Studies were carried out on the factors responsible for the change of pungency in 'Kagdugi'. During fermentation, the pH in 'Kagdugi' decreased to around to 4, while acidity increased continuously. In accordance with the decrease of pungency, the content of 4-methylthio-3-butenyl isothiocyanate (MTB-NCS) in the homogenate of 'Kagdugi' decreased gradually and on 3rd day, the optimum period for good flavor, below 5% of that in the homogenate of fresh radish. The decrease of MTB-NCS formation was accompanied by the gradual loss of 4-methylthio-3-butenyl glucosinolate (MTB-glucosinolate), which was found to disappear more rapidly than total glucosinolate. Also, the ascorbic acid-dependent myrosinase activity was observed to decline gradually with the fermentation time, although the ascorbic acid content varied above the concentration required for maximal enzyme activity. Thus, it was suggested that the decline of MTB-NCS may be related to the gradual losses of MTB-glucosinolate content and myrosinase activity which are both susceptible to the effect of acidic pH.
In order to establish the effective extraction and purification process of rapeseed protein, the extraction solvents were compared with one another ; and the residues of glucosinolate and phytate and the extraction yield of protein, which had been extracted by 1% sodium hexa mata-phosphate(SHMP) and purified through isoelectric precipitation, acid-washing and UF concentration, were investigated. As for the condition for extraction of rapeseed proteins, the solvent of 1% SHMP(pH 8.0) turned out the most appropriate ; so far as the purification process for the elimination of glucosinolate and phytate was concerned, the acid-washing twice or the process of the acid-washing once and UF concentration was considered the most effective. The yield and content of rapeseed protein were 37.1% and 75.3% respectively in the case of the acid-washing twice, 42.1% and 72.4% respectively in the case of the acid-washing once and UF concentraction, Consequently, with the elimination effects of glucosinolate and phytate put into consideration, the process of isoelectric precipitation, acid-washing once(pH 3.5), neutralizing(pH 7.5), UF concentration and then freeze drying proved the most effective purification process.
The study on the change of general properties of Kagdugi during dfermentation reveals that around the third day of fermentation, optimum for good flavor, the pH decreased to around or below 4 while the acidity increased gradually. The relative amount of thiocyanate in the radish Kimchi homogenate decreased to 73% after 1 day and after 3 days to 32% of control. And the content of indolylmethyl glucosinolate and total myrosinase activity in the tissue of radish Kimchi decreased gradually and on 3rd day to 25% and 4% of control, respectively. On the other hand the concentration of ascorbic acid in the radish Kimchi was found to vary around 1mM. Based on these results, the gradual decline of thiocyanate formation in the radish Kimchi homogenate is concluded to be caused by the gradual decomposition of indolylmethyl glucosinolate and the decline of myrosinase activity, which are directly affected by the change of pH during fermentation.

The edible roots of radish (Raphanus sativus L.) are consumed worldwide. For characterization and evaluation of the agronomic traits and health-promoting chemicals in radish germplasms, new germplasm breeding materials need to be identified. The objectives of this study were to evaluate the phenotypic traits and glucosinolate contents of radish roots from 110 germplasms, by analyzing correlations between 10 quantitative phenotypic traits and the individual and total contents of five glucosinolates. Phenotypic characterization was performed based on descriptors from the UPOV and IBPGR, and glucosinolate contents were analyzed using liquid chromatography-tandem mass spectrometry in multiple reaction monitoring mode (MRM). Regarding the phenotypic traits, a significant correlation between leaf length and root weight was observed. Glucoraphasatin was the main glucosinolate, accounting for an average of 71% of the total glucosinolates in the germplasms; moreover, its content was significantly correlated with that of glucoerucin, its precursor. Principal component analysis indicated that the 110 germplasms could be divided into five groups based on their glucosinolate contents. High levels of free-radical scavenging activity (DPPH) were observed in red radishes. These results shed light on the beneficial traits that could be targeted by breeders, and could also promote diet diversification by demonstrating the health benefits of various germplasms.
We evaluated the effects of three common cabbage cooking methods (blanching, steaming and microwaving) on glucosinolate and S-methylmethionine (SMM) content and total antioxidant capacity of cabbage leaves. We detected four glucosinolates, including glucoraphanin, sinigrin, glucobrassicin, and 4-methoxyglucobrassicin, by high-pressure liquid chromatography (HPLC). Cabbage contained high levels of SMM (192.85 mg/100 g dry weight), compared to other cruciferous vegetables. Blanching cabbage leaves for one to ten minutes decreased glucosinolate and SMM levels, whereas microwaving or steaming cabbage for 5-10 min preserved glucosinolate and SMM levels. 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and 2-2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging activities of cooked cabbage generally decreased as cooking time increased, but microwave cooking had a smaller negative effect on antioxidant activities than blanching or steaming. This study demonstrates that some domestic cooking methods, such as microwaving and steaming, can increase the bioaccessibility of glucosinolates and SMM, highlighting the positive role of cooking on the nutritional qualities of cabbage.
