An immunohistochemical study of the endocrine cells in the stomach of the Korean hedgehog(Erinaceus korean us) (고슴도치 위점막의 내분비세포에 관한 면역조직화학적 연구)

  • The gastric endocrine cells of the Korean hedgehog, Erinaceus korean us were studied immunohistochemically. Seven kinds of endocrine cells-, gastrin-, somatostatin-, 5-HT-, glucagon-, BPP-, motiIin-and GIP-immunoreactive cells- were identified in this study. The chracteristic findings of the regional distribution and relative frequency of them were examined. Gastrin-immunoreactive cells were very numerously detected only in the pyloric region. Somatostatin-immunoreactive cells were more numerous in the pyloric region than in the cardiac and fundic regions. 5-HT-immunoreactive cells were more numerous in the cardiac and pyloric regions than in the tundic one. Glucagon-immunoreactive cells were found few or rarely in the fundic and pyloric regions. BPP-imunoreactive cells were numerously distributed in the pyloric region, moderately in the fundic region and few in the cardiac region. Motilin-immunoreactive cells were found rarely or few in the fundic and pyloric regions. GIP-immnuoreactive Cells were detected onIy in the pyloric region.

Immunohistochemical study of the gastrointestinal endocrine cells in the Mongolian Gerbils, Meriones unguiculatus (몽골리안 저빌(Meriones unguiculatus)의 위장관 내분비 세포에 대한 면역조직화학적 연구)

  • In order to study the regional distribution and relative frequency of the immunoreactive endocrine cells in the gastrointestinal tract of the Mongolian gerbil, Meriones unguiculatus, the gastrointestinal tract was divided into 9 portions (cardia, fundus, pylorus, duodenum, jejunum, ileum, cecum, colon and rectum) and immunostained by immunohistochemical (PAP) method using 8 types of specific antisera against cholecystokinin (CCK)-8, gastrin, secretin, pancreatic polypeptide(PP), somatostatin, serotonin, glucagon and insulin. CCK-8-, gastrin-, somatostatin- and serotonin-immunoreactive cells were demonstrated in this study. These immunoreactive cells were found in the gastric gland regions of the pylorus or between parietal and chief cells of the fundus with round to spherical shaped, and in the interepithelial regions of the intestinal tract with spherical to spindle shaped except for jejunum where some of immunoreactive cells were also observed in the intestinal glands with round to spherical shaped. CCK-8-immunoreactive cells were restricted to the pylorus and duodenum with numerous and a few frequency, respectively. Gastrin-immunoreactive cells were restricted to the pylorus with numerous frequency. Similar to those of gastrin-immunoreactive cells, somatostatin-immunoreactive cells were restricted to pylorus with moderate frequency. Serotonin-immunoreactive cells were detected throughout whole gastrointestinal tract except for cardia and cecum with moderate or numerous frequency. However, no secretin-, PP-, glucagon- and insulin-immunoreactive cells were observed in this study. From these results, the appearance type, regional distribution and relative frequency of immunoreactive endocrine cells in the gastrointestinal tract of the Mongolian gerbils were somewhat lowered or restricted compared to those of other mammals and these differences were might be caused by feeding habits and species specification.

  • Histological changes, distributions and relative frequencies of bovine Sp-1/chromogranin (bCG)-, serotonin-, gastrin-, cholecystokinin-8(CCK-8)-, somatostatin-, S-100 protein-, polypeptide YY(PYY)- and glucagon-immunoreactive cells were investigated in the gizzard and pylorus of the chicken embryos from 10 days of incubation to hatching. Histologically, the pseudostratified columnar epithelium were observed from 10 days of incubation to 15 days of incubation, thereafter these epithelium were differentiated to simple columnar epithelium, gastric gland and/or mucosal gland. In the gizzard, bCG-immunoreactive cells were observed from 19 days of incubation and S-100 protein-immunoreactive cells were detected from 15 days of incubation to 18 days of incubation. No serotonin-, gastrin-, CCK-8-, somatostatin-, PYY- and glucagon-immunoreactive cells were found in this region. In the pylorus, bCG-, gastrin- and somatostatin-immunoreactive cells were observed from 16 days of incubation respectively, thereafter these cells were increased with ages. CCK-8-immunoreactive cells were detected on hatching and S-100 protein-immunoreactive cells were detected from 16 days of incubation to 18 days of incubation. No serotonin-, PYY- and glucagon-immunoreactive cells were observed in this region.

