Lysosomal storage diseases (LSDs) are a group of inherent diseases characterized by massive accumulation of undigested compounds in lysosomes, which is caused by genetic defects resulting in the deficiency of a lysosomal hydrolase. Currently, enzyme replacement therapy has been successfully used for treatment of 7 LSDs with 10 approved therapeutic enzymes whereas new approaches such as pharmacological chaperones and gene therapy still await evaluation in clinical trials. While therapeutic enzymes for Gaucher disease have N-glycans with terminal mannose residues for targeting to macrophages, the others require N-glycans containing mannose-6-phosphates that are recognized by mannose-6-phosphate receptors on the plasma membrane for cellular uptake and targeting to lysosomes. Due to the fact that efficient lysosomal delivery of therapeutic enzymes is essential for the clearance of accumulated compounds, the suitable glycan structure and its high content are key factors for efficient therapeutic efficacy. Therefore, glycan remodeling strategies to improve lysosomal targeting and tissue distribution have been highlighted. This review describes the glycan structures that are important for lysosomal targeting and provides information on recent glyco-engineering technologies for the development of therapeutic enzymes with improved efficacy. Many therapeutic glycoproteins have been successfully generated in plants. Plants have advantages regarding practical and economic concerns, and safety of protein production over other existing systems. However, plants are not ideal expression systems for the production of biopharmaceutical proteins, due to the fact that they are incapable of the authentic human N-glycosylation process. The majority of therapeutic proteins are glycoproteins which harbor N-glycans, which are often essential for their stability, folding, and biological activity. Thus, several glyco-engineering strategies have emerged for the tailor-making of N-glycosylation in plants, including glycoprotein subcellular targeting, the inhibition of plant specific glycosyltranferases, or the addition of human specific glycosyltransferases. This article focuses on plant N-glycosylation structure, glycosylation variation in plant cell, plant expression system of glycoproteins, and impact of glycosylation on immunological function. Furthermore, plant glyco-engineering techniques currently being developed to overcome the limitations of plant expression systems in the production of therapeutic glycoproteins will be discussed in this review.
Neopentyl glycoI(NPG) 의 함량변화에 따른 diglycidyl ether of bisphenol A (DGEBA)/4,4' -methylene dianiline (MDA) 계의 등온적 경화거동을 DSC를 이용하여 분석하였다. DGEBA/MDA 계의 경화속도를 증가시키기 위하여 NPG를 첨가하였다. NPG 함량에 관계없이 전화율 곡선이 s-자 모양이었으며, 이것은 DGEBA/MDA/NPG 계의 경화반응이 자촉매 반응 메커니즘에 따른다는 것을 의미한다. NPG의 함량이 증가함에 따라 경화속도가 증가하였으며, 이것은 NPG가 가지고 있는 두 개의 hydroxyl 기가 촉매로 작용하기 때문이다.
In metal-affinify aqueous two-phase systems, protein partitioning is affected by a variety of parameters such as pH, the number of surface-accessible histidines, and the amount and partition coefficient of metallated polythylene glyco(PEG) ligand. To enhance partitioning of proteins with surface-accessible histidines, we have synthesized and used a (Cu(II)-ininodiacetic acid)$_2$-PEG20,000($Cu(II)_2IDA_2$-PEG20,000) as well as Cu(II)IDA-PEG5,000 as an affinity ligand. The partition coefficient of $Cu(II)_2-IDA_2$-PEG20,000 in a PEG5,000/dextran two-phase system was 30.1, which corresponded to a 3.8-fold increase over that of Cu(II)IDA-PEG5,000. The partitioning experiments were performed on four proteins, horse cytochrome c, S. cerevisiae cytochrome c, horse myoglobin, and sheep myoglobin. Partitioning of proteins which convey surface-accessible histidines was enhanced dramatically by the addition of $Cu(II)_2IDA_2$-PEG20,000 ligand. These results demonstrate that enhanced partitioning of metal-binding proteins in an aqueous two -phase system can by achieved by using an appropriate metallated PEG ligand.
1. 섬유간 지방질 함량은 소와 닭의 근섬유의 경우에는 white muscle보다 red muscle에서 높게 나타났으나 돼지 근섬유의 경우에는 white muscle쪽이 높았다. 2. 근섬유간 지질의 분포를 살펴보면 중성지질이 가장 많이 차지하고 극성지질과의 조성비에 있어서는 동물의 종 및 muscle type에 따라 다르게 나타났다. 3. 지방관의 조성에 있어서는 불포화 지방산의 함량비가 두 muscle type 사이에서 다르게 나타났으며 특히 돼지의 근섬유간 지방질에는 고도불포화지방산 함량이 높은 특징을 보였다.

