Immunogenicity and Safety of Inactivated Influenza Vaccine in Healthy Korean Children and Adolescent (한국의 건강한 소아청소년을 대상으로 한 인플루엔자 사백신의 면역원성과 안전성 연구)

  • Purpose: This study aimed to evaluate the immunogenicity and safety of a trivalent inactivated influenza vaccine (TIV) among healthy Korean children and adolescents. Methods: From October to December 2008, 65 healthy patients aged 6 months to 18 years who visited Korea University Ansan Hospital for influenza vaccination were enrolled in this study. We measured the hemagglutinin inhibition antibody titers at baseline and 30 days after vaccinating enrollees with split influenza vaccine and calculated the seroprotection rates, geometric mean titers, and seroconversion rates. Local and systemic adverse events were assessed after vaccination. Results: The seroprotection rates against all three viral strains (A/H1N1, A/H3N2, B) were 87.7%, 89.2%, and 89.2% (${\geq}70%$), respectively; seroconversion rates were 44.6%, 73.8%, and 63.1% (${\geq}40%$), respectively; and seroconversion factors were 4.5, 8.4, and 10.5 (>2.5), respectively. The TIV immunogenicity was acceptable according to the CPMP (Committee for Proprietary Medicinal Products) criteria. Although 48 patients (73.8%) reported one or more adverse events, no severe adverse events such as anaphylaxis and convulsion were observed. Forty-two patients (64.6%) reported a local skin reaction, including redness (29.2%), pain (43.1%), or swelling (41.5%) of the injected site, and 26 (40.0%) reported a systemic reaction: fatigue (23.1%), myalgia (20.0%), headache (10.8%), arthralgia (10.8%), chills (9.2%), or fever (7.7%). Conclusions: This study shows that the immunogenicity of the TIV vaccine is acceptable. As there were no serious adverse events aside from local reactions and mild systemic reactions, this vaccine can be safely used among healthy Korean children and adolescents.

Evaluation of Ecological quality and establishment of ecological restoration guideline in landscape level of Mt. Moodeung National Park (무등산국립공원의 생태적 질 평가 및 복원 가이드라인 수립)

  • Ecological restoration is an eco-technology, which heals the nature damaged by human activity by imitating organization and function of the integrate nature and thereby provide an inhabitable space for diverse organisms. Such an ecological restoration has to be carried out by applying restoration plan prepared based on the results of diagnostic evaluation discussed in the diversified respects. This study aims to prepare an ecological restoration plan of the damaged forest ecosystem in Mt. Moodeung National Park. To arrive at the goal, first of all, we diagnosed quality of forest landscape established in Mt. Moodeung National Park based on natural (topography, climate, and distribution of vegetation) and artificial (land use, linear landscape element) factors. In addition, we evaluated the integrity of each zone divided by linear landscape element quantitatively based on geometric property and land use intensity. As the result of analysis, topography of Mt. Moodeung National Park tended to be depended on weathering property of parent rock and vegetation zones were divided to three vegetation zones. Based on land use pattern, deciduous broad-leaved forest, evergreen needle-leaved forest, and mixed forest occupied about 90% of Mt. Moodeung National Park. Mean score of forest landscape quality was shown in $69.86{\pm}11.41$. As a result, forest landscape elements in Mt. Moodeung National Park were influenced greatly by human activity and the degree was depended on topographic condition. This study suggested the synthetic restoration plan to improve ecological quality of Mt. Moodeung National Park based on the results of diagnostic evaluation.

Evaluation of the Bacteriological and Toxicological Safety for the Shellfish Growing Area in the Kamakman Area, Korea (가막만해역 패류의 세균학적·독물학적 안전성 평가)

  • To evaluate bacteriological and toxicological safety, the hygienic indicator bacterium and paralytic and diarrhetic shellfish toxins in the shellfish produced in the Kamakman Area from 2012 to 2016 were investigated. Fecal coliforms and E. coli of all 194 oyster samples tested did not exceed 230 MPN/100 g. The geometric mean of the fecal coliform analyzed with the oyster samples of harvesting period was 19.6 MPN/100 g, which was more stable than the non-harvesting period (26.5 MPN/100 g). For the toxicological evaluation of the Kamakman Area, 77 oyster samples and 350 mussel samples as an indicator were analyzed. Paralytic shellfish toxins were detected very low in the range of $40{\sim}46{\mu}g$/100 g in 13 mussel samples during late April and early June, but not in oyster samples. Diarrhetic shellfish toxin was detected in 2 of 180 samples, but it was found to be below the regulation value (0.16 mg OA equ./kg). Based on the bacteriological studies, it was confirmed that the shellfish produced in Kamakman area meets the standard of shellfish hygiene of the Food Sanitation Act and meets the Grade A of the shellfish production area of EU. As the results of the paralytic and diarrhetic shellfish toxin evaluation, it was confirmed that the Kamakman Area is also toxicologically safe for shellfish production.

