Identification of Novel Compound Heterozygous Mutations in the ACADS Gene of an Asymptomatic Korean Newborn with Short Chain Acyl-CoA Dehydrogenase Deficiency by Tandem Mass Spectrometry

  • Short-chain acyl-CoA dehydrogenase deficiency (SCADD; OMIM # 201470) is an autosomal recessive inborn error of mitochondrial fatty acid ${\beta}$-oxidation, presenting with a variety of clinical signs and symptoms. Developmental delay, hypertonia or hypotonia, ketotic hypoglycemia, and epilepsy are most frequently reported. In general, patients diagnosed through newborn screening have shown normal growth and development in contrast to those diagnosed as a result of clinically initiated evaluations. Here, the case of an asymptomatic Korean newborn with SCADD identified by tandem mass spectrometry is reported. The patient showed an elevated concentration of butyrylcarnitine detected on newborn screening. Urinary excretion of ethylmalonic acid was elevated by urine organic acid analysis. To confirm the diagnosis of SCADD, a direct sequencing analysis of 10 coding exons and the exon-intron boundaries of the ACADS gene were performed. Genetic analysis of ACADS showed the following novel compound heterozygous missense mutations: c.277C>A (p.Leu93Ile) on exon3 and c.682G>A (p.Glu288Lys) on exon6. These results will provide further evidence of mutational heterogeneity for SCADD.

Prenatal Population Screening for Fragile X Carrier and the Prevalence of Premutation Carriers in, Korea

  • Purpose: Fragile X carrier detection before or at early pregnancy through a wide screening program may not only confer a risk of having offspring with Fragile X syndrome (FXS), but may also confer a risk for Fragile X-associated primary ovarian insufficiency and Fragile X-associated tremor/ataxia syndrome. However, prior to the implementation of such a program, the carrier prevalence in a population and the availability of effective screening test should be evaluated. The aim of our study was to determine the prevalence of premutation carriers and to evaluate the feasibility of screening test. Materials and Methods: The blood samples were obtained from 8,641 pregnant women with no family history of mental retardation. We performed a three-primer CGG repeat primed (RP) PCR using the AmplideX$^{TM}$ FMR1 PCR kit (Asuragen, Inc. Austin, TX, USA). Samples showing full mutation alleles were reflexed to Southern blot analysis for methylation status and sizing. Results: Among the 8,641 women, we found 8 premutation carriers (1:1,090, 0.09%) and 46 women with an intermediate allele (1:190, 0.53%). No woman was found to carry the fully mutated allele. All the detected alleles were within the CGG repeat range of 8-117. Among the 8,641 samples, 29 and 30 CGG repeats represent 66.6% of all cases. The CGG RP PCR method provides robust detection of expanded alleles and resolves allele zygosity, thus minimizing the number of samples that require Southern blot analysis. Conclusion: This is the first study that has focused on the prevalence of FXS premutation carriers and FMR1 allele distribution in normal pregnant women. These data have important implications for population-based fragile X carrier screening in Korea.

The DNA Repair Gene ERCC6 rs1917799 Polymorphism is Associated with Gastric Cancer Risk in Chinese

  • Objective: Excision repair cross-complementing group 6 (ERCC6) is a major component of the nucleotide excision repair pathway that plays an important role in maintaining genomic stability and integrity. Several recent studies suggested a link of ERCC6 polymorphisms with susceptibility to various cancers. However, the relation of ERCC6 polymorphism with gastric cancer (GC) risk remains elusive. In this sex- and age-matched case-control study including 402 GC cases and 804 cancer-free controls, we aimed to investigate the association between a potentially functional polymorphism (rs1917799 T>G) in the ERCC6 regulatory region and GC risk. Methods: The genotypes of rs1917799 were determined by Sequenom MassARRAY platform and the status of Helicobacter pylori infection was detected by enzyme-linked immunosorbent assay. Odd ratios (ORs) and 95% confidential interval (CI) were calculated by logistic regression analysis. Results: Compared with the common TT genotype, the ERCC6 rs1917799 GG genotype was associated with increased GC risk (adjusted OR=1.46, 95%CI: 1.03-2.08, P=0.035). When compared with (GT+TT) genotypes, the GG genotype also demonstrated a statistical association with increased GC risk (adjusted OR=1.38, 95%CI: 1.01-1.89, P=0.044). This was also observed for the male subpopulation (GG vs. TT: adjusted OR=1.71, 95%CI: 1.12-2.62, P=0.013; G allele vs. T allele: adjusted OR=1.32, 95%CI: 1.07-1.62, P=0.009). Genetic effects on increased GC risk tended to be enhanced by H. pylori infection, smoking and drinking, but their interaction effects on GC risk did not reach statistical significance. Conclusions: ERCC6 rs1917799 GG genotype might be associated with increased GC risk in Chinese, especially in males.

