Intraspecific Molecular Phylogeny, Genetic Variation and Phylogeography of Reticulitermes speratus (Isoptera:Rhinotermitidae)

  • Population structure was investigated in Reticulitermes speratus populations in the Korean Peninsula and the Japanese Archipelago. All trees derived from analyses of the combined sequence dataset of two mitochondrial genes, COII and COIII, showed that R. speratus populations cluster into two major clades comprising the Korean/southern Japanese populations and the northern Japanese populations. Analysis of population genetic structure showed strong genetic partitioning between populations of the two clades. To understand historical migration routes and current distributions, the phylogeographic history of R. speratus was inferred from intra-/interspecific phylogeny and divergence times estimated between the clades of the phylogenetic tree. The estimated migration route and divergence time of ancestral R. speratus are congruent with recent paleogeographic hypotheses involving land-bridge connections between the Asian continent and the Japanese Archipelago. We suggest that ancestral R. speratus separated into northern and southern Japanese populations after its migration into the Japanese main islands from East China during the early Pleistocene via the East China Sea basin, which may have been exposed during that period. The Korean populations seem to have diverged recently from southern Japanese populations; this may explain the current distribution of R. speratus in the Japanese Arachipelago, and account for why it is restricted to northern areas of the Tokara Strait.

북방전복 (Haliotis discus hannai) 의 mitochondrial DNA 영역별 유전적 변이성 분석 (Analysis of genetic divergence according to each mitochondrial DNA region of Haliotis discus hannai)

  • 본 연구는 우리나라의 주요양식 품종인 북방전복을 대상으로 지금까지 전복류에서는 사용되지 않았던 mtDNA의 protein coding 영역 ND2, ND5, ND4, ND4L, ND6, ND1의 6개영역과 protein noncoding 영역인 12SrRNA(ribosomal RNA) 을 포함해 총 7개 영역을 이용하여 각 영역의 유전적 변이성 및 개체간 유전적 유연관계 등을 분석하여 각 영역별 특성을 파악하고 이러한 특성을 고려하여 유전학적 분석에 적합한 분자유전마커를 개발하였다. 유전적 변이성은 ND4 영역 (Haplotype diversity = 1.000, Nucleotide diversity = 0.010823) 이 가장 높게 나타났으며, 개체간의 유전적 차이는 ND2 및 ND1 영역이 각각 90% 및 87%로 유의적으로 명확히 구분할 수 있었다. 따라서 유전적 변이성이 가장 높은 ND4 영역과 영역내의 클러스터 간의 유전적 차이가 명확한 ND2 및 ND1 영역을 복합적으로 활용할 경우 북방 전복의 집단유전학 및 계통분류학 분석에 유용한 분자유전마커로 사용할 수 있을 것이라 생각된다.

배추좀나방(나비목: 집나방과)의 haplotype 다양성과 유전자 이동률 (Haplotype Diversity and Gene Flow of the Diamondback Moth, Plutella xylostella(L.) (Lepidoptera: Yponomeutidae), in Korea)

  • 국내 4개 지역으로부터 채집된 배추좀나방(Plutella xylostella)의 미토콘드리아 DNA중 COI 유전자 일부 (438 bp)의 염기서열을 결정, 유전적 다양도 및 유전자 이동정도를 파악함으로써 집단 유전적 구조 및 특성에 대하여 연구하였다. 총 21개체로부터 13개의 mtDNA haplotype을 얻었으며 이들의 변이는 0.3~1.4%로 다른 곤충을 대상으로 한 유사연구와 비슷한 크기를 나타내었으며 haplotype 다양도는 매우 높았다(평균 h=0.81). 지리적으로 먼 제주도의 개체군과 경남 김해 두 지역(11km 거리)의 개체군을 비교한 결과, 통계적으로 유의한 정도의 유전적 격리(p<0.05%)는 전혀 관찰되지 않았으며, 대신 상당한 정도의 세대당 암컷 이동률(Nm=2-30)을 보였다. 또한 GenBank에 등록된 하와이의 배추좀나방 haplotype은 본 연구에서 얻은 것들과 유전적으로 흡사하였다. 종합적으로, 국내 배추좀나방은 전체적으로는 많은 haplotype수에 기인한 적절한 크기의 유전적 분화율을 보유하고 있으며 국지적으로는 상당한 이동력에 의한 장거리 이동으로 개체군내 높은 haplotype 다양도를 보이며 동시에 지역간의 유전적 유사성을 나타낸다고 요약되었다.

