중금속 오염이 양서류 시원생식세포 발생에 미치는 영향 (Effects of the Heavy Metal Pollution on the Primordial Germ Cells of Developing Amphibia)

  • 양서류 수정란을 제 1 난할전에 중금속으로 처리하였던바 개구리의 시원생식세포수의 양전변화를 가져 왔다. 중금속의 일정량의 처리는 양서류 초기발생중에 시원생식세포 형성을 저하시켰다. 납 70ppm과 카드뮴 4ppm 이상에서 배양하면 9내지 12mm배에서 시원생식세포의 수적감소를 가져 왔으나 그후의 발생에서는 거의 정상군과같은 증가율을 보였다. 한편 수은 0.8ppm이상의 처리군에서는 생식 세포형성에 보다 심한 저하를 가져 왔으며 그후의 증가율도 대체로 낮았다. 위의 사실들은 자외선 조사 실험에서 얻은 결과와는 달리 양서류란에 있어서 중금속 처리는 조직에 심한 손상을 주는 다량투여에도 시원생식세포의 완전 배제에는 효과가 적다는 것을 암시한다.

YY1 and CP2c in Unidirectional Spermatogenesis and Stemness

  • Spermatogonial stem cells (SSCs) have stemness characteristics, including germ cell-specific imprints that allow them to form gametes. Spermatogenesis involves changes in gene expression such as a transition from expression of somatic to germ cell-specific genes, global repression of gene expression, meiotic sex chromosome inactivation, highly condensed packing of the nucleus with protamines, and morphogenesis. These step-by-step processes finally generate spermatozoa that are fertilization competent. Dynamic epigenetic modifications also confer totipotency to germ cells after fertilization. Primordial germ cells (PGCs) in embryos do not enter meiosis, remain in the proliferative stage, and are referred to as gonocytes, before entering quiescence. Gonocytes develop into SSCs at about 6 days after birth in rodents. Although chromatin structural modification by Polycomb is essential for gene silencing in mammals, and epigenetic changes are critical in spermatogenesis, a comprehensive understanding of transcriptional regulation is lacking. Recently, we evaluated the expression profiles of Yin Yang 1 (YY1) and CP2c in the gonads of E14.5 and 12-week-old mice. YY1 localizes at the nucleus and/or cytoplasm at specific stages of spermatogenesis, possibly by interaction with CP2c and YY1-interacting transcription factor. In the present article, we discuss the possible roles of YY1 and CP2c in spermatogenesis and stemness based on our results and a review of the relevant literature.

Germ Cell Development during Spermatogenesis and Taxonomic Values of Sperm Morphology in Septifer (Mytilisepta) virgatus (Bivalvia: Mytilidae)

  • Spermatogenesis and taxonomic values of mature sperm morphology of in male Septifer (Mytilisepta) virgatus were investigated by transmission electron microscope observations. The morphologies of the sperm nucleus and the acrosome of this species are the cylinder shape and cone shape, respectively. Spermatozoa are approximately 45-50 ${\mu}m$ in length including a sperm nucleus (about 1.26 ${\mu}m$ long), an acrosome (about 0.99 ${\mu}m$ long), and tail flagellum (about 45-47 ${\mu}m$). Several electron-dense proacrosomal vesicles become later the definitive acrosomal vesicle by the fusion of several Golgi-derived vesicles. The acrosome of this species has two regions of differing electron density: there is a thin, outer electron-dense opaque region (part) at the anterior end, behind which is a thicker, more electron-lucent region (part). In genus Septifer in Mytilidae, an axial rod does not find and also a mid-central line hole does not appear in the sperm nucleus. However, in genus Mytilus in Mytilidae, in subclass Pteriomorphia, an axial rod and a mid-central line hole appeared in the sperm nucleus. These morphological differences of the acrosome and sperm nucleus between the genuses Septifer and Mytilus can be used for phylogenetic and taxonomic analyses as a taxonomic key or a significant tool. The number of mitochondria in the midpiece of the sperm of this species are five, as seen in subclass Pteriomorphia.

