도라지 뿌리의 향기성분에 관하여 (Flavor Components in the Bellflower Roots (Platycodon glaucum Nakai))

  • 도라지 뿌리의 향기성분을 규명하고자 도라지 뿌리의 향기성분을 상압 수증기 증류법으로 포집하여 중성, 염기성, 약산성 및 산성성분으로 분획하였다. Diazomethane 법으로 methyl ester 화한 산성부분을 위시한 4부분은 모세관 column을 장치한 GC 및 GC-MS로 분석하였으며 함황화합물은 FPD 검출기로써 검출하였다. 도라지 뿌리로부터 aliphatic hydrocarbons류 6종, aromatic hydrocarbons류 10종, terpene hydrocarbons류 2종 alcohol류 12종, terpene alcohol류 8종 aldehyde류 17종 terpene aldehyde류 3종 ketone류 5종 ester류 5종, furan류 3종, thiazole류 2종, lactone류 2종, sulfide류 2종, phenol류 2종, 산류 12종 및 기타 5종을 위시하여 총 103종의 향기성분이 동정되었다. Caboxylic acid를 제외한 향기성분은 대부분 중성부분에서 동정되었고, 관능검사의 결과에서도 중성부분이 도라지 뿌리의 향기를 재현하는데 필요불가결한 것으로 나타났다. 관능검사의 결과, 중성부분에서 동정된 1-hexanal, trans-2-hexenal, 1-hexanol, cis-3-hexenol, trans-2-hexenol, 1-octenen-3-ol 등은 도라지 뿌리의 풋냄새의 주성분인 것으로 생각된다.

2008-2009년 국내 폐사 야생조류 및 동물 체내의 잔류농약 분석 (Analysis of residual pesticides in dead wild birds and other animals during 2008-2009 in Korea)

  • Pesticides are useful to eliminate harmful insects and grow crops however, misuse and abuse of pesticides may cause a death of wild birds, livestock, and companion animals. We analyzed residual pesticides in the ingesta and tissues from the dead wild birds, livestock, and companion animals which were suspected pesticide poisoning based on the diagnosis of the Animal Disease Diagnostic Center of National Veterinary Research and Quarantine Service (NVRQS). The samples were primarily brought to NVRQS from local communities and veterinary diagnostic laboratories. The 231 suspicious samples of pesticide poisoning based on the necropsy were analyzed by GC/NPD, GC/FPD, GC/ECD, or GC/MSD in 2008 and 2009. Pesticides were identified from the 55 samples of total 143 samples analyzed in 2008 and from the 34 samples of total 88 samples analyzed in 2009. The pesticide identification rates in 2008 were higher than the rates in 2009. It may have relevance to the increased samples with the outbreak of avian influenza in 2008 in Korea. The concentrations of pesticides found in the dead animals varied and exceeded the respective $LD_{50}$ of pesticides in many cases. Monochrotophos, phosphamidon, and methomyl were the most common pesticides found and those pesticides are high rank sales in Korea. The cause of pesticide poisoning in animal is assuming a misuse and an abuse of commonly used pesticides in Korea.

방오페인트에 함유된 유기주석물질 분석방법 연구 (Study of an analytical method for determining organotin compounds in anti-fouling paints)

  • An International Convention on the control of harmful anti-fouling system on ships(AFS Convention) was adopted on 5 October 2001 at Diplomatic Conference in London, and is expected to be presently effectuated with ratification of more than 25-member nations possessing about 25% of total world tonnage. This convention regulates the operation of harmful anti-fouling system and especially prohibits the use of organotin compounds contained in anti-fouling paint. Organotin compounds have a tendency to be easily extracted by specific solvents and have high polarity and low volatility as specific characteristics. This drives us to attempt of going through the process named derivatization that is required in analysis using a gas chromatography(GC). This study was conducted to determine the proper pre-treatment method, ethylation in comparison with hydridization on the analysis of tributyltin in organotin compounds and to verify the application of the method through the experimental analysis practically used anti-fouling paint and painted layer sample of the served ship.

우리나라 폐사 야생조류에서의 농약 분석 (Determination of pesticides in dead wild birds in Korea)

  • Pesticides are extensively used for the control of crop pests in agriculture and forestry. Organophosphate (OP) and carbamate pesticides are especially effective for the control of a variety of harmful insects. However, these cholinesterase inhibitors are also dangerous to non-target organisms (wildlife and other animals) because of their high acute toxicity. Most poisonings by pesticides occur as a result of misuse or accidental exposure, but intentional killing of unwanted animals also occurs. At the request of a local autonomous entity, we investigated wild bird poisonings by pesticides from 2003 to 2007. The 207 suspicious samples of pesticide poisoning based on the necropsy were analyzed by GC/NPD, GC/FPD, or GC/MSD. We looked for trends in the identification of pesticides in wild birds thought to have died from poisoning. Pesticides were determined in 59% of the total samples analyzed. Phosphamidon and monochrotophos were the most common pesticides identified, which amounted to 77% of the subtotal. Other OP and carbamate pesticides were also found in various concentrations from dead wild birds. The determined rates of pesticides were as high as 86% and 76% in 2003 and 2006, respectively, during an outbreak of avian influenza in Korea.

