은행나무와 곰솔에 처리된 인공산성비에 의한 잎의 형태변화 (Effect of simulated Acid rain on Foliar Structural of Changes of Ginkgo biloba and Pinus thunbergii)

  • 은행나무와 곰솔의 잎은 pH 3.2 이하의 인공산성비 처리구에서 괴사반점과 황화현상 및 조기노화 현상이 나타났다. 잎표면 구조의 관찰에서 은행나무 잎은 pH 2.4의 처리구에서 잎표면 왁스층의 침식이 심하게 일어난 반면 곰솔의 경우 pH 4.0 이하의 처리구에서 왁스층의 침식이 심하게 일어났다. 은행나무 잎은 pH 3.2 이하에서 표피세포와 해면조직에서 구조의 변화가 나타났으나 유관속조직은 정상적인 구조를 나타냈다. 엽육세포의 크기는 산도가 높아질수록 작아지고 세포간극이 커졌다. 곰솔의 잎은 표피세포보다 엽육조직과 유관속조직의 피해가 현저하였다. 은행나무의 기공의 크기와 모양은 인공산성비의 영향을 받지 않았으나 기공지수와 기공의 공극 크기는 높은 산도에서 증가하였다. 또한 인공산성비가 처리된 은행나무와 곰솔 잎의 기공들은 열려 있었다.

인공산성우가 상수리나무와 은행나무 유묘의 기공저항성, 엽의 Wettability 및 해부학적 변화에 미치는 영향 (Effects of Simulated Acid Rain on Stomatal Resistance, Wettability and Anatomical Changes in Quercus acutissima and Ginkgo biloba Seedlings)

  • 본(本) 연구(硏究)는 상수리나무(Quercus acutissima)와 은행나무(Ginkgo biloba) 유묘(幼苗)를 대상으로, 12주간 pH 4.0 또는 pH 2.5의 인공산성우(人工酸性雨)(Simulated acid rain ; SAR)에 노출(露出)시켜 기공저항성(氣孔抵抗性)과 엽(葉)의 Wettability 및 해부학적(解剖學的) 변화(變化)를 파악(把握)하고자 실행(實行)하였다. 대조구로는 pH 6.5의 증류수를 살포(撒布)하였다. 상수리나무와 은행나무의 기공저항성(氣孔抵抗性)은 두 수종(樹種) 모두 SAR처리에 의하여 높아졌으며, 색소(色素)(Water blue solution)의 부착정도 측정(測定)에 의한 상수리나무 엽(葉)의 Wettability는 SAR처리에 의하여 증가(增加)하였고, Contact angle측정(測定)에 의한 은행나무 엽(葉)의 Wettability 역시 SAR처리에 의하여 증가(增加)하였다. SAR을 처리(處理)한 상수리나무 엽조직(葉組織)에서 표피세포, 책상조직 및 해면상조직의 부분적(部分的)인 피해(被害)가 관찰되었고, 모용(毛茸)(Trichome)의 현저한 감소(減少)와 Wax층(層)의 침식이 SEM을 통해 관찰(觀察)되었다. 은행나무의 엽조직(葉組織)은 pH 4.0처리에서는 피해(被害)를 관찰(觀察)할 수 없었으나, pH 2.5처리에서는 표피세포 및 통도조직의 붕괴가 관찰(觀察)되었고, SEM에 의한 엽(葉) 표면조직(表面組織)의 관찰(觀察)에서는 현저한 변화(變化)를 찾아볼 수 없었다.

Screening of Antimicrobial Activity against Enterohemorrhagic escherichia coli 0157 : H7 from Plants in Korea

  • We screened the methanol extracts from 133 plant species growing in Korea for antimicrobial activity against enterohemorrhagic Escherichia coli 0157 : H7. Those are selected from three plant grouping ; traditional medicinal herbs, edible plants, and flowers. They were tested by disk diffusion assay. From evaluation of the inhibition zone diameter of microbial growth, we found that the flower extract of Rhododendron Schilpenbachii Max had the most significant antimicrobial activity against this bacteria. Extracts from most of the vegetables and plants did not show antimicrobial activity except for the leaves of Ginkgo biloba L. and seeds of Prunus Dallicina L. did not show antimicrobial activity except for the leaves of Ginkgo biloba L. and the seeds of Prunus sallicina L.

