늦반딧불이 Pyrocoelis rufa(딱정벌레목: 반딧불이과)의 미토콘드리아 DNA 염기서열 변이 (Mitochondrial DNA Swquence Variation of the Firefly, Pyrocoelia rufa(Coleoptera: Lampyridae), in Korea)

  • 국내 늦반딧불이(Pyrocoelia rufa)이 미토콘드리아 DNA중 COI 유전자 일부(403 bp)의 염기서열을 결정, 집단내 유전적 다양도, 지역적 변이, 계통유전적 관련에 대한 분석을 실시하였다. 남해, 부산, 무주, 용인 등 우리나라 4개 지역으로부터 채집된 총 26개체로부터 7개의 mtDNA haplotype을 얻었으며 이들의 변이는 0.2~1.2%이었다. 도심인 부산에서 채집한 늦반딧불이는 다른 산림 및 농업의 채집지역과 달리 하나의 haplotype으로 고정되어 있어 도시화에 따른 집단의 병목현상과 서식처 파편화가 심각했음을 보여주었다. 그러나 우리나라 최대의 반딧불이 서식처이자 보호지역으로 지정된 무주로부터 4개의 haplotype을 얻었으며 이들의 최대염기 치환율은 1.0%로 가장 높은 집단내 유전적 다양도를 나타내었다. 근해의 섬인 남해의 늦반딧불이는 상대적으로 낮은 haplotype 다양도(H=0.25)와 계통유전적으로 이질적인 haplotype(PR7)의 존재로 요약되었는데 이는 비교적 가까운 과거의 한반도 생물지리 역사 및 유전자 이동에 의해 나타난 현상으로 설명하였다. 계층적 유전분석 결과 무주-용인과 부산-남해 그룹의 형성은 역사적으로 두 그룹사이에 유전자 이동에 반한 장벽의 존재 가능성을 제시한다고 설명하였다.

mtDNA Cytochrome b 유전자에 기초한 한국재래염소의 계통유전학적 분석 (Phylogenetic Analysis of Korean Native Goats Based on the Mitochondrial Cytochrome b Gene)

  • 한국재래염소의 계통유전학적 위치를 확인하기 위해서 한국재래염소 4개 집단 48두를 공시한 후 mitochondrial DNA (mtDNA) 내부의 cytochrome b 유전자의 전체서열을 분석하였다. 또한 이 서열들을 이용하여 한국재래염소의 유전적 다양성을 확인하였고, 다른 나라의 여러 염소품종들과의 계통유전학적 분석을 수행하였다. 한국재래염소 cytochrome b 유전자 서열을 토대로 3개의 염기변이가 동정되었으며, 그 중 2개는 아미노산 치환을 일으키는 missense 변이로 확인되었다. 또한 4개의 haplotype으로 분류되었는데, 이 중 3개는 중국 재래염소 품종에서도 나타났으나 다른 나라의 품종에서는 확인되지 않았다. 계통유전학적 분석 결과 모든 재래흑염소는 4개의 clade를 형성하였으나, 5개의 야생염소와는 독립적인 그룹을 형성하였다. 한국재래염소는 mtDNA D-loop에 분류되는 여러 모계혈통 중 모계혈통-A로 추정되는 clade 1에 포함되었다. 한국재래염소에서 보여진 각각의 haplotype은 중국 재래염소품종들과 상대적으로 가까운 유전적 유연관계를 보였다. 기존 연구결과와 본 연구의 분석결과를 종합해보면 과거에 일부 중국 재래 염소품종이 한반도로 유입되어 한국재래염소의 기원 및 가축화에 영향을 주었을 것으로 사료된다.

Calcitonin Gene Related Peptide에 의한 치수미세순환 조절 (REGULATION OF PULPAL MICROCIRCULATION BY CALCITONIN GENE-RELATED PEPTIDE)

