Molecular mimicry is an attractive mechanism for triggering autoimmunity. In this review, we explore the potential role of evolutionary conserved bacterial proteins in the production of autoantibodies with focus on granulomatosis with polyangiitis (GPA) and rheumatoid arthritis (RA). Seven autoantigens characterized in GPA and RA were BLASTed against a bacterial protein database. Of the seven autoantigens, proteinase 3, type II collagen, binding immunoglobulin protein, glucose-6-phosphate isomerase, ${\alpha}$-enolase, and heterogeneous nuclear ribonuclear protein have well-conserved bacterial orthologs. Importantly, those bacterial orthologs are also found in human-associated bacteria. The wide distribution of the highly conserved stress proteins or enzymes among the members of the normal flora and common infectious microorganisms raises a new question on how cross-reactive autoantibodies are not produced during the immune response to these bacteria in most healthy people. Understanding the mechanisms that deselect auto-reactive B cell clones during the germinal center reaction to homologous foreign antigens may provide a novel strategy to treat autoimmune diseases.
The D-xylose operon in Escherichia coli is known to be regulated by a transcriptional activator protein, XyIR, which is responsible for the expression of both xylAB and xylFGH gene clusters. The XyIR was purified to homogeneity by using the maltose binding protein fusion expression and purification systems involving two chromatography steps. The purified XyIR protein was composed of two subunits of 45 kDa, which was determined by both sodium dodecyl sulfate polyacrylamide gel electrophoresis and gel filtration. The purified XyIR was specifically bounded to the xylA promoter, regardless of adding xylose to the reaction mixture, but binding of XylR was specifically bounded to the xylA promoter, regardless of adding xylose to the reaction mixture, but binding of XylR to the xylA promoter was enhanced by adding xylose. The enhanced binding ability of XyIR in the presence of xylose was not diminished by adding glucose. The presumed XyIR binding site is located between 120 bp to 100 bp upstream the xylA initiation codon.
가리비류 4종 - 큰가리비(Patinopecten yessoenensis), 주문진가리비(Chlamys swifti), 비단가리비(Chlamys ferreri ferreri), 해가리비(Amusium japonicum japonicum) -을 한국 5개 지역에서 채집하였고 중국산 비단가리비를 시장에서 구입하여 실험에 사용하였다. 총 7개 동위요소에 대한 starch gel 전기영동을 실시한 결과, 8개 유전자가 관찰되었다. 유전적 유사도는 비단가리비 3집단이 가장 가까운 관계를 보였고, 주문진가리비와 큰가리바가 유전적으로 서로 가까운 그룹으로 분류되었다. 이 그룹과 비단가리비가 0.595의 유사도를 보였으며 해가리비가 나머지 3종과는 유사도가 0.541로 가장 멀었다.
A horizontal starch gel electrophoresis for enzyme proteins extracted from 2 species of Korean viviparid snails; Cipangopaludina chinensis malleata and C. japonica was carried out in order to elucidate their genetic relationships. A total of 10 enzymes were employed in three different kinds of buffer systems. Two loci from each enzyme of alcohol dehydrogenase, esterase, glucose phosphate isomerase, isocitrate dehydrogenase, iditol dehydrogenase, malate dehydrogenase and peptidase(VL); and only one locus dach from two enzymes, glycerlo-3-phosphate dehydrogenase and phosphoglucomutase were detected; but, four loci from peptidase(LGG) were observed. Most of loci in two viviparid species showed homozygous monomorphic banding patterns and some of them were specific as genetic markers between two different species. However, EST-1, MDH-1, PEP(VL)-1loci showed polymorphic banding patterns. Foru populations of C. chinensis malleata were more closely clustered in a dendrogram within the range of genetic identity values of 0.928-1.00, and these clusters were lineated with C. japonica at the value of 0.355. In summarizing the above results, two viviparid snail species dmployed in this study mostly showed monomorphic enzyme protein banding patterns, and genetic differences specific between two species.
