Distribution Dynamics and Proposed Determinants: Exploring Morphological, Clinical Laboratory, and Lifestyle Factors in the Coexistence of Age-Related Skeletal Muscle Mass Loss and Obesity among Young Men: A Nationwide Cross-Sectional Study

  • PURPOSE: This study examined the distribution dynamics and proposed determinants, including morphological measurements, clinical laboratory tests, and lifestyle factors among young Korean men aged 20 to 29 years with the coexistence of age-related loss of skeletal muscle mass and obesity (CALSMO). METHODS: Six hundred and sixty-six participants were divided into two groups based on their skeletal muscle mass index, with 12 individuals categorized in the CALSMO group and the remaining 654 in the normal group. The proposed determinants variables consisted of three main components: morphological measurements, clinical laboratory tests, and lifestyle factors. The morphological measurement variables were height, weight, body mass index, waist circumference, and skeletal muscle mass index. The clinical laboratory tests were fasting glucose, triglyceride, total cholesterol levels, and systolic and diastolic blood pressure. The lifestyle factors considered were alcohol consumption and tobacco use. Complex sampling analysis was performed for the evaluation. RESULTS: The distribution dynamics were determined to be 1.81(1.02-3.18) %. Morphological factors, such as height, weight, body mass index, waist circumference, and skeletal muscle mass index, showed significant differences (p < .05). The clinical laboratory test variables, specifically the fasting glucose, triglyceride, and total cholesterol levels, also exhibited significant differences (p < .05). The lifestyle factor, alcohol consumption, also showed a significance (p < .05). CONCLUSION: This study provides insights into the distribution dynamics. The proposed determinants in young Korean individuals with CALSMO are height, weight, body mass index, waist circumference, skeletal muscle mass index, fasting glucose, triglyceride, total cholesterol levels, and alcohol consumption.

  • This laboratory has recently reported the synthesis and in vitro antitumor activity of PT(II) complexes containing ethylenediamine and diphosphine. In view of the reports of others, cisplatin is toxic to the kidney since the kidney's vulnerability to PT(II) complexes may originate in its ability to accumulate and retain platinum to a greater degree than other organs. The in vitro cytotoxicity of these synthetic PT(II) complexes on the primary cultured proximal tubular cells of rabbit kidney and renal cortical cells of human kidney was investigated. Three endpoints for cytotoxicity tests were evaluated:3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT), $^3H$-thymidine uptake and the glucose consumption tests. The rank order of sensitivity exhibited $^3H$-thymidine uptake>MTT>glucose consumption test. The agents with diphosphine leaving group were significantly less cytotoxic than cisplatin. Moreover, 1,2-bis(diphenylphosphino)ethane (DPPE) exhibited less cytotoxicity than 1.3-bis (diphenylphosphino)propane (DPPP) against on rabbit and human cultured kidney cells. Based on these results, the decreased nephrotoxicity of these new complexes over cisplatin appeared to be partially attributable to a leaving group of DPPP and DPPE. This novel class of platinum compound represents a valuable lead in the development of a "third-generation" agent.

Improvement in metabolic parameters in obese subjects after 16 weeks on a Brazilian-staple calorie-restricted diet

