Isolation of Angiotensin Converting enzyme inhibitors from Ripe Cucurbita moschata Duch

  • Angiotensin converting enzyme (ACE) inhibitor acts on the inhibition of ACE and causes a decrease in blood pressure. There have been several reports on screening of ACE inhibitors from natural food products and protein hydrolysates of various food sources. Ripe Cucurbita moschata Duch has been used as an oriental medicine in Korea. To isolate ACE inhibitors, crude water extracts of the edible portion of ripe Cucurbita moschata Duch were obtained after heating in water at 95$^{\circ}C$ for 2 h. Crude extracts were then filtered using PM-10 and YM-1 membranes. The membrane-filtered solution was loaded onto Sephadex G-15 column equlibrated with a phosphate buffer. Among the four major fractions of gel permeation chromatography, the second fraction had the highest inhibitory activity of 65%. Further purification of the fraction using reversed-phase HPLC with a $C_{18}$ column produced ACE inhibitors, which were identified as a mixture having molecular mass of 222 and 273 by Tandem mass spectrometry.

  • Copolymers with two different side groups, 2,2,2-trifluoroethoxy and p-chlorophenoxy groups, have been synthesized by reaction of poly(dichlorophosphazene) with their sodium salts, and characterized by elemental analysis, gel permeation chromatography, IR and NMR spectroscopies. The molecular weight measurement has shown that the weight average molecular weight, ${\bar{M}}_w$ of the copolymers are all above $10^6$ and their molecular weight distributions are monomodal and narrow (${\bar{M}}_w/{\bar{M}}_n<4$). Thermal properties of these copolymers measured by DSC and TGA exhibited to be intermediate between those of the two corresponding homopolymers.

  • A theory is given for determining the molecular weights of block copolymers from the experimental elution volume in GPC. Interaction effect between a sample of block copolymer and a column packing material as well as the size effect are separately considered for the first time applying the partial exclusion mechanism proposed by Dawkins. The molecular weight determination shows 6${\%}$ standard-deviation from the molecular weights measured by an osmometric method for eight block copolymers, which is much more improved result than other methods, e.g., the universal plot method (13 ${\%}$) and the Runyon's method (12 ${\%}$). The reason which explains the better result is that our theory takes into account the interaction effect correctly.

  • To find optimum conditions necessary in converting physical properties of any resin into those of others, eleven kinds of HIPS (High-Impact Polystyrene) resins were prepared. First physical properties of eleven samples divided into three groups are analyzed by a torque rheometer (named Plasti-Corder, Model No.: PLD 651) and GPC (Gel Permeation Chromatography), and then optimum conditions on conversion among samples are obtained by calculation from computer simulation so that any sample subjected to each group can show physical properties of other samples in its group. Even though the kind of plasticizer of any sample is different with others in its group, once optimum conditions on conversion among samples are met, it is found that physical properties of any sample are identical or similar to those of others in each group.

  • Novel polysilamethylenosilanes (PSMS) were prepared by Wurtz type co-condensation of various mixtures of 2,4,4,6-tetrachloro-2,6-dimethyl-2,4,6-trisilahept ane (TSH) and dimethyldichlorosilane (D). When TSH was incorporated more than 25 mole%, PSMS polymers were soluble in common organic solvents probably due to the polycarbosilane linkage brought from TSH. The molecular weights of the polymer were measured by gel permeation chromatography and showed higher molecular weight with high TSH content. The thermal gravimetric residues increased as TSH contents increased. These properties suggested that PSMS polymers could be useful as ceramic precursors for silicon carbide.

  • Triphenylvinylsilane (TPVS) containing vinyl acetate (VAc), butyl acrylate (BA), and Nmethylolacrylamide (NMA) copolymers were prepared by emulsion polymerization. The polymerization was performed at $80^{\circ}C$ in the presence of auxiliary agents and ammonium peroxodisulfate (APS) as the initiator. Sodium dodecyl sulphate (SDS) and Arkupal N-300 were used as anionic and nonionic emulsifiers, respectively. The resulting copolymers were characterized by using Fourier transform infrared spectroscopy (FTIR), gel permeation chromatography (GPC), and dynamic light scattering (DLS). Thermal properties of the copolymers were studied by using thermogravimetric analysis (TGA) and differential scanning calorimetry (DSC). The morphology of copolymers was also investigated by scanning electron microscopy (SEM) and then the effects of silicone concentrations on the properties of the TPVS-containing VAc-acrylic emulsion copolymers were discussed. The obtained copolymers have high solid content (50%) and can be used in weather resistant emulsion paints as a binder.

  • Natural(outdoor) and accelerated(artificial) weathering tests were performed to investigate their influence on polycarbonate. The polycarbonate materials were prepared of various formulations divided into three batches, with existing, development materials1, development materials2, containing mixture of UV additives. Weathering tests were carried out as outdoor weathering(Seosan, South Korea) and accelerated weathering(xenon-arc), and the results analysis were evaluated based on yellow-index(YI), scanning electron microscope(SEM/EDS), energy dispersive x-ray spectroscopy(XRD), and gel permeation chromatography(GPC). Among the three materials, processing method development materials1 with UV stabilizer was excellent in weathering. This study can provide basic data for standardization of development and performance evaluation on livestock barn roof.

Urokinase Conjugated with Water-Soluble Dextran

  • Urokinase, a plasminogen activator, was conjugated with dextran by the cyanogen bromide activation-coupling method. The resulting water-soluble conjugate was purified by gel permeation chromatography on Sephadex G-200. The maximal activity was obtained when the ratio of urokinase/dextran was 1/20 for the coupling. The final preparation showed 5 CTA units/mg conjugate, 300 CTA units/mg protein, 8.4 % activity retention, and 47 % protein retention. The urokinase-dextran conjugate had good thermal, pH and storage stabilities. In addition, it showed greater resistance to the inhibitory effect of human plasma than native urokinase. Also in vitro biological half-life of urokinase increased 40 times by this conjugation. In view of activity, excellent stability and increased half-life, the conjugate can be a potential fibrinolytic agent in an injectable form.

  • DWP40458 is a novel human thrombopoietin mutein with two additional N-linked glycosylation site. The thermal stability of DWP40458 in both solution and lyophilized form was studied in the temperature range of 4-50$^{\circ}C$, compared with recombinant human TPO (rhTPO). When the aggregation or degradation pattern of DWP40458 and rhTPO solution was characterized by using SDS-PAGE, gel permeation chromatography (GPC) and reverse phase HPLC, it was found that thermostability of DWP40458 was significantly different to rhTPO in the temperature at 25, 40, 40, 50$^{\circ}C$.   • Components of polysaccharides isolated from ascidian tunic were measuerd by gel filtration, electrophoresis and chemical analyses. The sulfated polysaccharides consisted in sulfate, protein, uronic acid and amino sugars. Hexosamines were composed of arabinose, xylose, glucose, galactose, glucuronic acid, N-acetylgalactosamine and N-acetylglucosamine by gas chromatography analysis. The galactose was predominant hexose after autoclave and nutrase digestion followed by DEAE-cellulose ion exchange chromatography and gel-permeation chromatography on Sephadex G-100 and G-25. FT-IR spectra of isolated polysaccharides from ascidian tunic and standard chondroitin sulfates have similar functional groups of the type of vibration and frequency. Molecular weight of isolated polysaccharides by autoclave represented more than 40 kDa by polyacrylamide gel electrophoresis. But the neutrase treatment group divided into three band. The highest molecular band group was shown more than 100 kDa, and the two low molecular band group were shown about 22 kDa and 5 kDa, respectively, compare to standard materials.
