군소 (Aplysia kurodai)의 중추신경계로부터 myomodulin A와 E의 정제 (Purification of Myomodulin A and Myomodulin E from the Central Nervous System of the Sea Hare, Aplysia kurodai)

  • 군소의 중추신경계로부터 myomodulin A (MMA)과 myomodlin E (MME)의 정제에 대해 보고하고자 한다. 500마리 군소의 중추신경계를 산조건으로 추출한 후 4종류의 HPLC column system에 적용하여 두가지 신경성 펩타이드를 정제하였다. HPLC 분리과정에서 얻어진 모든 분획의 활성을 조사하기 위하여 bioassay system으로 진주담치 (Mytilis edulis)의 ABRM을 이용하였다. 최종 정제된 신경성 펩타이드는 Edman 분해법을 이용한 아미노산 서열 분석기, 질량분석기와 화학합성을 통해 완전한 일차구조를 밝힐 수 있었다. 정제한 MMA와 MME의 일차구조는 각각 Pro-Met-Ser-Met-Leu-Arg-Leu-$NH_2$, (847.41 Da) 과 Gly-Leu-Gln-Met-Leu-Arg-Leu-$NH_2$, (830.50Da) 이었다. 또한 이들 합성 펩타이드는 진주담치의 ABRM에 대해 phasic contration의 조절활성을 나타내었다.

대장, 직장암 환자에서 화학방사선치료의 급성 부작용과 XRCC1 유전자 다형성과의 상관관계 (Relationship between XRCC1 Polymorphism and Acute Complication of Chemoradiation Therapy in the Patients with Colorectal Cancer)

  • 목적: 방사선치료와 항암제치료의 급성부작용은 환자 개인에 따라 차이가 많다는 것은 임상경험을 통하여 널리 알려져 있다. 그러나 아직 이를 미리 예측할 수 있는 인자로 알려진 것은 없다. XRCC1 유전자는 DNA base-excision repair에 관여하는 유전자로 알려져 있다 저자들은 대장 직장암 환자를 대상으로 방사선치료와 항암제치료로 인한 급성부작용과 XRCC1 유전자의 다형성이 관련이 있는지를 알아보고자 본 연구를 수행하게 되었다. 대상 및 방법: 1997년 7월부터 2003년 6월까지 인하대학교병원에서 치료를 받은 대장 직장암 환자 85명을 대상으로 하였다. 대장암이 2명, 5자결장암이 13명, 직장암이 71명이었다 병기는 B기가 22명, C기가 50명, D기가 8명이었고 절제 불가능한 경우가 6명이었다 방사선치료 범위는 골반강만 조사된 경우가 81명, extended field로 조사된 경우가 5명이었고 방사선량은 일일 1.8 Gy로 주 5회 조사하여 총 $30.6 Gy{\sim}59.4 Gy$ (중앙값: 54 Gy)를 조사하였다. 항암제치료는 전 환자에서 5FU를 근간으로 한 약제를 투여받았고 방사선 치료기간 중에 시행된 횟수는 1회가 24명, 2회가 45명이었고 17명은 동시에 투여 받지는 않았다. 치료의 급성부작용은 상부위장관과 하부위장관으로 나누어 기록하였고 증상이 전혀 없는 경우를 0, 증상이 있으나 투약이 필요하지 않은 경우를 1, 투약이 필요한 경우를 2, 투약에도 불구하고 증상이 심하여 치료의 휴식 또는 입원을 한 경우를 3으로 분류하였고 전 치료기간 중 최초, 최저 수치와 방사선치료기간 중 최초, 최저 백혈구, 혈소판 수치를 조사하였다. 환자의 동의 하에 혈액을 채취하여 림프구를 분리한 후 DNA를 추출하여 PCR-RFLP 방법으로 XRCC1 유전자의 코돈 194, 280, 399번 위치의 다형성을 분석하였다. 통계는 Chi-square, t-test, logistic regression, ANOVA를 사용하였다. 결과: 다변량 분석결과 상부위장관 부작용에 영향을 미치는 인자는 재발유무였고, 하부위장관의 부작용에 영향을 미치는 인자는 XRCC1 339 다형성, 방사선량, 방사선 중 항암제횟수 순이었다. 방사선 치료 중 백혈구 감소에 영향을 미치는 인자는 XRCC1 399 다형성, 194 다형성이었고, 혈소판 감소에 영향을 미치는 인자는 진단명, XRCC1 399 다형성이었다. 결론: 대장 직장암 환자에서 방사선치료와 항암제의 치료에 따른 정상조직의 급성부작용을 예측하는데 XRCC1 유전자의 코돈 399번의 다형성이 사용될 가능성이 있을 것으로 생각된다.