3종의 식물 추출물에 의한 백점충의 in vitro 구제효과 (In vitro eliminative effects three sorts of herbal extracts of against Ichthyophthirius multifiliis)

  • 3종의 식물성 추출물 및 포르말린의 시험관 내에서의 백점충에 대한 살충효과를 조사하였다. 복합생약제는 500배 희석액에서 1시간 이내에 충을 완전히 죽일수 있었으나, 여지초발효액은 100배의 희석액에서 1시간 이내에 모든 충을 죽일 수 있었다. 은행잎 추출물은 5,000배의 희석액으로 1시간 이내에 모든 충을 죽일 수 있었다. 대조약제로서 포르말린은 100ppm의 농도로 1시간 이내에 충을 모두 죽일 수 있었다. 이 결과로 은행잎 추출물이 살충효과가 가장 좋았다. 충에의 밀도에 따른 살충효과의 차이는 없었다. 또한 은행잎 추출물의 5,000배 희석액으로 2시간 동안 약욕한 후의 어류의 아가미 조직에 있어서 변화가 없었다.

Variation of Ginkgolides and Bilobalide Contents in Leaves and Cell Cultures of Ginkgo biloba L.

  • Ginkgolides (GK) and bilobalide are valuable compounds that belong to the lactone terpene. The contents of these metabolites were determined by HPLC from female and male tree of Ginkgo biloba L. The productivity of G. biloba cells was also compared with the corresponding individual trees. High variations in the ginkgolides and bilobalide were observed from different individuals, plant parts, and cultured cells. The ginkgolides and bilobalide contents were different depending on the plant parts. Callus was obtained from various plant tissues, and NAA was better at callogenesis than 2,4-D in both the female and male trees. The plants and their corresponding cells showed considerable variation in their ginkgolides and bilobalide concentrations. The ginkgolides and bilobalide contents were not correlated with the production between dominant trees and their corresponding cells. Light irradiation enhanced the production of GK-A and GK-B, however, the concentration of bilobalide decreased under dark conditions.

Cultures of Ginkgo biloba, Effect of Nutritional and Hormonal Factors on the Growth of Cultured Cells Derived from Ginkgo biloba

  • Calli and suspension cultures were obtained following inoculation of the explant from leaves of Ginkgo biloba L on the supplemented MS basal medium. The obtained calli and suspension cultured cells were able to produce detectable amounts of ginkgolides which are known as natural specific PAF antagonists. The production of ginkgolides in the calli and suspension cultured celles were identified using GC/MS, GC and HPLC with authentic ocmpounds. Since the production of ginkgolides A and B the calli and suspension cultured cells had been confirmed, effects of types and concentration of plant growth regulators, media and illumination on the induction and growth of the callus were studied. The concentrations of growth regulators for optimal callus were studied. The concentrations of growth regulators for optimal callus induction were studied. The concentrations of growth regulators for optimal callus induction were 1.0 to 2.0 mg/L for NAA and o.1 mg/L for kinetin. The growth of the Callus seemed to be more simnultaed with the combination of NAA and kinetin than NAA and BA with illumination at all concentration ranges of 1.0 to 4.0 mg/l for NAA and o.1 to 1.0 mg/L for kinetin or BA studied. Amogn 8 different media used, the induction rate of callus on Anderson, Eriksson, and Shenk and Hildebrant at 4 weeks after the innoculation was almost the same as that of MS. However, callus was rarely induced on Heller or White medium. Suspension cultures were easily initiated with 3 g of callus (fresh weight) derived from ginkgo leaves on supplemented MS medium. A typical growth curve of suspension cultured cells could be obtained by measuring the fresh weight of the suspension cultured cells at every 3 days. To improve the growth of suspension cultured cells of ginkgo, effects of concentrations of NAA, sucrose, phosphate ions and molar ratio of $NH_{4}^+\;to\;NO_{3}^-$ ions in the culture medium were studied. The maximum growth of the cells was achieved when the culture medium contained 1.0 mg/L of NAA, 30 g/L sucrose, 1.75 mM phosphate ions and 1:5 molar ratio of $NH_{4}\;to\;NO_{3}^-$ ions.

