Improvement of In Vitro Seed Germination in Chicory (Cichorium intybus L.) and Garland Chrysanthemum (Chrysanthemum coronarium L.) (치커리(Cichorium intybus L.)와 쑥갓(Chrysanthemum coronarium L.)의 기내 종자발아 개선)

  • A series of experiments were conducted to suggest efficient in vitro germination conditions to improve germination rate and aseptic seedling production in chicory and garland chrysanthemum. For this purpose, various levels of NaOCl concentration and soaking treatment time combinations were tasted. Fifteen minutes of soaking treatment with 5% NaOCl solution significantly improved germination and seedling formation percentages and prevented contamination in both chicory 'Precole' and 'Chiavari' and garland chrysanthemum 'Okiku 3'. There was no significant difference in germination response between MS and 1/2 MS medium compositions, but germination and seedling formation were delayed as sucrose concentration increased. Although petri-dish among various culture containers gave rise to improved germination responses in chicory 'Precole', there was no significant difference in germination responses among culture container types in chicory 'Chiavari' and garland chrysanthemum 'Okiku 3'. The ultrasonic treatment stimulated germination and seedling formation in both chicory cultivars but there was no effect in garland chrysanthemum 'Okiku 3'. The results suggested 15 minutes with 5% NaOCl for seed sterilization, 1/2 MS medium with no sucrose and petri-dish as proper medium composition and culture container, and 120 minutes with 5% NaOCl solution of ultrasonic treatment as promoting method for in vitro germination in chicory and garland chrysanthemum.

  • The factors influencing somatic embryo maturation, high frequency somatic embryo germination, and plantlet formation were studied in Terminalia chebula Retz. Maturation of somatic embryo were influenced by a number of factors such as in vitro culture passage, concentrations of sucrose, levels of abscisic acid (ABA), basal media and media additive combinations. Maximum frequency of somatic embryo maturation ($57.22{\pm}2.02$), was obtained on MS medium supplemented with 50 g/l sucrose. Different factors such as strengths of MS nutrients, plant growth regulators, media additives and their combinations controlling somatic embryo germination and plantlet formation were studied. High frequency of germination and plantlet formation ($58.80{\pm}1.47$) were achieved by subsequent subculture of mature somatic embryos on MS medium containing 30 g/l sucrose and 0.5 mg/l benzyl-adenine (BA). However, although duration of in vitro passage of the callus tissue was critical, contribution of the combinations of plant growth regulators and media additives showed nugatory effect on somatic embryo maturation and germination as evident from variable responses.

  • The present study was carried out to investigate the effect of Ni on germination, cell elongation, ${\alpha}-amylase$ activity, contents of chlorophyll and protein in radish were determined in the water culture. As the concentration of Ni was increased in the water culture, germination of radish was 55% by Ni 10 mg/kg and 30% by Ni 20 mg/kg. The ratio of cell elongation injury was 50%, by two days after Ni 20 mg/kg treatment. The injury ratio of ${\alpha}-amylase$ activity was 45% in the same condition and as the time goes on, inhibition of ${\alpha}-amylase$ activity were slightly decreased. Contents of chlorophyll a and b were decreased two days after treatment and chlorophyll a was more inhibited than chlorophyll b. Also changes of the protein contents was slightly decreased. Activity of ${\alpha}-amylase$ was decreased at germination stage, contents of chlorophyll a and b were decreased at growing stage.

Effect of Sowing Time on Germination and Early Seedling Growth of Quercus floribunda Lindl.

  • Quercus floribunda (Tilonj oak) is among the five species of Quercus and an evergreen tree found in Kumaun Himalayan region. Timing of germination is a crucial event determining the success of seedling establishment and survival. The aims of the study were to investigate the effect of sowing date on the germination and morphological responses of Tilonj oak (Quercus floribunda), cultivated during the month of August-September, with supplementary irrigation. The experiment was conducted by sowing seeds at two dates with one month interval in glass house conditions at DSB Campus, Nainital, Uttarakhand, India. Present study revealed that higher germination percentage (46.67 %) was recorded at sowing time $S_2$ as compared to the sowing time $S_1$ (32.86 %). Germination percentage as well as seedling growth were affected by sowing date though the differences were insignificant. At both the sowing dates, highest germination percentage was recorded for large seeds (32.86-46.67 %) followed by medium (31.43-33.33 %) and lowest germination (6.67-7.14 %) was recorded for small size seeds. In all parameters assessed, sowing time $S_2$ had the best performance in combination with large seed size. Thus, September is suitable month for best germination, growth and seedling vigour and large seed size is recommended to silviculturists and tree planters. The information on seed germination and seedling growth is vital both for conservation and rehabilitation of degraded lands.

Seed Germination in Lettuce Affected by Light Quality and Plant Growth Regulators (상추 종자의 발아에 있어 광질 및 생장조절물질의 영향)

  • Lettuce, a typical light-induced seed germination type, exhibits different germination responses according to cultivars, light quality, and plant growth regulator (PGR) treatments. Germination rates in most tested cultivars were over 85% under both white and red light, and were slightly decreased by blue light. Although photo-inhibition in germination was observed from most cultivars by far-red light, 'Cheongguangcheongchima', 'Okdol', and 'Manchudaecheongchima' could be classified as photo-insensitive lettuce cultivars by exhibiting the germination rates as 78,63, and 48% under for-red light, respectively. 6-Benzylamino purine (BAP) and kinetin promoted seed germination and normal seedling production under far-red light, but ethephon did not show any positive effects. Cytokinins such as BAP, kinetin, thidiazuron (TDZ), and zeatin overcame photo-inhibition of seed germination even the concentration of below $50\;mg{\cdot}L^{-1}$. However, auxins such as IBA, 2,4-D, and NAA failed to overcome the far-red light-induced photo-inhibition.

