Leptin is produced by adipocytes and its role in the regulation of lipid metabolism, feed intake, productive and reproductive performance of domestic animal species has been greatly stressed and extensively investigated in recent years. This study was conducted to develop a radioimmunoassay (RIA) for the estimation of plasma bovine leptin and to determine plasma leptin concentration in fattening Japanese Black cattle (Wagyu) and its crossbreds at commercial farms. Relationships of plasma leptin with plasma vitamin A and age of crossbred cattle were also determined. Recombinant bovine leptin (rbleptin) was produced by the E. coli overexpressed leptin as a GST (glutathione S-transferase)-fusion protein. Then antiserum against bovine leptin was obtained by its immunization in rabbits. Using this antiserum, a bovine specific RIA was developed and plasma leptin level was determined in 120 crossbred fattening cattle (WagyuHolstein, 50:50) at commercial farms. The plasma leptin level increased with the age of cattle and its level was greater in the crossbred heifers than in the steers. Plasma vitamin A level was negatively correlated with plasma leptin level in crossbred heifers and steers. This relationship was stronger in heifers than in steers. Plasma leptin was gradually increased with advancing age in fattening Wagyu cattle. In conclusion, development of a bovine specific RIA to estimate plasma leptin will contribute to better understanding of the role of leptin in cattle.
A biocontrol bacterium Bacillus licheniformis N1 grown in nutrient broth showed no chitinolytic activity, while its genome contains a gene which encodes a chitinase. The gene for chitinase from B. licheniformis N1 was amplified by PCR and the deduced amino acid sequence analysis revealed that the chitinase exhibited over 95% identity with chitinases from other B. licheniformis strains. Escherichia coli cells carrying the recombinant plasmid displayed chitinase activity as revealed by the formation of a clear zone on chitin containing media, indicating that the gene could be expressed in E. coli cells. Chitinase gene expression in B. licheniformis N1 was not detected by RT-PCR analysis. The protein was over-expressed in E. coli BL21 (DE3) as a glutathione S-transferase fusion protein. The protein could also be produced in B. subtilis 168 strain carrying the chitinase gene of N1 strain. The crude protein extract from E. coli BL21 carrying GST fusion protein or culture supernatant of B. subtilis carrying the chitinase gene exhibited enzyme activity by hydrolyzing chitin analogs, 4-methylumbelliferyl-$\beta$-D-N,N'-diacetylchitobioside and 4-methylumbelliferyl-$\beta$-D-N,N',N"-triacetylchitotrioside. These results indicated that even though the chitinase gene is not expressed in the N1 strain, the coding region is functional and encodes an active chitinase enzyme. Furthermore, B. subtilis 168 transformants expressing the chitinase gene exhibited antifungal activity against Fulvia fulva by suppressing spore germination. Our results suggest that the proper engineering of the expression of the indigenous chitinase gene, which will lead to its expression in the biocontrol strain B. licheniformis N1, may further enhance its biocontrol activity.
Src-Homology 2 (SH2) domains have a capacity to bind phosphotyrosine-containing sequence context and play essential roles in various cellular signaling pathways. Due to the specific nature of the binding between SH2 domains and their counterpart proteins, inhibitors of SID domain binding have drawn extensive attention as a potential candidate for therapeutic agents. Here, we describe the binding assay system to screen for the ligands or blockers of the SH2 domains with an emphasis on the $p56^{lck}$ SH2 domain. In our assay system, SID domains expressed and purified as fusion proteins to Glutathione-S-transferase (GST) were covalently attached to 96-well microtitre plates through amide bond formation, which were subsequently allowed to bind the biotinylated phosphotyrosine (pY)containing synthetic pep tides. The binding of biotinylated pY peptides was detected by the horseradish peroxidase (HRP)-conjugated streptavidin. Using the various combinations of SH2 domain-pY peptides, we observed that: (1) The binding of pY-peptides to its counterpart SH2 domain is concentration-dependent and saturable; (2) The binding is highly specific for a particular combination of SH2 domain-pY peptide pair; and (3) The binding of Lck SH2-cognate pY-peptides is specifically competed by the nonbiotinylated peptides with expected relative affinity. These results indicate that the established assay system detects the SH2-pY peptide interaction with reproducible sensitivity and specificity and is suitable for screening the specific inhibitors of $p56^{lck}$ SH2 function.

4-lBB molecule is expressed on the surface of activated CD4$\^$+/ and CD8$\^$+/ T cells. We generated a panel of anti-4-1 B5 murine mAbs using a fusion protein consisting of the extracellular domain of human 4-1 BB fused to Glutathione S-transferase. The binding activity against cell surface 4-1 BB molecule was assessed by flow cytometry analysis. These studies showed that several anti-4-1 BB mAbs bound to 10-30% of CD4$\^$+/ and CD8$\^$+/T cells in PHA or Con A stimulated PBLs, although these mAbs interacted with only, l-2% of CD4$\^$+/ and CD8$\^$+/ T cells in normal PBLs, indicating the specificity of mAbs to the 4-l BB molecule on activated CD4$\^$+/ and CD8$\^$+/ T cells. Next, we examined the effect of an anti-4-l BB mAb (4B4-1-1) on allogeneic mixed lymphocyte reactions (MLRs). The data indicated that the antibody significantly inhibited the proliferative response at higher concentrations. When tested with several T cell mitogens, the antibody had no stimulatory or inhibitory effects on the mitogen-mediated T cell proliferation. These data suggest that 4-1 BB molecule may play a role in the regulation of antigen-mediated immune response.
