Objectives: This experiment investigated the antimicrobial activity of Hwangryunheadok-tang and extracts of Scutellariae radix , Phellodendri cortex , Coptis rhizome , and Gardenia jasminoides against Salmonella typhimurium . Methods: After spreading S. typhimurium on a bacterial culture medium plate, antimicrobial activity was tested by dripping diluted Hwangryunheadok-tang or extracts of Scutellariae radix , Phellodendri cortex , Coptis rhizome , or Gardenia jasminoides (80 μl, diluted 100, 50, 10, and 1%) onto the plate, followed by culture for 16 to 72 hours. The minimal inhibitory concentration (MIC) was tested by dripping the minimum dilution that showed antimicrobial activity (80, 60, 40, and 20 μl) and measuring the density. The antimicrobial activity of Hwangryunheadok-tang and four extracts showed continuous antibacterial activity against S. typhimurium throughout the experiment. Result: 1. S. typhimurium . (Standard Microorganism, ATCC) 1) The Hwangryunheadok-tang and extracts of Scutellariae radix , Phellodendri cortex , and Coptis rhizome showed antibacterial activity in the undiluted solutions and in 50% dilutions. Gardenia jasminoides extract showed potency only in the undiluted solution. The antimicrobial potency of the undiluted solution was increased when the volume of inoculation was increased, but no difference was noted when the culture time was extended. All the extracts showed antimicrobial potency against S. typhimurium . 2. S. typhimurium isolated from diarrhea patients 1) When compared to the standard microorganism experiment on S. typhimurium , the MICs of the five extracts were increased. However, whereas the antimicrobial potency of doxycycline was lost entirely against bacteria isolated from patients with diarrhea, the antimicrobial potency of all the extracts was diminished but did not disappear. 2) The antimicrobial activity of Hwangryunheadok-tang and four extracts was continuous even when the culture time was extended to 16, 24, and 72 hours. Conclusions: The Hwangryunheadok-tang and four kinds of extracts have antimicrobial activity against S. typhimurium . Comparison of a standard microorganism with S. typhimurium isolated from diarrhea patients showed that the antimicrobial activity of all the extracts was better than that of antibiotics. Further studies should focus on the value and benefits of Hwangryunheadok-tang , and the four kinds of extracts as clinical treatments.
In this study, we conducted the anti-HIV-1 enzymes assay in vitro and its active components were predicted by QSAR in silico for the elucidation of action mechanism on anti-HIV of natural resources. The extracts of Gardenia jasminoides var. radicans Makino were tested for their inhibitory effects on the reverse transcriptase (RT), protease and ${\alpha}$-glucosidase. In the enzyme inhibition assay, the methanol extracts of Gardenia jasminoides var. radicans Makino stem showed a strong activity of 32.5% on the enzyme activity to cleave an oligopeptide, resembling one of the cleavage sites in the viral polyprotein which can only be processed by HIV-1 protease. Moreover the methanol extracts of stem exhibited alpha-glucosidase inhibitory activities of 26.1%. The methanol extracts ($100{\mu}g/ml$) of stem showed a weak activity of 13.4% on anti-HIV-1 RT using Enzyme Linked Oligonucleotide Sorbent Assay (ELOSA) method. However, all extracts of leaf and stem didn't exhibit the HIV-1-induced cytopathic effects with IC (inhibitory concentration) of $100{\mu}g/ml$ in HIV-1-infected human T-cell line. From these results, it is suggested that Gardenia jasminoides var. radicans Makino extracts may possibly be involved in the inhibition of reverse transcriptase, protease and alpha-glucosidase but can't vitally concerned with the viral replication in vitro.

Died Gardenia jasminoides fruit is used as a dye in the food and clothes industries in Asia. The present study investigated the anti-inflammatory effects of aqueous extract of G. jasminoides fruits (GJ) in BV-2 microglial cells. GJ inhibited lipopolysaccharide-induced nitric oxide (NO) secretion, inducible nitric oxide synthase (iNOS) expression, and reactive oxygen species production, without affecting cell viability. Furthermore, GJ increased the expression of heme oxygenase-1 (HO-1) in a dose-dependent manner. Moreover, the inhibitory effect of GJ on iNOS expression was abrogated by small interfering RNA-mediated knock-down of HO-1. In addition, GJ induced nuclear translocation of nuclear factor E2-related factor 2 (Nrf2), a transcription factor that regulates HO-1 expression. GJ-mediated expression of HO-1 was suppressed by LY294002, a phosphoinositide 3-kinase (PI-3K) inhibitor, and SB203580, a p38 kinase inhibitor, but not by the extracellular signal-regulated kinase (ERK) inhibitor PD98059 or c-Jun N-terminal kinase (JNK) inhibitor SP600125. GJ also enhanced the phosphorylation of Akt and p38. These results suggest that GJ suppresses the production of NO, a pro-inflammatory mediator, by inducing HO-1 expression via PI-3K/Akt/p38 signaling. These findings illustrate a novel molecular mechanism by which extract from G. jasminoides fruits inhibits neuroinflammation.