Mitochondrial DNA Sequence Variation of the Mason Bee, Osmia cornifrons (Hymenoptera: Apidae)

  • In order to understand geographic genetic variation and relationship among populations of the mason bee (Osmia cornifrons Radoszkowsky), which is used as pollinator for apple tree, we sequenced a portion of mitochondrial (mt) COI gene, which corresponds to "DNA Barcode" region (658 bp) from 81 O. cornifrons individuals collected over eight localities in Korea. The sequence data revealed overall moderate to low genetic diversity within species, with a maximum sequence divergence of 0.76%. Geographically, two haplotypes (BAROC01 and BAROC02) were widespread with a frequency of 82.7%, whereas several haplotypes were found in a locality as a single individual, suggesting that haplotype distribution can be summarized as coexistence of a few widespread haplotypes and several regionally restricted haplotypes. Overall, high rate of per generation female migration (Nm=$1.1{\sim}$infinite) and low level of geographic subdivision ($F_{ST}=0{\sim}0.315$) among localities were characteristic. Although two populations (p < 0.026) were genetically subdivided from the remaining localities, no clear polarity was observed. Taken together, the nature of genetic divergence of the mason bee populations is characterized as one that possessing moderate to low genetic diversity, high gene flow, and wide spread haplotypes with ahigh frequency, concordant with the capability of dispersal in connection with the lack of historical biogeographic barriers.

벼에서 RAPDs 분석에 의한 양친의 유전적 유사성과 잡종강세의 상관 (Relationship Between Heterosis and Genetic Divergence in Tongil-type Rice(Oryza sativa L.))

  • RAPDs 분석에 의한 양친의 유전적 유사성 정도가 F1 잡종강세 예측에 효율적으로 이용될 수 있는지 알아보고자 통일형 6 품종(밀양42, IR747, 백운찰벼, 삼강벼, 수원307, 이리356) 간 반이면 교배 F1 15조합을 작성하였다. 양친과 F1을 공시하여 조사한 수량 등 8개의 특성과 RAPDs 분석을 통해 구한 양친의 유전적거리와의 관계를 조사하였다. 120개의 Operon 프라이머를 6개 품종의 DNA 증폭에 이용한 결과 3367개의 밴드가 발생되었으며 이중 168개가 6개 품종 중 1개 이상의 품종에서 변이를 보였다. 6품종간 유전적 거리는 최소 0.157(백운찰벼와 수원 307호 간), 최대 0.383(삼강벼와 이리356호 간)의 분포를 보였다. 잡종강세의 정도는 정조수량(129%), 수당립수(125%), 수장(109%) 등이 높은 잡종강세를 보였고, 유전적거리는 수당립수와 통계적으로 유의한 부의 상관을 보였지만, 정조수량, 천립중 등과는 유의한 상관을 보이지 않았다.

Genetic Variation and Population Structure of Crepidiastrum lanceolatum (Compositae)

  • Crepidiastrum lanceolatum ($H_{OUTT}$) $N_{AKAI}$ (Compositae) is distributed in East Asia including Korea. Genetic diversity and population structure of six C. lanceolatum populations in Korea and two populations in Japan were determined using genetic variation at 19 allozyme loci. The percent of polymorphic loci within the enzymes was 42.1%. Genetic diversity at the species level and at the population level was low (Hes : 0.077; Hep : 0.068, respectively), where-as the extent of the population divergence was relatively low ($G_{ST}$ : 0.093). One of the most striking features of this study was the more significant difference within populations than among populations. An indirect estimate of the number of migrants per generation (Nm : 2.44) indicated that gene flow was moderate among eight populations of the species. In addition, analysis of fixation indices revealed a slight heterozygosity deficiency in some populations and at some loci. Narrow geographic ranges, short-lived perennial herbaceous, and small population sizes are mainly associated with the low level of genetic variation.n.