  • 본 연구에서는 감각성 neuropeptide인 CCRP의 치수혈류 조절에 관해 교감신경과의 유기적 관계를 연구함으로써 CGRP의 치수혈류 조절기전을 밝히고자 하였다. 열두 마리의 전신마취된 고양이에서 실험하였으며 CGRP를 혈관을 통해 전신적 또는 국소적으로 투여하였다. 견치에서 치수혈류의 변화를 측정하고 paired t-test로 통계분석 하였으며 $95\%$ 수준에서 유의성을 검증하였다. CGRP $(0.31{\mu}g/kg)$를 전신정맥으로 주사시, 전신혈압에 현저한 영향을 나타내면서 치수혈류는 평균 $68.85\%$의 일차적인 증가와 감소를 보였고 이차적으로 다시 평균 $161.8\%$ 증가하였다가 감소하였다. CCRP를 저용량 $(0.03{\mu}g/kg)$으로 국소적으로 투여시, 치수혈류는 평균 $2.92\%$의 미약한 증가를 나타내었다. 교감신경을 전기자극시 (10Hz, 4V, 1.5ms), 전신혈압은 영향을 받지 않으면서 치수혈류가 유의하게 평균 $57.88\%$ 감소하였다. 교감신경 자극으로 치수혈류가 저하되어 있는 동안 주입한 CCRP는 저하된 치수혈류를 유의하게 회복시켰다. CGRP의 이 치수혈류 증가 효과는 $CGRP_{8-37}$에 의해 효과적으로 차단되었다.

The Combined Effect of Gamma Knife Irradiation and p53 Gene Transfection in Human Malignant Glioma Cell Lines

  • Objective: The purpose of this study is to elucidate in vitro responses to combined gamma knife irradiation and p53 gene transfection on human malignant glioma cell lines. Methods: Two malignant human glioma cell lines, U87MG (p53-wild type) and U373MG (p53-mutant) were transfected with an adenoviral vector containing p53 (MOI of 50) before and after applying 20Gy of gamma irradiation. Various assessments were performed, including, cell viability by MTT assay; apoptosis by annexin assay; and cell cycle by flow cytometry, for the seven groups: mock, p53 only, gamma knife (GK) only, GK after LacZ, LacZ after GK, GK after p53, p53 after GK. Results: Cell survival decreased especially, in the subgroup transfected with p53 after gamma irradiation. Apoptosis tended to increase in p53 transfected U373 MG after gamma irradiation (apoptotic rate, 38.9%). The G2-M phase cell cycle arrest markedly increased by transfecting with p53, 48 hours after gamma knife irradiation in U373 MG (G2-M phase, 90.8%). Conclusion: These results suggest that the in vitro effects of combined gamma knife irradiation and p53 gene transfection is an augmentation of apoptosis and G2-M phase cell cycle arrest, which are more exaggerated in U373 MG with p53 transfection after gamma knife irradiation.

SDC4 Gene Silencing Favors Human Papillary Thyroid Carcinoma Cell Apoptosis and Inhibits Epithelial Mesenchymal Transition via Wnt/β-Catenin Pathway

  • As the most common type of endocrine malignancy, papillary thyroid cancer (PTC) accounts for 85-90% of all thyroid cancers. In this study, we presented the hypothesis that SDC4 gene silencing could effectively attenuate epithelial mesenchymal transition (EMT), and promote cell apoptosis via the $Wnt/{\beta}-catenin$ signaling pathway in human PTC cells. Bioinformatics methods were employed to screen the determined differential expression levels of SDC4 in PTC and adjacent normal samples. PTC tissues and adjacent normal tissues were prepared and their respective levels of SDC4 protein positive expression, in addition to the mRNA and protein levels of SDC4, $Wnt/{\beta}-catenin$ signaling pathway, EMT and apoptosis related genes were all detected accordingly. Flow cytometry was applied in order to detect cell cycle entry and apoptosis. Finally, analyses of PTC migration and invasion abilities were assessed by using a Transwell assay and scratch test. In PTC tissues, activated $Wnt/{\beta}-catenin$ signaling pathway, increased EMT and repressed cell apoptosis were determined. Moreover, the PTC K1 and TPC-1 cell lines exhibiting the highest SDC4 expression were selected for further experiments. In vitro experiments revealed that SDC4 gene silencing could suppress cell migration, invasion and EMT, while acting to promote the apoptosis of PTC cells by inhibiting the activation of the $Wnt/{\beta}-catenin$ signaling pathway. Besides, $si-{\beta}-catenin$ was observed to inhibit the promotion of PTC cell migration and invasion caused by SDC4 overexpression. Our study revealed that SDC4 gene silencing represses EMT, and enhances cell apoptosis by suppressing the activation of the $Wnt/{\beta}-catenin$ signaling pathway in human PTC.

