Camphor Inhibits Adipocyte Differentiation via Its Impact on SMO-dependent Regulation of Hedgehog Signaling (Camphor의 Hedgehog 신호 SMO 조절을 통한 지방구세포 분화 억제효과)

  • In this study, we examined inhibition of adipocyte differentiation associated with the administration of camphor, a substance identified in extracts of the flowering plant Chrysanthemum indicum L. (CI). No camphor-mediated cytotoxicity was observed over a period of 1-10 days in studies targeting cells of the 3T3-L1 adipocyte-like line. Experiments that featured siRNA-mediated suppression of the transmembrane proteins Patched (PTCH) and Smoothened (SMO) resulted in inhibition and activation of differentiation, respectively. Interestingly, inhibition of PTCH typically activates SMO protein targeting and serves to activate hedgehog (HH)-mediated signaling. The results of our study suggest that activation of HH-mediated signaling can inhibit adipocyte differentiation. Furthermore, expression of glioma-associated oncogene homologue 1 (Gli1) was detected by flow cytometry in 62.7±1.5% of cells in response to administration of Lactobacillus rhamnosus (KCTC 3237) and in 60.4±2.2% of cells in response to camphor; these levels are higher than those detected in undifferentiated controls (24.9±3.1%). No change in the state of fermented camphor was identified by gas chromatography-mass spectrometry (GC-MS), but a 15.41% quantitative increase was confirmed in KCTC 3237. Overall, we conclude that administration of camphor resulted in overexpression of SMO and modulated the differential expression of Gli1. Animal studies focused on the impact of camphor as an agent to counteract obesity might be considered in the future. Indeed, camphor and similar physiologically active compounds from fermented CI might be developed as new and effective treatments for obesity.

  • The Third Assessment Report of the Intergovernmental Panel on Climate Change (IPCC) concluded that many collective observations gave a aspect of a global warming and other changes in the climate system. Future earth observation using satellite data should monitor global climate change, and should contribute to social benefits. Especially, human activities has given the big impacts to earth environment This is a very complex affair, and nature itself also impacts the clouds, namely the seasonal variations. JAXA (former NASDA) has the plan of the Global Change Observation Mission (GCOM) for monitoring of global environmental change. SGLI (Second Generation GLI) onboard GCOM-C (Climate) satellite, which is one of this mission, is an optical sensor from Near-UV to TIR. This sensor is the GLI follow-on sensor, which has the various new characteristics. Polarized/multi-directional channels and 250m resolution channels are the unique characteristics on this sensor. This sensor can be contributed to clarification of coastal change in sea surface. This paper shows the introduction of the unique aspects and characteristics of the next generation satellite sensor, SGLIIGCOM-C, and shows the preliminary research for this sensor.

  • Release of hazardous and flammable gas is a significant contributor to risk. The ignition of flammable gas clouds can lead to explosion accidents in the offshore installations. A gas detector, which is one of active protect systems, brings the module into a safe state through emergency shut down processes and reduces the damage by eliminating the dangerous releases. It is critical to understand the gas release, the wind field, and the complex geometry of installations to determine gas detector placement. In this paper, the Gas detector Location Index (GLI) which is a novel index for optimal detector location determination to efficiently prevent explosion accident using probabilistic approach.

Inhibitory Effect of Lactic Acid Bacteria-fermented Chrysanthemum indicum L. on Adipocyte Differentiation through Hedgehog Signaling (감국의 유산균 발효물이 hedgehog 신호를 통한 지방구세포 분화 억제효과)

  • In this study, we describe the inhibition of adipocyte differentiation by the lactic acid bacteria (LAB) fermentation product of Chrysanthemum indicum L. (CI) extract to control obesity. Preparation of LAB-fermented products was performed to overcome the cytotoxicity of CI extract. During fermentation and 3T3-L1 cell line experiment, cytotoxicity was not induced in the CI fermentation products over 1 day in culture. Fermented materials from highly proliferative cultures were selected for treatment of 3T3-L1 cells and for comparison with unfermented control groups. Cell survival and undifferentiated cell populations were decreased differentiation population in all experimental groups compared with controls, as measured using fluorescence-activated cell sorting analysis. Akt pathway activity increased upon treatment with these fermented extracts in 3T3-L1 cells. Gli2 depleted at the protein level in association with adipocyte differentiation. LAB KCTC 3115- and 3109-fermented extract treatment caused controlled Gli2 protein accumulation. Moreover, KCTC 3115 and 3109 were found to reduce C/EBPα and FAS was depleted, whereas pACC was increased at the protein level upon treatment with the fermentation products of each of the four LAB used in this study. With Lactococcus lactis subsp. lactis KCTC 3115 fermentation, the regulation of adipose differentiation and hedgehog signaling were also suppressed, thereby inhibiting the differentiation of progenitor cells. The basis for the activation of hedgehog signaling may provide insights into the treatment of obesity and the inhibition of adipocyte differentiation.