The effects of microbubbles on glucosinolate accumulation and growth of watercress (Nasturtium officinale R. Br.) were investigated. Watercress plant at the 4th mature leaf stage (2 weeks old) were exposed to microbubbles or non-microbubbles generated in an Otsuka-house nutrient solution for 3 weeks in a controlled environment culture room. Stem length of the watercress grown under the microbubbles was 41% shorter than that of the non-microbubbles, showing significantly different. However, shoot fresh and dry weights, root length, leaf length, leaf width, SPAD, and quentum yield of the watercress were not significantly different between treatments. Glucoiberin, glucobrassicin, gluconapin, gluconasturtiin of the watercress grown under microbubbles, excepted for 4-methoxyglucobrassicin, were significantly higher than those of the watercress grown in non-microbubbles. In addition, watercress grown under microbubbles for 3 weeks contained 85% (${\mu}mol/g$ DW) and 65% (${\mu}mol/plant$) more total glucosinolate, respectively. Results indicated that microbubbles generated in a deep flow technique hydroponics system could increase the accumulation of glucosinolate without growth reduction.
Glucosinolates are hydrolyzed by the enzyme myrosinase and are mainly found in cruciferous vegetables such as Chinese cabbage (Brassica campestris L. ssp. pekinensis). lsothiocyanates (ITCs) are glucosinolate degradation products with reported anticarcinogenic properties. Korean Chinese cabbage in the form of 'kimchi' is a staple part of the Korean diet. In this study, we examined the effects of storage temperature and duration on glucosinolate, ITC, soluble sugar, and organic acid levels in kimchi. Changes in pH and the impact of various parts of the Korean Chinese cabbage being used during the preparation of the dish were also assessed. Extracted ITC levels, analyzed via gas chromatography (GC) and GC/mass spectrometry (GC/MS), were higher in the midrib parts than in the cabbage leaves after storage at both 4 and $20^{\circ}C$. During storage, organic acid levels increased while soluble sugars were depleted. The pH initially increased (after 1 day at $20^{\circ}C$, and 1 week at $4^{\circ}C$), but subsequently decreased over time at both temperatures. Glucosinolate and ITC levels increased in the beginning of storage but then generally fell during further storage. Our data suggest that acidity-related reduction in myrosinase activity during storage may decrease glucosinolate and ITC levels. The changes in these levels depended on the storage conditions and the Korean Chinese cabbage parts used for the kimchi preparation.
Purpose: The objective of this study is to evaluate the effect of cultivation conditions on the growth and glucosinolate content of Chinese cabbage and kale. Methods: Chinese cabbage and kale were grown in three different cultivation conditions, including a plant factory, greenhouse, and open field. Samples were collected at two harvesting times (10 d and 20 d after transplanting the seedlings). Nine growth parameters (plant height, plant width, number of leaves, petiole diameter, SPAD readout, leaf length, leaf width, stem diameter, and plant weight) were measured immediately after harvesting, and the samples were freeze-dried and stored until the glucosinolate content was analyzed. Mean values of the growth parameters and glucosinolate contents were evaluated using Duncan's multiple range tests. Results: The results indicated that the plant parameters of the Chinese cabbage and kale were greater for plants grown in the plant factory and greenhouse. The plant height, width, and weight showed significant differences in the Duncan's multiple range tests at a 5% level. The plant factory also produced greater contents of most of the glucosinolates. Conclusions: Three different cultivation conditions significantly affected the growth and glucosinolate contents of Chinese cabbage and kale. Further study is necessary to investigate other functional components and different vegetable varieties.

Compositional changes in pungent components of Dolsan Leaf Kimchi during fermentation were investigated. Major volatile compounds identified in the kimchi were 3-isothiocyanate-1-propene(allyl isothiocyanate, AITC) di-2-propenyl disulfide, 1-methoxy-2-butanol, 4-isothiocyanate-1-butene and dimethyl trisulfide. The contents of allyl isothiocyanate and 4-isothiocyanate-1-butene decreased, while dimethyl trisulfide increased during fermentation and storage. 1-methoxy 2-butanol increased at the initial stage of fermentation, showing highest at 2~3 days, and decreased thereafter. Di-2-propenyl disulfide decreased after 5 days and increased after 10days of storage. Total glucosinolate content increased by 3days and decreased from 4days of storage.
The objective of this study was to investigate the technical feasibility of producing toxicant-free rapeseed by germination. To this end, rapeseed(Brassica napus L.)was germinated at $25^{\circ}C$ for 120 hours, and the chemical compositions-glucosinolates and free sugers-were determinated in every 24 hours during germination. The amount of glucosinolates in rapeseed measured by UV method was very close to that measured by GLC method. The glucosinolates were considerably abundant in rapeseed before germination, and the total content was found to be 13.6 mg/g. Rapeseed showed the lowest glucosinolate content in 72 hours during germination, and it gradually increased glucosinolate content from 96 hours. Free suger content in rapeseed before germination was as follows : 3.03 mg/g of fructose, 2.97 mg/g of glucose and 5.63 mg/g of sucrose. Raffinose and stachyose were not detected, and in general free sugars were gradually decreased during germination. However, sucrose was increased in the early period of gremination and decreased in the later period.