  • Benfuresate의 잔류분석을 위해 GC-FPD(S-mode)를 이용하여 분석조건을 설정 중 열적 불안정으로 인해 열분해 산물과 모 화합물이 혼재됨이 확인되었다. 가스크로마토그라프를 이용한 열분해의 주된 요인은 주입구의 온도와 주입구내의 기화 정도인 것으로 판단되었으며, 주입구를 Chauhan과 Debre 가 고안한 on-column으로 교체 후 분석한 결과 분해산물이 없는 단일 봉우리의 크로마토그램을 얻을 수 있었고, 절대량으로 0.6-4ng의 정량 범위 내에서 유의성 높은 검량선을 얻을 수 있는 주입구 온도는 20$0^{\circ}C$였다.

  • For gas chromatographic determination with the sulfur-specific flame photometric detector, nine ${\beta}$-lactam antibiotics without ${\alpha}$-amino group were esterified with borontrifluoride-methanol complex and then N-benzoylated with benzoyl chloride. The gas chromatographic separation of these products was successfully carried out on various silicon polymers (OV-1, OV-101, OV-17, OV-225, and QF-1) coated on the acid washed, silanized diatomite. The structure of the esterified and N-benzoylated product was confirmed by mass spectromer.

광양만에서 TBTCl (Tributyltin Chloride) 내성세균의 분리 및 분해활성 (Isolation and Degradation Activity of a TBTCl (Tributyltin Chloride) Resistant Bacteriain Gwangyang Bay)

  • 본 연구에서 우리는 광양만의 해수와 표층 퇴적물에서 TBTCl 내성세균의 개체수를 조사하였다. 광양만에서 TBTCl 내성세균의 개체수는 해수에서 $2.5{\times}10^3-3.8{\times}10^3$ cfu/mL 범위였으며, 표층 퇴적물에서 TBTCl 내성세균의 개체수는 $3.2{\times}10^5-9.1{\times}10^5$ cfu/g 범위였다. 광양만에서 TBTCl 내성세균의 종조성은 Vibrio spp. (19.2%)가 가장 높은 우점종으로 나타났고, Bacillus spp. (16.2%), Aeromonas spp. (15.2%), Pseudomonas spp. (13.1%), Klebsiella spp. (11.1%), Alteromonas spp. (9.1%), Pantoea spp. (6.1%), Proteus spp. (3.0%), Listeria spp. (2.0%), unidentified (5.0%)의 순으로 우점하였다. 또한 11개의 대표적인 TBTCl 내성균주는 여러 중금속들(Cd, Cu, Hg 및 Zn)에도 내성을 나타내었다. 이들 중에서 가장 강한 TBTCl 내성을 보이는 T7 균주를 선별하여, API 20NE등을 이용하여 본 균주의 형태학적, 생리학적 및 생화학적 특성들을 조사하였다. T7 균주는 16S rRNA gene 염기서열 분석에 의해 Pantoea 속으로 동정되어 Pantoea sp. T7으로 명명되었다. 또한 본 균주는 $500{\mu}M$의 TBTCl 농도에서도 60시간 배양 후에 정상 균주 성장의 50.7%까지 증식하였다. Pantoea sp. T7의 생물학적 TBTCl 분해활성은 GC-FPD 분석에 의해 측정되었는데, $100{\mu}M$의 TBTCl 농도에서 배양 40시간 후에 TBTCl 제거 효율은 62.7%로 나타났다.

Determination of Mono-, Di-, and Tri-Butyltin Compounds in Seawater of Several Bays in Korea

  • The method of study describes the determination of the nanogram amounts of mono-, di-, and tri-butyltin compounds in sea water. The procedure is based on 1) the conversion of tin compounds to non-ionic species by sodium tetraethyl borate as an alkylation reagent, 2) one-step ethylation and 3) extraction. No further purification or concentration of the extract was required before GC-FPD (Flame Photometric Detector). The detection limits for mono-, di- and tri-butyltin were 12.50, 6.02, and 4.19 ng/L. The linear range (0-120 ng/L) was solely dependent on the detector response height. Sea water samples (n=24) were obtained from Masan Bay, Busan, Ullsan, Gunsan, Sockcho, Yeasu, Donghae, Inchun, Kanghwa, and Ahsan in Korea. The samples were collected between June, 1992 to July, 1992 and the distribution of monobutyltintrichloride (MBTC), dibutyltindichloride (DBTC), and tributyltinchloride (TBTC) in sea water was recorded. The concentrations of butyltin compounds in seawater were found to be in the range of N.D. (not detected)-61.93 ng/L for MBTC, N.D.-32.16 ng/L for DBTC, and N.D.-55.76 ng/L for TBTC.