  • The duplex system has been considered as an important slip-transfer mechanism to evaluate the evolution of orogenic belts. Duplexes are generally found in the hinterland portion of fold-thrust belts and accommodate large amounts of total shortening. Thus, understanding its geometric and kinematic evolution can give information to evaluate the evolution of the entire orogenic belt. Duplexes are recognized as closed-loop thrust traces on map view, indicating higher connectivity than imbricate fans. As originally defined, a duplex is an array of thrust horses which are surrounded by thrust faults including the floor and roof thrusts, and imbricate faults between them. Duplexes can accommodate regional layer-parallel shortening and transfer slip from a floor thrust to a roof thrust. However, an imbricate fault is not the only mean for layer-parallel shortening (LPS) and displacement transfer within duplexes. LPS cleavages and detachment folds can also play the same role. From this aspect, a duplex can be divided into three types; 1) fault duplex, 2) cleavage duplex and 3) fold duplex. Fault duplex can further be subdivided into the Boyer-type duplex, which was firstly designed duplex system in the 1980s that widely applied most of the major fold-thrust belts in the world, and connecting splay duplex, which has different time order in the emplacement of horses from those of the Boyer-type. On the contrary, the cleavage and fold duplexes are newly defined types based on some selected examples. In the Korean Peninsula, the Yeongwol area, the western part of the Taebaeksan Zone of the Okcheon Belt, gives an excellent natural laboratory to study the structural geometry and kinematics of the closed-loops by thrust fault traces in terms of a duplex system. In the previous study, the Yeongwol thrust system was interpreted by alternative duplex models; a Boyer-type hinterland-dipping duplex vs. a combination of major imbricate thrusts and their connecting splays. Although the high angled beds and thrusts as well as different stratigraphic packages within the horses of the Yeongwol duplex system may prefer the later complicate model, currently, we cannot choose one simple answer between the models because of the lack of direct field evidence and time information. Therefore, further researches on the structural field investigations and geochronological analyses in the Yeongwol and adjacent areas should be carried out to test the possibility of applying the fold and cleavage duplex models to the Yeongwol thrust system, and it will eventually provide clues to solve the enigma of formation and its evolution of the Okcheon Belt.

Estimation of Soil Moisture Using Sentinel-1 SAR Images and Multiple Linear Regression Model Considering Antecedent Precipitations (선행 강우를 고려한 Sentinel-1 SAR 위성영상과 다중선형회귀모형을 활용한 토양수분 산정)

  • This study is to estimate soil moisture (SM) using Sentinel-1A/B C-band SAR (synthetic aperture radar) images and Multiple Linear Regression Model(MLRM) in the Yongdam-Dam watershed of South Korea. Both the Sentinel-1A and -1B images (6 days interval and 10 m resolution) were collected for 5 years from 2015 to 2019. The geometric, radiometric, and noise corrections were performed using the SNAP (SentiNel Application Platform) software and converted to backscattering coefficient of VV and VH polarization. The in-situ SM data measured at 6 locations using TDR were used to validate the estimated SM results. The 5 days antecedent precipitation data were also collected to overcome the estimation difficulty for the vegetated area not reaching the ground. The MLRM modeling was performed using yearly data and seasonal data set, and correlation analysis was performed according to the number of the independent variable. The estimated SM was verified with observed SM using the coefficient of determination (R2) and the root mean square error (RMSE). As a result of SM modeling using only BSC in the grass area, R2 was 0.13 and RMSE was 4.83%. When 5 days of antecedent precipitation data was used, R2 was 0.37 and RMSE was 4.11%. With the use of dry days and seasonal regression equation to reflect the decrease pattern and seasonal variability of SM, the correlation increased significantly with R2 of 0.69 and RMSE of 2.88%.

Granite Dike Swarm and U-Pb Ages in the Ueumdo, Hwaseong City, Korea (경기도 화성시 우음도 일원의 화강암 암맥군과 U-Pb 연령)

