This study was conducted to investigate effects of cryopreservation by vitrification on the seed germination rate and growth and physiological properties of seedlings of Maackia amurensis. Cryopreservation significantly decreased the germination rate of seeds of M. amurensis, but the reduction of germination rate was mitigated by the treatment of cryoprotectant (plant vitrification solution, PVS2) before plugging into liquid nitrogen and fast thawing rate after cryopreservation. Long-term PVS2 exposure decreased seed germination rate, whereas cryopreservation time didn't have influence on seed germination rate. In addition, growth and physiological properties of seedlings were not affected by PVS2 exposing time and cryopreservation time. Therefore cryopreservation could be widely used as a technique of long-term ex situ conservation without any damage and deterioration of cells or tissues of the forest seeds. However, in order to increase the effect of cryopreservation, we have to develope the lower toxic cryoprotectant and suitable techniques to the structural or chemical properties of a variety of seeds.
Tabulis [Astilboides tabularis (Hemsl.) Engl.] has the potential for becoming a useful horticultural crop. This study was conducted to classify the seed dormancy types and to identify the germination conditions that improve the horticultural usefulness of this plant. We found that A. tabularis (Hemsl.) Engl. produced undifferentiated embryos and had a low germination rate of under 50%. In addition, water submersion led to moisture absorption. Therefore, A. tabularis seeds were designated as morphologically dormant. The germination rate was highest (86.3%) at $30^{\circ}C$ under light conditions. Higher temperatures generally led to an acceleration of the germination process, regardless of light condition. To improve the germination rate, seeds were submerged in various concentrations of growth regulators, such as $GA_3$ and kinetin, as well as minerals such as $KNO_3$ and KCl. An analysis of germination characteristics at $30^{\circ}C$ under light conditions revealed that treatment with 200 and $500mg{\cdot}L^{-1}GA_3$ led to an excellent germination rate of 97.0%. Treatment with $100mg{\cdot}L^{-1}GA_3$ led to a better germination rate than the control as well. However, treatment with kinetin, $KNO_3$, and KCl had no effect on germination, regardless of concentration.

Germination rate and early growth in crop such as rice, soybean, and perilla were observed after irradiation of $^{\gamma}$-ray (Co-60) in order to determine the effects of low does radiation. The low dose radiation was able to improve the early growth in crops and their agricultural charaters. Germination rate of 2Gy-irradiatied rice seeds was high and also were seeding height and fresh weight of the 0.5 Gy-irradiated. Germination rate and early growth of soybean were high in 4Gy-irradiated group. Perilla gew ot so promisingly after after low dose irradiation, however there slightly increasing effects on germination rate, seeding height and fresh weightat 2Gt-, 1Gy-, and 1Gy irradiated group, respectively.
This study was carried out to establish optimal conditions for breaking dormancy of Agastache rugosa O. Kuntze seeds. A series of experiments according to seed maturity and treatment with plant growth regulators were performed to improve germination percentage and synchronize germination of the seeds. In addition, it was conducted to test whether the useful effect of seed treatment before sowing leads to healthy seedling and early vigorous growth. The average seed size was 1.85 mm (length) × 0.82 mm(width). The seed size was much smaller than other vegetable seeds. Seeds colorappeared dark brown, the shape of the seeds was oval and the weight of 1,000seeds was 352.8 mg. The optimum germination temperature was 22℃. Light exposure during germination did not affect germination promotion, suggesting that A. rugosa seeds are a kind of dark germinating seeds. Seed dormancy lasted for 40 days after harvesting, and GA3 treatment of dormant seeds could break dormancy. There were significant differences in germination percentage and rate according to the maturity of seeds. The germination percentage of mature seeds was 10 - 18% higher than that of immature seeds, and germination rate was 2 days faster. GA3 treatment during growth regulator treatment improved germinability, but BAP or ethephone treatment did not. The optimal growth regulator concentration of for germination was the combination treatment of 100 mM GA3 + 100 mM BAP.
The effect of salinity on the seed germination and seedling growth of Pinus densiflora for. erecta Uyeki were studied under the controlled conditions in the growth chamber. The seeds were sorted into three classes in weight such as large ($15.49\pm$1.63mg), medium($10.61\pm$1.38mg), and small ($6.57\pm$1.33mg) to determine the role of seed weight in germination and seedling growth of the pine. Polymorphic seeds of the pine were germinated an salinity range of 0 to 1.5% NaCl under various temperature ($10^{\circ}$$-25^{\circ}$ with $5^{\circ}$C interval of constant temperature, and $10^{\circ}$$-20^{\circ}$C, $15^{\circ}$$-25^{\circ}$C of alternating temperature) in order to determine their germinability and seeding growth. In control plot, there was little difference of germination percentage among the seed weight classes, but in saline plot, the larger seeds generally had a higher percentage and rate of germination. There occurred synergistic interaction between salinity and temperature in the germination and the increase of temperature enhanced germination of seeds at the same salinity level. Alternating temperature regimes of $15^{\circ}$-$25^{\circ}$C yielded maximum germination and no germination was occurred at $10^{\circ}C$. The germination at alternating temperature showed higher germination percentage than at constant temperature. The percentage and of germination decreased drastically with increased salinity level to 1.00%; no germination was occurred at 1.50% salinity level. The growth fo sddelings from larger seeds was better than that of smaller seeds at the same salinity and temperature. The hypocotyl and radicle were more sensitive than cotyledon to the increased salinity stress.
We checked the emergence rate and early growth characteristics of young rice plants according to the germination length and seed damage ratio in direct seeding. The emergence rate was reduced according to the longer germination length and higher seed damage ratio. The emergence rate was further reduced under high-temperature conditions. The leaf number did not show a significant difference according to germination length and seed damage ratio. However, shoot and root dry weights were reduced according to the germination length and seed damage ratio. The root dry weight was highly correlated with the shoot dry weight. Based on these results, we concluded that a longer germination length and seed damage could induce the loss of nutrients and affect poor root growth, consequently inducing poor emergence and growth in the early growth stage of rice seeds. Therefore, in rice direct seeding, seed conditions of less than 1 mm of germination length should be checked for a sufficiently high standing rate and adequate growth characteristics after seeding.
In the study, the feasibility of deposit soil in river and stream bed as a subsitute for conventional plant soil was investigated through the analysis of soil characteristics and germination/growth rate for 75 days. Proper mixtures among sediments from various places were compared to optimize the germination and growth rate of plant in the ratio of 2 : 1 and 3 : 1 (sand : deposit). From the results, it could be concluded that the sediment mixed 3 : 1 showed most favorable germination and growth conditions for pansy and the sediment containing enough amount of silt and clay components showed most favorable conditions for marigold. Consequently, the feasibility of sediments as a substitutional plant soil was evaluated to be high, once the sediments were properly mixed for each specific plant.