Increased Primary Cilia in Idiopathic Pulmonary Fibrosis

  • Primary cilia are solitary, non-motile, axonemal microtubule-based antenna-like organelles that project from the plasma membrane of most mammalian cells and are implicated in transducing hedgehog signals during development. It was recently proposed that aberrant SHH signaling may be implicated in the progression of idiopathic pulmonary fibrosis (IPF). However, the distribution and role of primary cilia in IPF remains unclear. Here, we clearly observed the primary cilia in alveolar epithelial cells, fibroblasts, and endothelial cells of human normal lung tissue. Then, we investigated the distribution of primary cilia in human IPF tissue samples using immunofluorescence. Tissues from six IPF cases showed an increase in the number of primary cilia in alveolar cells and fibroblasts. In addition, we observed an increase in ciliogenesis related genes such as IFT20 and IFT88 in IPF. Since major components of the SHH signaling pathway are known to be localized in primary cilia, we quantified the mRNA expression of the SHH signaling components using qRT-PCR in both IPF and control lung. mRNA levels of SHH, the coreceptor SMO, and the transcription factors GLI1 and GLI2 were upregulated in IPF compared with control. Furthermore, the nuclear localization of GLI1 was observed mainly in alveolar epithelia and fibroblasts. In addition, we showed that defective KIF3A-mediated ciliary loss in human type II alveolar epithelial cell lines leads to disruption of SHH signaling. These results indicate that a significant increase in the number of primary cilia in IPF contributes to the upregulation of SHH signals.

  • Game Rating Board(GRB) of Korea set up in 29 October, 2006 has been 7th years and it will disappear in 23 November, 2013. For the first time the 'GRB' of Korea has the goal which held the nonofficial transfer of rating classification for game. But it had the justification which become more distant from the nonofficial transfer due to the affairs of 'See Story' in August, 2006. As a result, because of rapid normalization of game rating rather than the systematic transfer from 'Video Rating Board' of Korea cause no preparation of scientific professional rating review. It showed the entry-level appearance though stressing profession from the beginning of setup. Specially more transaction expense were increased because of no consistency and predictability for game review. However, it could be overcome by preparation for definite global standard and scientific rating system. Consequently technological possibility of BMM Korea based on 'Gaming Laboratory Institute(GLI)' standard expected to be alternative in the technical review of the adult arcade game.

Primary Cilia, A Novel Bio-target to Regulate Skin Pigmentation (바이오 안테나인 일차 섬모 조절을 통한 피부 미백 기술)

  • The primary cilium protrudes from the cell body like a bio-antenna that has many receptors, channels and signaling molecules to sense and response to external stimuli. The external environment such as ultraviolet irradiation, temperature, humidity, gravity and shear stress always influences skin. Skin responds to external stimuli and differentiates by making melanin, collagen and horny layer. Ciliogenesis participates in developmental processes of skin, such as keratinocyte differentiation and hair formation. And it was reported that skin pigmentation was inhibited when ciliogenesis was induced by sonic hedgehog-smoothened-GLI2 signaling. When skin is exposed to ultraviolet irradiation, alpha-melanocyte stimulating hormones (${\alpha}$-MSH) increase melanin synthesis through activation of the cAMP pathway in melanocytes. We observed that ${\alpha}$-MSH and cAMP production inducers inhibited ciliogenesis of melanocytes. Therefore, we thought that regulation of ciliogenesis is potential candidate target for the development of agents to treat undesirable hyperpigmentation of skin. As a result, we found out that an ethanol extract of Glycyrrhiza glabra (EGG) root and 3,4,5-trimethoxy cinnamate thymol ester (TCTE, Melasolv) significantly inhibit melanin synthesis of normal human melanocyte by inducing primary cilium formation. This study proposed new theory to regulate skin pigmentation and cosmetic components for skin whitening.