Screening of Precancerous Gastric Lesions by Serum Pepsinogen, Gastrin-17, Anti-Helicobacter Pylori and Anti-Caga Antibodies in Dyspeptic Patients over 50 years Old in Guilan Province, North of Iran

  • Background: The aim of this study was to investigate the value of serum gastric markers to differentiate between patients with precancerous lesions and nonatrophic chronic gastritis. Materials and Methods: Serum samples of 128 patients with dyspepsia who were candidates for endoscopic examination were tested for pepsinogen (PG I and PG II), PG I/II ratio, gastrin 17(G-17), anti-Helicobacter pylori (anti-H pylori ) and anti-CagA antibodies. Two sample t-tests, chi-square tests and Pearson's correlation analyses were used for analysis using SPSS (version 20). Results: PGI, PG I/II ratio values were decreased significantly in the precancerous lesion group (0.05, 0.001 respectively). The frequency of H pylori infection was significantly (p=0.03) different between the two groups ofthe study. Conclusions: We suggest PGI and the PG I/II ratio as valuable markers for screening of premalignant gastric lesions.

  • Regional distribution and relative frequency of endocrine cells in ten portions of the gastrointestinal tract of the Korean native cattle were observed by immunocytochemical methods using specific antisera against chromogranin, serotonin, somatostatin, glucagon, bovine pancreatic polypeptide(BPP), motilin, gastric inhibitory polypeptide(GIP), neurotensin, secretin, gastrin and substance P. The results observed are summarized as follows: In the abomasum, chromogranin-, serotonin-, somatostatin-, motilin-, glucagon-, gastrin-, and substance P-immunoreactive cells were found. Chromogranin-and serotonin-immunoreactive cells were more numerous in the fundic region than pyloric region. Somatostatin- and gastrinimmunoreactive cells were numerous in the pyloric region than in the fundic region. In the small intestine, chromogranin-, serotonin-, somatostatin-, glucagon-, BPP-, motilin-, gastrin-, GIP-, neurotensin-, secretin-, and substance P-immunoreactive cells were detected. Chromogranin-, somatostatin-, GIP- and secretin-immunoreactive cells were most numerous in the duodenum, while BPP-, motilin-, glucagon-, neurotensin- and substance P-immunoreactive cells were rarely seen in the small intestine. In the large intestine, chromogranin-, serotonin- and BPP-immunoreactive cells were widely distributed and most numerous in the rectum. Somatostatin-, glucagon- and substance P-immunoreactive cells were rarely seen in the large intestine.

Chongkukjang Mucilage Stimulates Immunohistochemical Activities of Gastrointestinal Tract in Rats

  • We investigated the effect of a viscous substance from chongkukjang (chongkukjang mucilage) on immunohistochemical reactions in rat gastrointestinal (GI) tracts, Rats fed a steady diet of chongkukjang mucilage showed an increase in the immunoreactive densities of gastrin and serotonin in the pyloric region of their stomachs and duodenal villi, The number of gastrin and serotonin immunoreactive cells was significantly higher in the experimental group than in the control group. Feeding on dietary chongkukjang mucilage increased the immunohistochemical densities of $CD4^+$ and $CD8^+$ lymphocytes in the mucosa and submucosa of the rats' gastroduodenal region. The universal nitric oxide synthase (uNOS)-immunoreactive neurons and nerve fibers were strongly stained in the vascular walls of the submucosa and myenteric plexus in rats fed the test diet. The results indicate that the intake of chongkukjang mucilage could increase mucosal immune activity, gastrointestinal motility, and blood circulation in the GI tract.

  • To investigate the regional distribution and relative frequency of the neurotensin-, pancreatic polypeptide(PP)- and gastrin/cholecystokinin(Gas/CCK)-immunoreactive cells in the gastrointestinal tract of the bullfrog(Rana catesbeiana) with developmental stages, group of bullfrogs subdivided into the tadpole with hindlegs, metamorphosed bullfrog with tail, 2 weeks after metamorphosed bullfrog and adult bullfrog, were stained by immunohistochemical methods (PAP methods). Neurotensin-immunoreactive cells were observed from the pylorus of the metamorphosed bullfrog with tail, but these cells were not detected after that periods. PP-immunoreactive cells were detected from the adult bullfrog in the pylorus, duodenum and ileum. These cells were most predominant in the pylorus. Gas/CCK-immunoreactive cells were observed from the adult bullfrog in the pylorus. According to these results, most of immunoreactive cells in the gastrointestinal tract of the bullfrog were appeared after the complete metamorphosed periods, in which the complete differentiation of structure of gastrointestinal tract were occurred, and variable changes of the regional distribution and relative frequency with developmental stages were observed.

  • The appearance of some gastrointestinal endocrine cells in the Meckel's diverticulum (MD) of the bean goose, Anser fabalis Latham was observed using specific antisera against serotonin, gastrin, cholecystokinin (CCK)-8, glucagon, secretin, somatostatin and human pancreatic polypeptide (HPP) with the peroxidase antiperoxidase (PAP) method. Among these specific antisera, serotonin-, gastrin-, CCK-8-, somatostatin- and HPP-immunoreactive cells were demonstrated in this study. Serotonin-, gastrin- and somatostatin-immunoreactive cells were detected at moderate frequency and CCK-8- and HPP-immunoreactive cells was rare and low frequencies, respectively. These immunoreactive cells were located in the superficial epithelium, intestinal crvpt and intestinal glands with spherical or spindle shaped cells having long cytoplasmic processes (open typed-cell). Mucosal layer of MD was composed of simple columnar epithelium and numerous intestinal glands. In addition, numerous lymphatic tissues were also demonstrated. In conclusion, histological profiles of MD were similar to any parts of the large intestine, especially the cecum, but the appearance, distribution and relative frequency of gastrointestinal endocrine cells were similar to those of upper parts of the small intestine. Although the exact digestive functions were unknown, the finding that the appearance, distribution and relative frequency of gastrointestinal endocrine cells in MD is similar to small intestine may be considered as distinct evidence that this organ may have some digestive functions.

