Effect of Saponin and Non-saponin of Panax Ginseng on the Blood Pressure in the Renovascular Hypertensive Rats (신성고혈압백서에서 혈압에 미치는 고려홍삼사포닌과 비사포닌의 효과)

  • The effect of saponin and non-saponin of Panax Red Ginseng on the blood pressure and nitric oxide production were investigated in the conscious free moving one-kidney, one-clip Goldbaltt hypertensive (lK, 1C-GBH) rats. Mean blood pressure in the control and lK, 1C-GBH rats was decreased by the administration of ginseng saponin (100 mg/kg, i.v.). The hypotensive effect induced by ginseng saponin was reached maximum at 2-4 minutes and was slowly recovered to the initial level of blood pressure. Also ginseng saponin induced reflex tachycardia in the conscious both rats. Contrast to the response induced by ginseng saponin, hypotensive effect induced by non-saponin of panax ginseng is minimal. Plasma nitric oxide concentration was increased by the treatment of ginseng saponin (100 mg/kg, i.p for 5 days) in both rats. It has been shown by western blotting that the expression level of the protein for endothelial nitric oxide synthase (eNOS) in the aorta of rats was not increased by the treatment of ginseng saponin (100 mg/kg, i.p). However, eNOS activity in aortic homogenates of both rats were increased by the treatment of ginseng saponins. From the above results, the hypotensive effect of saponin was greater than that of non-saponin of Panax Red Ginseng. The lowering effect of blood pressure by ginseng saponin may be due to the increase of plasma nitric oxide concentration via the increase of endothelial nitric oxide synthase activity in the renovascular hypertensive and control rats.

  • To investigate the effect of ginseng extracts on the changes of flllidity and lysis of liposome reverse phase evaporation vesicle (REV) was employed as model membrane and methylene blue was used as photosensitizer. Fluoresence polarization (P vaule) that represented fluidity of liposome was increased by photooxidation. All of the ginseng saponin inhibited the increasing rate of P value; the order of effect was ginseng water extract>biol saponin>triol saponin>crude saponin. In trapped G-6-P% measurement for lysis of liposome, ginseng water extract and crude saponin promoted the lysis of liposome. Therefore, we thought that ginseng extracts acted as both antioxidant and detergent.

Content and Composition of Saponin Compounds of Panax Species (Panax(인삼)속 식물의 사포닌화합물 함량 및 조성)

  • The content and composition of saponin compounds of Panax species were analyzed according to their species, region and processing type of red and white ginseng. The species employed were Korean-, Chinese-, Japanese red ginsengs, and Korean white ginseng of Panax ginseng, American- and Canadian ginsengs of Panax quinquefolium, and Panax notoinseng. Twelve main saponin components in the ginseng were identified and quantified using TLC and HPLC. All three species had remarkably different content and composition. However, within each species they were similar. Twelve major ginsenosides were determined in P. ginseng, eight in p. quinquefolium, and six in P. notoginseng. Of the components of P ginseng Rf, $Rh_1$, $Rh_2$ and Ra were not detected in P quinquefolium, and $Rb_2$, Rc, Rf, $Rh_2$, Ra and Ro not detected in P. notoinseam. Crude saponin content and protopanaxadiol/protopanaxatriol saponin ratio were compared. They were 4.81~5.24% and 1.27~ 1.45 in p. ginsengs, 7.01~7.25% and 2.12~ 2.15 in p. quinquefolium, 9.80% and 0.99 in P. notoineng. The prosapogenin and sapogenin content were different among the Panax species.

  • The effect of ginseng saponin on aflatoxin(AF) production by Aspergillus parasiticus NRRL2999 and the mutagenicity of produced aflatoxin. The production of aflatoxin were decreased by the addition of ginseng saponin and the most effective concentration was 0.05%. The ratio of aflatoxin $B_{1}$ and aflatoxin $G_{1}$ were not changed by the addition of ginseng saponin. For the nutagenicity test, Ames method were adopted. Mutagenicity of mycelial aflatoxin was decreased by the addition of ginseng saponin on TA98, but not changed on TA100. Mutagenicity of excreted aflatoxin to broth was slightly increased by the saponin on TA98, but decreased on TA100.

  • Crude saponin and ginsenosides in commercial white ginsengs such as six kinds of whole ginsengs, five kinds of tail roots and one undergrade raw ginseng were analyzed. The contents of crude saponin and ginsenosides in whole ginsengs were found to be 2.7 to 4.6% and 1.0 to 1.7%, respectively. On the other hand, those of tail roots were 3.3 to 7.2% and 1.3 to 4.4%, respectively. The content ratio of PD to PT saponin in whole ginsengs showed a little variation as 0.73 to 0.92, while those of tail roots showed a greater variation in the range of 0.72 to 1.75, indicating that tail roots contain higher content of PD saponin than whole ginsengs did.

  • Ginseng saponin had an effect on the proliferation and viability of cultured murine thymocytes. When the thymocytes were cultured in various concentrations of ginseng saponin, the number of thymocytes increased at $10^{-5}$% ginseng saponin but decreased at $10^{-5}$%. There was little change in the number of thymocytes when cultured in IL 2(Interleukin 2), a factor known for its influence on the proliferation and maturation of thymocytes. When the thymocytes were cultured in various concentrations of IL 2 with $10^{-5}$% ginseng saponin, the number of total cells increased at 1.5% or 3% IL 2 when cultured for 9 hours, or at 6% IL 2 for 12, 24, or 48 hours. But there was little change in the number of viable cells. In vitro, ginseng saponin had an effect on the activity of ADA(Adenosine Deaminase), an enzyme known to affect the production of IL 2. There was a 25% increase in the activity of ADA in the presence of $10^{-5}$% ginseng saponin.