본 연구에서는 한국인 아동의 우식치아로부터 S. mutans를 분리하고, acid stress하에서 분리한 S. mutans의 내산성 능력과 관련된 단백질을 규명하고자 하였다. 2D gel electrophoresis를 수행한 결과, acid stress동안 elongation factor Ts, hypothetical protein, putative amino acid ABC transporter, adenylate kinase, fructokinase, Putative 40K cell well protein precursor, peptide deformylase, shikimate 5-dehydrogenase, mannose-6-phosphate isomerase, threonine synthase, putative dTDP-glucose-4,6-dehydratase의 발현량이 뚜렷이 증가하였으며 이들 단백질은 acid stress에 관여하는 단백질들로 추정된다.
Rhizopus oryzae의 카드뮴 적응 및 해독기작과 이에 관련된 세포내 생리 생화학적 변화를 조사하였다. R. oryzae는 카드뮴을 첨가 배양하였을 때 카드뮴 영향하에서는 carbohydrate metabolic pathway에 관련된 효소 활성(MDH, GPI)이 촉진되고 과산화물 제거에 관여하는 효소가 새롭게 유도(CAT2)된 반면, lactate를 이용하는 효소(LDH, ADH)의 활성이 감소된 사실은 중금속 영향하에서 세포의 성장과 에너지 공급을 위해 에너지 수율이 낮은 lactate를 이용하는 경로보다는 에너지 수율이 높은 TCA cycle 경로에 작용하는 효소들과 독성과산화물 제거에 관여하는 효소의 더 많은 derepression이 필요하다는 것을 알 수 있었다.
목 적: 본 연구진은 선행연구를 통하여 생쥐의 미성숙 난자와 성숙 난자 사이에 차이 나게 발현하는 유전자(DEGs)의 목록을 보유하고 있는데, 그 중에서 pentose phosphate pathway (PPP)에 필수적 효소인 Ribose 5-phosphate isomerase A (Rpia)를 선택하여 본 연구를 수행하였다. 난자 성숙 과정에 관련된 Rpia의 기능을 알아보기 위한 기초연구로서 생쥐와 돼지의 난소에서 Rpia의 발현을 비교분석 하였다. 연구방법: 생쥐의 각 조직에서 11개의 MII-selective DEGs의 발현을 RT-PCR방법으로 확인하여 난소에서 강하게 발현하는 4개의 유전자를 선택하였고, 다시 이들 4개 유전자 중 난자에서 높게 발현하는 Rpia를 선택하여 생쥐 및 돼지의 난자, 난구세포, 과립세포에서의 발현을 비교분석 하였다. 돼지 Rpia 염기서열은 밝혀져 있지 않아 EST clustering 기법을 통해 동정하였다. 결 과: EST clustering 기법으로 찾아낸 돼지 Rpia 염기서열은 GenBank에 등록하였고 (Accession Number EF213106), 이를 근거로 primer를 작성하여 RT-PCR을 수행하였다. Rpia 유전자는 생쥐에서는 난자 특이적으로 발현하는 반면 돼지에서는 난자, 난구세포, 과립세포에서 모두 발현하는 차이점을 발견하였다. 결 론: 본 연구는 생쥐와 돼지의 난소에서 Rpia유전자 동정에 대한 첫 보고로서, 본 연구결과로부터 생쥐와 돼지의 COCs는 서로 다른 경로로 포도당의 대사가 일어나는 것을 알 수 있었다. 따라서 이와 같은 차이점이 두 종의 난자를 체외 배양할 때 나타나는 난자 성숙률의 차이를 가져오는 기전 중의 하나가 아닐까 추측된다. 난자 성숙을 조절하는 기전을 연구함과 동시에 체외에서 난자 성숙이 어려운 종의 최적의 IVM (in vitro maturation)조건을 찾기 위해서는 앞으로 난자와 주변세포의 포도당 대사과정에 미치는 Rpia의 기능에 대한 후속연구가 필요할 것으로 사료된다.