  • The Middle Jurassic granite dike swarm intruding into the Paleoproterozoic banded gneiss is pervasively observed in Ueumdo, Hwaseong City, mid-western Gyeonggi Massif. Based on their cross-cutting relationships in a representative outcrop, there are four dikes (UE-A, UE-C, UE-D, UE-E), and depending on the direction, there are three granite dike groups, which are NW- (UE-A dike), NW to WNW- (UE-C dike), and NE-trending (UE-D and UE-E dikes). These granite dikes are massive, medium-to coarse-grained biotite granites, and their relative ages observed in outcrops are in the order of UE-A, UE-D (=UE-E), and UE-C. The geometric analysis of the dikes indicates that the UE-A and UE-C dikes intrude under approximately NE-SW trending horizontal minimum stress fields. The UE-A dike, which showed a relatively low average SiO2 content by major element analysis, is a product of early magma differentiation compared to other dikes; therefore, it is consistent with the relative age of each dike. The 206Pb/238U weighted mean ages for each dike obtained from SHRIMP zircon U-Pb dating were calculated to be 167 Ma (UE-A), 164 Ma (UE-C), 167 Ma (UE-D), and 167 Ma (UE-E), respectively. The samples of the UE-A, UE-D, and UE-E dikes showed very similar ages. The UE-C dike shows the youngest age, which is consistent with the results of the relative age in the outcrops and major element analysis. Therefore, the granite dikes intruded into the Middle Jurassic (approximately 167 and 164 Ma), coinciding with those of the Gyeonggi Massif, where the Middle Jurassic plutons are geographically widely distributed. This result indicates that the wide occurrence of the Middle Jurassic plutons on the Gyeonggi Massif was formed as a result of igneous activity moving in the northwest direction with the shallower subduction angle of the subducting oceanic plate during the Jurassic.

Immunogenicity and Safety of a Haemophilus influenzae Type b Polysaccharide-Tetanus Toxoid Conjugate Vaccine (PRP-T: HiberixTM) in Korean Infants (우리나라 영아에서 PRP-T(HiberixTM)백신의 면역원성 및 안전성에 대한 연구)

  • Purpose : Four kinds of Haemophilus influenzae type b protein conjugate vaccines, PRPD, PRP-T, PRP-OMP and PRP-CRM197, have been developed, and PRP-T vaccines are currently produced by two manufacturer, $ActHib^{(R)}$ by Aventis and $Hiberix^{TM}$ by GlaxoSmith-Kline Biologicals. The purpose of this study is to evaluate the immunogenicity and safety of $Hiberix^{TM}$ in Korean infants. Methods : Seventy-three healthy infants(43 male infants) were recruited for this study after parental informed consent was obtained. Each infant was vaccinated at 2, 4 and 6 months of age with the study vaccine. At each visit, infants were also immunized with DTaP, trivalent oral polio vaccine and hepatitis B vaccine when indicated. The serum anti-PRP antibody was measured at prevaccination, 2 month later after the 2nd dose, and 1 month later after the 3rd dose by the ELISA method. The local and systemic adverse reactions of vaccination were monitored for 3 consecutive days after each immunization. Immunogenicity of vaccine was evaluated in infants who received all the scheduled immunization and the adverse reactions were evaluated for infants who received at least one dose of the study vaccine. Results : Among seventy three infants, enrolled in this study; sixty three(37 male infants) completed all the scheduled immunizations. The geometric mean titer(GMT) of anti-PRP antibodies at prevaccination was 0.17 ${\mu}g/mL$(95% confidence interval[CI]; 0.13~0.22). The GMT of anti-PRP antibodies increased to 4.14 ${\mu}g/mL$(95% CI; 2.65~6.48) at 2 month later after the 2nd dose of PRP-T and 14.65 ${\mu}g/mL$(95% CI; 10.83~19.81) at 1 month later after the 3rd dose. Anti-PRP antibody ${\geq}0.15$ ${\mu}g/mL$, was observed in 98.4%(95% CI; 91.8~100) after 2 doses and 100%(95% CI; 100~100) after 3 doses. Anti-PRP antibody ${\geq}1.0$ ${\mu}g/mL$, was obtained in 77.8%(95% CI; 67.5~88.0) after 2 doses, and 98.4%(95% CI; 95.3~100) after 3 doses. Most of the adverse reaction after vaccination were mild. Irritability, the most common systemic reaction, was observed in 45.5%, followed by drowsiness(30.5%), poor feeding(26.7%) and fever(5.6%). Among the local reactions tenderness was observed in 7.9%, redness(${\geq}5$ mm) in 2.8% and swelling(${\geq}5$ mm) in 1.8%. Conclusion : The PRP-T vaccine used in this study was highly immunogenic and safe in Korean young infants. The finding that high GMT and high frequency of infants with a protective titer achieved after 2 doses is consistent with the previous studies which were done with a PRP-T vaccine of other manufacturer. This study suggests that the immunization schedule of PRP-T vaccine for Korean infants may need re-evaluation.