  • BACKGROUND/OBJECTIVES: The standard pattern of Brazilian food consumption is based on the combination of rice and beans served together in the main meals. This study assessed the effects of Brazilian-staple calorie-restricted (BS-diet) dietary advice, with brown rice and beans, on metabolic parameters, body composition, and food intake in overweight/obese subjects. SUBJECTS/METHODS: Twentyseven subjects were randomly assigned to a conventional-type calorie-restricted diet (CT-diet) (n = 13) or a BS-diet (n = 14). Glucose metabolism, lipid profile, anthropometric and body composition parameters, and food intake were measured before and after 16 weeks. Paired t-tests/Wilcoxon tests were used for comparison of differences from baseline and unpaired t-tests/Mann-Whitney tests were used for comparison of differences between the groups. RESULTS: After16 weeks, both groups showed reductions in weight and waist circumference (P < 0.02), and the BS-diet group showed a decrease in body fat (P = 0.0001), and significant improvement in glucose metabolism (fasting plasma glucose, glucose and insulin areas under the curve, Cederholm index, and HOMA2-$%{\beta}$) ($P{\leq}0.04$) and lipid profile (cholesterol, triacylglycerol, LDL-c, VLDL-c, and cholesterol/HDL-c ratio) ($P{\leq}0.05$). In addition, the BS-diet group showed significant improvement in HOMA2-$%{\beta}$, compared to the CT-diet group (P = 0.03). The BS-diet group also showed a significant reduction in energy, lipids, carbohydrate, and cholesterol intake ($P{\leq}0.04$) and an increase in fiber intake ($P{\leq}0.001$), while the CT-diet group showed a significant reduction in intake of energy, macronutrients, PUFA, and cholesterol ($P{\leq}0.002$). CONCLUSIONS: These results demonstrate the benefits of the BS-diet on metabolic parameters in obese subjects.

Association of coffee consumption with type 2 diabetes and glycemic traits: a Mendelian randomization study

  • BACKGROUND/OBJECTIVES: Habitual coffee consumption was inversely associated with type 2 diabetes (T2D) and hyperglycemia in observational studies, but the causality of the association remains uncertain. This study tested a causal association of genetically predicted coffee consumption with T2D using the Mendelian randomization (MR) method. SUBJECTS/METHODS: We used five single-nucleotide polymorphisms (SNPs) as instrumental variables (IVs) associated with habitual coffee consumption in a previous genome-wide association study among Koreans. We analyzed the associations between IVs and T2D, fasting blood glucose (FBG), 2h-postprandial glucose (2h-PG), and glycated haemoglobin (HbA1C) levels. The MR results were further evaluated by standard sensitivity tests for possible pleiotropism. RESULTS: MR analysis revealed that increased genetically predicted coffee consumption was associated with a reduced prevalence of T2D; ORs per one-unit increment of log-transformed cup per day of coffee consumption ranged from 0.75 (0.62-0.90) for the weighted mode-based method to 0.79 (0.62-0.99) for Wald ratio estimator. We also used the inverse-variance-weighted method, weighted median-based method, MR-Egger method, and MR-PRESSO method. Similarly, genetically predicted coffee consumption was inversely associated with FBG and 2h-PG levels but not with HbA1c. Sensitivity measures gave similar results without evidence of pleiotropy. CONCLUSIONS: A genetic predisposition to habitual coffee consumption was inversely associated with T2D prevalence and lower levels of FBG and 2h-PG profiles. Our study warrants further exploration.

A Study of Health-Related Habit and Hematological Index of Male Workers Residing in Ulsan City

  • This study was conducted by surveying 616 male workers living in Ulsan City regarding their health status based on lifestyles such as alcohol consumption, smoking and exercising as well as physical measurements and biochemical tests. The average height, weight and BMI(body mass index, $kg/m^2$) of the subjects was 170.9cm, 70.2kg and 24.2, respectively. The rate of drinking was $80.9\%$ and the rate of smoking was $53.4\%$. Seventy four percent of subjects responded that they exercise regularly. The results of the blood biochemical tests revealed that the average hemoglobin concentration was 14.7g/dl, and the levels of GPT(glutamic pyruvic transaminase) and GOT (glutamic oxaloacetic transaminase) were 32.74unit/l, 26.99 unit/l, respectively. The average hemoglobin concentration for the subjects aged in the 50s was 14.39g/dl, which was significantly lower than those in the 20s(14.81g/dl), 30s(14.69g/dl) and 40s(14.73g/dl). The blood glucose level and the cholesterol level also increased with age. Also investigated was the blood pressure of the subjects increased with age,. and there was a significant increase(p < 0.05) for the subjects in the 50s compared to those in the 20s. The frequency of alcoholic beverages was significantly correlated with systolic/ diastolic blood pressure(p < 0.05) and $\gamma-GTP(gamma\;glutamyl\; transpeptidase)$(p<0.01). The duration of smoking showed a negative correlation(p < 0.05) with the hemoglobin and positive correlations with diastolic blood pressure, cholesterol and $\gamma-GTP(p<0.01)$. The study shows that blood pressure, blood glucose level, cholesterol level, GOT, GPT and $\gamma­GTP$ level, increase with age, which indicates higher possibility of degenerative diseases, calling for nutritional education in terms of advisable lifestyles regarding eating habits, alcohol consumption, smoking and regular exercise.

