임신 중 내분비계 장애물질 Bisphenol A의 양수 내 농도와 임신결과와의 상관관계 (Amniotic Fluid Bisphenol A Level and Its Relationship with Pregnancy Outcomes)

  • Bisphenol A(BPA)는 약한 estrogen 활성도를 보이는 화학물질로서 착상 전 배아나 태아에 영향을 미쳐, 출생 후 그들의 신체발달에 변화를 초래한다고 알려져 있다. 본 연구는 인체에서 임신 중기에 양수 내 BPA 농도를 측정하였으며, 측정된 농도에 따라 임신결과에 영향을 미치는지 여부를 알아보고자 하였다. 임신 15주에서 20주 사이에 유전학적 적응증으로 양수천자 검사를 시행 받았던 120명의 임신부 양수를 연구대상으로 ELISA 방법으로 농도를 측정하였다. 양수천자의 가장 많은 적응증은 35세 이상의 고령 임신부였다. BPA는 모든 양수에서 검출이 가능하였으며, 그 농도는 최저치 0.89 ng/mL부터 최고치 37.13 ng/mL까지의 분포를 보였고, 평균치는 7.24 ng/mL이었다. 양수 내 BPA 농도가 여러 임신결과들과 관련성이 있는지를 알아보기 위해 임신부 나이 35세 이상 대 미만, 남자 태아 대 여자 태아, 분만 당시 재태 연령 37주 이상 대 미만, 출생 당시 신생아 체중 2.5 kg 이상 대 미만으로 각각 나누어, 두 군간에 농도를 비교한 결과, 통계학적으로 유의한 차이는 없었다. 본 연구는 한국인 임신부를 대상으로 최초로 양수 내 BPA 농도에 관한 보고이다. 본 연구결과 BPA 농도는 임신부 나이나 태아성별에 따라 차이를 보이지 않았으며, 조산이나 저체중아의 발생과도 관련이 없음을 알 수 있었다. 향후 산전 BPA 노출이 배발생에 영향을 미치는지에 대한 연구가 필요하다고 사료된다.

Comparative physiological and proteomic analysis of leaf in response to cadmium stress in sorghum

  • Cadmium (Cd) is of particular concern because of its widespread occurrence and high toxicity and may cause serious morpho-physiological and molecular abnormalities in in plants. The present study was performed to explore Cd-induced morpho-physiological alterations and their potentiality associated mechanisms in Sorghum bicolor leaves at the protein level. Ten-day-old sorghum seedlings were exposed to different concentrations (0, 100, and $150{\mu}M$) of $CdCl_2$, and different morpho-physiological responses were recorded. The effects of Cd exposure on protein expression patterns in S. bicolor were investigated using two-dimensional gel electrophoresis (2-DE) in samples derived from the leaves of both control and Cd-treated seedlings. The observed morphological changes revealed that the plants treated with Cd displayed dramatically altered shoot lengths, fresh weights, and relative water content. In addition, the concentration of Cd was markedly increased by treatment with Cd, and the amount of Cd taken up by the shoots was significantly and directly correlated with the applied level of Cd. Using the 2-DE method, a total of 33 differentially expressed protein spots were analyzed using MALDI-TOF/TOF MS. Of these, treatment with Cd resulted in significant increases in 15 proteins and decreases in 18 proteins. Significant changes were absorbed in the levels of proteins known to be involved in carbohydrate metabolism, transcriptional regulation, translation and stress responses. Proteomic results revealed that Cd stress had an inhibitory effect on carbon fixation, ATP production and the regulation of protein synthesis. In addition, the up-regulation of glutathione S-transferase and cytochrome P450 may play a significant role in Cd-related toxicity and stress responses. Our study provides insights into the integrated molecular mechanisms involved in response to Cd and the effects of Cd on the growth and physiological characteristics of sorghum seedlings. The upregulation of these stress-related genes may be candidates for further research and use in genetic manipulation of sorghum tolerance to Cd stress.

