The effects of glycine on cell growth and L-omithine production were investigated in shake-flask and jar fermentor cultures of a citrulline auxotrophic mutant, Brevibacterium ketoglutamicum BK 1046. In the shake-flask culture, the optimal concentration of glycine for L-ornithine production was found to be 20 g/l. In the jar fermentor culture with the glycine at an initial concentration of 20 g/l, L-ornithine production increased by 28%, compared to that of the culture with no glycine added. 37 g/l of L-ornithine was produced when additional feeding of glycine (5 g/l) was made. This was a significant improvement in L-ornithine production compared to that (ca. 24 g/l) of the corresponding batch culture conducted without glycine. According to the stoichiometric analysis with the batch fermentation results, the experimental and theoretical L-ornithine yields based on the glucose consumption were 0.24 and 0.59, respectively. This indicates that the performance of L-ornithine fermentation can further be improved by the supplementation of glycine and the development of a mutant strain possessing a higher growth yield.
본 연구는 엄격한 시간제약 차량경로문제에 대하여 유전자알고리즘과 휴리스틱 기법을 이용하여 최적해를 산출하는 것이다. 문제해결을 위해 수리적 모형을 구성하고, ILOG-CPLEX를 이용하여 최적해를 산출하였다. 임의 생성방법과 세이빙 휴리스틱을 적용한 초기해 생성, 실행불가능해의 교정과 유전자 알고리즘 종료 후 2-opt, Or-opt 등 해교정 및 해개선을 위한 과정이 추가된 하이브리드 유전자 알고리즘을 구축하여 엄격한 시간제약이 있는 차량경로 문제에 적용하여 솔로몬 예제와 비교하고, 제안한 알고리즘의 해공간탐색능력, 수렴성, 휴리스틱 기법의 효과를 확인하였다.
Glycine soja Sieb. et Zucc., a predominantly selfing annual, has been served as a reservoir of germplasm for soybean, G. max (L.) Merr., cultivar improvement. This study describes the levels and distribution of genetic variation within and among 22 Korean populations of G. soja using starch gel electrophoresis. The species maintains very similar levels of genetic variability within populations observed in most other annuals. At the population level, the mean percent of polymorphic loci (P) was 32.6%, mean number of allele per locus (A) was 1.32, and mean expected heterozygosity (He) was 0.112. In addition, total genetic diversity (HT) calculated only for polymorphic loci was 0.347. However, significant differences in allele frequencies among populations were found for all loci (P<0.001 in each case) and, on average, about 70% of the total variation in the species is common to all populations. Indirects estimate of the number of migrants per generation (Nm=0.58, calculated from mean GST) indicates that gene flow is low among Korean populations of the species. In addition, analysis of fixation indices revealed a substantial heterozygote deficiency in most populations and at all loci. This indicates that most populations sampled may have been substructed largely due to inbreeding (predominantly selfing) and restricted gene flow, coupled with founder effect and genetic drift. Considering a high genetic divergence among populations, it is recommended that several Korean populations of the species should be preserved, especially such as populations in the eastern and southeastern Korean peninsula with high variation.
Genus Taraxacum has been widely used as a folkloric medicine for treatment of diverse diseases. The genetic diversity and relationship among 32 accessions belonging to five Taraxacum species (T. mongolicum T. coreanum, T. coreanum var. flavescens, T. officinale and T. laevigatum) which collected from field, mountain, island and seaside of Korea were evaluated using ISSR markers. A total of 142 ISSR loci detected in the overall species were all polymorphic loci (100%) and interspecies polymorphisms obtained from Korean native and naturalized species were 98.2% and 94.5%, respectively. The genetic similarity matrix (GSM) among 32 accessions ranged from 0.025 to 0.860 with an average of 0.303. According to the clustering analysis, the Korean native species and naturalized species were divided two major clusters. In addition, the different species were divided into independent groups except for the T. coreanum and T. coreanum var. flavescens, and all the 32 accessions could be classified into 7 categories. The study findings indicate that Taraxacum accessions have a high genetic diversity and the dandelion accessions as breeding materials can be effectively utilized for the improvement of Taraxacum breeding.
Tamarind (Tamarindus indica L.) is one of the multipurpose tree species distributed in the tropical and sub-tropical climates. It is an important fruit yielding tree that supports the livelihood and has high social and cultural values for rural communities. The vegetative, reproductive, qualitative, and quantitative traits of tamarind vary widely. Characterization of phenotypic and genetic structure is essential for the selection of suitable accessions for sustainable cultivation and conservation. This study aimedto examine the genetic relationship among the collected accessions of sweet, red, and sour tamarind by using Random Amplified Polymorphic DNA (RAPD) primers. Nine accessions were collected from germplasm gene banks and subjected to marker analysis. Fifteen highly polymorphic primers generated a total of 169 fragments, out of which 138 bands were polymorphic. The polymorphic information content of RAPD markers varied from 0.10 to 0.44, and the Jaccard's similarity coefficient values ranged from 0.37 to 0.70. The genetic clustering showed a sizable genetic variation in the tamarind accessions at the molecular level. The molecular and biochemical variations in the selected accessions are very important for developing varieties with high sugar, anthocyanin, and acidity traits in the ongoing tamarind improvement program.

