Aryl hydrocarbon receptor nuclear translocator (Arnt) belongs to bHLH-PAS protein family. Here, we study the role of Arnt in both cell growth and glucose metabolism. Our results demonstrated that the absence of Arnt does affect ATP homeostasis but not cell growth in monolayer-cultured mouse hepatoma cells. ATP level of Arnt defective BpRc1 hepatoma cells is less than that of wild type hepatoma cells in both normoxia and hypoxia. BpRc1 cells also fail to increase the expression of glycolytic enzymes in response to hypoxia. Our results suggest that Arnt is essential for glucose metabolism and ATP production but not for cell growth.
This study was carried to investigate the antidiabetic and lipid metabolism of water extract paecilomyces japonica(PJ) in Streptozotocin (STZ) induced diabetic rats. Diabetes were induced by intravenous injection of STZ at a dose of 42mg/kg dissolved in citrate buffer. The water extract of paecilomyces japonica were orally administrated once a day for 7 days at a dose of 500mg/kg or 1,000mg/kg. The contents of serum glucose, triglyceride(TG), total cholesterol were significantly decreased in PJ treated group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucose-6-phosphate dehydrogenase(G-6-PDH), glucokinase(GK) were significantly increased, but activity of glucose-6-phoshatase(G-6-Pase) was significantly decreased in PJ treated group compared to the those of STZ-control group. These results indicated that water extract of paecilomyces japonica would have antidiabetic and lipid metabolism effect in STZ-induced diabetic rats.
This present study was undertaken to evaluate the effects of 5 % chicory extract on serum glucose and lipid metabolism in diabetic rats treated with streptozotocin (STZ). The experimental subjects were divided into 4 groups. : No-fiber , cellulose, insulin, and chicorygroup. The animals were fed ad libitum each of the experimental diets for 4 weeks. The food intake and food efficiency ratio in chicory group was significantly higher than in no-fiber, cellulose, and inulin groups. The reduction of body weight was also significantly lower. The wet weights of cecum and cecal contents were significantly increased in rat fed chicory extract. Total glycated hemoglobin was significantly decreased by chicory extract feeding whereas serum total cholesterol . LDL-choelsterol, and HDL-choelsterol levels were significantly increased. But there were no differences between HDL-choesterol/total cholesterol ratios, LDL-cholesterol/HDL-cholesterol ratios, and atherogenic index. After 10 -hour fast, the levels of hepatic triglyceride and phospholipid were significantly higher in the chicory group than any in other groups. These results indicated that chicory extract is an effective therapeutic regimen for control of metabolic deragements in diabetics.
To investigate the metabolism of various substrates in preimplantation bovine embryos, uptake of glucose and pyruvate, and lactate production were measured in single IVF-derived bovine embryos by a non-invasive method. When the embryos were incubated for 5 h in culture medium supplemented with 1 mM glucose and 0.4mM pyruvate as substrates at each developmental stage, glucose uptake was increased with more advanced developmental stages while pyruvate uptake was decreased. Total lactate producton of 2-cell embryos was significantly higher than that of blastocysts (p<0.05). Both of glucose uptake and lactate production in normal morulae produced in vitro was significantly high compared to the degenerated embryos(p<0.05). The results obtained in the study suggest that pyruvate as an exogenous substrate may be support in bovine embryos until 8-cell stage, whereas glucose may be effective as an energy source after morula stage. In addition, it was proven thatlactate was not effective as an energy source in preimplantation development of IVF-derived bovine embryos.
Sleep-disordered breathing (SDB) is associated with increased cardiovascular and cerebrovascular morbidity. Epidemiological and clinic-based studies have shown that SDB is related to impaired glucose tolerance and increased insulin resistance, independent of obesity. Despite of a consistent association between SDB and impaired glucose-insulin metabolism, the mechanism underlying this relationship has not been fully elucidated. It is recognized that hypoxemia and hypercapnia that occur in SDB provoke sympathetic nervous activity and catecholamine, epinephrine and norepinephrine, and cortisol are released. Sympathetic hyperactivity and increased catecholamines can impair glucose homeostasis by increasing glycogenolysis and gluconeogenesis, which can result in increased circulating insulin levels and increased risk of insulin resistance. A prospective study is needed to investigate the causal relationship between SDB and impaired glucose-insulin metabolism in a healthy population without diabetes, hypertension and obesity as etiologic risk factors.