국내에서 유행한 Respiratory Syncytial 바이러스의 염기서열 및 계통분석 (Sequence and Phylogenetic Analysis of Respiratory Syncytial Virus Isolated from Korea)

  • Respiratory Syncytial virus (RSV) is an important cause of acute lower respiratory tract infections in human, with infants and young children being particularly susceptible. In the temperate zones, sharp annual outbreaks of RSV occur during the colder months, in both the northern and the southern hemisphere. RSV is unusual in that it can repeatedly reinfect individuals throughout life and infect babies in the presence of maternal antibody. RSV isolates can be divided into two subgroups, A and B, on the basis of their reactions with monoclonal antibodies, and the two subgroups are also distinct at the nucleotide sequence level. The specific diagnosis of RSV infection was best made by isolation of virus in tissue culture, identification of viral antigen, or by specific serologic procedures. Recently, rapid detection of RSV and analysis of RSV strain variation became possible by development of methods of reverse transcription and polymerase chain reaction amplification. In this study, to determine the genetic diversity of RSV found in Korea, 173 bp and 164 bp spanning selected regions of the RSV F and SH genes were enzymatically amplified and sequenced, respectively. Eight for F gene and three for SH gene were detected in 66 nasopharyngeal swap samples tested. Two major antigenic subgroups, A and B were confirmed from Korean samples (seven for subgroup A and one for subgroup B). At the nucleotide level of the F gene region, Korean subgroup A strains showed 95-99% homologies compared to the prototype A2 strain of subgroup A and 93-100% homologies among Korean subgroup A themselves. For the SH gene region, Korean subgroup A strain showed 97.5% homology compared to the prototype A2 strain of subgroup A, and Korean subgroup B strain showed 97% homology compared to the prototype 18537 strain of subgroup B. Most of base changes were transition and occured in codon position 3, which resulted in amino acid conservation. Using the maximum parsimony method, phylogenetic analysis indicated that Korean RSV strains formed a group with other RSV strains isolated from the United States, Canada, the Great Britain and Australia.

Association of the Porcine Cluster of Differentiation 4 Gene with T Lymphocyte Subpopulations and Its Expression in Immune Tissues

  • Cluster of differentiation 4 (CD4) is mainly expressed on $CD4^+$ T cells, which plays an important role in immune response. The aim of this study was to detect the association between polymorphisms of the CD4 gene and T lymphocyte subpopulations in pigs, and to investigate the effects of genetic variation on the CD4 gene expression level in immune tissues. Five missense mutations in the CD4 gene were identified using DNA pooling sequencing assays, and two main haplotypes (CCTCC and AGCTG) in strong linkage disequilibrium (with frequencies of 50.26% and 46.34%, respectively) were detected in the population of Large White pigs. Our results indicated that the five SNPs and the two haplotypes were significantly associated with the proportions of $CD4^-CD8^-$, $CD4^+CD8^+$, $CD4^+CD8^-$, $CD4^+$ and $CD4^+/CD8^+$ in peripheral blood (p<0.05). Gene expression analysis showed the mRNA level of the CD4 gene in thymus was significantly higher than that in lymph node and spleen (p<0.05). However, no significant difference was observed between animals with CCTCC/CCTCC genotype and animals with AGCTG/AGCTG genotype in the three immune tissues (p>0.05). These results indicate that the CD4 gene may influence T lymphocyte subpopulations and can be considered as a candidate gene affecting immunity in pigs.

Performance Evaluation of Karan Fries and Karan Swiss Cattle under Closed Breeding System