  • Population screening of newborns is an extremely important and informative diagnostic approach that allows early identification of babies who are predisposed to the development of a number of serious diseases. Some of these diseases are known and have effective treatment methods. Neonatal screening enables the early diagnosis and subsequent timely initiation of therapy. This helps to prevent serious complications and reduce the percentage of disability and deaths among newborns and young children. Primary immunodeficiency diseases and primary immunodeficiency syndrome (PIDS) are a heterogeneous group of diseases and conditions based on impaired immune system function associated with developmental defects and characterized by various combinations of recurrent infections, development of autoimmune and lymphoproliferative syndromes (genetic defects in apoptosis, gene mutation Fas receptor or ligand), granulomatous process, and malignant neoplasms. Most of these diseases manifest in infancy and lead to serious illness, disability, and high mortality rates. Until recently, it was impossible to identify children with PIDS before the onset of the first clinical symptoms, which are usually accompanied by complications in the form of severe coinfections of a viral-bacterial-fungal etiology. Modern advances in medical laboratory technology have allowed the identification of children with severe PIDS, manifested by T- and/or B-cell lymphopenia and other disorders of the immune system. This review discusses the main existing strategies and directions used in PIDS screening programs for newborns, including approaches to screening based on excision of T-cell receptors and kappa-recombination excision circles, as well as the potential role and place of next-generation sequencing technology to increase the diagnostic accuracy of these diseases.

구기자-맥문동 전통주 제조용 진균 발효제와 알콜발효 효모의 선발 및 최적 발효조건 (Screening of Fungal Nuruk and Yeast for Brewing of Gugija-Liriope tuber Traditional Rice Wine and Optimal Fermentation Condition)

  • 구기자와 맥문동을 이용하여 새로운 생리기능성 전통주를 개발하고자 항고혈압 활성이 보고된 구기자 3품종과 항통풍성을 가진 맥문동 2품종과 시판 Saccharomyces cerevisiae 3종류와 시판 2종류의 발효제를 이용하여 발효시켜 구기자-맥문동 전통주 제조에 적합한 알콜 발효 효모로 Saccharomyces cerevisiae C-2와 당화효소를 생성하는 진균 발효제로 SJ누룩을 최종 선발 하였다. 또한 이들을 이용한 발효 최적조건으로 구기자-맥문동 첨가량과 발효 최적시간 등을 검토하였다. 최종적으로 장명 구기자와 맥문동 1호를 덧밥에 각각 3%씩 첨가하고 Saccharomyces cerevisiae C-2와 SJ 누룩으로 $25^{\circ}C$에서 5일간 발효시켜 고품질의 기호도가 높고 항고혈압 활성과 항통풍성을 가진 새로운 구기자-맥문동 전통주를 개발하였다.

Screening of Multiple Abiotic Stress-Induced Genes in Italian Ryegrass leaves

  • Cold, salt and heat are the most critical factors that restrict full genetic potential, growth and development of crops globally. However, clarification of genes expression and regulation is a fundamental approach to understanding the adaptive response of plants under unfavorable environments. In this study, we applied an annealing control primer (ACP) based on the GeneFishing approach to identify differentially expressed genes (DEGs) in Italian ryegrass (cv. Kowinearly) leaves under cold, salt and heat stresses. Two-week-old seedlings were exposed to cold ($4^{\circ}C$), salt (NaCl 200 mM) and heat ($42^{\circ}C$) treatments for six hours. A total 8 differentially expressed genes were isolated from ryegrass leaves. These genes were sequenced then identified and validated using the National Center for Biotechnology Information (NCBI) database. We identified several promising genes encoding light harvesting chlorophyll a/b binding protein, alpha-glactosidase b, chromosome 3B, elongation factor 1-alpha, FLbaf106f03, Lolium multiflorum plastid, complete genome, translation initiation factor SUI1, and glyceraldehyde-3-phosphate dehydrogenase. These genes were potentially involved in photosynthesis, plant development, protein synthesis and abiotic stress tolerance in plants. However, this study provides new insight regarding molecular information about several genes in response to multiple abiotic stresses. Additionally, these genes may be useful for enhancement of abiotic stress tolerance in fodder crops as well a crop improvement under unfavorable environmental conditions.

Gene Cloning and Nucleotide Sequence of Human Dihydrolipoamide Dehydrogenase-Binding Protein