The Safety and Immunogenicity of a Trivalent, Live, Attenuated MMR Vaccine, PriorixTM (MMR(Measles-Mumps-Rubella) 약독화 생백신인 프리오릭스주를 접종한 후 안전성과 유효성의 평가에 관한 연구)

  • Purpose : This multi-center, open-label, clinical study was designed to evaluate the safety and immunogenicity of a trivalent, live, attenuated measles-mumps-rubella(MMR) vaccine, $Priorix^{TM}$ in Korean children. Methods : From July 2002 to February 2003, a total of 252 children, aged 12-15 months or 4-6 years, received $Priorix^{TM}$ at four centers : Han-il General Hospital, Kyunghee University Hospital, St. Paul's Hospital at the Catholic Medical College in Seoul, and Korea University Hospital in Ansan, Korea. Only subjects who fully met protocol requirements were included in the final analysis. The occurrence of local and systemic adverse events after vaccination was evaluated from diary cards and physical examination for 42 days after vaccination. Serum antibody levels were measured prior to and 42 days post-vaccination using IgG ELISA assays at GlaxoSmithKline Biologicals (GSK) in Belgium. Results : Of the 252 enrolled subjects, a total of 199 were included in the safety analysis, including 103 from the 12-15 month age group and 96 from the 4-6 year age group. The occurrence of local reactions related to the study drug was 10.1 percent, and the occurrence of systemic reactions was 6.5 percent. There were no episodes of aseptic meningitis or febrile convulsions, nor any other serious adverse reaction. In immunogenicity analysis, the seroconversion rate of previously seronegative subjects was 99 percent for measles, 93 percent for mumps and 100 percent for rubella. Both age groups showed similar seroconversion rates. The geometric mean titers achieved, 42 days pos-tvaccination, were : For measles, in the age group 12-15 months, 3,838.6 mIU/mL [3,304.47, 4,458.91]; in the age group 4-6 years, 1,886.2 mIU/mL [825.83, 4,308.26]. For mumps, in the age group 12-15 months, 956.3 U/mL [821.81, 1,112.71]; in the age group 4-6 years, 2,473.8 U/mL [1,518.94, 4,028.92]. For rubella, in the age group 12-15 months, 94.5 IU/mL [79.56, 112.28]; in the age group 4-6 years, 168.9 IU/mL [108.96, 261.90]. Conclusion : When Korean children in the age groups of 12-15 months or 4-6 years were vaccinated with GlaxoSmithKline Biologicals' live attenuated MMR vaccine ($Priorix^{TM}$), adverse events were limited to those generally expected with any live vaccine. $Priorix^{TM}$ demonstrated excellent immunogenicity in this population.

  • Purpose : To reduce the number of injections necessary to vaccinate young infants, various combined vaccines have been developed. The $Comvax^{TM}$ manufactured by Merck & Co. is a combination of Hepatitis B and PRP-OMP conjugate Haemophilus influenzae Type b vaccine. The purpose of this study is to evaluate the immunogenicity and safety of $Comvax^{TM}$ in Korean infants. Methods : The infants who were vaccinated at 0 months of age with Hepatitis B vaccine, were recruited for this study after parental informed consent was obtained. The subjects were vaccinated with $Comvax^{TM}$ at 2 and 4 months of age. At each visit, infants were also immunized with DTaP, inactivated poliovirus vaccine, and pneumococcal vaccine when indicated. The serum anti-PRP and anti-HBs were measured at 2 months after the 1st dose(4 months age), and the 2nd dose(6 months age) by the ELISA and chemiluminescent microparticle immunoassay method, respectively. The local and systemic adverse reactions of vaccination were monitored for 3 consecutive days after each immunization. Results : Among sixty-five healthy infants(35 male infants) enrolled in this study; fifty eight(32 male infants) completed the scheduled immunizations. The geometric mean titers (GMTs) of anti-PRP at 2 months after the 1st dose and the 2nd dose were 1.96 ${\mu}g/mL$ (95% CI; 1.38~2.78) and 10.02 ${\mu}g/mL$ (95% CI; 7.04~14.26), respectively. Anti-PRP ${\geq}1.0$ ${\mu}g/mL$, was obtained in 63.2%(95% CI; 53.75~72.65) after 1 dose, and 96.6%(95% CI; 93.05~100) after 2 doses. The GMTs of anti-HBs were 38.32 mIU/mL(95% CI; 22.42~65.51), and 101.17 mIU/mL(95% CI; 65.94~155.25) at 2 month after the 1st dose and 2nd dose of $Comvax^{TM}$, respectively. Anti-HBs ${\geq}10$ mIU/mL was observed in 73.7%(95% CI; 65.07~82.33) after 1 dose and 94.8%(95% CI; 90.45~99.15) after 2 doses. Most of the adverse reactions after vaccination were mild. Irritability, the most common systemic reaction, was observed in 24.8%, followed by drowsiness(19.2%), poor feeding(19.2%) and fever(7.2%). Among the local reactions tenderness was observed in 25.6%, redness(${\geq}5$ mm) in 19.2% and swelling(${\geq}5$ mm) in 4.8%. Conclusion : The $Comvax^{TM}$ vaccine was highly immunogenic for PRP and safe in Korean infants. Although the hepatitis B vaccine component was administered at 0, 2, 4 months, this study showed good immunogenicity against HBsAg.
