Culturable Fungal Endophytes Isolated from the Roots of Coastal Plants Inhabiting Korean East Coast

  • Twelve plant species were collected from the east coast of Korea to identify culturable endophytes present in their roots. The fungal internal transcribe spacer (ITS) region (ITS1-5.8SrRNA-ITS2) was used as a DNA barcode for identification of fungi. A total of 194 fungal strains were identified and categorized into 31 genera. The genus Penicillium accounted for the largest number of strains, followed by the genus Aspergillus. Furthermore, using 5 statistical methods, the diversity indices of the fungi were calculated at the genus level. After comprehensive evaluation, the endophytic fungal group from Phragmites australis ranked highest in diversity analyses. Several strains responsible for plant growth and survival (Penicillium citrinum, P. funiculosum, P. janthinellum, P. restrictum, and P. simplicissimum), were also identified. This study provides basic data on the sheds light on the symbiotic relationship between coastal plants and fungi.

A new distribution of Dalbergia hupeana Hance (Fabaceae) in Korea and its taxonomic characteristics (우리나라 황단나무의 신분포 및 분류학적 특징)

  • A new distribution of Dalbergia hupeana has been reported at Mt. Yudal in Mokpo-si, Korea. It was previously thought to be restricted to China, but is now recognized as a new addition to Korean flora. Species identification was confirmed according to morphological characteristics and DNA sequences. The genus Dalbergia is clearly distinguished from other leguminous tree species in Korea due to its diadelphous (5+5) stamens. Here, we describe the characteristics of the genus and species and speculate about whether its origins are native or introduced in Korea.

Identification and Characteristics of Extreme Halophilic bacteria Isolated from a Saltern in Korea (한국 염전으로 부터 분리한 고도 호염성 세균의 동정 및 특성)

  • Extremely halophilic bacteria isolated from salterns at Mado, Kyunggido, Korea, were identified and investigated on their salt requirements. The results have shown that six strains were identified to be belonged to the genus Halobacterium and three strains identified as the fenus Halococcus. Among them, the optimal NaCl concentration for growth of Halobacterium sp. EH10 was at 4.2M and no growth occurs below 2.0M NaCl. The strain, EH10, is nonmotile and showed acid production from glucose, fructose and maltose while H. salinarum is motile and does not produce acid from any carbohydrates. On the other hand, the strain EH10 does not utilize readily glucose while a number of sugars are readily utilized for growth with acid production by H. saccharovorum. Thus, the isolate, EH10, was classified into the genus Halobacterium and could be a novel species of the genus by its main morphological and physiological features including G+C content. The optimal temperature for growth of the isolate, EH10, was 50.deg.C. But this strain did not grow when NaCl was replaced with KCl.

Taxonomy and phylogeny of the genus Cryptomonas (Cryptophyceae, Cryptophyta) from Korea

  • The genus Cryptomonas is easily recognized by having two flagella, green brownish color, and a swaying behavior. They have relatively simple morphology, and limited diagnostic characters, which present a major difficulty in differentiating between species of the genus. To understand species delineation and phylogenetic relationships among Cryptomonas species, the nuclear-encoded internal transcribed spacer 2 (ITS2), partial large subunit (LSU) and small subunit ribosomal DNA (rDNA), and chloroplast-encoded psbA and LSU rDNA sequences were determined and used for phylogenetic analyses, using Bayesian and maximum likelihood methods. In addition, nuclear-encoded ITS2 sequences were predicted to secondary structures, and were used to determine nine species and four unidentified species from 47 strains. Sequences of helix I, II, and IIIb in ITS2 secondary structure were very useful for the identification of Cryptomonas species. However, the helix IV was the most variable region across species in alignment. The phylogenetic tree showed that fourteen species were monophyletic. However, some strains of C. obovata had chloroplasts with pyrenoid while others were without pyrenoid, which used as a key character in few species. Therefore, classification systems depending solely on morphological characters are inadequate, and require the use of molecular data.

  • Species boundaries among the Alcyonacean soft coral, the genus Dendronephthya, are often obscured by inter- and intraspecific morphological variations. In the present study, we attempted to infer the genetic relationships of eight dendronephthians based on their molecular characters, the internal transcribed spacer (ITS) regions of ribosomal DNA, and then compared this result together with the random amplified polymorphic DNA (RAPD) data from our previous investigation. Dendronephthya. putteri and D. suensoni formed a divaricate form - VI grade specific clade, whereas D. castanea, D. gigantea, D. aurea and D. spinifera, formed a umbellate and glomerate form - IV and III grade specific clade. Therefore, we confirmed that the main characters the growth form and the anthocodial grade and formula, are important in identification of the species in dendronephthians despite some problems. Also, the relationships of the growth form are clarified as the glomerate form is much closer to the umbellate form than to the divaricate form based on two sets of independent molecular data. However, we cannot determine the molecular markers which limit the species boundaries among this genus with ITS sequences.

