Prediction of glucose concentration in the interstitial fluid (ISF) based on mid-infrared absorption spectroscopy was examined at the glucose fundamental absorption band of 1000 - 1500/cm (10 - 6.67 um) using multi-component analysis. Simulated ISF samples were prepared by including four major ISF components. Sodium lactate had absorption spectra that interfere with those of glucose. The rest NaCl, KCl and $CaCl_2$ did not have any signatures. A preliminary experiment based on Design of Experiment, an optimization method, proved that sodium lactate influenced the prediction accuracy of glucose. For the main experiment, 54 samples were prepared whose glucose and sodium lactate concentration varied independently. A partial least squares regression (PLSR) analysis was used to build calibration models. The prediction accuracy was dependent on spectrum preprocessing methods, and Mean Centering produced the best results. Depending on calibration sample sets whose sodium lactate had different concentration levels, the standard error prediction (SEP) of glucose ranged $17.19{\sim}21.02\;mg/dl$.
This study examined whether dietary supplementation with sea tangle alters the intestinal morphology of streptozotocin-induced diabetic rats and affects the glucose absorption rate. Forty male Sprague-Dawley rats were divided into 2 groups and fed either a control (AIN76-based) diet or a sea tangle-supplemented diet. After 3 weeks, 10 rats in each group received an intramuscular injection of streptozotocin (45 mg/kg BW), and feeding was continued for 3 additional weeks. Dietary supplementation with sea tangle resulted in a lower fasting plasma glucose level compared with the control diet in diabetic rats. Scanning electron micrographs revealed serious damage to the jejunal villi of diabetic rats fed the control diet, whereas supplementation with sea tangle alleviated the damage. In a separate experiment, 20 male Sprague-Dawley rats were divided into 2 groups and fed either a control diet or a sea tangle-supplemented diet for 5 weeks, and fasted rats were subjected to in situ single-pass perfusion. The glucose absorption rate determined in the absence of digesta was decreased by 34% in the jejunum of rats fed a sea tangle diet compared with those fed a control diet. In conclusion, sea tangle supplementation lowered glucose absorption rate, altered intestinal morphology, and appeared to protect villi from damage caused by diabetes mellitus.
The mechanisms responsible for the antidiabetic activity of both the white ginseng radix (Ginseng Radix Alba, GRA) and the rootlet (Cinseng Radix Palva, GRP) were investigated. After a four week oral administration, the fasting blood glucose levels in the GRA- and GRP-treated groups were lower when compared to the control group. To elucidate the hypoglycemic mechanism(s) of the ginseng radices, glucose absorption from the small intestine, hepatic hexokinase and glucose-6-phosphatase activities, in addition to PPAR-${\gamma}$ expression in adipose tissue were examined. The results strongly suggest that GRA can improve hyperglycemia in KKAy mice, possibly by blocking intestinal glucose absorption and inhibiting hepatic glucose-6-phosphatase, and GRP through the upregulation of adipocytic PPAR-$\gamma$ protein expression as well as inhibiting intestinal glucose absorption.
This study was described for measuring clinically relevant levels of glucose in undiluted plasma and whole blood by near-infrared (NIR) spectroscopy. Result from an initial measurement of major blood components powder was over-lapped the absorption bands of glucose at 1500-1600 nm. However, the NIR data of blood components were clearly separated by principle component analysis (PCA) space. By the use of partial least squares (PLS) regression, glucose concentrations in undiluted plasma and whole blood could be determined with standard errors of prediction (SEP) of 15 mg/dl and 76 mg/dl, respectively. Although these blood components possessed strong absorption bands that overlapped with the absorption bands of glucose, successful calibration models could be carried out.
A method is described for measuring clinically relevant levels of glucose in a pH 7.4 phosphate buffer by nearinfrared (NIR) absorption spectroscopy. Three types of NIR spectrometer, dispersive type, photo-diode array (PDA) type, and fourier transform (FT) type spectrometer were used and the performance was compared. Spectra were collected with a cuvette cell or quartz liquid fiber of 1 mm or 2 mm optical pathlength as transmittance method. Glucose absorption band appeared at second overtone, first overtone, and combination region for all systems. By use of the multivariate technigue of partial least squares (PLS) regression, glucose concentrations can be determined with a 16, 44, and 9.1 mg/d l standard error of prediction for dispersive type, photo-diode array type, and fourier transform type system, respectively. Sensitivity of spectrometer was evaluated by absorbance for the difference of 10 mg/d l glucose. Three absorption bands, second overtone, first overtone, and combination region were suited to three types systems, dispersive type, photo-diode array type, and fourier transform type systems, respectively. This investigation showed that three types NIR spectrometer were proper method for identification and quantitative analysis of glucose and possible for noninvasive blood glucose monitoring.
The purpose of the present study was to investigate the effect of medium chain fatty acid salts, reported as enhancers in insulin nasal absorption, on the rectal absorption of insulin in rats. The serum glucose and remained insulin level in perfusate were measured after rectal recirculation of insulin with or without sod. laurate, sod. caprate and sod. caprylate in situ. The addition of sod. laurate or sod. caprate reduced serum glucose concentration considerably. Sod. caprate (1.0%) showed the greatest promoting effect on the decrement of serum glucose. Eudispert hv hydrogels containing insulin with medium chain fatty acid salts were, thereby, prepared and evaluated. The release rate of insulin from Eudispert hv hydrogels was reduced with an increase in the content of Eudispert hv, and was raised with increasing NaOH concentration. Ten percent Eudispert hv hydrogels were offered for the rectal administration of insulin. The addition of 1.0% sod. caprate reduced serum glucose concentration remarkably after rectal administration of 10% Eudispert hv hydrogels containing insulin. The level of glucose decrement was greater by 30% compared to subcutaneous administration of insulin solution. From the above findings, Eudispert hv hydrogels would be used as useful rectal delivery systems of insulin.