징거미새우, Macrobrachium nipponense 정소 구성세포의 미세구조와 기능 (Ultrastructure and Role of Somatic Cells in Macrobrachium nipponense Testis)

  • 징거미새우의 정소는 간췌장의 배면 측상부에서 심장사이에 위치하며, 한 쌍의 소엽으로 이루어져 있고 각 소엽의 선단은 서로 연결되어 있다. 그리고 각 소엽은 수많은 세정관이 결합조직으로 연결되어 있다. 각 세정관은 생식세포와 지주세포가 밀착되어 있는 생식세포대를 제외한 부분은 낮은 입방상피에 의해 내강이 형성되어 있고, 세정관의 제일 바깥은 단층편평상피로 둘러싸여 있다. 세정관 사이에는 결합조직 외에 Leydig cell-like cell도 관찰된다. 단층편평상피세포는 얇은 측면으로 연결되어 있고 종종 이웃한 세포와 포개져 있다. 지주세포는 생식세포에 비해 그 수가 현저히 적고, 대부분 생식세포대의 가장자리에 위치하고 있다. 지주세포의 세포질은 기저판과 접해 있어 기저면과 생식세포 사이는 지주세포의 원형질에 의해 분리되어 있다. 지주세포의 핵은 대부분 각져 있고, 크고 현저한 인이 핵의 중심부에 위치하고 있다. 낮은 입방상피세포는 세정관의 기저판과 접하고 있는 기저면과 일부는 생식세포대와 내강 사이에 위치하고 있다. 입방상피세포의 세포질에서는 횡 방향의 크리스테가 발달된 미토콘드리아, 층상구조의 조면소포체 그리고 Golgi 복합체들이 매우 발달되어 있다. 그리고 조면소포체와 Golgi 복합체의 볼록한 형성면 사이에는 전이소낭이, Golgi 복합체의 성숙면에서는 분비소낭이 관찰되며, 상피세포의 선단에 수많은 홈들이 존재하는 점으로 보아 exocytosis에 의해 내강으로의 물질 분비가 이루어지는 것으로 판단된다.

Ultrastructures of Germ Cells and the Accessory Cells During Spermatogenesis in Male Gomphina veneriformis (Bivalvia: Veneridae) on the East Sea of Korea

  • The ultrastructures of germ cells and the accessory cells during spermatogenesis and mature sperm ultrastructure in male Gomphina veneriformis, which was collected on the coastal waters of Yangyang, East Sea of Korea, were investigated by transmission electron microscope observations. The morphology of the spermatozoon has a primitive type and is similar to those of other bivalves in that it contains a short midpiece with four mitochondria surrounding the centrioles. Accessory cells are observed to be connected to adjacent germ cells, they contain a large quantity of glycogen particles and lipid droplets in the cytoplasm. Therefore, it is assumed that they are involved in the supplying of the nutrients for germ cell development, while any phenomena associated with phagocytosis of undischarged, residual sperms by lysosomes in the cytoplasm of the accessory cells after spawning was not observed in this study. The morphologies of the sperm nucleus type and the acrosome shape of this species have a cylindrical and modified long cone shape, respectively. In particular, the axial filaments in the lumen of the acrosome, and subacrosomal granular materials are observed in the subacrosomal space between the anterior nuclear fossa and the beginning part of axial filaments in the acrosome. The spermatozoon is approximately $50-55{\mu}m$ in length including a long sperm nucleus (about $7.80{\mu}m$ in length), an acrosome (about $1.13{\mu}m$ in length) and tail flagellum ($40-45{\mu}m$). The axoneme of the sperm tail flagellum consists of nine pairs of microtubules at the periphery and a pair at the center. The axoneme of the sperm tail shows a 9+2 structure. Some charateristics of sperm morphology of this species in the family Veneridae are (1) acrosomal morphology, (2) the number of mitochondria in the midpiece of the sperm,. The axial filament appears in the acrosome as one of characteristics seen in several species of the family Veneridae in the subclass heterodonta, unlikely the subclass pteriomorphia containing axial rod instead of the axial filament. As some characteristics of the acrosome structures, the peripheral parts of two basal rings show electron opaque part (region), while the apex part of the acrosome shows electron lucent part (region). These charateristics belong to the family Veneridae in the subclass heterodonta, unlikely a characteristic of the subclass pteriomorphia showing all part of the acrosome being composed of electron opaque part (region). Therefore, it is easy to distinguish the families or the subclasses by the acrosome structures. The number of mitochondria in the midpiece of the sperm of this species are four, as one of common characteristics appeared in most species in the family Veneridae.