Phylogenetic relationships of Coreanomecon (Papaveraceae: Papaveroideae), an endemic genus in Korea, using DNA sequences

  • Coreanomecon is a monotypic and endemic genus in Korea, distributed mainly in the southern regions. Coreanomecon is morphologically similar to Hylomecon by producing red latex, easily distinguished from Chelidonium, which produces yellow latex. Coreanomecon were merged into Hylomecon or Chelidonium depending on the authors. To understand the phylogenetic relationship of Coreanomecon, DNA sequences of chloroplast rbcL and matK and nuclear Internal Transcribed Spacer (ITS) regions were determined from the species of Papaveroideae (Papaveraceae) in Korea and analyzed with the Maximum Parsimony and Bayesian methods. Phylogenetic analyses of Papaveroideae suggest that Coreanomecon is sister to the clade of Chelidonium and Stylophorum in the ITS data and that it is sister to Hylomecon in the chloroplast (cpDNA) data. A constraining analysis using the Shimodaira-Hasegawa test (S-H test) suggested that the ITS data do not reject the sister relationship of Coreanomecon and Hylomecon. The S-H test also suggested that the cpDNA data is compatible with the placement of Coreanomecon as a sister to the clade of Chelidonium and Stylophorum. Although the conflicting phylogenetic results may stem from insufficient phylogenetic signals, they may also be associated with hybridization between Hylomecon and an ancestor of Stylophorum and Chelidonium. The results of this study suggest that Coreanomecon is a distinct lineage as an endemic genus, supporting the morphological data.

  • The genus Helina Robineau-Desvoidy, 1830 is one of the largest groups belonging to the subfamily Phaoniinae in the family Muscidae, comprising more than 540 species worldwide. In this taxonomic study, the genus Helina was recorded first time from the Korea with following three unrecorded species: H. deleta (Stein, 1914), H. evecta (Harris, 1780), and H. subpubiseta Xue, 1986. Taxonomic information, morphological photographs, and an identification key to the Korean species were provided.

New records of the genus Spirogyra (Zygnemataceae, Conjugatophyceae) in Korea

  • Spirogyra is a zygnematalean green algal genus that is ubiquitous in a broad range of freshwater habitats throughout the world. Samples collected throughout Korea from October 2004 to July 2015 were examined using light microscopy. Morphological characteristics (e.g., size of vegetative cells, number of chloroplasts in each cell, type of end walls of adjacent cells, details of conjugation, shape of female gametangia, dimensions and shape of zygospores, color and ornamentation of median spore walls) were used as diagnostic characteristics for species identification. In this study, five species of Spirogyra (i.e., S. emilianensis Bonhomme, S. jaoensis Randhawa, S. pascheriana Czurda, S. weberi var. farlowii (Transeau) Petlovany, and S. weberi var. grevilleana (Hassall) Kirchner) were described as newly recorded in Korea.

Six Korean New Records of the Nais Species (Annelida, Clitellata, Naididae)

  • The oligochaete worms in the genus Nais are common to freshwater habitats. These worms have a highly-developed prostomium and pigmented eye spots. All species have hair chaetae, bifid or single pointed needle chaetae on the dorsal side and ventral chaetae beginning in II with bifid crotchets. Most species of Nais genus live in areas where bottom is covered by sand, gravel or organic matter in aquatic plants. Currently, 26 species of Nais have been recorded worldwide. From the recent faunal studies, 3 species of Nais have been newly reported in Korea. Here, we newly report 6 species of Nais with diagnosis, illustrations and identification keys to Korean Nais species: N. behningi, N. pseudobtusa, N. simplex, N. bretscheri, N. stolci and N. elinguis.

Isolation and Identification of Superior Bifidobacterium strains from Korean Feces

  • For isolating and identifying Bifidobacterium spp. originating from Korea, feces were sampled from healthy Korean infants nursery school and postpartum care center. Through the use of gram staining and microscopic examination for cell morphology, 87 bacterial strains presumed to be the Bifidobacterium strains were isolated from 59 Koreans. To identify the Bifidobacterium strains at the genus level, these bacteria were then analyzed using the TLC method. As a result, 29 of the isolated strains were confirmed as members of the genus Bifidobacterium. 29 Bifidobacterium strains were tested acid, bile salts and oxygen tolerance and investigated antioxidative effect specially. And determined the superiority of 5 strains out of 29 Bifidobacterium strains. Finally, the selected bifidobacterium was identified with using designed 16S-ITS rDNA primer.
