Optimization of Ferric Chloride Induced Carotid Artery Thrombosis Model in a Rat: Effect of Ginkgo biloba Extracts

  • Animal models are important tools in thrombosis research and preclinical drug development. In recent studies, ferric chloride ($FeCl_3$) has been widely used to induce arterial thrombosis in a variety of species. The purpose of this study was to find an optimal concentration of $FeCl_3$ and validate this model suited better for thrombosis research. A small piece of filter paper, soaked in $FeCl_3$ solution (10, 20 or 35%, v/v, in distilled water) was topically applied on the carotid artery of SD rats to measure the time to occlusion (TTO) and thrombus weight (TW) to ascertain 35%, as an optimal $FeCl_3$ concentration ($8.63{\pm}0.92min$; p =0.000, $0.79{\pm}0.03mg/mm$; p =0.000, respectively). To validate this experimental model, Ginkgo biloba special extract EGb761 (5, 10 or 30 mg/kg) as a reference agent administered by peritoneal route for 1h prior to the induction of thrombosis, showed significantly delayed TTO in a dose dependent manner ($18.50{\pm}2.17$, $29.17{\pm}1.83$, and $38.00{\pm}1.79min$, respectively) and significantly reduced TW and repaired collagen fibre in the injured vessel compare to vehicle group. Our results provide a simple, reproducible and well controlled in vivo screening system to induce thrombosis in rats by the topical application of 35% $FeCl_3$ to assess the efficacy of the new anti-thrombotic agents.

육미지황탕가미방(六味地黃湯加味方)이 흰쥐의 기억능력과 중추신경계 유전자 발현에 미치는 영향 (The effect on gene expression profile of rat hippocampus caused by administration of memory enhancing herbal extract)

  • The herbal extract (YMT_02) is a modified herbal extracts from Yukmijihwangtang (YMJ) to promote memory-enhancing. The YMJ extracts has been widely used as an anti-aging herbal medicine for hundred years in Asian countries. The purpose of this study is to; 1) quantitatively evaluate the memory-enhancing effect of YMT_02 by hehavior task, 2) identify candidate genes responsible for enhancing memory by cDNA microarray and 3) assess the anti-oxidant effect of YMT_02 on PC12 cell. Memory retention abilities are addressed by passive avoidance task with Sprague-Dawley (SD) male rat. Before the training session, the rats are subdivided into four groups and administrated with YMT_02, Ginkgo biloba, Soya lecithin and normal saline for 10 days. The retention test was performed. 24 hours after the training session. The retention time of the YMT_02 group was significantly (p<0.05) delayed $({\sim}100%)$, whereas Ginkgo biloba and Soya lecithin treatment delayed 20% and 10% respectively. The hippocampi of YMT_02 and control group were dissected and mRNA was further purified. After synthesizing cDNA using oligo-dT primer, the cDNA were applied and mRNA was further purified. After synthesizing cDNA using oligo-dT primer, the cDNA were applied to Incyte rat GEMTM 2 cDNA microarray. The microarray results show that prealbumin(transthyretin), phosphotidy lethanolamine N-methyltransferase, and PEP-19 are expressed abundantly in the YMT_02 treated group. Especially, PEP-19 is a neuron-specific protein, which inhibits apoptotic processes in neuronal cell. On the other hand, transcripts of RAB15, glutamate receptor subunit 2 and CDK 108 are abundant in control group. Besides, neuronal genes involved in neuronal death or neurodegeneration such as neuronal-pentraxin and spectrin are abundantly expressed in control group. Additionally, the YMT_02 shows an anti oxidative effect in the PC12 cell. The list of differentially expressed genes may implicate further insight on the action and mechanism behind the memory-enhancing effect of herbal extracts YMT_02, for example, anti-apoptotic, anti-oxidative, and neuroprotective effects.