Genetic parameters for dairy performance traits were estimated, breeding values for the traits of all breeding sires and cows were predicted and the genetic trends were estimated using the breeding values in the Central Cattle Breeding Station (CCBS). A total of 3,801 records for Bangladeshi Local, 756 records for Red Sindhi and 959 records for Sahiwal covering the period from 1961 to 1997 were used in this analysis. Traits considered were total milk production per lactation (TLP), lactation length (LL) and daily milk yield (DMY). The genetic parameters were estimated by the REML using MTDFREML program. The breeding values were predicted by a best linear unbiased prediction (BLUP). In all sets of data, the genetic trends for the dairy performance traits were computed as averages of breeding values for cows born in the particular year. The estimates of heritability for TLP (0.26 and 0.27) and DMY (0.28 and 0.27) were moderate in Bangladeshi local and Red Sindhi breed, respectively. Furthermore, the heritability estimate for LL (0.24) was moderate in Red Sindhi. The estimates of heritabilities for all traits were low in Sahiwal. The repeatability estimate was high for TLP, moderate for LL and moderate to high for DMY. All variances estimated in Bangladeshi Local were low, comparing the respective values estimated in both Red Sindhi and Sahiwal. On the other hand, additive genetic variances for the three traits were estimated very low in Sahiwal. The genetic trends for the three dairy production traits have not been positive except for the recent trend in Bangladeshi Local.
Nowadays, Unmanned Combat Air Vehicle(UCAV) has become an important aircraft system for the national defense. For its efficiency and survivability, shape optimization of UCAV is an essential part of its design process. In this paper, shape optimization of UCAV was processed for aerodynamic performance improvement and Radar Cross Section(RCS) reduction using Multi Objective Genetic Algorithm(MOGA). Lift and induced drag, friction drag, RCS were calculated using panel method, boundary layer theory, Physical Optics(PO) approximation respectively. In particular, calculation applied Radar Absorbing Material(RAM) was performed for the additional RCS reduction. Results are indicated that shape optimization is performed well for improving aerodynamic performance, reducing RCS. Further study will be performed with higher fidelity tools and consider other design segments including structure.
Reciprocal crosses were conducted between three strains of Taiwan Country chickens, developed in the National Chung-Hsing University, and two strains of Single Comb White Leghorns, developed in the Taiwan Livestock Research Institute. Traits studied were growing performances, laying performances, egg quality traits and traits concerning disease resistance, including resistance to Marek's disease virus and immune responses to Newcastle disease virus vaccine and to sheep red blood cell. Results indicated that laying performances of Taiwan country chickens were much inferior to White Leghorns, but they matured earlier, their eggs had better shell strength and larger proportion of yolk, and their general disease resistance was much better than White Leghorns. Heterosis were found in laying performances and egg quality traits. The heterosis in laying traits was so large that the hybrid laid as many eggs and as large eggs as did pure strains of White Leghorns. Strategies on the improvement of native chickens and the utilization of genetic merits of native chickens were also discussed.
To date, many viable smart base isolation systems have been proposed. In this study, a novel friction pendulum system (FPS) and an MR damper are employed as the isolator and supplemental damping device, respectively. A fuzzy logic controller (FLC) is used to modulate the MR damper. A genetic algorithm (GA) is used for optimization of the FLC. The main purpose of employing a GA is to determine appropriate fuzzy control rules as well to adjust parameters of the membership functions. To this end, a GA with a local improvement mechanism is applied. Neuro-fuzzy models are used to represent dynamic behavior of the MR damper and FPS. Effectiveness of the proposed method for optimal design of the FLC is judged based on computed responses to several historical earthquakes. It has been shown that the proposed method can find appropriate fuzzy rules and the GA-optimized FLC outperforms not only a passive control strategy but also a human-designed FLC and a conventional semi-active control algorithm.
In this review, we discuss the ways in which our understanding of the controls of nitrogen use efficiency applied to crop improvement has been increased through the development of molecular physiology studies using transgenic plants or mutants with modified capacities for nitrogen uptake, assimilation and recycling. More recently, exploiting crop genetic variability through quantitative trait loci and candidate gene detection has opened new perspectives toward the identification of key structural or regulatory elements involved in the control of nitrogen metabolism for improving crop productivity. All together these studies strongly suggest that in the near future nitrogen use efficiency can be improved both by marker-assisted selection and genetic engineering, thus having the most promise for the practical application of increasing the capacity of a wide range of economically important species to take up and utilize nitrogen more efficiently.