새로운 Platinum (II) Complex [Pt (II)(trans-d-dach)(DPPE)] $(NO_3)_2$의 항암효과 및 신독성에 관한 연구 (Antitumor Activity and Nephrotoxicity of the Novel Platinum(II) Coordination Complex)

  • 일부 malignant tumor에 Pt-complex의 일상 응용 과정에서 신장독성등의 심한 부작용이 문제점으로 지적되고 있다. 이 연구에서는 기존의 cisplatin보다 항암효과는 우수하면서, 부작용을 감소시킨 새로운 Pt-complex의 개발에 역점을 두었다. 본 연구에서 합성한 Pt(Ⅱ) complex는 carrier ligand로서 1,2-diaminocyclohexane(dach)을 사용하였고, leaving group으로는 diophosphine류인 1,2-bisdiphenylphosphinoethane(DPPE)을 도입하였으며, 물에 대한 용해도를 높이기 위해 dinitrate로 만들었다. 새로이 합성한 [Pt(Ⅱ)(trans-d-dach)(DPPE)]$(NO_3)_2$는 원소 분석, IR 및 $^{13}C-NMR$ 분석 data에 의하여 위의 물질임이 확인되었다. MTT assay method에 의한 항암활성 연구를 통하여 P-388, L-1210 lymphocytic leukemia cell과 SK-OV3 난소암세포에서 항암효과가 인정되었으며, 이 항암효과는 대조 약물로 사용된 cisplatin과 유사하였다. 토끼의 신세뇨관 세포와 인체의 신피질 세포를 이용한 cytotoxity 및 thymidine 섭취율과 인체 신피질 조직 배양을 이용한 glucose consumption 실험을 통하여 모두 cisplatin보다 신장독성이 현저히 감소되었다. 이상의 결과로 보아 Pt(Ⅱ)complexes는 carrier ligand와 leaving group의 선택에 따라 항암활성의 증가와 신독성의 감소를 일으키는 요인으로 보여지며, 이 연구에서 만들어진 Pt(Ⅱ)complex는 앞으로 다각적인 검토를 거쳐 새로운 항암화학요법제로 개발될 가능성이 있을 것으로 생각된다.

Comprehensive Cross-sectional Study of Sarcopenia in Young Korean Women: Assessing Body Dimensions, Clinical Indicators, and Behavioral Traits for Hazardous Components and Proportional Analysis

  • PURPOSE: This research investigated clinical hazardous components and analyzed the proportion of sarcopenia among young Korean women. METHODS: The cross-sectional study included 1,236 women aged 20 to 29 years, categorized into two groups according to their skeletal muscle mass index (SMI). Of these, 20 participants were placed in the sarcopenia group, while 1,216 were included in the normal group. The analysis involved hazardous components including body dimensions, clinical indicators, and behavioral trait variables: height, weight, body mass index, waist circumference, skeletal muscle mass index, systolic and diastolic blood pressure, blood laboratory tests assessing fasting glucose, triglycerides, total cholesterol, as well as smoking habits and alcohol consumption. Complex sampling analysis was used to analyze the proportion and hazardous components of sarcopenia. RESULTS: The proportion of sarcopenia was at 1.76% (95% of CI: 1.08-2.83). Anthropometric measurements, such as height, BMI, and WC, exhibited significant differences between the groups (p < .05). However, there was no significant difference in weight (p > .05) between the two groups. Among the clinical indicators, SBP, DBP, FBG, serum triglycerides, and total TC found to be significant hazardous components for sarcopenia within both groups (p < .05). Smoking status as a behavioral trait was significant as well (p < .05), unlike alcohol consumption (p > .05). CONCLUSION: This study discerned both the proportion of sarcopenia and the hazardous components associated with it among community-dwelling women of a young age.