Conserved domains are defined as recurring units in molecular evolution and are commonly used to interpret the molecular function and biochemical structure of proteins. Herein, the ADP-glucose pyrophosphorylase (AGPase) amino acid sequences of three species of the Ipomoea genus [Ipomoea trifida, I. triloba, and I. batatas (L.) Lam. (sweetpotato)] were identified to investigate their physicochemical and biochemical characteristics. The molecular weight, isoelectric point, instability index, and grand average of hyropathy markedly differed among the three species. The aliphatic index values of sweetpotato AGPase proteins were higher in the small subunit than in the large subunit. The AGPase proteins from sweetpotato were found to contain an LbH_G1P_AT_C domain in the C-terminal region and various domains (NTP_transferase, ADP_Glucose_PP, or Glyco_tranf_GTA) in the N-terminal region. Conversely, most of its two relatives (I. trifida and I. triloba) were found to only contain the NTP_transferase domain in the N-terminal region. These findings suggested that these conserved domains were species-specific and related to the subunit types of AGPase proteins. The study may enable research on the AGPase-related specific characteristics of sweetpotatoes that do not exist in the other two species, such as starch metabolism and tuberization mechanism.
Poly (vinylidene fluoride) (PVDF)의 소수성 중공사막 표면에 계면중합하여 복합막을 제조하였다. Piperazine (PIP)과 trimesoyl chloride (TMC)의 농도변화, polyethylene glycol (PEG)의 함량변화에 따라 막을 제조하였으며, 막의 특성 평가를 위해 100 ppm의 NaCl, $CaSO_4$, $MgCl_2$ 용액과 NaCl과 $CaSO_4$를 혼합하여 제조한 300 ppm의 공급액에 대한 막의 투과도와 배제율을 알아보고자 하였다. TMC를 사용하여 계면중합하였을 때 막의 투과도와 배제율이 가장 높게 나타났으며, 0.1~1 wt%로 TMC 농도를 변화시켜 가며 실험을 수행한 결과 0.1 wt%일 때 NaCl 100 ppm에 대해 투과도 48.3 LMH ($L/m^2{\cdot}hr$)와 배제율 59%로 가장 높은 값을 나타내었다. 또한, 투과도를 향상시키기 위해 annealing처리와 piperazine에 PEG를 첨가하여 실험을 수행하였다. 실험결과 처리하지 않은 막에 비해 투과도는 전체적으로 향상되는 모습을 나타냈지만 배제율이 감소하는 경향을 나타내었다.
본 연구에서는 CHO 세포를 이용해 세포를 별도의 적응기간 없이 무혈청 배지에서 배양했을 때 세포의 증식이 중단되는 원인을 찾고 배지 첨가 성분을 통해 이를 개선하고자 했다. 현재 개발된 무혈청 배지는 아직까지 혈청을 대체할 만한 성분을 포함하고 있지 않다. 때문에 무혈청 배지에 적응되지 않은 비적응 세포의 경우 계대 배양에 한계가 있다. 이런 한계가 나타나는 원인은 다양할 것으로 생각이 되지만 혈청의 부재로 인해 세포가 받게 되는 스트레스와 그로 인한 세포주기의 정지가 가장 근본적인 원인으로 생각된다. 무혈청 배지에서 세포가 받는 스트레스의 정도를 알아보고 배양 환경과 첨가물에 따른 ROS 농도의 변화를 측정하기 위해 배지와 세포의 ROS 농도를 측정하였다. ROS 농도를 측정한 결과 무혈청 상태에서 세포내 ROS가 엄청난 양으로 증가하는 것을 알 수 있었다. 이것은 혈청이 항산화능력을 갖고 있어서가 아니라 세포가 무혈청 환경에서 극심한 스트레스상태에 놓이기 때문인 것으로 생각된다. 이렇듯 증가한 ROS가 세포의 증식이 멈추게 되는 원인 중 하나로 생각되고, 항산화제를 첨가한 경우에도 증식력이나 ROS의 농도에 큰 차이가 없었던 것으로 미루어 보아 근본적으로 혈청과 같은 강력하게 증식을 촉진하는 성분을 배지에 첨가해야 할 것으로 여겨진다. ROS 이외에 세포의 증식이 멈추는 또 다른 원인으로 세포사멸의 여부를 확인했다. 무혈청 배지에서 배양한 적응세포와 비적응 세포 모두 특별한 세포사멸의 징후가 나타나지 않았다. 또한 무혈청 배지에서 증식이 멈춘 세포를 회수해 다시 혈청배지에서 배양한 경우 곧바로 증식력이 회복되기 때문에 대규모의 세포사멸은 발생하지 않는 것으로 생각된다. 위와 같은 현상들은 모두 혈청이 없기 때문에 발생하는 것으로 혈청을 대체할 수 있는 첨가물을 배지에 더해주면 세포의 증식이 개선될 것이다. 그래서 몇 가지 첨가물을 이용해 세포의 증식력에 변화가 나타나는지 알아보았다. 첨가물을 이용한 실험에서 IGF-I의 경우 장기간 배양에서 세포의 수를 안정적으로 유지하고 계대 횟수를 증가시키는 효과를 보였다. 이는 IGF-I이 어느정도 세포의 증식을 유지시켜주는 역할을 하기 때문인 것으로 생각된다. 무혈청 배지에서 비적응 CHO 세포의 계대 배양에 한계가 있는 것은 세포주기가 멈추기 때문인 것으로 생각된다. 세포주기가 멈추는 growth factor와 같이 세포의 증식을 지속적으로 유도할 수 있는 물질이 무혈청 배지에서는 부족하기 때문인 것으로 생각되고, IGF-I과 같은 첨가물을 통해 극복할 수 있는 문제라고 여겨진다.