Ginkgo Biloba Extract가 C3H 마우스 섬유육종의 저산소세포 분획에 미치는 영향 (The Effect of Ginkgo Biloba Extract on Hypoxic Fraction of C3H Mouse Fibrosarcoma)

  • 목적 : C3H 마우스의 섬유육종에 있어서 저산소 분획 및 대사상태에 미치는 Ginkgo biloba extract (GBE)의 영향을 알아보기 위해 본 실험을 시행하였다. 방법 : C3H 마우스의 우측 대퇴부에 이식한 섬유육종이 6mm가 되었을 때 $TCD_50$를 이용하여 저산소 세포 분획을 계산하였다. 방사선은 GBE (100mg/kg) 1회 투여시는 1시간 후에, 2회 투여시는 24시간 간격으로 2회 투여후 1시간 경과후 방사선을 조사하였으며, 산소공급이 충분한 상태와 결찰에 의한 저산소 상태에서 각각 조사하였다. 종양의 대사 상태 변화를 알아보기 위해 GBE 투여 전과 투여 후 1시간에 각각 ^{31}P$ 핵자기 공명 분광분석법을 시행하였다. 결과 : 종양 치료방사선량은 결찰에 의한 저산소 상태시 81.7 (77.7 - 86.0) Gy, 정상 혈류상태시는 69.6 (66.8 - 72.5) Gy 였으며, GBE 1회 투여한 경우에는 67.5 (64.1 - 71.1) Gy, GBE 2회 투여시는 62.2 (59.1 - 65.5) Gy 로서, 2회 투여시 현저히 감소하였다. 이로부터 계산한 저산소 세포 분획은 정상 혈류 상태에서는 $10.6{\%}$, GBE 1회 투여시는 $7.2{\%}$로 다소 감소하였고, GBE 2회 투여시는 $2.7{\%}$로 현저히 감소하였다. ^{31}P$ 핵자기공명분광분석결과 PCr/Pi 비는 대조군 $0.27{\pm}0.04$, GBE 1회 투여시는 $0.40{\pm}0.04$, 2회 투여시는 $0.71{\pm}0.04$로 GBE 2회 투여시 종양의 대사상태가 현저히 호전되었다. 결론 : 이상의 결과를 종합해 볼 때, GBE 의 투여에 의해 혈류량과 산소 및 영양분의 공급이 증가함에 따라, 저산소세포분획이 감소하였고, 종양의 대사 상태가 호전되어, 종양의 방사선에 대한 감수성이 높아졌다고 판단되었다.

은행잎의 염색성 및 자외선 차단 효과 (Dyeability and UV-blocking Effect of Dyed Fabrics with Ginkgo Extract)

  • Ginkgo leaves, which have been known as medical materials, were selected as new natural dyes in this study, in which it was examined whether they have the function of UV-blocker or not. Cotton, linen & Hanji(Korean traditional paper) were dyed with Ginkgo. Al, Cr, Cu, Fe and Sn were selected as mordants. UV-blocking rate of dyed samples was measured after dyeing and mordant treatment. The results of this study are as follows. First, most surface color of dyed samples was yellow except that Hanji mordanted with Cu was yellowish red. Second, dyeing repetition had positive correlations with K/S values of cotton ($r=.\;758^{**}$), linen ($r=.\;500^*$) and Hanji ($r=.\;819^{**}$), because K/S values were increased according to dyeing repetition. Third, solar UV-blocking rates had positive correlations with dyeing repetitions (p<0.01), because UV-blocking rates had increased according to dyeing repetition. Fourth, UV-blocking ability of Hanji was highest among samples, as UV-blocking rates of cotton and linen dyed after 3 dyeing repetition were up to 93%, Hanji's values were up to 98%. Therefore, it was proven that Ginkgo extracts used in this experiment as dyestuff have excellent dyeing ability and high UV-blocking ability. It is hoped that this work will lead to further research to confirm the physiological effects when human wear the clothes made from Hanji.