Identification of growth trait related genes in a Yorkshire purebred pig population by genome-wide association studies

  • Objective: The aim of this study is to identify genomic regions or genes controlling growth traits in pigs. Methods: Using a panel of 54,148 single nucleotide polymorphisms (SNPs), we performed a genome-wide Association (GWA) study in 562 pure Yorshire pigs with four growth traits: average daily gain from 30 kg to 100 kg or 115 kg, and days to 100 kg or 115 kg. Fixed and random model Circulating Probability Unification method was used to identify the associations between 54,148 SNPs and these four traits. SNP annotations were performed through the Sus scrofa data set from Ensembl. Bioinformatics analysis, including gene ontology analysis, pathway analysis and network analysis, was used to identify the candidate genes. Results: We detected 6 significant and 12 suggestive SNPs, and identified 9 candidate genes in close proximity to them (suppressor of glucose by autophagy [SOGA1], R-Spondin 2 [RSPO2], mitogen activated protein kinase kinase 6 [MAP2K6], phospholipase C beta 1 [PLCB1], rho GTPASE activating protein 24 [ARHGAP24], cytoplasmic polyadenylation element binding protein 4 [CPEB4], GLI family zinc finger 2 [GLI2], neuronal tyrosine-phosphorylated phosphoinositide-3-kinase adaptor 2 [NYAP2], and zinc finger protein multitype 2 [ZFPM2]). Gene ontology analysis and literature mining indicated that the candidate genes are involved in bone, muscle, fat, and lung development. Pathway analysis revealed that PLCB1 and MAP2K6 participate in the gonadotropin signaling pathway and suggests that these two genes contribute to growth at the onset of puberty. Conclusion: Our results provide new clues for understanding the genetic mechanisms underlying growth traits, and may help improve these traits in future breeding programs.

Establishment of a Pancreatic Cancer Stem Cell Model Using the SW1990 Human Pancreatic Cancer Cell Line in Nude Mice

  • Aim: To establish a pancreatic cancer stem cell model using human pancreatic cancer cells in nude mice to provide a platform for pancreatic cancer stem cell research. Materials and Methods: To establish pancreatic cancer xenografts using human pancreatic cancer cell line SW1990, nude mice were randomly divided into control and gemcitabine groups. When the tumor grew to a volume of $125mm^3$, they treated with gemcitabine at a dose of 50mg/kg by intraperitoneal injection of 0.2ml in the gemcitabine group, while the mice in control group were treated with the same volume of normal saline. Gemcitabine was given 2 times a week for 3 times. When the model was established, the proliferation of pancreatic cancer stem cells was observed by clone formation assay, and the protein and/or mRNA expression of pancreatic stem cell surface markers including CD24, CD44, CD133, ALDH, transcription factors containing Oct-4, Sox-2, Nanog and Gli, the key nuclear transcription factor in Sonic Hedgehog signaling pathway was detected by Western blot and/or RT-PCR to verify the reliability of this model. Results: This model is feasible and safe. During the establishment, no mice died and the weight of nude mice maintained above 16.5g. The clone forming ability in gemcitabine group was stronger than that of the control group (p<0.01). In gemcitabine group, the protein expression of pancreatic cancer stem cell surface markers including CD44, and ALDH was up-regulated, the protein and mRNA expression of nuclear transcription factor including Oct-4, Sox-2 and Nanog was also significantly increased (P<0.01). In addition, the protein expression of key nuclear transcription factor in Sonic Hedgehog signaling pathway, Gli-1, was significantly enhanced (p<0.01). Conclusions: The pancreatic cancer stem cell model was successfully established using human pancreatic cancer cell line SW1990 in nude mice. Gemcitabine could enrich pancreatic cancer stem cells, simultaneously accompanied by the activation of Sonic Hedgehog signaling pathway.

  • Unmanned Aerial Vehicle (UAV) imagery are being assessed for analyzing within field spatial variability for agricultural precision management, because UAV imagery may be acquired quickly during critical periods of rapid crop growth. This study refers to the derivation of barley and wheat growth prediction equation by using UAV derived vegetation index. UAV imagery was taken on the test plots six times from late February to late June during the barley and wheat growing season. The field spectral reflectance during growing period for the 5 variety (Keunal-bori, Huinchalssal-bori, Saechalssal-bori, Keumkang and Jopum) were measured using ground spectroradiometer and three growth parameters, including plant height, shoot dry weight and number of tiller were investigated for each ground survey. Among the 6 Vegetation Indices (VI), the RVI, NDVI, NGRDI and GLI between measured and image derived showed high relationship with the coefficient of determination respectively. Using the field investigation data, the vegetation indices regression curves were derived, and the growth parameters were tried to compare with the VIs value.
