Comparative Genetic Diversity in Natural and Hatchery Populations of Indian Major Carps (C. catla and L. rohita)

  • This study deals with the characterization of three populations (two hatchery and one natural) of Indian major carps Catla catla and Labeo rohita from different locations in India. The genetics of Indian major carps has been completely obscure and this is the first report on comparative allozyme variations in natural and hatchery population. The total 10 biochemical genetic markers used to measure interspecific and intraspecific level of diversity. The allele frequency data indicate different level of genetic variability in three populations. The hatchery population exhibited least polymorphism, low level of heterozygosity and genetic diversity.

On the Negative Estimates of Direct and Maternal Genetic Correlation - A Review

  • Estimates of genetic correlation between direct and maternal effects for weaning weight of beef cattle are often negative in field data. The biological existence of this genetic antagonism has been the point at issue. Some researchers perceived such negative estimate to be an artifact from poor modeling. Recent studies on sources affecting the genetic correlation estimates are reviewed in this article. They focus on heterogeneity of the correlation by sex, selection bias caused from selective reporting, selection bias caused from splitting data by sex, sire by year interaction variance, and sire misidentification and inbreeding depression as factors contributing sire by year interaction variance. A biological justification of the genetic antagonism is also discussed. It is proposed to include the direct-maternal genetic covariance in the analytical models.

Association Study Between Genetic Polymorph isms in Interleukin-1 Gene Family and Adult Periodontitis in Korean

  • Adult periodontitis (AP) is a chronic inflammatory disease whose etiology is not well defined. Some studies suggested that the clinical characteristics of this disease may be in part explained by genetic factors, and some attempts to find genetic markers for this disease were successful. The interleukin-1 (IL-1) gene family as one of genetic factors may influence the expression of adult periodontitis. The aim of present study is to investigate the frequencies of genetic polymorphisms in the IL-1 gene family encoding three genes (IL-1A, IL-1B and IL-1RN) in Korean AP patients and periodontically healthy controls. There were no significant differences in genotype and allele frequencies of these polymorph isms between two groups, respectively. However, -511 polymorphism of IL-1 B gene was significantly associated with mean pocket depth (MPD, mm) value in AP patients (P<0.05). Therefore, our results suggest that -511 polymorphism in the IL-1B gene may be useful as a genetic marker for the severity of AP in Koreans.

  • Cleft lip and/or palate are common birth defects in humans and the causes including multiple genetic and environmental factors are complex. Combinations of genetic, biochemical, and embryological approaches in the laboratory mice are used to investigate the molecular mechanisms underlying normal craniofacial development and the congenital craniofacial malformations including cleft lip and/or palate. Both forward and reverse genetic approaches are used. The forward genetic approach involves identification of causative genes and molecular pathways disrupted by uncharacterized mutations that cause craniofacial malformations including cleft lip and/or cleft palate. The reverse genetic approach involves generation and analyses of mice carrying null or conditional mutations using the Cre-loxP mediated gene targeting techniques.

Number of Calves Produced at Specified Age as a Measure of Reproductive Performance in Beef Cattle under Artificially-Inseminated Breeding Scheme

  • Reproductive abilities in beef cattle herds are receiving increased attention due to recent rises in production costs. To achieve more efficient management, a measure of fertility, namely the number of calves produced at k yr of age ($NCP_k$), was developed and its genetic parameters were estimated from Japanese Black cows by restricted maximum likelihood procedures. The k examined were distributed from 2 to 10 yr of age and $NCP_2$ averaged 1.077 calves over 43,536 cows. The averages increased by approximately 0.9 calf with each additional 1 yr increment in k. Heritabilities of $NCP_k$ were estimated to be low ranging from 0.083 to 0.162, which seemingly suggested a difficulty of genetic improvement. However, large genetic variation and high accuracy were observed in predicted breeding values of $NCP_k$. For example, the breeding values of $NCP_7$ were predicted between -0.303 and +0.213 with average accuracy of 0.607 for cows with observations. Genetic correlations among different k were generally high and positive (0.474 to 0.995). The analyses showed that at least $NCP_4$ was required to maintain the genetic correlations of 0.8 or higher with subsequent $NCP_k$. Also $NCP_5$ maintained the genetic correlations of 0.9 or higher with subsequent $NCP_k$ The results suggested some possibilities for $NCP_k$ to be a selection criterion considering its genetic variation, high accuracy and consistency with subsequent performance.