Streptomyces chibaensis J-59 xylanase는 CSL 배지(1% corn steep liquor, 0.15% glucose, 0.1% $MgSO_4{\cdot}7H_2O$, pH 7.0)에서 1% xylan을 효소생산 유도물질로 첨가하였을 경우에 생산(0.83 unit/ml) 되었으나, xylose를 비롯한 각 종 탄소원을 포함하는 배지에서는 효소가 생산되지 않았다. S. chibaensis로 부터 생산된 세포외 xylanase를 $(NH_4)_2SO_4$ 분획침전, DEAE-Sephadex A-50 column chromatography, Sephadex G-200 gel filtration 과정으로 약 29%의 수율로 20배 정제하였다. 정제한 xylanase는 SDS-PAGE 및 Sephadex G-200 gel filtraion에서 분자량이 모두 25,000 Da으로 나타나 monomenc enzyme으로 확인되었다. 금속이온에 대한 영향은 $Fe^{3+}$, $Hg^{2+}$, $Cu^{2+}$, sodium dodecyl sulfate, N-bromosuccinide 등에서는 아주 강한 저해를 받았고, $Mn^{2+}$, $Zn^{2+}$에서는 약간의 저해를 받았다. 반면에 $Mg^{2+}$는 효소활성을 증가시켰다. Xylanase에 의한 xylooligo 당의 생산양상을 TLC로 조사한 결과 xylobiose, xylotriose 및 xylotetrose가 주 생산물이었으며, 반응 1시간 이후부터 소량의 xylose가 생성되었다. 따라서 본 효소는 전형적인 endotype xylanase로 확인되었으며 새로운 기능성식품인 자일로 올리고당(xylooligo-saccharides)의 제조에 이용할 수 있을 것이다.

Although the arthritis symptoms observed in the K/BxN model have been shown to be dependent on the functions of T and B cells specific to the self antigen glucose-6-phosphate isomerase, less is known about the in vivo roles of $CD4^{+}CD25^{+}$ regulatory T($T_{reg}$) cells in the pathology of K/BxN mice. We determined the quantitative and functional characteristics of the $T_{reg}$ cells in K/BxN mice. These mice contained a higher percentage of $Foxp3^+\;T_{reg}$ cells among the $CD4^+$ T cells than their BxN littermates. These $T_{reg}$ cells were anergic and efficiently suppressed the proliferation of $na\ddot{i}ve$$CD4^+$ T cells and cytokine production by effector $CD4^+$ T cells in vitro. Antibody-mediated depletion of $CD25^+$ cells caused K/BxN mice to develop multi-organ inflammation and autoantibody production, while the symptoms of arthritis were not affected. These results demonstrate that despite the inability of the $T_{reg}$ cells to suppress arthritis development, they play a critical role protecting the arthritic mice from systemic expansion of autoimmunity.

Anaerobic rumen fungi have been regarded as good genetic resources for enzyme production which might be useful for feed supplements, bio-energy production, bio-remediation and other industrial purposes. In this study, an expressed sequence tag (EST) library of the rumen anaerobic fungus Neocallimastix frontalis was constructed and functional genes from the EST library were analyzed to elucidate carbohydrate metabolism of anaerobic fungi. From 10,080 acquired clones, 9,569 clones with average size of 628 bp were selected for analysis. After the assembling process, 1,410 contigs were assembled and 1,369 sequences remained as singletons. 1,192 sequences were matched with proteins in the public data base with known function and 693 of them were matched with proteins isolated from fungi. One hundred and fifty four sequences were classified as genes related with biological process and 328 sequences were classified as genes related with cellular components. Most of the enzymes in the pathway of glucose metabolism were successfully isolated via construction of 10,080 ESTs. Four kinds of hemi-cellulase were isolated such as mannanase, xylose isomerase, xylan esterase, and xylanase. Five $\beta$-glucosidases with at least three different conserved domain structures were isolated. Ten cellulases with at least five different conserved domain structures were isolated. This is the first solid data supporting the expression of a multiple enzyme system in the fungus N. frontalis for polysaccharide hydrolysis.