Cis-Diaminocyclohexan을 배위자로 하는 배금(II)착체의 선택적 세포독성 (Selective Cytotoxicity Platinum (II) Complex Containing Carrier Ligand of cis-1,2-Diaminocyclohexane)

  • The use of cisplatin is limited by severe side effects such as renal toxicity. Our platinum-base drug discovery is aimed at developing drugs capable of diminishing toxicity and improving antitumor activity. We synthesized new Pt (II) complex analogue [Pt (cis-DACH)(DPPP)]. 2NO$_3$ (PC) containing cis-1,2-diaminocyclohexane as a carrier ligand and 1,3-bis(diphenylphosphino) propane as a leaving group. Furthermore, nitrate was added to improved the solubility. In this study, its structure was determined and its antitumor activity against SKOV-3 and NIH-OVCAR-3 human ovarian adenocarcinoma, and in vitro cytotoxicity was determined against primary cultured rabbit kidney proximal tubular and renal cortical cells of human kidney using colorimetric MTT assay. PC demonstrated acceptable antitumor activity against SKOV-3 and NIH-OVCAR-3 human ovarian adenocarcinoma and significant activity as compared with that of cisplatin. The toxicity of PC was found quite less than that of cisplatin using MTT and $^3$H-thymidine uptake tests in rabbit proximal tubular cells and human kidney cortical cells. PC was used for human cortical tissue in 7 weeks hitoculture by the glucose-consumption tests. We determined that the new platinum drug has lower nephrotoxicity than cisplatin. Based on these results, this novel platinum (II) complex compound (PC) represent a valuable lead in the development of a new anticancer chemotherapeutic agent capable of improving antitumor activity and low nephrotoxicity.

여성형유방증 발생에 영향을 미치는 통합적 요인분석 (Integrated Factor Analysis of the Influences on the Development of Gynecomastia)

  • 본 연구는 여성형유방증을 진단받은 환자와 정상 대조군 사이의 혈액검사 결과 및 생활 습관을 비교하여 여성형유방증의 발생에 영향을 미치는 요인을 분석하였다. 위험인자 간의 평균차이검정은 독립표본 t검정과 카이제곱검정을 시행하였으며 로지스틱 회귀분석을 통해 여성형유방증 발생에 영향을 미치는 다양한 예측 인자들의 상대적 위험비를 산출하였다. 나이, HGB, HCT, TC, Glucose, ALT, 음주, 탈모약, 전립선 비대증 치료제가 유의한 차이를 보이는 변수로 확인되었다. 특히 탈모약은 위험비 단변량분석에서 15.79로 가장 높게 나타났으며 HGB는 0.37배로 감소하였다. 이러한 결과는 여성형유방증 발생에 이들 요인이 중요한 역할을 할 수 있음을 시사하며, 향후 여성형유방증의 진단 및 관리를 위한 기초자료로 활용될 수 있을 것이다.

In Vitro Cytotoxicity of a Novel Platinum(II) Coordination Complex Containing Diaminocyclohexane and Dichloropropane

  • We have synthesized a novel platinum(II) coordination complex containing cis-1,2-diaminocyclohexane (DACH) as a carrier ligand and 1,3-dichloropropane (DCP) as a leaving group. A new series of [Pt(cis- DACH)(DCP)](PC) was evaluated for its cytotoxic activity on MKN-45 human gastric adenocarcinoma cells and normal primary cultured kidney cells. The new platinum complex has demonstrated high efficacy in the cytotoxicity against MKN-45/P, MKN-45/ADM and MKN-45/CDDP cell-lines. The cytotoxicity of PC against rabbit proximal renal tubular cells, human renal cortical cells and human renal cortical tissues, determined by MTT assay, the $[^3H]-thymidine$ uptake and glucose consumption tests, was found to be quite less than those of cisplatin. Based on these results, this novel platinum(II) coordination complex appears to be better for improving antitumor activities with low nephrotoxicity and is a valuable lead in the development of new, clinically available anticancer chemotherapeutic agents.