Fast kinetics of transient pH changes and difference spectrum formation have been investigated following mixing of ADP/ATP with partially purified plasma membrane PM-ATPase of the pathogenic yeast Candida albicans in the presence of five nutrients: glucose, glutamic acid, proline, lysine, and arginine and two analogs of glucose: 2-deoxy D-glucose and xylose. Average $H^+$- absorption to release ratio, indicative of population of ATPase undergoing complete hydrolytic cycle, was found to be 0.27 for control. This ratio varied between 0.25 (proline) to 0.36 (arginine) for all other compounds tested, except for glucose. In the presence of glucose, $H^+$- absorption to release ratio was exceptionally high (0.92). While no UV difference spectrum was observed with ADP, mixing of ATP with ATPase led to a large conformational change. Exposure to different nutrients restricted the magnitude of the conformational change; the analogs of glucose were found to be ineffective. This suppression was maximal in the case of glucose (80%); with other nutrients, the magnitude of suppression ranged from 40-50%. Rate of $H^+$- absorption, which is indicative of E~P complex dissociation, showed positive correlation with suppression of conformational change only in the case of glucose and no other nutrient/analog. Mode of interaction of glucose with plasma membrane $H^+$-ATPase thus appears to be strikingly distinct compared to that of other nutrients/analogs tested. The results obtained lead us to propose a model for explaining glucose stimulation of plasma membrane $H^+$-ATPase activity.
This study was conducted to investigate the effects of Chinese medicine granule (CMG, including Cortex Phellodendron, Atractylodes Rhizome, Agastache Rugosa and Gypsum Fibrosum) on absorption and transport of glucose in porcine small intestinal brush border membrane vesicles (BBMVs) under heat stress. Forty-eight 2-month-old Chinese experimental barrows were screened according to weight and litter origin, and then allotted to three groups and treated as follows: Normal temperature control group (NTCG; $23^{\circ}C$), high temperature control group (HTCG; $26^{\circ}C$ for 19 h, $40^{\circ}C$ for 5 h); Chinese medicine granule anti-stress group (CMGG; $26^{\circ}C$ for 19 h, $40^{\circ}C$ for 5 h) (n = 16 per group). The results showed that high temperature treatment decreased (p<0.05) the growth performance and intestinal glucose absorption but there was no change (p>0.05) in the expression of SGLT1 and GLUT2 genes in the small intestine of pigs compared with the NTCG. Dietary supplementation with CMG improved the growth performance, and increased the activity of disaccharidases in duodenum and jejunum of heat stressed pigs (p<0.05). CMG treatment increased (p<0.05) the protein levels of SGLT1 and GLUT2 in the small intestine, and up-regulated (p<0.05) the expression of SGLT1 and GLUT2 genes in the duodenum and jejunum but without changing (p>0.05) them in the ileum compared with the HTCG. These results indicated that CMG treatment significantly improved porcine growth performance, and increased intestinal glucose absorption and transport by BBMVs under heat stress, in addition to up-regulating the expression of SGLT1 and GLUT2 genes in porcine duodenum and jejunum.

Among non-invasive blood glucose detection technologies, the optical technique is a method that uses light reflection, absorption, and scattering characteristics when passing through a biological medium. It reduces pain or discomfort in measurement and has no risk of infection. So it is becoming a major flow of blood glucose detection research. Among them, near-infrared spectroscopy has a disadvantage in that the complexity increases when analyzing signals detected due to interferences between proteins and acids that share a similar absorption function with blood glucose molecules. In this study, a non-invasive sensor system with multiple near-infrared bands was designed and manufactured to alleviate the deterioration of blood glucose detection function that may occur due to skin absorption of near-infrared rays. A blood survey was conducted to verify the system, and the degree of blood glucose response in the blood was collected as spectral data, and the results of this study were quantitatively verified in terms of correlation between the data and blood glucose.
The experiment was carried out to investigate the absorption profile of blood glucose in mice administered to silkworm powder MeOH extract. The mice was injected to oral load of maltose, sucrose and lactose(2 g/kg) and silkworm powder MeOH extract at the same time. After injecting the sample, blood glucose concentration was measured at 0, 30, 60, 120, 180 and 240 minutes. Blood glucose lowering effect of silkworm powder for loading maltose was 69% in postprandial 30 minutes. After the postprandial 60 minutes, the glucose was absorbed slowly. Total amount of blood glucose absorption in mice administered to maltose were 560.7 mg/dl during 240 minutes. That of silkworm powder MeOH extract marked 534.7 mg/dl. Total amount of blood glucose from oral loading sucrose reached to 508.9 mg/dl. That of loading silkworm powder MeOH extract were 468.8. But, silkworm powder was not inhibited lactose absorption. As a above results, silkworm powder inhibits the transient rising of blood glucose after postprandial 30 minuts through inhibition of ${\alpha}$-glucosidases. In case of starvation silkworm powder don't promote the hypoglycemia. In addition, silkworm powder induces the delay absorption of glucose without loss of it.