후박 및 은행잎 추출물의 향균, 향염 및 세포활성도에 미치는 영향 (BIOLOGICAL EFFECT OF MAGNOLIA AND GINKGO BILOBA EXTRACT TO THE ANTIMICROBIAL, ANTIINFLAMMATORY AND CELLULAR ACTIVITY)

  • Periodontal therapy for treatment of periodontitis involves the elimination of bacterial plaque and elimination of the anatomic defects by regenerative procedure. The purpose of this study was to evaluate on the biological effect of magnolia and Ginkgo biloba extract to the antimicrobial, antiinflammatory and cellular activity. Antimicrobial assay was performed with the diffusion method of the extract by measuring of growth inhibitory zone of B. cereus from blood agar plate. Effect of the extract to cellular activity of gingival fibroblast were examined using MTT method and measured the result with optical density on 570nm by ELISA reader. Inhibitory effects of $PGE_2$ production from gingival fibroblast was performed with the addition of $IL-l{\beta}$ and the extract to the well and examined to the product of $PGE_2$ from cell by ELISA reader. In vivo anti-inflammatory effect was performed with injection examined with clinically and histologically for their extent of mecrosis and inflammation. Antimicrobial activity of Magnolia extract showed significantly higher activity than that of control. However, GBE did not showed significant activity to compare with control, and mixture of Magnolia and GBE extract showed significantly higher activity than that of control. The effect of cellular activity to gingival fibroblast showed no significant differences of between control and Magnolia extract. However, GBE showed significantly higher rate of cellular activity to compare with control and even to PDGF-BB, and also showed same degree of cellular activity even though mixed with Magnolia extract. The inhibitory effect of $PGE_2$ production showed significantly reduction of $PGE_2$ production to compare with control, but its inhibitory effect was not much strong to compare with Indomethacin. In vivo, antiinflammatory effect of Magnolia extract to P. gingivalis injection of Hamster buccal check showed significantly reduction of inflammatory cell infiltration and tissue necrosis, but GBE showed no effect on the inhibition of inflammatory process. These results suggested that Magnolia and GBE extract possessed different kind of biological activity and also can be compensated on their activity with each other for elimination of bacterial plaque and anatonical defect.

열처리에 따른 은행 외종피 추출물의 생리활성 (Physiological Activities of Ginkgo biloba Sarcotesta Extract with Heat Treatment)

  • 은행 가공산업에서 부산물로 제거되는 은행 외종피의 활용성을 증대시키고자 다양한 생리활성에 미치는 열처리의 영향을 살펴보았다. 은행 외종피를 분리하여 $130^{\circ}C$에서 2시간 열처리한 후 물, 70% 에탄올 및 80% 메탄올 추출물을 제조하여 생리활성변화를 살펴본 결과, ABTS와 DPPH 자유라디칼 소거활성은 80% 메탄올 추출물을 제외하고, 열처리 후 증가하는 경향을 보였으며, 물 추출물에서 각각 14.95 mg AAE/g과 7.36 mg TE/g으로 높게 나타났다. ${\alpha}$-Glucosidase 저해 활성은 물 추출물을 제외하고 열처리 시 감소하는 경향을 보였으며, 열처리 전 80% 메탄올 추출물에서 98.66%로 가장 높게 나타났다. Angiotensin converting enzyme I 저해 활성은 추출물 간의 유의적인 차이가 없었으며(P>0.05), 열처리 후 모두 감소하였다. 산화질소 생성억제 활성은 열처리 시 증가하였으며, 물 추출물($200{\mu}g/mL$)이 $12.33{\mu}M$로 LPS 처리구에 비해 산화질소 생성량을 72.39% 감소시켰다. 지방구 축적억제 활성은 세포독성을 보이지 않는 농도에서 열처리 시 감소하였다. 유방암 세포주에 대한 항암 활성은 열처리 후 증가하는 경향을 보였다. 이상의 결과로부터 은행 외종피 추출물의 다양한 생리활성을 확인하였으며, 특히 열처리 후 항산화 활성과 유방암 세포주에 대한 증식억제 활성이 크게 증가하였으며, 추후 생리활성물질의 규명 연구가 필요하다고 판단된다.

