Mechanisms of Salt Tolerance in Crop Plants I. Physiological Responses of Barley, Rye and Italian Ryegrass Seed Germination to NaCl Concentrations (작물의 내염성 기작연구 I. 염분농도에 따른 보리, 호밀, 이탈리안 라이그래스 종자의 발아생리반응)

  • This experiment was conducted to elucidate the salt tolerance mechanism of crop plants. Plants used in this experiment were rye, barley, Italian ryegrass. Seeds were treated at saline condition and the saline conditions were controlled by NaCl. The results were summerized as follows. In rye, germination ratio and speed were above 90% and 80% at NaCl concentrations below 0.5% but germination ratio was seriously decreased at NaCl concentration above 0.6%. In barley germination ratio was not less than 80% at NaCl concentration below 0.8%. In italian rye grass germination ratio was decreased gradually by increasing NaCl concentration but germination speed was seriously decreased at NaCl concentration above 0.6% as not more than 7.5%. Water absorption rate was low in barley than rye and it was decreased by increasing NaCl concentration and duration of absorption. In barley, carbohydrate content was decreased slightly in response to increased NaCl concentration and content of reducing sugar was independent of NaCl concentration. In rye, reducing sugar content was increased until 0.3% of NaCl and increased seriously after 20 hours. $\beta-Amylase activity was high at control and 0.6% lot of NaCl, but it was not general tendency.

Presown Seed Treatments to Promote Seed Germination of Zoysiagrass (Zoysia japonica) (한국잔디 종자의 발아율 향상을 위한 파종전 종자처리)

  • Dormancy of intact zoysiagrass seeds hinders its seed propagation in which presowing seed treatment must be done to overcome it. The study was done to remodel the presowing treatments of zoysiagrass seeds by determining the treatment effects of seedcoat softening, prechilling, drying and combined light treatment with them or during germination stage. Seedcoat softening with different chemicals (NaOH 20%, KOH 20%, $H_2SO_4\;8M, acetone 20%) and periods (10, 30, 60 min.), prechilling with different periods at 3$^{\circ}C$, and drying with 4 hours at 35$^{\circ}C$ was done. Light sources were also applied with 660 nm red LED light, commercial fluorescent and incandescent light and darkness. Germination test was done in alternative day and night temperature of 35 and 2$0^{\circ}C$ to check the daily germination rates. With seeds treated with NaOH 20% for 30 minutes, red and incandescent light showing similar responses inclined their germination rates compared to dark conditions. Drying resulted in its higher rate than non-drying but fluorescent light treatment forced during drying more enhanced the rate than red light. While mean germination rate considering all the seedcoat softening treatment periods was the greatest in $H_2SO_4$ in comparison with NaOH, KOH and acetone, NaOH treatment for 30 minutes showed the best rate among the all the treatment levels. In the conditions of sowing them to no-mulching field, presowing treatment procedure of zoysiagrass seeds was remodeled: the seedcoat softening treatment with NaOH 20% for 30 minutes was done, washed, and chilled at 3$^{\circ}C$ for 2 weeks before dried under 4 hour fluorescent light illumination at 35$^{\circ}C$.

Studies on Germination Rates and Early Seedling Growth Characteristics by Different Storing Temperatures, Durations, and Methods in Aralia cordata var. continentalis (Kitagawa) Y.C.Chu (독활 종자의 저장온도, 기간 및 방법에 따른 발아율과 유묘 생육특성 연구)

  • This study was conducted to improve the managing and storing methods of the seeds of Aralia cordata var. continentalis (Kitagawa) Y.C.Chu, to examine the viability and the germination ability of seeds with different storing conditions and methods, and to develop new ways to propagate and have better healthy seedling. Therefore, the germination rate, days required for germinating seeds, and early growth responses of Aralia cordata var. continentalis (Kitagawa) Y.C.Chu were investigated with different storing temperatures, durations and methods. The germination rate was higher in stratified storage than that in dry storage condition. The highest germination rate was with outdoor temperature at 30 days after stratified storage. The days required for germinating seeds were less than 10 days with the treatment of $25^{\circ}C$ and outdoor temperature in stratified storage. In dry condition, they were shorter with $4^{\circ}C$ and $25^{\circ}C$ than those with $-20^{\circ}C$ and outdoor temperature. Leaf number of seedling was higher in stratified storage compared to that in dry condition, while it was not clearly different according to storage temperatures and durations. Leaf length and leaf width of seedling was not difference among the treatment of storage methods, temperatures, and durations. Stem length of seedling was higher in stratified storage than those in dry condition, while root length was not clearly different among the treatments. It would be assumed that temperatures, methods and durations of storage could affect much to the germination rate and the early seedling growth response.

