Glehnia littoralis is known as an edible and medicinal plant using green loaves and mature roots of plant. In the present paper, the influence of plant growth regulators on callus induction and plant regeneration was investigated. Callus induction and regeneration occurred from leaf and petiole explants in Glehnia littoralis. Optimal condition of plant growth regulators for callus induction from leaf and petiole explants was MS basal medium supplemented with 2mg/L 2,4-D and 2mg/L BA. The frequency of callus induction was higher in petiole explant than leaf. When the callus was cultured on MS basal medium supplemented with 0∼1 mg/L IAA, 0∼1mg/L NAA and 0∼2mg/L BA for about 65 days, the most effective plant growth regulators on plant regeneration from callus were 1mg/L NAA and 2mg/L BA. The plantlets acclimatized successfully and grown in vermiculite matrix.

Glehnia littoralis Fr. Schmidt (Umbelliferae) is a short-lived herbaceous species that are mostly distributed throughout East Asia. Although G. littoralis has been regarded as ecologically important one, there is no report on population structure in Korea. Starch gel electrophoresis was used to investigate the allozyme variation and genetic structure of Korean populations of this species. A high level of genetic variation was found in G. littoral is populations. Nine enzymes revealed 18 loci, of which 12 were polymorphic (66.7%). Genetic diversity at the species and population levels were 0.159 and 0.129, respectively. The sexual and asexual reproduction, high fecundity, and colonization process are proposed as possible factors contributing to genetic diversity. An indirect estimate of the number of migrants per generation (Nm = 1.45) indicated that gene flow was not extensive among Korean populations of this species. It is suggested that the ability of vegetation and artificial selection may have played roles in shaping the population structure of this species. we recommend that a desirable conservation population should be included at least 30 plants per population and especially those with high variation.
To develop a new natural antibiotics from Koran plant resources for dealing with the current situation regarding the rapid increase in antibiotic-resistant pathogen, the in vitro inhibitory activities of essential oils from the young leaves of Glehnia littoralis (Umbelliferae) as well as its main constituents were evaluated against susceptible and resistant species of Streptococcus pneumoniae. The essential oil fraction of G. littoralis and its main components, $\alpha-and\;\beta-pinene$, exhibited significant inhibitory activities against the antibiotic-susceptible and resistant strains of S. pneumoniae, with MICs (minimum inhibiting concentrations) ranging from 4.0mg/ml to 16mg/ml. No remarkable differences were shown between the susceptible and resistant strains. Moreover, the disk diffusion test disclosed that these inhibitory activities were dosedependent. Furthermore, data from the checkerboard titer test with FICIs (fractional inhibiting concentration indices) from 0.15 to 0.28 indicated synergisms between norfloxacin and $\alpha-or\;{\beta}-pinene$ in activity against S. pneumoniae KCCM49629 and S. pneumoniae CCARM4059.
Glehnia littoralis Fr. Schmidt et Miquel is an important medicinal plants in East Asian countries. This plant species naturally distributed in Korea, Japan, China, and Taiwan, but it is a rare plants living in the coastal dune in Korea. To investigate the genetic variation, genetic diversity and genetic evolutionary relationships of 14 different geographical G. littoralis, ITS sequence and random amplified polymorphic DNA (RAPD) were analyzed. On the basis of ITS sequences, it was clearly showed that the ITS1 and ITS2 sequences among 14 populations are identical regardless of geographical origin excepting 2 bp in pair-wise comparison of ITS1. Furthermore, RAPD results also showed that 14 different geographical G. littoralis produce various polymorphic patterns without critical relationship among neighboring regions. These combined results suggest that the geographical variation and genetic evolution of G. littoralis is stable and provide important information on genetic diversity, and conservation of this rare plant species in situ and ex situ.
The experiment was carried out to clarify the effects of air temperature, light exposure and UV light on the growth and anthocyanin syntheses of Glehnia littoralis Fr. Schm. The major pigment of purfied aglycone of Glehnia littoralis Fr. Schm. was identified as cyanidin by TLC and HPLC analysis. The anthocyanin Syntheses was promoted at low temperature and in the treatment of light exposure (12hrs a day) for 4-6 days after shading. The plants grown under UV-cut film had less anthocyanin content those tose under UV-transmission film.