  • The 490 and 380 performance records of Karan Fries and Karan Swiss cows developed through crossbreeding followed by inter-se mating were evaluated for production, reproduction performance and disposal rate. Duration of study (1982-92) was grouped into five periods (1982-83; 1984-85; 1986-87; 1988-89; 1990-92) and each year was divided into four seasons (Winter: Dec.-Jan.; Spring: Feb.-March; Summer: April-June; Rainy: July-Sept.; Autumn: Oct.-Nov). Data were also classified according to sire and level of inbreeding. The least squares means of FLY (first lactation yield -305 days), FLL (first lactation length), FSP (first service period), FDP (first dry period), FCI (first calving interval ), MY/FLL (milk yield per day of first lactation length), MY/FCI (milk yield per day of first calving interval), EBV (expected breeding value) and EBE (expected breeding efficiency) were $3,173{\pm}82$ kg, $34611{\pm}$days, $143{\pm}11$ days, $75{\pm}6$ days, $423{\pm}11$ days, $10.6{\pm}0.2$ kg, $8.9{\pm}0.2$ kg, $3,380{\pm}26$ kg, and $88.2{\pm}1.3$% respectively in Karan Fries. Corresponding estimates in Karan Swiss cows were $2,616{\pm}82$ kg, $328{\pm}8$ days, $148{\pm}12$ days, $103{\pm}9$ days, $435{\pm}13$ days, $8.9{\pm}0.2$ kg, $7.2{\pm}0.3$ kg, $2,924{\pm38}$ kg and $86.0{\pm}3.8$% respectively. The effect of sire was significant on FLY, MY/FLL, MY/FCI and EBV in both the herd (p<0.01). The 10 and 15% sires showed superiority (FLY) over herd average of 11 and 7% in Karan Fries and 32 and 21% in Karan Swiss cattle respectively. Inbreeding has adversely and significantly (p<0.05) affected the FLY, MY/FLL, MY/FCI and survivability of Karan Fries females; FDP and disposal through culling of Karan Swiss heifers. The Karan Fries heifers with inbreeding above 12.5% performed 16% lower FLY to herd average. The effect of season of calving was significant on FLL, FSP and FCI (p<0.05) in Karan Fries. Summer calvers ($361{\pm}12$) were have higher lactation length and autumn calvers ($329{\pm}14$) had minimum. Period of calving significantly influenced the FSP, FCI, MY/FLL and MY/FCI in Karan Fries and FLY, MY/FLL and MY/FCI in Karan Swiss. Production efficiency traits in Karan Fries herd witnessed higher yield in the last two periods whereas, Karan Swiss heifers showed fluctuating performance. The findings suggested judicious use of available genetic variability, keeping of inbreeding under safer level and managemental interventions for the consistent improvement of both herds.

한의과대학 본초학 교육과정의 개정 및 보완을 위한 설문조사 연구 (Survey on Revision and Complements for the Current Curriculum of Herbology)

  • Objects: This study was conducted to investigate the current educational environment of herbology and to develop a future-oriented curriculum for oriental medicine. The questionnaire used in this research was drawn up based on the current curriculum referring to the current curriculum of herbology and pharmacognosy. Methods: The survey was carried out presenting the questionnaires to a total 12,754 of the students and doctors of oriental medicine through e-mailing five times; of these, 2,074 replied. Results: 1. Among the respondents, about 97% agreed that it was necessary to revise and complement the current curriculum of herbology. 2. The respondents felt that the assigned lecture time of subject was "sufficient" (19%), "insufficient" (39%) and "average" (39%), respectively, and the level of lecture was "insufficient" (37%) or "average" (43%) respectively. According to priority, it showed that the contents which needed complement in lecture were discrimination of medicinal herbs (24%), practical use of action and indications (23%), and correlation with modern disease (21%). In theoretical lectures, 69% of the respondents agreed on the introduction of natural scientific methods 3. In practice, 51% of the respondents replied that the lecture time for practice was insufficient. The contents which needed to be complemented in practice were as follows: audio-visual materials for discrimination of medicinal herbs (22%), concrete exercise for the processing of medicinal herbs (21%), and attempts for the objective discrimination of medicinal herbs using instruments (microscope, analytical instrument, residual pesticide, heavy metal, genetic analysis) (16%). 70% replied that the discrimination of medicinal herbs of high price and rarity was "none or insufficient". 4. 56% replied that it was necessary to introduce and practice physicochemical analysis, and they showed higher requests according to the increase of their educational level. However, 86% replied that they had never experienced concrete attempts for objective discrimination of medicinal herbs, which seemed to indicate that, excepting some schools, practice exercise was rarely performed. Conclusions: According to results, it seems that an urgent review on the current course of herbology and a workshop on the process of experimental practice for professors is needed.