  • Laboratory experiements were conducted to evaluate water uptake and osmotic potential of soybean seeds associated with germinability. Bonus, Wayne, Essex and Pickett were selected for this study. Large and small seeds from the four varieties were included in water uptake measurements at $25^{\circ}C$ There were significant differences in water uptake during germination due to seed size and cultivars at optimum moisture condition, -6 and -15 bars soil moisture. As water stress increased, the water uptake of Pickett and Essex were superior to Bonus and Wayne, and small seeds were superior to large seeds. The seed moisture content at germination was 60.8% on a fresh weight basis under optimum moisture condition. The minimum moisture contents necessary for the germination were 50.2% at -6 bars and 50.9% at -15 bars. There were significant differences among varieties in seed osmotic potential during germination, although these differences depended on imbibition time. The average osmotic potential ranged from -32.0 bars after 4 hours imbibition to -11.2 bars at the beginning of germination. The correlation coefficient between seedling length and osmotic potential was not significant after any period of imbibition, suggesting that osmotic potential is not directly associated with seedling growth. However, osmotic potential is closely related to water uptake capacity of soybean seed.

Influence of Salinity Treatment on Seed Germination and Polyamine Synthesis in Barnyard Grass(Echinochloa hispidula) (강피종자의 발아와 폴리아민 생합성에 대한 염류의 영향)

  • To illuminate the physiological response to salinity, barnyard grass (Echinochloa hispidula) was germinated with high concentration of NaCl and KCL. Duration and promptness of seed germination were observed. Under salt stress, lipid peroxidation and polyamine biosynthesis were also analyzed. It appeared that high salt treatments per se did not provoke an inhibition of germination although the process of germination was significantly delayed. In context of lipid peroxidation and polyamine biosynthesis, we would imply that barnyard grass is tolerant to salinity. The increase in lipid peroxidation and putrescine content was prolonged only for 1 day after saline treatment. It could be concluded that these early acciimulation of putrescine and production of lipid peroxide seems to be associated with salt tolerance in the short-term. The physiological interest of these responses was discussed.

THE EFFECTS OF X-RAY IRRADIATION ON THE GERMINATION AND GROWTH OF SEEDLING IN GinKgo biloba L.

  • Ginkgo biloba L., which was distributed in Korea, was x-rayed with 100, 250, 500, 1000, 2000, 6000R. The results of responses about germination, morphological variations in seedling and early stage growth may be summarized as follows : 1. The germination percent was decreased with X-ray increasing dose from 500R except the slight stimulation of 100R. 2. The germination $LD_{50}$ seemed to be 765R. 3. The seedling height was depressed significantly but only slightly in 100R. 4. The loaf index was increased proportionally to high dose but T/R ratio was reduced apparently. 5. The dry or fresh weight decreased from 250R. It was highly significant except 100R. 6. Many morphological variations were indicated in plants.

Effect of Growth Medium and Extract Sterilization on a Seedling Bioassay of Alfalfa Autotoxicity (생장배지 및 추출액의 멸균이 알팔파 Autotoxicity 의 유묘 생물검정에 미치는 영향)

  • Autotoxic substance(s) from alfalfa(Medicago sativa L.) plants reduces germination and growth of adjacent new alfalfa after alfalfa. The autotoxic chemical(s) in alfalfa are clearly unknown. Our objective was to improve the sensitivity of an alfalfa seedling bioassay for evaluating autotoxic leaf extracts. We determined critical extract concentrations that inhibit seed germination and seedling growth, compared two different culture media, and evaluated the effects of extract sterilization on the sensitivity of the assay, by using streptomycin and autoclaving method. An agar medium in petri plate gave better responses of germination and seedling growth to the extracts than using filter paper in the plate. On agar medium, the concentration of extract required to reach 50% inhibition of root length was 2.7 g $kg^{-1}$, and of germination and hypocotyl length were 3.8 and 9.9 g $kg^{-1}$, respectively. Leaf extracts with 100 ppm streptomycin stimulated germination significantly compared to Leaf extract alone but reduced root length of control by 43%. Root length was more sensitive to the autotoxin(s) than was germination or hypocotyl length. These results suggest that agar medium mixed with extract and sterilization by autoclaving could be improved the consistency and precision of bioassay, and that root length was the best parameter of autotoxic effect of alfalfa leaf extract.

