituents of GR. Following procedure described in the oriental medicinal reference, GR and GL were heated at $140^{\circ}C{\sim}240^{\circ}C$ in GC oven. And then, the content of GL in the processed GR and GL was analyzed by HPLC. GL was transformed to $18-{\beta}-glycyrrhetic\;acid\;mono-{\beta}-D-glucuronide(GM)$ and glycyrrhetic acid(GA) by processing at $170^{\circ}C$. Determination of the content of GL increasing the heating temperature showed that GL was decomposed by heat above $150^{\circ}C$. It was also found that the content of GL in GR processed by heat above $170^{\circ}C$ remakeably decreased.

Effects of Glycyrrhizae Radix on Repeated Restraint Stress-induced Neurochemical and Behavioral Responses

  • Glycyrrhizae radix (GR) is an herbal medicine that is commonly used in the East Asia for treating a variety of diseases, including stomach disorders. The objective of the present study was to examine the anti-stress effects of GR on repeated stress-induced alterations of anxiety, learning and memory in rats. Restraint stress was administered for 14 days (2 h/day) to the rats in the Control and GR groups (400 mg/kg/day, PO). Starting on the eighth day, the rats were tested for spatial memory on the Morris water maze test (MW) and for anxiety on the elevated plus maze (EPM). We studied the changes of the expressions of cholineacetyl transferase (ChAT) and tyrosine hydroxylase (TH) in the locus coerleus (LC) using immunohistochemistry. The results showed that the rats treated with GR had significantly reduced stress-induced deficits on their learning and memory on the spatial memory tasks. In addition, the ChAT immunoreactivities were increased. Gor the EPM, treatment with GR increased the time spent in the open arms (p<0.001) as compared to that of the control group. Moreover, GR treatment also normalized the increases of the TH expression in the LC (p<0.001). In conclusion, administration of GR improved spatial learning and memory and reduced stress-induced anxiety. Thus, the present results suggest that GR has the potential to attenuate the behavioral and neurochemical impairments caused by stress.

감초가 면역반응에 미치는 영향 (I) - 50% 메탄올 엑스의 면역조절작용 - (Effect of Glycyrrhizae Radix on the Immune Responses(I) - Immuno-regulatory Action of 50% Methanol Extract -)

  • These experiments were conducted to investigate the effects of Glycyrrhizae Radix extract(GR) on histamine synthesis, lymphocyte blastogenesis in C57BL/6J mice splenocytes, IL-1 production, $Ca^{2+}$ uptake by macrophage-like P388D$_{1}$ cells and plaque forming cell assay against SRBC. Histamine contents, lymphocyte blastogenesis, IL-1 activity, $Ca^{2+}$ uptake and plaque forming cell were determined by enzyme isotope method, [$^{3}$H]-thymidine incorporation, C3H/HeJ mouse thymocytes proliferation, the addition of 5 $\mu$Ci/ml $^{45}Ca^{2+}$ to P388D$_{1}$ cell suspension and assay to sheep red blood cell, respectively. Cytotoxicity, which was expressed as 50% mortality, was occurred by the addition of GR(10$^{-3}$g/ml). Histamine production in mouse spleen cell culture was significantly increased by 48 hour incubation added 0.25$\mu\textrm{g}$/ml of Con A. Con A-dependent T-lymphocyte proliferation was also enhanced by the addition of 0.25 $\mu\textrm{g}$/ml of Con A. GR depressed histamine contents at 10$^{-9}$~10$^{-4}$g/ml. and Con A (0.25 $\mu\textrm{g}$/ml) dependent T-lymphocyte proliferation at 10$^{-5}$~10$^{-4}$g/ml. IL-1 activity was significantly decreased by 10$^{-8}$~10$^{-4}$g/ml of GR. $Ca^{2+}$ uptake was not changed by GR, but antibody production markedly increased at 10.0~50.0 mg/kg of GR. From the above results, it is suggested that GR have immuno-regulatory action; GR decreased cell-mediated immune response and increased antibody production by B lymphocyte at high doses.