국내에 자생하는 한국잔디 종자의 변이 특성 (Characteristics of Seed Variation of Zoysiagrasses (Zoysia spp.) Native to South Korea)

  • 본 연구는 국내에서 자생하고 있는 241개체의 한국잔디를 지역별로 수집하여 종자 형태적 형질을 조사했으며, 한국잔디의 종자 형태적 특성 및 변이를 알아보고자 연구를 수행하였다. 수집된 한국잔디 241개체 중에서 중지가 99개체로 41% 이상이 수집되었으며, 다양한 형태의 종자 변이를 보였다. 크게 갯잔디형과 들잔디형으로 두 개 그룹으로 나뉘었으며, I과 II군은 화서당 종자수가 30.1개와 17.5개, 종자길이는 4.6과 5.2 mm으로 갯잔디형이 포함되었고, III과 IV군은 화서당 종자수가 59.2개와 45.8개, 종자길이는 3.3과 3.4 mm으로 들잔디형 특성을 나타내었다. 정확한 종간교잡의 정도를 파악하기 위해서는 추가적인 생육특성 및 분자생물학적 연구가 필요하다고 판단되었다.

토끼에서 oxytetracycline의 정맥 및 근육 투여시의 약물동태학 (Pharmacokinetics of oxytetracycline in rabbits after intravenous and intramuscular administrations)

  • The study was carried out to determine the pharmacokinetic parameters after intravenous(iv) and intramuscular(im) administration (10mg/kr) in healthy rabbits. The results obtained through the experiments were summarized as follows: 1. Bioassay (Bacillus cereus 11778) was evaluated very useful for the determination of oxytetracycline(OTC) in the rabbit serum and tissues, with the detection limit of $0.125{\mu}g/ml$. 2. The pharmacokinetic profiles of OTC (10mg/kg, iv) in rabbits were best described with a two compartment open model $(C=29.5e^{-4,3t}{\pm}3.6^{-0.2t})$, whereas that of OTC (10mg/kg, im) showed a one compartment curve fitting. 3. Following iv administration, a rapid distribution phase was predominant [$t_{\frac{1}{2}}({\alpha}):1.43{\pm}0.98hr$ (♂), $0.5{\pm}0.1hr$(♀)], and then more slow elimination phase ensued [$t_{\frac{1}{2}}({\beta}):4.52{\pm}0.76hr$(♂), $7.32{\pm}2.52hr$(♀)]. Other computer generated pharmacokinetic values were as follows:C1 [$67.76{\pm}18.59ml/kg/h$(♂), $76.03{\pm}22.98ml/kg/h$ (♀)] Vd [$257.74{\pm}180.47ml/kg$ (♂), $92.33{\pm}23.62$ (♀)] AUC [$25.6{\pm}4.44mgh/L$ (♂), $39.6{\pm}12.13mgh/l$ (♀)]. There were no statistical significance between both sexes for all the parameters at the confidence level of 95%. 4. After im administaration, the absorption from the injection sites was very rapid [ Ka:$0.18{\pm}0.03h^{-1}$ (♂), $0.24{\pm}0.02h^{-1}$ (♀)] followed by a monoexponential elimination fashion. The time to peak blood level (Tmax) were calculated $1.64{\pm}0.15hr$ and $1.34{\pm}0.24hr$, in the male and female, respectively. The peak levels (Cmax) at the corresponding time were $1.69{\pm}0.23{\mu}g/ml$ (♂) and $2.08{\pm}0.16{\mu}g/ml$ (♀), with no statistical differences (p>0.05).