Effect of Simulated Acid Rain on Germination, Growth, Acid Buffering Capacity and Nutrient Leaching in Impatiens balsamina L. and Tagetes patula L. (산성비가 봉선화(Impatiens balsamina L.) 및 만수국(Tagetes patula L.)의 발아, 생장, 완충능력 및 양료용탈에 미치는 영향)

  • This study was conducted to investigate the effect of simulated acid rain (SAR) on germination, growth, acid buffering capacity and nutrient leaching in Impatiens balsamina L. and Tagetes patula L.. In both species, germination was not inhibited at pH 4.0, but the germination rate decreased at the lower pH of 3.0, showing higher rate in Inpatiens balsamina L. than Tagetes patula L.. As the pH decreases, the growth of radicle was markedly decreased than that of hypocotyl in both species. The plant height, root length, leaf area, total dry weight, relative growth rate and net assimilation rate were inhibited by SAR. The acid buffering capacity in the leaves were increased at pH 4.0, on the other hand, it was shown a tendency to decrease at pH 2.0 in both species. As the pH levels decreased from 5.6 to 2.0, the nutrient leaching from leaves was significantly increased in both species. Based on the results, there are a great difference in the responses to SAR between the two species. In general, Tagetes patula L. represented a higher tolerance to SAR than Impatiens balsamina L.. These results suggested that interspecific variation in the acid buffering capacity and nutrient leaching from leaves may be responsible for the interspecific susceptibility to SAR.

The Allelopathic Effects of Lantana camara on Seed Germination and Growth of Selected Bioassay Species

  • The allelopathic effects of Lantana camara L. (Family:Verbenaceae) on germination and seedling establishment of some agricultural crops and weed species have been identified. Aqueous extracts of dry leaves and contaminated soil where L. camara is grown were used to verify allelopathic effect on seed germination of five bioassay species; Raphanus sativas, Capsicum annum, Lycopersicum esculantem, Crotalaria juncia and Chromoleana odorata. Fifty seeds from each bioassay species were placed in a petri dish containing leaf extracts or contaminated soil, and seed germination were examined after 3 days. The plant house experiments were carried out to evaluate the impact of L. camara contaminated soil and leaf debris using L. esculantem as the indicator plant. Seed germination of L. esculentem, C. junica and Capsicum annum was significantly inhibited by L. camara contaminated soil. However, the degree of inhibition varied among the bioassay species. The aqueous extract of dry leaves of L. camara was highly phytotoxic and it significantly reduced seed germination of all bioassay species. There was a decline in plant height, leaf area and shoot dry weight of tomato only in early growth stages when grown in L. camara contaminated soils. However, incorporation of leaf debris into soil affected the vegetative growth of tomato in early stages when the leaf debris concentration was increased. Growth recovered at the latter part of the life cycle. On the basis of these results it can be concluded that the allelochemicals in L. camara contaminated soils are harmful to the seed germination of crop species. The adverse effect was present only during the early growth stages and it did not suppress the latter part of the plant growth. These responses are attributed to allelopathic effects which need confirmation under field conditions.

Effect of Presowing Seed Treatments on Germination and Seedling Emergence in Taraxacum platycarpum (민들레 종자의 파종전 처리가 발아율 및 입모율에 미치는 영향)

  • Several presowing seed treatments have been used to facilitate the seed germination and seedling emergence. The study was performed to design a model of presowing seed treatments in Taraxacum platycarpum. Germination and seedling growth responses to temperature, drying, chilling, $GA_3$ and priming were investigated to know the optimum germination conditions and presowing treatment. The optimum temperature condition for germination and seedling growth were $15{\sim}20^{\circ}C$ at dark condition. The seeds exposed to infra red light, however, did not germinate. The moisture content of seeds dried at $35^{\circ}C$ for 3.5 hours was returned to the level of unimbibed seeds. The germination of seeds dried under dark condition was better than those dried under infra red light. Prechilling treatment imposed for 6 weeks showed higher germination rate than $GA_3$ treatment or priming. In field test, the highest field emergence was observed in the seeds dried after prechilled for 6 weeks and followed by drying followed by $GA_3$ > low temperature > $KNO_3$ priming. Imbibition with 0.1 mM $GA_3$ solution for 2 hours then drying at 35 for 3.5 hour under dark condition was effective to the practical presowing treatment for Taraxacum seeds.

  • Growth responses of lettuce seeds germinated in aqueous solutions of $SO_2$ were investigated for mode of action studies of this gas. The concentration and pH of $SO_2$ solution greatly affected the growth of germinating seeds. Root growth was more sensitive to $SO_2$ than hypocotyl growth. The sensitivity of total seedling growth lay between root and hypocotyl growth. Growth inhibition of germinating seeds appeared more serioius when $SO_2$ was added during the early stage of germination.