Amino Acid, Amino Acid Metabolite, and GABA Content of Three Domestic Tomato Varieties

  • To determine the nutritional value of domestic tomatoes, the levels of amino acids, amino acid metabolites, and the bioactive compound ${\gamma}-aminobutyric-acid$ (GABA) were analyzed in three domestic tomato varieties (Rafito, Momotaro, and Medison). Eighteen free amino acids were found, and total free amino acid content was 3,810.21~4,594.56 mg/100 g (dry weight). L-glutamic acid (L-Glu) was the most abundant amino acid, ranging from 1,866.60 mg/100 g for Momotaro to 2,417.45 mg/100 g for Medison. The next most abundant amino acids were L-glutamine (L-Gln) and L-aspartic acid (L-Asp). The three tomato varieties had a good balance of all the essential amino acids except tryptophan. Total essential amino acid content was 274.26~472.71 mg/100 g (dry weight). The following amino acid metabolites were found: L-carnitine (L-Car), hydroxylysine (Hyl), o-phosphoethanolamine (o-Pea), phosphoserine (p-Ser), ${\beta}-alanine$ (${\beta}-Ala$), N-methyl-histidine (Me-His), ethanolamine (EtNH2),and L-citrulline(L-Cit). Large quantities of GABA were found in all three varieties: 666.95-868.48 mg/100g (dry weight). These results support the use of these tomato varieties as nutritious food materials.

A DPL (DNA/peptide/liposomes) Tripartite Complex Effective for Transfection in Serum

  • A short peptide corresponding to the nuclear localization signal (NLS) of human immunodeficiency virus (HIV)-l Tat protein, Arg-Lys-Lys-Arg-Arg-Gln-Arg-Arg-Arg, was employed to improve the efficiency of cellular uptake of nucleic acids. The peptide was first mixed with a reporter plasmid and then with cationic liposomes to form a tripartite complex of DNA/peptide/liposomes (DPL). Transfection efficiency of the DPL complex was compared with that of the conventional DNA/liposomes (DL) complex. When the DPL complex was formed with various cationic liposomes, DOTAP/DOPE (DP) liposome exhibited superior transfection efficiency to other liposomes tested in vitro. With the inclusion of the peptide, the DPL complex showed much enhanced transfection in various cancer cell lines. Particularly, transfection of the DPL complex in serum increased cellular uptake of a transgene up to 2 fold when compared with that in a serum free condition. Further, when the DPL complex was infused through the ureteric route of a rat, transfection efficiency was shown to be better in reporter gene expression than that obtained with the DL complex. This study shows that the DPL complex that is easy to formulate can be employed for much enhanced cellular uptake of a trans gene.

  • Antibodies were raised against fusion proteins of the N-terminus and a region containing the GDNQ (Gly-Asp-Asn-Gln) polymerase motif of the L (polymerase) protein of sonchus yellow net virus (SYNV). Immunoblot analyses using these antibodies revealed the presence of the L protein in purified SYNV preparations and in nuclear extracts from infected tobacco. The serological analyses and detection in a polyacrylamide gels suggested that the L protein is present in at least a 20 fold lower abundance than the G, N, M1 and M2 proteins, and has size corresponding to a molecular weight of over 200 kDa as predicted from nucleotide sequence data. Electron microscopy with gold-labelled antibodies was used to localize the N, M2, and G proteins of SYNV in thin sections of infected tissue. When sections of SYNV-infected tissue were treated with antisera against total SYNV proteins and N protein, gold label could be detected in both the viroplasms and in virus particles. With the anti-M2 protein antiserum, the gold label was strongly localized in the viroplasms but only limited labelling of the virus particle sonly. Limited labelling of the L protein was observed in the viroplasms and the virus particles, presumably because of the low abundance of L protein in the tissues.

A Mutagenic Study of β-1,4-Galactosyltransferases from Neisseria meningitidis

  • N-terminal His-tagged recombinant $\beta$-1,4-galactosyltransferase from Neisseria meningitidis was expressed and purified to homogeneity by column chromatography using Ni-NTA resin. Mutations were introduced to investigate the roles of, Ser68, His69, Glu88, Asp90, and Tyr156, which are components of a highly conserved region in recombinant $\beta$-1,4 galactosyltransferase. Also, the functions of three other cysteine residues, Cys65, Cys139, and Cys205, were investigated using site-directed mutagenesis to determine the location of the disulfide bond and the role of the sulfhydryl groups. Purified mutant galactosyltransferases, His69Phe, Glu88Gln and Asp90Asn completely shut down wild-type galactosyltransferase activity (1-3%). Also, Ser68Ala showed much lower activity than wild-type galactosyltransferase (19%). However, only the substitution of Tyr156Phe resulted in a slight reduction in galactosyltransferase activity (90%). The enzyme was found to remain active when the cysteine residues at positions 139 and 205 were replaced separately with serine. However, enzyme reactivity was found to be markedly reduced when Cys65 was replaced with serine (27%). These results indicate that conserved amino acids such as Cys65, Ser68, His69, Glu88, and Asp90 may be involved in the binding of substrates or in the catalysis of the galactosyltransferase reaction.