Germination Responses and Early Growth of Allium thunbergii by Temperature and Shading Level (온도와 차광수준에 따른 산부추의 발아반응 및 초기생육)

  • This study was carried out to determine the effects of environment controls (temperature and shading level) on germination responses and early growth of Allium thunbergii. Germination experiment was performed by pre-treatment (with low temperature and wetting treatments for 0, 20, 40 and 60 days) and temperature controls (5, 10, 15, 20, 25 and 30). And growth experiment was performed by containers (128 and 200 cavities containers) and shading level (full sunlight (control), 35%, 50% and 75% shading). Germination rate of A. thunbergii seeds were, 20 days of seed pre-treatment, the highest at $10^{\circ}C$ (81.7%) and the more temperature went up, the more germination rate went down. As a result of surveying container and shading treatments, the height, leaf area, leaf length, leaf aspect ratio (L/W) were higher under 50% shading of 128 (24.2cm, $2.76cm^2$, 22.3cm and 223.4, respectively) and 200 (22.6cm, $2.29cm^2$, 19.4cm and 190.5, respectively) cavities container. The root was grown well under full sunlight. Specially, fresh weight of shoot (leaves+stem) was higher under 50% shading of 128 (0.241g) and 200 (0.212g) cavities container. As a result of surveying the whole experiment, A. thunbergii seeds need to pre-treatment (with low temperature and wetting treatments for 20~40 days) for high germination rate. And it is judged better growth and higher yield by maintaining 50% shading of 200 cavities container.

Growth Responses at Different Growth Stage of Pinus densiflora Seedlings to Enhanced Uv-B Radiation (자외선-B 증가에 따른 소나무 유묘의 생장 단계별 생장 반응)

  • This study was carried out to investigate the growth responses of Pinus densiflora seedlings to enhanced UV-B environment for 16 weeks in the field condition. The seedlings were treated with one of three levels of UV-B dosages - ambient UV-B, ambient + 3.2, and ambient + 5.2 KJ m$^{-2}$day$^{-1}$ and the irradiation was performed at the stage before the germination, the fully expanded cotyledon, and the primary needles grown more than 0.8cm in length of the seedlings, respectively. Enhanced UV-B irradiation reduced the height and the root collar diameter growth, and dry mass production of the seedling, and T/R ratio was increased by the UV-B treatment. Difference in seedling growth was observed by difference in time of the UV-B treatment. Among the seedlings which were treated with ambient - 3.2 KJ m$^{-2}$day$^{-1}$, height and root collar diameter growth was relatively high in the seedling received the UV-B treatment at the stage before the germination. The lowest dry mass production was observed in the seedlings received the UV-B at stage of cotyledon both in two levels of enhanced UV-B treatment. Chlorophyll concentration was reduced by enhanced UV-B irradiation, and chlorophyll a/b ratio was increased by the UV-B treatment.

  • We have examined the effects of low concentrations of MNNG, alkylating agent, in survival and mutagenesis in Aspergillus nidulans. Pretreatments of cells with nontoxic and submutagenic doses of MNNG did not reduce the cytotoxic and mutagenic effects of exposure to a high concentration of drug. The results imply that adaptive responses on survival and mutagenesis for MNNG treatments do not occur in Aspergillus nidulans. In the first mitotic cell cycle during germination, the sensitivity to MNNG has been investigated at hourly time interval, and compared with that for UV irradiation. In both UV and MNNG treatments, the sensitivity increased till S cell stage, and decreased while DNA replication continued. Different from that show for UV irradiation, lethality to MNNG reached to the maximum at G2 cell stage.