Effect of Cingulate Cortical Ablation on Gastric Acid Secretion and Plasma Gastrin Concentration in Rats (흰쥐의 위산분비 및 혈장 Gastrin농도에 미치는 대상회전 제거의 영향)

  • It has been recently reported that cingulate cortex mar facilitate gastric acid secretion, but its facilitatory mechanism on the gastric acid secretion is still unclear. This study was undertaken to investigate the facilitatory mechanism of the cingulate cortex upon gastric acid secretion in rats. Twenty·three male albino rats were divided into the cingulate(N= 13) and the operated control(N= 10) groups. The cingulate group in which cingulate cortex was removed by suction through a slit-shaped opening on each side of, and parallel to, the sagittal suture. In the operated control group, the surgical procedure was ended with the skull opening and the incision of dura mater. The gastric juice was collected via a chronic gastric cannula after 24 hours of fast, with water ad libitum. The juice was collected continuously for 6 hours, starting 3 hours prior to the injection of gastric secretagogue, pentagastrin$(12\;{\mu}g/kg)$ or histamine dihydrochloride $(320\;{\mu}g/kg)$. Three one·hour samples were obtained before ana after the administration of each secretagogue. The two agents were injected separately and subcutaneously at intervals of 1 week, the blood samples were drawn from the abdominal aorta for the radioimmunoassay of postprandial plasma gastrin concentration in response to the forced feeding of 10% cod liver oil. 1) After pentagastrin administration, the volume of gastric juice tended to decrease, but its acidity tended to increase in the cingulate group compared with those of the operated control group. However, there was no any difference in the acid output between the two groups. 2) Histamine-stimulated acid output and volume of the gastric juice of the cingulate group decreased significantly compared with those of the operated control group, while there was not significantly different in the acidity between the two groups. 3) Before pentagastrin or histamine administration, any change was not observed in the gastric acid secretion following the cingulate cortical ablation. 4) Postprandial plasma gastrin concentration in response to the forced feeding of 10% cod liver oil was insignificantly lower in the cingulate group than in the operated control group. It is inferred from the above results that the cingulate cortex exerts a facilitatory influence upon the histamine-stimulated gastric acid secretion in rats, and its influence may not be mediated by the stimulation of gastrin secretion.

A Study on the Abnormalities of the Various Endocrine Functions Associated with Chronic Renal Failure (만성신부전에 수반된 각종 내분비기능 변화에 관한 연구)

  • In an attempt to evaluate the various serum hormonal changes in terminal renal failure, the fasting $T_4,\;T_3,\;rT_3$, gastrin, secretin prolactin, and aldosterone were measured by radioimmunoassay in 12 patients with chronic renal failure, who were admitted to Chosun University Hospital from January to June, 1981. From the analysis the following results were obtained. 1. Mean values of serum hormonal concentration in 10 normal control were as follows: $T_4,\;12.93{\pm}2.00ug/ml$; $T_3,\;113.0{\pm}28.7ng/ml$; $rT_3,\;0.11{\pm}0.10ng/ml$; gastrin, $100.0{\pm}47.1pg/ml$; secretin, $32.46{\pm}11.45pg/ml$; prolactin, $11.0{\pm}3.6ng/ml$; aldosterone, $137.0{\pm}58.5pg/ml$, 2. Mean values of serum hormonal concentration in 12 chronic renal failure were as follows: $T_4,\;7.34{\pm}2.43ug/ml$; $T_3,\;71.0{\pm}19.1ng/ml$; reverse $T_3,\;0.38{\pm}0.19ng/ml$; gastrin, $162.5{\pm}40.2pg/ml$; secretin, $107.50{\pm}20.48pg/ml$; prolactin, $34.0{\pm}17.2ng/ml$; Aldosterone, $86.5{\pm}19.8pg/ml$. 3. In chronic renal failure group, serum T 4, T 3 and adosterone level were significantly lower than those of the control group, but serum $rT_3$, gastrin, secretin and prolactin were significantly higher than those of the control group. 4. In the view of the correlation between serum hormonal concentions and serum creatinine levels in patients with chronic renal failure, $rT_3$, gastrin, secretin and prolactin showed increasing tendency(positive correlations), whereas $T_4,\;T_3$ and aldosterone showed decreasing tendency (negative correlations) with increment of serum cteatinine levels. And so, we observed the negative correlation between $T_3\;and\;rT_3$.