Klebsiella pneumoniae가 생산하는 Glutamine synthetase의 정제 및 특성 (Purification and Characterization of Glutamine synthetase of Klebsiella pneumoniae)

  • K.pneumoniae의 nif 유전자 발현에 미치는 glnA 산물 (GS)의 영향을 살펴보기 앞서 K.pneumoniae의 GS를 분리.정제하여 그 성질을 조사하였다. K.pneumoniae의 GS는 분자량 약 600,000의 12개 동일 subunit으로 구성되어져 이었으며 최적 반응 pH 및 온도는 각기 pH 7.0, $37^{\circ}C$ 였고 pH 5-8사이에서 활성의 변화가 크지 않았으며 $57^{\circ}C$에서 10분간 열처리 할 때 50의 활성 감소가 나타났다. 또 암모니아 농도의 변화에 따라 adenylylation-deadenylylation 기작에 의한 활성조절기작이 E.coli GS와 동일 했으며 anti-E.coli GS 항체를 이용한 immunodiffusion에서도 K.pneumoniae GS가 E.coli GS와 혈청적으로 매우 유사함이 드러났다.

Characterization of New Antihypertensive Angiotensin I-Converting Enzyme Inhibitory Peptides from Korean Traditional Rice Wine

  • This study describes the characterization of a new angiotensin I-converting enzyme (ACE) inhibitory peptide from a Korean traditional rice wine. After purification of the ACE inhibitor peptides with ultrafiltration, Sephadex G-25 column chromatography, and successively $C_{18}$ and SCX solid-phase extraction, reverse-phase HPLC, and size exculsion chromatography, two types of the purified ACE inhibitors with $IC_{50}$ values of 0.34 mg/ml and 1.23 mg/ml were finally obtained. The two purified ACE inhibitors (F-1 and F-2) were found to have two kinds of novel oligopeptides, showing very little similarity to other ACE inhibitory peptide sequences. The amino acid sequences of the two purified oligopeptides were found to be Gln-Phe-Tyr-Ala-Val (F-1) and Ala-Gly-Pro-Val-Leu-Leu (F-2), and their molecular masses were estimated to be 468.7 Da (F-1) and 357.7 Da (F-2), respectively. They all showed a clear antihypertensive effect on spontaneously hypertensive rats at a dosage of 500 mg/kg.

An Active Site Arginine Residue in Tobacco Acetolactate Synthase

  • Acetolatate synthase(ALS) catalyzes the first common step in the biosynthesis of valine, leucine, isoleucine in plants and microorganisms. ALS is the target of several classes of herbicides, including the sulfonylureas, the imidazolinones, and the triazolopyrimidines. To elucidate the roles of arginine residues in tobacco ALS, chemical modification and site-directed mutagenesis were performed. Recombinant tobacco ALS was expressed in E. coli and purified to homogeneity. The ALS was inactivated by arginine specific reagents, phenylglyoxal and 2,3-butanedione. The rate of inactivation was a function of the concentration of modifier. The inactivation by butanedione was enhanced by borate, and the inactivation was reversible on removal of excess butanedione and borate. The substrate pyruvate and competitive inhibitors fluoropyruvate and phenylpyruvate protected the enzyme against inactivation by both modifiers. The mutation of well-conserved Arg198 of the ALS by Gln abolished the enzymatic activity as well as the binding affinity for cofactor FAD. However, the mutation of R198K did not affect significantly the binding of FAD to the enzyme. Taken together, the results imply that Arg198 is essential for the catalytic activity of the ALS and involved in the binding of FAD, and that the positive charge of the Arg is crucial for the interaction with negatively charged FAD.

A novel de novo mosaic mutation in PHEX in a Korean patient with hypophosphatemic rickets

  • X-linked hypophosphatemic rickets is caused by loss-of-function mutations in PHEX, which encodes a phosphate-regulating endopeptidase homolog. We report a 26-year-old man with X-linked hypophosphatemic rickets who showed decreased serum phosphate accompanied by bilateral genu valgum and short stature. He had received medical treatment with vitamin D (alfacalcidol) and phosphate from the age of 3 to 20 years. He underwent surgery due to valgus deformity at the age of 14 and 15. Targeted gene panel sequencing for Mendelian genes identified a nonsense mutation in PHEX (c.589C>T; p.Gln197Ter) and a mosaic pattern where only 38% of sequence reads showed the variant allele. This mutation was not found in his mother, who had a normal phenotype. This is a case of a sporadic nonsense mutation in PHEX and up to date, this is the first case of a mosaic mutation in PHEX in Korea.