  • The pyruvate dehydrogenase complex (PDC), a member of $\alpha$-keto acid dehydrogenase complex, catalyzes the oxidative decarboxylation of pyruvate with the formation of $CO_2$, acetyl-CoA, NADH, and $H^+$. This complex contains multiple copies of three catalytic components including pyruvate dehydrogenase (E1), dihydrolipoamide acetyltransferase (E2), and dihydrolipoamide dehydrogenase (E3). Two regulatory components (E1-kinase and phospho-E1 phosphatase) and functionally less-understood protein (protein X, E3BP) are also involved in the formation of the complex. In this study, we have partially cloned the gene for E3BP in human. Nine putative clones were isolated by human genomic library screening with 1.35 kb fragment of E3BP cDNA as a probe. For investigation of cloned genes, Southern blot analysis and the construction of the restriction map were performed. One of the isolated clones, E3BP741, has a 3 kb-SacI fragment, which contains 200 bp region matched with E3BP cDNA sequences. The matched DNA sequence encodes the carboxyl-terminal portion of lipoyl-bearing domain and hinge region of human E3BP. Differences between yeast E3BP and mammalian E3BP coupled with the remarkable similarity between mammalian E2 and mammalian E3BP were confirmed from the comparison of the nucleotide sequence and the deduced amino acid sequence in the cloned E3BP. Cloning of human E3BP gene and analysis of the gene structure will facilitate the understanding of the role(s) of E3BP in mammalian PDC.

Cloning and Expression of Lactate Dehydrogenase H Chain Gene in Adipose Tissues of Korean Cattle

  • To understand molecular mechanisms that regulate deposition and release of intramuscular fat, a fasting-induced clone was identified by differential screening from cDNA library of adipose tissues of Korean cattle. The clone had a total length of 1,319 nucleotides coding for 334 amino acids. It was identified as one encoding L-lactate dehydrogenase H chain (LDH-B). Comparison of the deduced amino acid sequences of bovine LDH-B with those of pig, human, rat, and mouse showed 98%, 98%, 97%, and 96% identity, respectively. Food deprivation for 48 h increased mRNA levels of LDH-B gene in adipose tissues of Korean cattle compared to fed- and 6 h refed- tissues. The expression of obese mRNA was examined for individual adipose tissue from several fat depots. Fasting induced expression of LDH-B gene in subcutaneous adipose tissues, but it did not affect expression levels in abdominal, perirenal and intramuscular tissues. Results demonstrate that induction of LDH-B gene during fasting may represent a metabolic shift from anaerobic state to aerobic predominance in fasted adipose tissues and that its responses to fasting are different among several adipose tissues.

Genetic Analysis of Growth Response to Cold Water Irrigation in Rice

  • This study was carried out to obtain the basic information for breeding cold-tolerant rice varieties with high yield-productivity through wide crosses between indica and japonica rice. Genetic analysis was conducted using 55 F$_1$s obtained from half-diallel crosses among eleven cultivars of various origin including indica and japonica rice. Screening for cold tolerance was done with cold-water irrigation after transplanting until ripening stage. Both general combining ability (GCA) and specific combining ability (SCA) effects were highly significant in all characters associated with dry matter accumulation at 30 and 50days after cold-water irrigation (DAC). The variance of GCA was much larger than that of SCA in plant height, shoot dry weight per plant (DWP), crop growth rate (CGR) and cold-water response index (CRI) of these characters except CRI of shoot dry weight per plant. The DWP, CGR and CRI of these characters of Gaochan 102, Tong88-7 and TR22183 were markedly higher than those of the others. GCA effects of these varieties on DWP, CGR and their CRI were also higher than those of the others, indicating that they are useful as promising parents for breeding cold-tolerant varieties. Analysis of genetic parameters for 11$\times$11 half-diallel F$_1$s revealed that inter-locus gene interaction were concerned in the expression of plant height at 50 DAC, CRI of DWP at 50 DAC, and CRI of CGR, and that intra-locus gene interaction for plant height and the other characters were partial dominance and over-dominance, respectively. Narrow-sense heritability (h$^2$$_{N}$) was the highest in plant height as 0.729, and the lowest in CRI of DWP at 30 DAC as 0.048, suggesting that selection for cold tolerance will be quite effective in case that the selection criterion is the performance itself.f.

Insights into the in vitro germicidal activities of Acalypha indica

  • Background and purpose: This study was carried out to learn more about the potential prophylactic or antibacterial activity of the plant Acalypha indica against selective pathogenic bacteria. Experimental: The test organisms were Sarcina lutea IFO 3232, Bacillus subtilis IFO 3026, Pseudomonas denitrificans, Escherichia coli IFO 3007, Klebsiella pneumoniae ATTC 10031, Xanthomonas campestris IAM 1671, and Proteus vulgaris. Leaf, stem, and bud powder of Acalypha indica were dissolved in various solvents, and the extracts were tested for antimicrobial activity through the disc diffusion method. GC-MS profiling was performed to characterize active chemical compounds in the essential oil of Acalypha indica. Results: The ethanol extract showed the highest activity against all bacteria, while the petroleum ether extract yielded the highest zone of inhibition against Proteus vulgaris ($11.83{\pm}1.75mm$). The minimum inhibitory concentration (MIC) of the ethyl acetate extract against Bacillus subtilis was 16 µg/mL. Phytochemical screening by GC-MS revealed a total of 12 bioactive compounds. Conclusion: Extracts of Acalypha indica may be useful in formulating and synthesizing new antibacterial drugs.