The effects of the standardized extracts of Ginkgo biloba on steroidogenesis pathways and aromatase activity in H295R human adrenocortical carcinoma cells

  • Objectives Aromatase inhibitors that block estrogen synthesis are a proven first-line hormonal therapy for postmenopausal breast cancer. Although it is known that standardized extract of Ginkgo biloba (EGb761) induces anti-carcinogenic effects like the aromatase inhibitors, the effects of EGb761 on steroidogenesis have not been studied yet. Therefore, the effects of EGb761 on steroidogenesis and aromatase activity was studied using a H295R cell model, which was a good in vitro model to predict effects on human adrenal steroidogenesis. Methods Cortisol, aldosterone, testosterone, and $17{\beta}$-estradiol were evaluated in the H295R cells by competitive enzyme-linked immunospecific assay after exposure to EGb761. Real-time polymerase chain reaction were performed to evaluate effects on critical genes in steroid hormone production, specifically cytochrome P450 (CYP11/ 17/19/21) and the hydroxysteroid dehydrogenases ($3{\beta}$-HSD2 and $17{\beta}$-HSD1/4). Finally, aromatase activities were measured with a tritiated water-release assay and by western blotting analysis. Results H295R cells exposed to EGb761 (10 and $100{\mu}g/mL$) showed a significant decrease in $17{\beta}$-estradiol and testosterone, but no change in aldosterone or cortisol. Genes (CYP19 and $17{\beta}$-HSD1) related to the estrogen steroidogenesis were significantly decreased by EGb761. EGb761 treatment of H295R cells resulted in a significant decrease of aromatase activity as measured by the direct and indirect assays. The coding sequence/Exon PII of CYP19 gene transcript and protein level of CYP19 were significantly decreased by EGb761. Conclusions These results suggest that EGb761 could regulate steroidogenesis-related genes such as CYP19 and $17{\beta}$-HSD1, and lead to a decrease in $17{\beta}$-estradiol and testosterone. The present study provides good information on potential therapeutic effects of EGb761 on estrogen dependent breast cancer.