Extracellular ATP Induces Apoptotic Signaling in Human Monocyte Leukemic Cells, HL-60 and F-36P

  • Extracellular adenosine 5'-triphosphate (ATP) affects the function of many tissues and cells. To confirm the biological activity of ATP on human myeloid leukemic cells, F-36P and HL-60, cells were treated with a variety of concentrations of ATP. The stimulation with extracellular ATP induced the arrest of cell proliferation and cell death. from the analysis of Annexin-V staining and caspase activity by flow cytometry. The Annexin-V positive cells in both cell lines were dramatically increased following ATP stimulation. The expression of P2 purinergic receptor genes was confirmed, such as P2X1, P2X4, P2X5, P2X7 and P2Y1, P2Y2, P2Y4, P2Y5, P2Y6, P2Y11 in both leukemic cell lines. Interestingly, ATP induced intracellular calcium flux in HL-60 cells but not in F-36P cells, as determined by Fluo-3 AM staining. Cell cycle analysis revealed that ATP treatment arrested both F-36P and HL-60 cells at G1/G0. Taken together, these data showed that extracellular ATP via P2 receptor genes was involved in the cell proliferation and survival in human myeloid leukemic cells, HL-60 and F-36P cells by the induction of apoptosis and control of cell cycle. Our data suggest that treatment with extracellular nucleotides may be a novel and powerful therapeutic avenue for myeloid leukemic disease.

Role of Kupffer Cells in the Vasoregulatory Gene Expression during Hepatic Ischemia/Reperfusion

  • Hepatic microcirculatory failure is a major component of reperfusion injury in the liver. Recent data provided some evidence that endothelium-derived vasoconstrictors and vasodilators may be functionally important to the control of the total hepatic blood flow under these conditions of circulatory failure. Since Kupffer cells provide signals that regulate the hepatic response in ischemia/reperfusion (I/R), the aim of this study was to investigate the role of Kupffer cells in the I/R-induced imbalance of vasoregulatory gene expression. Rats were subjected to 60 min hepatic ischemia, followed by 5 h of reperfusion. The Kupffer cells were inactivated by gadolinium chloride ($GdCl_3$, 7.5 mg/kg body weight, intravenously) 1 day prior to ischemia. Liver samples were obtained 5 hrs after reperfusion for RT-PCR analysis of the mRNA for genes of interest: endothelin-1 (ET-1), its receptors $ET_A and ET_B$, endothelial nitric oxide synthase (eNOS), inducible nitric oxide synthase (iNOS) and heme oxygenase-1 (HO-1). ET-1 mRNA expression was increased by I/R. mRNA levels for $ET_A$ receptors showed no change, whereas $ET_B$ receptor transcripts increased in the I/R group. The increases in ET-1 and $ET_B$ mRNA were not prevented by the $GdCI_3$ pretreatment. The mRNA levels for iNOS and eNOS significantly increased within the I/R group with no significant difference between the I/R group and the $GdCl_3$-treated I/R group. HO-1 mRNA expression significantly increased in the I/R group and this increase was attenuated by $GdCI_3$. In conclusion, we have demonstrated that an imbalance in hepatic vasoregulatory gene expression occurs during I/R. Our findings suggest that the activation of Kupffer cells is not required for I/R-induced hepatic microvascular dysfunction.

Recent advances in Bayesian inference of isolation-with-migration models

  • Isolation-with-migration (IM) models have become popular for explaining population divergence in the presence of migrations. Bayesian methods are commonly used to estimate IM models, but they are limited to small data analysis or simple model inference. Recently three methods, IMa3, MIST, and AIM, resolved these limitations. Here, we describe the major problems addressed by these three software and compare differences among their inference methods, despite their use of the same standard likelihood function.

  • A comparative study on ground boundary conditions for the airfoil in ground effect has been carried out. The objective of the present study is to clarify effects of the ground boundary conditions so that it will be helpful to analyse results of wind tunnel tests using the fixed ground board or the image method. A low Mach number preconditioned Navier-Stokes solver using the overlap grid method has been applied. It has been turned out that results with the symmetric boundary condition are almost the same to those with the moving boundary condition. Results with the fixed ground boundary show discrepancy to those with the moving boundary condition when flow separation on the ground board takes place.