Glehnia littoralis (GL) is widely used as an oriental medicine for cough, fever, stroke and other disease conditions. However, the anti-cancer properties of GL on MCF-7 human breast cancer cells have not been investigated. In order to elucidate anti-cancer properties and underlying cell death mechanisms, MCF-7cells ($5{\times}10^4/well$) were treated with Glehnia littoralis root extract at 0-400 ug/ml. A hot water extract of GL root inhibited the proliferation of MCF-7 cells in a dose-dependent manner. Analysis of the cell cycle after treatment of MCF-7 cells with increasing concentrations of GL root extract for 24 hours showed significant cell cycle arrest in the G1 phase. RT-PCR and Western blot analysis both revealed that GL root extract significantly increased the expression of p21 and p27 with an accompanying decrease in both CDK4 and cyclin D1. Our reuslts indicated that GL root extract arrested the proliferation of MCF-7 cells in G1 phase through inhibition of CDK4 and cyclin D1 via increased induction of p21 and p27. In summary, the current study showed that GL could serve as a potential source of chemotherapeutic or chemopreventative agents against human breast cancer.

This study was carried out to investigate the growth responses of medicinal plants such as Glehnia littoralis, Cryptotaenia japonica, Angelica gigas, Reynoutria japonica, Saururus chinensis, Houttuynia cordata, Ligularia fischeri, and Aralia cordata under 70%-shading and full sunlight conditions on a rooftop condition using artificial soil as a growth media. Glehnia littoralis, Cryptotaenia japonica, Ligularia fischeri, Saururus chinensis, and Houttuynia cordata showed better growth on plant height, leaf length, leaf width, petiole length and leaf number under full sunlight conditions on a rooftop condition. Cryptotaenia japonica and Houttuynia cordata had high value of Hunter's a (red-green) under full sunlight, while had lower value of L(lightness) and b(blue-yellow) than those of 70% shading condition. As an index of plant stress response, Glehnia littoralis, Reynoutria japonica, Houttuynia cordata, Ligularia fischeri showed Fv/Fm values 0.79~0.84 under full sunlight in August, indicating low stress on plant growth. Therefore they seemed to be suitable medicinal plats for rooftop conditions. Ligularia fischeri and Reynoutria japonica showed better growth under 70% shading treatment in August, while showed high growth response under full sunlight conditions in September. The 70% shading treatment was effective for the growth of Angelica gigas, Reynoutria japonica, and Aralia cordata. Angelica gigas and Reynoutria japonica, however, showed better plant growth under full sunlight during summer. The results showed that Glehnia littoralis, Cryptotaenia japonica, Ligularia fischeri, Saururus chinensis, and Houttuynia cordat seemed to be suitable medicinal plants for rooftop garden where there is a full sunlight condition.
Petiole explant of Glehnia littoralis Fr. Schmidt was in vitro cultured MS plant medium(DUCHEFA co.) supplemented with various plant growth regulators and examined to find out their optimum combination and concentration for plantlet regeneration. We investigated optimal condition for efficient plant regeneration through adventitious shoot formation on MS plant medium with various kinds of plant growth regulators. Embryogenic calli and adventitious shoot formation were greatly influenced by plant growth regulators. Embryogenic calli induction showed a good response on MS medium supplemented with NAA and BA than others. Especially, combination of 1.0 mg/L NAA and 0.5 mg/L BA on MS medium led to the greatest frequency in adventitious shoot. The results suggest that plant regulator selection be important factor to achieve an efficient regeneration.

This study was conducted to compare the contents of fatty acids and phytosterols in Glehnia littoralis according to habitat areas. In leaves of G. littoralis collected from Yeong-deok, fatty acids contents was higher than that of the other aeras, while there was no difference in contents of roots and seeds according to habitat areas. Average phytosterols contents of 10 habitat areas were $3.8{\mu}g\;g^{-1}$ (campestreol) $16.8{\mu}g\;g^{-1}$ (stigmasterol) and $3.7{\mu}g\;g^{-1}$ ($\beta$-sitosterol), respectively. G. littoralis collected from Uljin showed the highest phytosterols contents than that of the other aeras.
Background: The present study was conducted to investigate the fatty acid content and habitat conditions of Glehnia littoralis in the Korean midwest region. Methods and Results: The emergence area of G. littoralis was 36.1 m from the coastline at an elevation of 4.2 m and slope of 4.3%. The soil of the habitat was sandy, the pH was 8.4, organic matter content was 0.4%, $P_2O_5$ content was 9.1 mg/kg and the content of K, Ca, Mg and Na were 0.09, 9.31, 0.43 and $0.23cmol^+/kg$, respectively. Other plants growing in this area with G. littoralis included 16 taxa. Plant with high coverage and frequency were Carex kobomugi, Elymus mollis, Imperata cylindrica var. koenigii, Lathyrus japonicus, Calystegia soldanella and Carex pumila. In the seeds of G. littoralis, the maximum fatty acid content was 65.5% oleic acid, 28.7% linoleic acid, 4.3% palmitic acid, 1.5% stearic acid, and 0.2% linolenic acid. Conclusions: In G. littoralis of Korean midwest, the scope of collection and cultivation for medicinal purposes was broad, because differences in associated vegetation, soil composition, and fatty acid content owing to habitat conditions was low.