  • The effect of Korean Ginseng saponins (total saponin, PD saponin and PT saponin) on the serotonin type 3 receptor, which is known to be involved in nausea and vomiting following anticancer chemotherapy or the general anesthesia, was investigated. after in vitro transcribed recombinant serotonin type 3 receptor in the Xenopus laevis oocyte, classic two electrodes voltage clamp technique was used. All of ginseng saponins inhibited the response of the agonist, serotonin, on the serotonin type 3 receptor in a dose-dependent manner. PT saponin showed to have the inhibitory effect more than 2 times as potent as PD saponin. Total saponin shifted the serotonin dose response plot to the right (EC$\_$50/, 0.70$\pm$0.17 $\mu$M into 3.57$\pm$1.42 $\mu$M, and Hill coefficient, 2.14$\pm$0.60 into 1.52$\pm$1.00). Ginseng saponin did not change the reversal potential (∼0 mV) of serotonin type 3 receptor. These results suggest that Korean ginseng saponin may have the inhibitory effect on serotonin type 3 receptor.

  • We have studied the effect of ouabain, tool ginseng saponin, panax saponin C (protopanaxatriol derivative) and ginsenoside $Rb_{1}$ (protopanaxadiol derivative) on $Na^+,K^+$-ATPase and $Mg^{++}$-ATPase activities were determined by the method of Robinson and ATPase activities were determined by the method of King. The $Na^+,K^+$-ATPase activities were inhibitied by 90.1% and 51.1% respectively at the concentration of $10^{-3}M$ and $10^{-4}M$ ouabain. The results are consistent with those of Robinson. The $Na^+,K^+$-ATPase activities were increased by 14.3% and 10.0% respectively at the concentration of $10^{-4}$g/ml and $10^{-5}$g/ml total ginseng saponin. Panax saponin C obtained by the method of Han and ginsenoside $Rb_{1}$ obtained by the method of Shibata were used. The $Na^+,K^+$-ATPase activities were increased in the presence of panax saponin C and the increased activity with panax saponin C was greater than that with total ginseng saponin. On the other hand ginsenoside $Rb_{1}$ showed an inhibitory effect on $Na^+,K^+$-ATPase. Total ginseng saponin, panax saponin C and ginsenoside $Rb_{1}$ had no effect on $Mg^{++}$-ATPase. Therefore, it may be concluded that total ginseng saponin has dual effects on microsomal $Na^+,K^+$-ATPase, that is, panax saponin C exhibits stimulatory action, whereas ginsenoside $Rb_{1}$ shows inhibitory action.

Inhibition of MMP-13 mRNA expression by ginseng saponin in fetal rat calvarial cells (백서 태자 두개관세포에서 인삼 사포닌에 의한 MMP-13 mRNA 발현 억제)

  • There is a potential role of collagenase-3 in alveolar bone loss and periodontal disease progression, we need to develope or find chemotherapeutic drugs or herbal agents which may regulate the expression of MMP-13. Ginseng saponin, one of the major components of Korea ginseng(panax ginseng) root, has many various biologic effects, such as cytotoxic effect, tumoricidal effects, cytokine regulations, and protein biosynthesis effect. The purpose of this study was to determine the effects of Korea red ginseng saponin on MMP-13 gene expression in osteoblasts. The experimental groups were cultured with ginseng saponin in concentration of 1.0, 10, 25, 50, 100, 250 and $500{\mu}g/ml$ for MTT assay. Primary rat calvarial cells were pre-treated for 1 hour with ginseng saponin(100 ${\mu}g/ml$) and then stimulated with $IL-1{\beta}(1.0ng/ml)$ and PTH(10 nM). MMP-13 gene expression was evaluated by RT-PCR. The results were as follows: Ginseng saponin was cytotoxic to osteoblast at concentration exceeding $250{\mu}g/ml$ for longer than 24 hours in tissue culture(p<0.01). In RT-PCR analysis, steady state MMP-13 mRNA levels were increased approximately 350% by $IL-1{\beta}$, and 400% by PTH when normalized to untreated control. $IL-1{\beta}-indued$ MMP-13 mRNA expression was reduced 50% by pretreatment with ginseng saponin. But ginseng saponin didn't inhibit MMP-13 expression from PTH stimulated cells. This results suggest that ginseng saponin Inhibit $IL-1{\beta}-indued$ MMP-13 mRNA expression.

  • Total saponin from Korean red ginseng changed thermodynamic parameters of membranes from dipalmitoylphosphatidylcholine (DPPC) and ghost erythrocytes of human. In liposomes from DPPC, temperature of the main transition (Lb'-La) in liquid-crystalline phase increases by 0.2$^{\circ}C$ in average, but enthalpy does not change. Total saponin at a concentration of smaller than $10^5$% "stabilizes" the timid bilayers. At larger than 0.07 of saponin/DPPC ratio, saponin leads to an exclusion of the bound lipid molecules from the main phase transition into lamella liquid crystalline La-phase. Total saponin influences specifically all erythrocyte membrane transitions in a concentration-dependent manner, i.e. on the structures of all the main membrane skeleton proteins. A high structural specificity of saponin with membrane proteins, could be a base of specificity of physiological response of not only erythrocytes, but also other cells.her cells.