흰쥐를 이용한 Methidathion의 경구투여 및 피부도포 후 뇨 중 대사물질 측정 (Determination of Urinary Metabolites of Methidathion after Oral Administration and Dermal Application to Rats)

  • Methidation을 흰쥐에 경구투여 및 피부도포 후 뇨 중 대사물질을 GC/MS로 분석하였고, 투여경로에 따른 뇨 중 대사물질의 차이를 비교하였으며, 뇨 중 dialkyl phosphte의 시간별 배설량과 그 외 대사물질을 측정하여 다음과 같은 결과를 얻었다. Methidathion을 경구투여 및 피부도포 후 뇨 중 대사물질을 측정한 결과 유기인계 농약의 공통적인 대사물질인 dialkyl phosphtes 중 세 가지 물질인 DMP, DMTP 및 DMDTP로 동일하게 배설되었다. GC/MS로 주요조각이온을 확인한 결과 DMP는 m/z=153, 127, 109, DMTP는 m/z=198, 142, 109 그리고, DMDTP는 m/z-158, 125, 93에서 분자이온을 확인할 수 있었다. Methidathion을 경구투여 및 피부도포 후 뇨 중 dialkyl phosphtes의 시간별 배설량을 측정한 결과 경구투여의 경우 DMP는 12시간 이내에 79.2%, DMTP는 24시간 이내에 93.3% 그리고, DMDTP는 12시간 이내에 83.0%가 배설되었고, 피부도포의 경우 DMP는 24시간 이내에 71.1%, DMTP와 DMDTP는 48시간 이내에 각각 82.8%, 87.7%가 배설되었다. 이 결과 경구투여 한 실험에서는 48시간 이내에 뇨 중 dialkyl phosphtes가 완전히 배설되는 반면 피부도포 한 실험에서는 각 개체의 상태에 따라 대사물질들이 늦게는 168시간까지 배설이 지연됨을 알 수 있었다. 이상의 결과를 종합하면, 흰쥐에 methidathion을 경구투여 및 피부도포 후 뇨 중 대사물질을 측정한 결과 dialkyl phosphate 중 DMP, DMTP로 검출되었으며, 경구 투여와 피부도포 시 뇨 중 대사물질의 차이는 없었으나, 경구투여보다 피부도포의 경우 배설이 더 지연되었다. 결론적으로, methidathion의 뇨 중 대사물질인 DMP, DMTP 및 DMDTP는 이 농약의 생체모니터링 지표물질로서 사용될 수 있을 것으로 생각된다.

악취성분중 황화합물에 대한 기기분석법의 적용 (Application of Analytical Instrument Method for Determining Level of Malodorous Sulfur Compounds.)

  • The analytical instrument method was applied to analyze malodorous sulfur compounds emitted from industrial fields. Six factories and two sites which release malodorous substances into ambient air were selected to determine the level of hydrogen sulfide($H_2S$), methylmercaptan(MeSH), dimethyl sulfide($Me_2S$), and dimethyl disulfide($Me_2S_2$) using automated thermal desorption system (STD400) and GC-FPD in summer and fall seasons of 1999. The Air sampler for ATD400 uses a small pump to draw sample and a mass flow controller to adjust sample amount without using a dilution apparatus. The trap temperature of ATD400 reached to $-80^{\circ}$ by supplying liquid nitrogen and $H_2S$ can be analyzed under this condition. The recovery rates of $H_2S$, MeSH, $Me_2S$, and $Me_2S_2$ of odorous sulfur compounds standard were shown 98.2%, 93.6%, 98.2%, 99.4% respectively. The concentrations of $Me_2S$ at outside boundary of G market, L factory, and J factory were 0.018ppm, 0.021ppm, 0.032ppm in summer, respectively. The concentration of $H_2S$ at Nanjido landfill was 1.167ppm in summer, but that of $H_2S$ was not detected in fall because of soil covering. The concentration of H2S and $Me_2S_2$ at inside of Chonggye stream were 0.564ppm and 1.045ppm in summer, while those of H2S and Me2S2 were 0.285ppm and 0.465ppm in fall, respectively.