Cytokine으로 처음 밝혀진 macrophage migration inhibitory factor (MIF)는 다양한 생물학적 활성 및 효소적 활성을 지닌 단백질로 알려지고 있다. 아직까지 MIF의 서로 다른 활성간의 연관성 및 효소적 기능이 세포내에서 어떤 역할을 하는지는 명확하게 밝혀지지 않았다. 본 연구에서는 MIF가 지닌 효소적 활성 중 tautomerase활성에 대하여 좀더 자세하게 연구하였다. 먼저, GST 융합 시스템을 이용하여 MIF의 정제 조건을 확립하였다. 이를 통해서 얻어진 GST (glutathione S-transferase)-MIF와 MIF 간의 활성을 비교하여 N-말단의 GST에 의하여 MIF의 tautomerase활성이 완전히 저해되는 것을 확인하였다. 다음으로, GST 대신 N-말단에 존재하는 매우 짧은 아미노산 잔기가 MIF의 tautomerase 활성을 저해하는 지를 조사하고자, GST-MIF의 GST를 thrombin으로 제거한 tMIF를 사용하였다. 그 결과 tMIF도 GST-MIF와 마찬가지로 tautomerase 활성이 완전히 저해된다는 사실을 확인할 수 있었다. 추가로 N-말단 tag에 의한 MIF의 효소적 활성 저해가 단백질의 구조적인 변환에 의한 것인지를 조사하기 위하여 단백질 cross-linking 연구를 수행하였다. 단백질 cross-linking 결과 tMIF도 MIF와 같이 정상적으로 oligomer를 형성하는 것을 확인하였다. 이들 결과는 MIF의 N-말단 $P^{2}$를 중심으로 하는 소수성 활성부위의 노출이 tautomerase활성에 결정적인 역할을 한다는 것을 시사한다. 또한 본 연구결과는 세포내에서 MIF가 다른 단백질과의 상호결합에 의해서도 충분히 tautomerase활성이 저해될 수 있을 것이라는 가능성을 제시한다.lelopathy 효과가 있음을 밝혀냈다. 특히 화본과 사료작물의 경우 italian ryegrass, 콩과 사료작물의 경우는 purple alfalfa가 생장억제효과가 높게 나타났다. 영향을 미치지 못했다. 이상의 결과를 종합하면 현재의 체형, 체형에 대한 인식, 매스컴의 영향 등이 체형에 대항 불만족이라는 스트레스로 인지되고, 이러한 이지된 스트레스와 체중조절 행동에 대한 태도가 체중조절 행동의도를 갖도록 하는 것으로 보인다. 특히 다이어트 행동에 대한 태도는 행동결과에 대한 신념이 주요 변수였음을 감안할 때 체형에 대한 불만족을 유발하는 요인들과 감안할 때 체형에 대한 불만족을 유발하는 요인들과 다이어트 행동 결과에 대한 신념이 주요 변수였음을 감안할 때 체형에 대한 불만족을 유발하는 요인들과 다이어트 행동 결과에 대한 신념이 체중조절 행동을 유발하는 주요 요인인 것으로 판단된다. 따라서 과잉 체중조절의 확산을 억제하기 위해서 현재 자신의 체형과 바람직한 체형의 기준에 대한 바른 인식을 갖도록 하는 것이 필요하다. 그리고 체형에 대한 사회적 인식이 중용한 스트레스 요인임을 감안하여 사회 전반에 만연되어 있는 바람직한 체형에 대한 인식의 변화를 위한 노력이 행해져야 한다. 또한 신념이 행동의도에 큰 영향을 미친다는 결과에 따라 다이어트 행동 결과에 대한 신념이 올바른 지식에 근거한 바람직한 신념으로 변화될 수 있도록 하여야 할 것이다. 또한 본 연구에서는 영양지식이 다이어트 행동의도에 아무런 영향을 미치지 않은 것으로 나타났다. 그러나 단기간에 실시한 영양지식의 교육이 행동의 변화까지는 영향을 미치지 않았으나 태도에는 영향을 미친 것을 참고하여 장기적인 목표를 갖고 영양 지식의 보급에도 힘을 기울여야 하겠다.큰 것으로 보인다.의 경우 압흔과 함께 pitting 이 관찰되며, Ormco Stainless Steel의
KIF5는 2분자의 kinesin heavy chain (KHC)과 2분자의 kinesin light cham (KLC)으로 구성되며 미세소관과 직접 결합한다. KIF5는 여러가지 세포 내 소기관을 이동시키나 KIF5가 이동시키는 운반체가 어떻게 특이적으로 결합하는지는 아직 밝혀지지 못하였다. 본 연구에서 효모 two-hybrid system을 사용하여 KLC1의 tetratricopeptide repeats (TRP) 부위와 결합하는 세포 내의 단백질을 분리하였다. 결과 KLC1와 특이적으로 결합하는JNK/stress-activated protein kinase-associated protein 1 (JSAP1/JIP3)을 분리하였다. 이러한 결합은 KLC1의 TRP1, 2 영역과 JSAP1의 leucine zipper 영역이 결합에 관여하며, 또한 효모 two-hybrid assay에서 JSAP1은 KLC2와 결합하지만 신경세포에서 발현하는 KIF5A, KIF5C 그리고 모든 세포에서 발현하는 KIF5B와는 결합하지 않았다. 단백질간의 결합을 pull-down assay로 확인한 결과 KLC1은 glutathione S-transferase (GST)와는 결합하지 않으나 GST결한 JSAP1과는 결합하였다. 또한 생쥐의 뇌 파쇄 액으로부터 JSAP1 항체로 면역침강을 행한 결과 KLC1은 JSAP1과 같이 침강하였다. 이러한 결과들은 KLC1은 JSAP1과 결합하며, JSAP1은 KLC1의 수용체로세포 내 KIF5의 수송의 매개 단백질로 작용함을 시사한다.