  • 우리나라 해역에 서식하고 있는 농어 2형(점이 없는 계통과 점이 있는 계통)의 유전학적 분류 및 특징을 조사하기 위하여 isozyme 분석을 실시한 결과, 2형에 있어서 4개의 유전자좌의 분기가 관찰 되었으며 이중 Pt-1 유전자좌는 완전 분기되어 있는 것으로 확인되었다. 총 유전적 변이 보유량은 2형 모두 동일하였으나 점이 없는 계통은 다형 변이 유전좌의 비가 높았으며, 점이 있는 계통은 변이 유전자좌의 비가 높은 것으로 나타났다. 2형에 있어서 평균 이형 접합체율의 기대치에 차이가 나타나 점이 없는 계통과 점이 있는 계통은 유전적으로 집단구조가 다름이 확인 되었다. 2형간의 유전적 분화 정도를 나타내는 유전적 거리(D)의 값은 0.12036이었으며, 분화연도는 $6.2{\times}10^5$년전 인 것으로 추정되었다.

Mitochondrial DNA Sequence Variations and Genetic Relationships among Korean Thais Species (Muricidae: Gastropoda)

  • Thais Roding, 1798, commonly known as rock-shell, is among the most frequently found gastropod genera worldwide on intertidal rocky shores including those of Japan, China, Taiwan and Korea. This group contains important species in many marine environmental studies but species-level taxonomy of the group is quite complicated due to the morphological variations in shell characters. This study examined the genetic variations and relationships among three Korean Thais species based on the partial nucleotide sequences of mitochondrial cox1 gene fragments. Phylogenetic trees from different analytic methods (maximum parsimony, neighbor-joining, and maximum likelihood) showed that T. bronni and T. luteostoma are closely related, indicating the most recent common ancestry. The low sequence divergence found between T. luteostoma and T. bronni, ranging from 1.53% to 3.19%, also corroborates this idea. Further molecular survey using different molecular marker is required to fully understand a detailed picture of the origin for their low level of interspecific sequence divergence. Sequence comparisons among conspecific individuals revealed extensive sequence variations within the three species with maximum values of 2.43% in T. clavigera and 1.37% in both T. bronni and T. luteostoma. In addition, there is an unexpectedly high level of mitochondrial genotypic diversity within each of the three Korean Thais species. The high genetic diversity revealed in Korean Thais species is likely to reflect genetic diversity introduced from potential source populations with diverse geographic origins, such as Taiwan, Hong Kong, and a variety of different coastal regions in South China and Japan. Additional sequence analysis with comprehensive taxon sampling from unstudied potential source populations will be also needed to address the origin and key factors for the high level of genetic diversity discovered within the three Korean Thais species studied.

  • This paper deals with the fuzzy modeling for the complex and uncertain system in which conventional and mathematical models may fail to give satisfactory results. mGA(messy Genetic Algorithm) has more effective and adaptive structure than sGA with respect to using changeable-length string and VEGA(Virus Evolution Genetic) Algorithm) can search the global and local optimal solution simultaneously with reverse transcription operator and transduction operator. Therefore in this paper, the optimal fuzzy model is obtained using Virus-messy Genetic Algorithm(Virus-mGA). In this method local information is exchanged in population so that population may sustain genetic divergence. To prove the surperioty of the proposed approach, we provide the numerical example.

유전 알고리즘의 조기수렴 저감을 위한 연산자 소인방법 연구 (On Sweeping Operators for Reducing Premature Convergence of Genetic Algorithms)

  • GA (Genetic Algorithms) are efficient for searching for global optima but may have some problems such as premature convergence, convergence to local extremum and divergence. These phenomena are related to the evolutionary operators. As population diversity converges to low value, the search ability of a GA decreases and premature convergence or converging to local extremum may occur but population diversity converges to high value, then genetic algorithm may diverge. To guarantee that genetic algorithms converge to the global optima, the genetic operators should be chosen properly. In this paper, we analyze the effects of the selection operator, crossover operator, and mutation operator on convergence properties, and propose the sweeping method of mutation probability and elitist propagation rate to maintain the diversity of the GA's population for getting out of the premature convergence. Results of simulation studies verify the feasibility of using these sweeping operators to avoid premature convergence and convergence to local extrema.