This study was conducted to investigate the quality of seeds, the germination rates and the days required for germination, to examine the patterns of protein expressions during the germination and to improve the techniques of managing and storing seeds and viability of the seeds of Lithospermum erythrorhizon Sieb. et Zucc. After collecting and harvesting seeds, they were classified to white and brown colors of seed coat through testing their seed size, weight, and quality. The germination rates, the days required for germination, and the protein expressions were examined with different colors of seed coats, storing temperatures and durations by treating the different plant growth regulators and primings. One hundred seed weight of white color was heavier about 1.17 g than those of brown one about 0.81 g. The germination rates in white color of seed coat was higher, 3.05 ~ 5.75%, than those in brown one. Its rates were decreased with getting longer in storage durations. There was no big differences on germination rates between storage temperatures. The plant growth regulator of $GA_3$ and Kinetin was affected to improve the seed germination. $GA_3$ increased the seed germination clearly at 25 ppm level, while kinetin increased it gradually from 25 to 100 ppm levels. In germination by seed primings, PEG6000 made higher germination rate with increasing their levels, whereas $KNO_3$ increased the germination until 100 mM level and then decreased it with 200 mM unlike PEG6000. The protein expressed during the seed germination were appeared more and clearer bands in the seed after germination, especially 20 ~ 30 kDa, compared to those in the seed before germination. These results showing more and clearer bands were positively related to the germination rates which were different by seed colors, storage temperatures and durations, and plant growth regulators and primings.

This study was conducted to improve the postharvest storage techniques of managing and storing seeds, to test qualities and viabilities of the seeds and to examine the germination rate and the protein expression of Achyranthes japonica Nakai. The seeds collected from different areas of Je-Cheon and Gwang-Ju were stored with different temperatures and durations. Two plant growth regulators and two seed priming were treated to investigate their effect on the germination rates and the days required for germination. The weight of one hundred seed collected in Gwang-Ju was heavier than those in Je-Cheon. Seed length collected in Gwang-Ju was also longer about 5.12 mm than those in Je-Cheon about 4.90 mm and seed width was longer in Gwang-Ju than those in Je-Cheon. The rates of seed germination in two different collection areas were higher about 2.9 to 13.0% in Gwang-Ju compared to those in Je-Cheon. Comparing its rates with the storing temperatures and durations, they were not clearly different in between $4^{\circ}C$ and $25^{\circ}C$ and they also were gradually decreased with getting longer storing durations. The germination rates treated by plant growth regulators were higher with $GA_3$ than those with Kinetin. The highest seed germination rate was appeared at 50 ppm of $GA_3$. Comparing its rates with different seed priming, they were relatively higher with $KNO_3$ than those with PEG6000. In protein expression patterns between before the germinating and after the germinating of seeds, more and clear bands were appeared in the seed after the germination compared to those before the germination of seeds, especially 10 ~ 20 kDa. These results showing more and clear bands were more clearly appeared in Gwang-Ju compared to Je-Cheon. Comparing the protein expression with plant growth regulators and seed primings, $GA_3$ was better expression than those with Kinetin and $KNO_3$ was better than those with PEG6000. More and clear bands were closely related to the germination rates of seeds and more detailed studies would be required.