Expression Patterns of Germ Cell-specific Phosducin-like 2 during Testicular and Ovarian Development in Chickens

  • Phosducin (PDC) is a photoreceptor cell-specific protein that is phosphorylated by cyclic nucleotide-dependent protein kinase. PDC and PDC-like proteins (PDCL, PDCL2, and PDCL3) are members of a conserved family of small thioredoxin-like proteins that modulate the ${\beta}$- and ${\gamma}$-subunits of G-proteins. In mammals, Pdc, Pdcl, and Pdcl3 genes show ubiquitous expression; however, Pdcl2 gene expression is limited to the testis and ovary. The aim of the present study was to examine the expression patterns of chicken Pdcl2 (cPdcl2) during testicular and ovarian development. Protein sequence comparisons performed using the CLUSTAL X program revealed that the amino acid sequences and potential phosphorylation sites of cPDCL2 and mammalian PDCL2 proteins were highly conserved. Quantitative real-time PCR analysis revealed that cPdcl2 was differentially expressed in the testis and ovary. Specifically, cPdcl2 expression was detected at low levels in the ovary at all time points. In the testis, cPdcl2 expression was detected at low levels until 5 weeks of age. At 8 weeks of age, however, cPdcl2 showed increased expression levels in the testis. Using in situ hybridization, we detected high levels of cPdcl2 expression in the testis, particularly in the spermatocytes and round spermatids. In summary, our data describe expression patterns of germ cell-specific Pdcl2 during testicular and ovarian development in chickens.

Deficiency of Bloom's Syndrome Protein Causes Hypersensitivity of C. elegans to Ionizing Radiation but Not to UV Radiation, and Induces p53-dependent Physiological Apoptosis

  • Caenorhabditis elegans him-6 mutants, which show a high incidence of males and partial embryonic lethality, are defective in the orthologue of human Bloom's syndrome protein (BLM). When strain him-6(e1104) containing a missense him-6 mutation was irradiated with ${\gamma}$-rays during germ cell development or embryogenesis, embryonic lethality was higher than in the wild type, suggesting a critical function of the wild type gene in mitotic and pachytene stage germ cells as well as in early embryos. Even in the absence of ${\gamma}$-irradiation, apoptosis was elevated in the germ cells of the him-6 strain and this increase was dependent on a functional p53 homologue (CEP-1), suggesting that spontaneous DNA damage accumulates due to him-6 deficiency. However, induction of germline apoptosis by ionizing radiation was not significantly affected by the deficiency, indicating that HIM-6 has no role in the induction of apoptosis by exogenous DNA damage. We conclude that the C. elegans BLM orthologue is involved in DNA repair in promeiotic cells undergoing homologous recombination, as well as in actively dividing germline and somatic cells.

넙치, Paralichthys olivaceus 자어의 원시생식소와 신장의 발달에 미치는 Polychlorinated Biphenyls (PCBs)의 영향 (Effect of PCBs on the Primitive Gonad and Kidney Development of the Larval Stage of Olive Flounder, Paralichthys olivaceus in Culture Farm)

  • 양식산 넙치를 대상으로 아치사 만성농도의 PCBs가 자어기의 원시생식소와 신장의 발달에 미치는 영향을 살펴보았다. 원시생식소 발달 과정은 4단계; 원시생식세포(primodial germ cell, PGC) 단계, 생식융기 (genital ridge type, GRT) 단계, 원시생식소 I형(primitive gonad type I, PGT I) 단계 및 원시생식소 II형(primitive gonad type II, PGT II) 단계로 구분할 수 있었다. 시원생식세포는 부화 후 3일에 발견되었고, 원시 생식소는 대조구에서 부화 후 24일에 나타나며 처리구에서는 부화 후 22일에 나타났다. 신장 발달과정은 4단계; 단일관형 중신관(unitubular type of mesonephric duct, UTMD) 단계, 분지형 중신관(the branched mesonephric duct, BMD) 단계, 세뇨관 형성(convoluted tubule formation CTF) 단계 및 사구체 출현(glomerulus appearance, GA) 단계로 구분할 수 있었으며, 신장의 구조적 완성시기는 대조구와 처리구 모두 부화 후 25∼30일로 나타났다. 하지만 원시 생식소와 신장 모두 대조구와 처리구에서 유의한 차를 보이지 않았다 (P>0.05). 그러므로, 넙치 자어기의 원시 생식소와 신장의 발달에 따른 PCBs(3.0 $\mu\textrm{g}$/L)의 영향은 없었다.