Land plants produce glucose (C6H12O2) through photosynthesis by utilizing carbon dioxide (CO2), water (H2O), and light energy. Glucose can be stored in various polysaccharide forms for later use (e.g., sucrose in fruit, amylose in plastids), used to create cellulose, the primary structural component of cell walls, and immediately metabolized to generate cellular energy, adenosine triphosphate, through a series of respiratory pathways including glycolysis, the tricarboxylic acid cycle, and oxidative phosphorylation. Additionally, plants must metabolize glucose into amino acids, nucleotides, and various plant hormones, which are crucial for regulating many aspects of plant physiology. This review will summarize the biosynthesis of different plant hormones, such as auxin, salicylic acid, gibberellins, cytokinins, ethylene, and abscisic acid, in relation to glucose metabolism.

Glucose 농도에 따른 바성장속도의 변화는 기질저해형식 (substrate inhibition type) 의 성장모델로 나타낼 수 있었다. 초기 glucose 농도 4g/f까지는 최대 세포밀도의 증가를 보여 주었다. Glucose는 세포사망속도에 큰 영향을 나타내었고 glucose 농도와 비사망속도 간에는 반비례의 관계가 성립됨을 보였다. Glucose 놓도가 증가될수록 glucose의 비소비속도가 증가되 었고, 초기 glucose 농도의 증가는 lactate의 총괄 비 생산속도의 증대를 가져왔다. 암모늄 이온의 총괄비 생산속도는 초기 Glucose의 농도에 의존하 였지만 암모늄 이온의 전반적인 생성속도는 초기 glucose 농도에 거 의 무관함을 보여 주었다.
Background: Ginsenoside Rg3 is one of the main active ingredients in ginseng. Here, we aimed to confirm its protective effect on the heart function in transverse aortic coarctation (TAC)-induced heart failure mice and explore the potential molecular mechanisms involved. Methods: The effects of ginsenoside Rg3 on heart and mitochondrial function were investigated by treating TAC-induced heart failure in mice. The mechanism of ginsenoside Rg3 for improving heart and mitochondrial function in mice with heart failure was predicted through integrative analysis of the proteome and plasma metabolome. Glucose uptake and myocardial insulin sensitivity were evaluated using micro-positron emission tomography. The effect of ginsenoside Rg3 on myocardial insulin sensitivity was clarified by combining in vivo animal experiments and in vitro cell experiments. Results: Treatment of TAC-induced mouse models with ginsenoside Rg3 significantly improved heart function and protected mitochondrial structure and function. Fusion of metabolomics, proteomics, and targeted metabolomics data showed that Rg3 regulated the glycolysis process, and Rg3 not only regulated glucose uptake but also improve myocardial insulin resistance. The molecular mechanism of ginsenoside Rg3 regulation of glucose metabolism was determined by exploring the interaction pathways of AMPK, insulin resistance, and glucose metabolism. The effect of ginsenoside Rg3 on the promotion of glucose uptake in IR-H9c2 cells by AMPK activation was dependent on the insulin signaling pathway. Conclusions: Ginsenoside Rg3 modulates glucose metabolism and significantly ameliorates insulin resistance through activation of the AMPK pathway.
Cancer cells are known to show increased rates of glycolysis metabolism. Based on this, PET studies using F-18-fluorodeoxyglucose have been used for the detection of primary and metastatic tumors. To account for this increased glucose uptake, a variety of mechanisms has been proposed. Glucose influx across the cell membrane is mediated by a family of structurally related proteins known as glucose transporters (Gluts). Among 6 isoforms of Gluts, Glut-1 and/or Glut-3 have been reported to show increased expression in various tumors. Increased level of Glut mRNA transcription is supposed to be the basic mechanism of Glut overexpression at the protein level. Some oncogens such as src or ras intensely stimulate Glut-1 by means of increased Glut-1 mRNA levels. Hexokinase activity is another important factor in glucose uptake in cancer cells. Especially hexokinase type II is considered to be involved in glycolysis of cancer cells. Much of the hexokinase of tumor cells is bound to outer membrane of mitochondria by the porin, a hexokinase receptor. Through this interaction, hexokinase may gain preferred access to ATP synthesized via oxidative phosphorylation in the inner mitochondria compartment. Other biologic factors such as tumor blood flow, blood volume, hypoxia, and infiltrating cells in tumor tissue are involved. Relative hypoxia may activate the anaerobic glycotytic pathway. Surrounding macrophages and newly formed granulation tissue in tumor showed greater glucose uptake than did viable cancer cells. To expand the application of FDG PET in oncology, it is important for nuclear medicine physicians to understand the related mechanisms of glucose uptake in cancer tissue.