  • 목적 : Ginkgo biloba extract (GBE)는 혈액순환개선효과가 있어 기존의 방사선민감제와 달리 혈류저항이 높은 악성종양에서 저산소세포 분획을 줄일 수 있으나 아직까지는 실제 임상에서 사용되는 저선량 분할 방사선조사시에도 GBE가 방사선조사 효과의 증대를 나타내는지 밝혀지지 않았다. 이에 본 연구에서는 저선량 분할 방사선조사시 GBE가 방사선조사효과증대를 나타내는지, 또한 정상조직과 악성종양의 혈류량 분포에 어떠한 영향을 미치는지 알아보고자 하였다. 대상 및 방법 : 통상적인 범위의 저산소세포 분획을 가진 C3H 마우스의 섬유육종(FSall)을 마우스 우측하지에 이식한 후 종양직경이 7 mm에 도달하였을 때 마우스 체중 kg당 100 mg의 GBE를 방사선조사 25시간 전과 매회 1시간 전에 복강내 투여 후 방사선을 조사하였다. 3 Gy 내지 12 Gy의 방사선을 일회 조사시 GBE투여군과 방사선단독조사군의 종양성장지연기간을 비교하여 GBE에 의한 방사선효과 증강율을 구하였다. 3 Gy씩 10회의 분할 방사선조사시 방사선 단독조사군의 방사선량-종양성장지연기간(tumor growth delay, TGD)의 관계식을 구하여 두 실험군의 선형회귀직선 기울기의 비율, 즉 GBE에 의한 방사선효과 증강율을 구하였다. 같은 종양을 이용하여 레이저 도플러 혈류측정기로 정상근육과 종양의 혈류량을 측정함으로써 저혈류 분포의 차이가 존재하는지 확인하였고, GBE 투여에 의한 혈류량분포의 변화 여부를 관찰하였다. 결과 : 3 Gy 내지 12 Gy의 방사선을 일회 조사시 GBE투여군의 종양성장지연기간이 방사선단독조사군에 비하여 의미있게 증가되었으며(p<0.05), GBE에 의한 방사선효과 증강율은 3일 종양성장지연 기준으로 1.16이었다. 3 Gy씩 10회의 분할 방사선조사시 방사선 단독조사군의 방사선량-종양성장지연기간(tumor growth delay, TGD)의 관계는 TGD $(days)=0.26{\times}D$ (Gy)+0.13, GBE 병용군의 경우 TGD $(days)=0.30{\times}D$ (Gy)+0.13이었고 두 실험군의 직선 기울기의 비율, 즉 GBE에 의한 방사선효과 증강율은 1.19 ($95\%$ 신뢰구간: $1.13\~1.27$)로 나타났다. 같은 종양을 이용하여 레이저 도플러 혈류측정기로 혈류량을 측정한 결과, 혈류량의 분포는 모두 Poisson 분포와 비슷한 양상을 보였다. 정상근육의 혈류량은 평균 10.15 mL/100 g/min, 종양의 혈류량은 평균 7.78 mL/100 g/min으로 종양의 평균 혈류량이 근육에 비하여 낮게 나타났고(p=0.001), 2 mL/100 g/min 이하의 저혈류 분포는 정상근육에 비하여 종양에서 높게 나타났다($0.5\%$$5.2\%$, p=0.005). 정상근육에서는 GBE 투여에 의하여 혈류량 분포에 큰 변화가 없었던 반면, 종양에서는 2 mL/100 g/min 이하의 저혈류량 분포가 감소하였고, 평균 혈류량은 $23.5\%$ 증가되었다(p=0.0004). 결론 : GBE는 방사선 일회 조사시 뿐만 아니라 분할조사시에도 방사선치료의 효과를 유의하게 증가시켰다. 또한 정상근육에 비하여 종양의 혈류량을 선택적으로 증가시킴이 확인되었다.

Antithrombotic and Antiplatelet Effects of Cordyceps militaris

  • Cordyceps is a genus of ascomycete fungi and is well known as one of the important medical fungi in Chinese, Korea, and other Asian countries, because of its various beneficial effects on human health. The pharmacological activities of Cordyceps extract are mainly focused on anti-cancer, anti-metastatic, and immune modulating effects. In the present study, we investigated whether the antiplatelet effect of ethanol extract of cultured Cordyceps militaris (CMEE) with FeCl3-induced arterial thrombosis model. We observed that CMEE exhibited a significant inhibitory effect against ADP and collagen-induced platelet aggregation. However, there were no significant differences in prothrombin time (PT) and activated partial thromboplastin time (aPTT). These results suggest that antithrombotic activity of CMEE is related to antiplatelet effect rather than anticoagulation effect, and CMEE may be a positive effect on improving blood circulation against vessel injury and occlusion.

The Effect of Antioxidant-complex on Oxygen Free Radical Generating and Scavenging System in Rats

  • To elucidate the effect of antioxidant complex containing $\beta-carotene$, vitamin E, vitamin C, Ginkgo Biloba leaf extract and selenium on oxygen :tree radical production and detoxification system, rats were fed normal diet and normal diet with antioxidant complex 0.1%, 0.3% and 0.5% for 3 weeks. Feed efficiency ratio, changes in body weight, weight gain and amounts of feces of rat are similar in four groups. Liver weight per body weight and hepatic lipid peroxide weight increased in 0.5% group. However, hepatic glutathione contents in all antioxidant complex added groups were significantly increased compare with normal control group. On the other hand, the activity of xanthine oxidase was a little increased due to the amounts of antioxidant complex. Superoxide dismutase and gutathione peroxidase activity of 0.1% antioxidant complex added group were increased about $10{\sim}20%$ in comparison to normal control group. These results suggest that the supplementation of antioxidant complex 0.1% to basal diet may reduce the hepatic damage caused by free radicals.