In Vitro Peroxynitrite Scavenging Activity of 6-Hydroxykynurenic Acid and Other Flavonoids from Gingko biloba Yellow Leaves

  • As part of our research on phytochemicals that exert protective effects against diseases related to reactive nitrogen species, we have evaluated the scavenging activity of the yellow leaves of Ginkgo biloba on $ONOO^{-}$. The methanol extract and ethyl acetate fraction obtained from yellow leaves of G. biloba evidenced a marked scavenging activity on authentic $ONOO^{-}$. Repeated column chromatography of the active ethyl acetate soluble fraction on silica gel, Sephadex LH-20, and RP-18, resulted in the purification of 15 known compounds, including sciadopitysin (1), ginkgolide B (2), bilobalide (3), isoginkgetin (4), kaempferol (5), luteolin (6), protocatechuic acid (7), bilobetin (8), amentoflavone (9), ${\beta}-sitosterol$ glucopyranoside (10), kaempferol 3-O-rhamnopyranoside (11), kaempferol 3-O-glucopyranoside (12), kaempferol $3-O-[{6^{'}-O-p-coumaroyl-{\beta}-D-glucopyranosyl(1{\rightarrow}2)-{\alpha}-L-rhamnopyranoside]$ (13), kaempferol 3-O-rutinoside (14), and 6-hydroxykynurenic acid (15). Among the compounds isolated, flavonoids (5, 6 and 11-14), protocatechuic acid (7), and 6-hydroxykynurenic acid (15) all exhibited marked scavenging activities on authentic $ONOO^{-}$. The $IC_{50}$ values of 5-7, 11-14 and 15 were as follows: $2.86{\pm}0.70,\;2.30{\pm}0.04,\;2.85{\pm}0.10,\;5.60{\pm}0.47,\;4.16{\pm}1.65,\;2.47{\pm}0.15,\;3.02{\pm}0.48,\;and\;6.24{\pm}0.27\;{\mu}M$, respectively. DL-Penicillamine ($IC_{50}=4.98{\pm}0.27\;{\mu}M$) was utilized as a positive control. However, the other compounds (1-4, 8-10) exerted no effects against $ONOO^{-}$.

비만세포에서 은행잎 플라보놀에 의한 Th2 Cytokine 발현 및 신호전달 억제 기전 효과 (Suppressive effects of Th2 cytokines expression and the signal transduction mechanism in MC/9 mast cells by flavonol derived from Ginkgo biloba leaves)

  • 은행잎 유래 플라보놀 성분이 갖는 항아토피 활성에 대해 입증된 바는 드물다. 이 논문에서는 MC/9 비만세포에서의 은행잎 플라보놀의 항아토피 효과를 조사하기 위하여 ELISA와 Real-time PCR, western blot으로 은행잎 플라보놀을 분석하였다. 분석 결과 IL-13, MIP-1a의 생성량은 농도 의존적으로 현저하게 감소되었으며 IL-4, IL-5, IL-13, TNF-a 유전자 발현량은 25, 50, $100{\mu}g/m{\ell}$의 농도에서 효과적으로 억제되었다. western blot 분석 결과 c-jun과 NFAT-1단백질 발현이 감소되었음을 확인하였다. 이러한 결과들은 은행잎 유래 플라보놀 성분이 NFAT-1, c-jun 전사인자의 전사를 억제함으로써 MC/9 비만세포에서의 Th2 싸이토카인의 생성을 감소시키는 효과를 갖는 것을 나타낸다. 따라서 은행잎 유래 플라보놀 성분이 아토피 피부염 치료제로서 활용가능성이 있음을 보고하고자 한다.