ADFP promoter polymorphism associated with marbling score in Korean cattle

  • Marbling score (MS) is the major trait that affects carcass quality in beef cattle. In this study, we investigated the association between genetic polymorphisms of the adipose differentiation-related protein gene (ADFP) and carcass traits in Korean cattle (also known as Hanwoo). Using direct DNA sequencing in 24 unrelated Korean cattle, 25 novel polymorphisms were identified within all exons and their flanking regions of ADFP, including the promoter region (1.5 kb). Among them, 21 polymorphic sites were selected for genotyping in the beef cattle (n = 425). Statistical analyses revealed that one promoter polymorphism (c.-56-18A > G) was associated with MS (P = 0.009). The 'A' allele of c.-56-18A > G exerted a lowering effect on MS, e.g., the lowest MS was found in 'A/A' (MS = 2.09 ${\pm}$ 1.23), intermediate in 'A/G' (MS = 2.11 ${\pm}$ 1.31), and the highest in 'G/G' (MS = 2.47 ${\pm}$ 1.47). Our findings suggest that these polymorphisms in ADFP might be important genetic factors involved in carcass quality in beef cattle.

ZNF435, a Novel Human SCAN-containing Zinc Finger Protein, Inhibits AP-1-mediated Transcriptional Activation

  • Zinc finger transcription factor genes are a significant fraction of the genes in the vertebrate genome. Here we report the isolation and characterization of a human zinc finger-containing gene, ZNF435, from a fetal brain cDNA library. ZNF435 cDNA is 1290 base pairs in length and contains an open reading frame encoding 349 amino acids with four C2H2-type zinc fingers at its carboxyl terminus and a SCAN motif at its amino terminus. RT-PCR results showed that ZNF435 was expressed in all tested tissues. A ZNF435-GFP fusion protein was located in the nucleus and the four zinc fingers acted as nuclear localization signals (NLSs). ZNF435 was found to be capable of homo-association, and this effect was independent of its zinc fingers. Furthermore, ZNF435 proved to be a transcription repressor as its overexpression in AD293 cells inhibited the transcriptional activities of AP-1.

  • 패턴 인식 분야에는 지수적 탐색 공간을 가진 최적화 문제가 많이 있다. 이를 해결하기 위해 부 최적해를 구하는 순차 탐색 알고리즘이 사용되어 왔고, 이들 알고리즘은 국부 최적점에 빠지는 문제점을 안고 있다. 최근 이를 극복하기 위해 유전 알고리즘을 사용하는 사례가 많아졌다. 이 논문은 특징 선택, 분류기 앙상블 선택, 신경망 가지치기, 군집화 문제의 지수적 탐색 공간 특성을 설명하고 이를 해결하기 위한 유전 알고리즘을 살펴본다. 또한 향후 연구로서 가치가 높은 주제들에 대해 소개한다.

  • The objective of this study is the development of a size and shape discrete optimum design algorithms, which is based on the genetic algorithms and the fuzzy theory. This algorithms can perform both size and shape optimum designs of plane and space trusses. The developed fuzzy shape-GAs (FS-GAs) was implemented in a computer program. For the optimum design, the objective function is the weight of structures and the constraints are limits on loads and serviceability. This study solves the problem by introducing the FS-GAs operators into the genetic.

Genetic Analysis of TGFA, MTHFR, and IFR6 in Korean Patients Affected by Nonsyndromic Cleft Lip with or without Cleft Palate (CL/P)

  • Nonsyndromic cleft lip with or without cleft palate (CL/P) is a common craniofacial birth defect that is the result of a mixture of genetic and environmental factors. While studies have identified a number of different candidate genes and loci for the etiology of CL/P, the results have not been consistent among different ethnic groups. To study the genetic association of the candidate genes in Korean patients affected by CL/P, we genotyped 97 nonsyndromic CL/P patients and 100 control individuals using single nucleotide polymorphic markers at the MTHFR, TGFA, and IRF6 genes. We report that the T3827C marker at TGFA showed significant association with nonsyndromic CL/P, but all the other markers tested were not significantly associated with nonsyndromic CL/P in Korean patients.