몰운대 탐방로 주변 식생의 생태적 특성 평가 (Assessment on Ecological Characteristics of Vegetation in the Trail of Adjacent to Molundae)

  • The objective of this study is to analyze the flora and forest vegetation of trails with high visitor density at Molundae. Nine quadrats of $20{\times}20m$ were selected for the survey. The survey was conducted from April to October 2010. The obtained results are summarized as follows. Plot1, plot2, plot3, plot4, plot6, and plot7 were located at slopes of $5{\sim}20^{\circ}$, 17~40m above sea level, and were formed with the colony of Japanese black pine(Pinus thunbergii Parl) and Japanese black pine(Pinus thunbergii Parl)-white oak(Quercus aliena Blume). Tree layer had the height of 8~20m, and the coverage of 50~70%, while subtree layer had the height of 3-8m, and the coverage 30~80%. On the other hand, shrub layer had the height of 2~4m, and the coverage of 10~30%, and herb had the height of 0.2~1m and coverage 5~20%. Plot5, plot8, and plot9 were located at the summit areas of 57~78m above sea level, and $0^{\circ}$ slope. Japanese black pine(Pinus thunbergii Parl) formed a community there. Tree layer was 8~20m high, and covered 60~70%, of the area, and subtree layer was 6~8m high, and coverage 30~40%. Shrub layer had the height of 2~6m, and the coverage of 30%, while herb layer had the height 0.2~2m, and the coverage 20-80%. Succession does not occur in the surveyed areas which have high visitor density. Artificially planted sawtooth oak(Quercus acutissima) trees were found to disturb succession and formation of multi-layer vegetation, resulting in the ecologically unstable forest. Therefore, the researcher suggested the strategy of managing the vegetation in the conclusion. This study has the limit in that the plots selected for the survey reflected only part of various trails in the Molundae area. It is necessary to suggest the vegetation management plans by selecting more diverse trail areas in consideration of the visitor density and behaviors, and analyzing the changes in vegetation quantitatively in order to manage the vegetation in urban areas damaged by visitors more effectively.

Stem Cells and Cell-Cell Communication in the Understanding of the Role of Diet and Nutrients in Human Diseases

  • The term, "food safety", has traditionally been viewed as a practical science aimed at assuring the prevention acute illnesses caused by biological microorganisms, and only to a minor extent, chronic diseases cause by chronic low level exposures to natural and synthetic chemicals or pollutants. "food safety" meant to prevent microbiological agents/toxins in/on foods, due to contamination any where from "farm to Fork", from causing acute health effects, especially to the young, immune-compromised, genetically-predisposed and elderly. However, today a broader view must also include the fact that diet, perse (nutrients, vitamins/minerals, calories), as well as low level toxins and pollutant or supplemented synthetic chemicals, can alter gene expressions of stem/progenitor/terminally-differentiated cells, leading to chronic inflammation and other mal-functions that could lead to diseases such as cancer, diabetes, atherogenesis and possibly reproductive and neurological disorders. Understanding of the mechanisms by which natural or synthetic chemical toxins/toxicants, in/on food, interact with the pathogenesis of acute and chronic diseases, should lead to a "systems" approach to "food safety". Clearly, the interactions of diet/food with the genetic background, gender, and developmental state of the individual, together with (a) interactions of other endogenous/exogenous chemicals/drugs; (b) the specific biology of the cells being affected; (c) the mechanisms by which the presence or absence of toxins/toxicants and nutrients work to cause toxicities; and (d) how those mechanisms affect the pathogenesis of acute and/or chronic diseases, must be integrated into a "system" approach. Mechanisms of how toxins/toxicants cause cellular toxicities, such as mutagenesis; cytotoxicity and altered gene expression, must take into account (a) irreversible or reversal changes caused by these toxins or toxicants; (b)concepts of thresholds or no-thresholds of action; and (c) concepts of differential effects on stem cells, progenitor cells and terminally differentiated cells in different organs. This brief Commentary tries to illustrate this complex interaction between what is on/in foods with one disease, namely cancer. Since the understanding of cancer, while still incomplete, can shed light on the multiple ways that toxins/toxicants, as well as dietary modulation of nutrients/vitamins/metals/ calories, can either enhance or reduce the risk to cancer. In particular, diets that alter the embryo-fetal micro-environment might dramatically alter disease formation later in life. In effect "food safety" can not be assessed without understanding how food could be 'toxic', or how that mechanism of toxicity interacts with the pathogenesis of any disease.