Genipin, aglycone of geniposide isolated from fruits of Gardenia jasminoides, was transformed into blue pigments through reaction with glycine and methylamine. The blue pigments formed from glycine-reacted genipin were passed through Bio-Gel P-2 resin yielding fractions GG1 and GG2, and those from methylamine-reacted genipin were separated into fractions GM1-GM4. The first eluted higher molecular-weight fractions, GG1 and GM1, had higher tinctorial strength than the later eluted lower molecular-weight fractions, GG2 and GM2-GM4, respectively. $^1H-NMR$ spectra of GG1 and GM1 showed very broad peaks indicating that structures of the pigments were highly polymeric. $^1H-NMR$ spectra of GG2, GM3, and GM4 showed several sharp peaks at aliphatic and aromatic regions with accompanying broad peaks, although the spectrum of GM2 was rather simple. Determination of the structural and physical nature of the isolated pigments is in progress.
This research was designed to determine what components of Gardenia jasminoides play a major role in inhibiting the enzymes related antidepressant activity of this plant. In our previous research, the ethyl acetate fraction of G. jasminosides fruits inhibited the activities of both monoamine oxidase-A (MAO-A) and monoamine oxidase-B (MAO-B), and oral administration of the ethanolic extract slightly increased serotonin concentrations in the brain tissues of rats and decreased MAO-B activity. In addition, we found through in vitro screening test that the ethyl acetate fraction showed modest inhibitory activity on dopamine-${\beta}$ hydroxylase (DBH). The bioassay-guided fractionation led to the isolation of five bio-active compounds, protocatechuic acid (1), geniposide (2), 6'-O-trans-p-coumaroylgeniposide (3), 3,5-dihydroxy-1,7-bis(4-hydroxyphenyl) heptanes (4), and ursolic acid (5), from the ethyl acetate fraction of G. jasminoides fruits. The isolated compounds showed different inhibitory potentials against MAO-A, -B, and DBH. Protocatechuic acid showed potent inhibition against MAO-B ($IC_{50}$$300{\mu}mol/L$) and DBH ($334{\mu}mol/L$), exhibiting weak MAO-A inhibition (2.41 mmol/L). Two iridoid glycosides, geniposide ($223{\mu}mol/L$) and 6'-O-trans-p-coumaroylgeniposide ($127{\mu}mol/L$), were selective MAO-B inhibitor. Especially, 6'-O-trans-p-coumaroylgeniposide exhibited more selective MAO-B inhibition than deprenyl, well-known MAO-B inhibitor for the treatment of early-stage Parkinson's disease. The inhibitory activity of 3,5-dihydroxy-1,7-bis (4-hydroxyphenyl) heptane was strong for MAO-B ($196{\mu}mol/L$), modest for MAO-A ($400{\mu}mol/L$), and weak for DBH ($941{\mu}mol/L$). Ursolic acid exhibited significant inhibition of DBH ($214{\mu}mol/L$), weak inhibition of MAO-B ($780{\mu}mol/L$), and no inhibition against MAO-A. Consequently, G. jasminoides fruits are considerable for development of biofunctional food materials for the combination treatment of depression and neurodegenerative disorders.
Iridoid glycosides (mainly geniposide) and crocetin derivatives (crocins) are the two major active constituents in Gardenia jasminoides Ellis. In the present study, geniposide, crocins, crocin-1 and crocetin were separated from gardenia chromatographically. Then, mice were orally administrated with geniposide (400 mg/kg b.w.), crocins (400 mg/kg b.w.), crocin-1 (400 mg/kg b.w.) and crocetin (140 mg/kg b.w.) once daily for 7 days with $CCl_4$. Hepatoprotective properties were evaluated by biochemical parameters: Administration of geniposide, crocins, crocin-1and crocetin significantly lowered serum alanine transaminase (ALT), aspartate transaminase (AST) and alkaline phosphatase (ALP) levels in $CCl_4$-treated mice. The reduced glutathione (GSH) levels and antioxidant enzymes (SOD and CAT) activities were also increased by geniposide, crocins, crocin-1 and crocetin. Histopathological examination of livers showed that these components reduced deformability, irregular arrangement and rupture of hepatocyte in $CCl_4$-treated mice. These biochemical results and liver histopathological assessment demonstrated that geniposide, crocetin derivatives and crocetin show comparative beneficial effects on $CCl_4$-induced liver damage via induction of antioxidant defense. Therefore, contents of geniposide and crocetin derivatives should be both considered for hepatoprotective efficacy of Gardenia jasminoides Ellis.
Natural pigments have many applications like colouring agent, pigments, food additives, and antiseptics. At present, instead of synthetic pigments that have contributed to the development of industry, many kinds of natural pigments have been developed. The constituents of gardenia fruits, Gardenia jasminoides ELLIS, are traditionally known as herb medicine and natural dyes/pigments due to the customer is needs. The fruits produce yellow carotenoid pigments and iridoid compounds. The two main components in the yellow pigments are called crocin and crocetin. The extraction mode of yellow pigment from Gardenia is depended upon the extraction time, temperature, and volume of solvent. Red pigments or blue pigments formed from geniposide and amino acids have been reported a lot. Geniposide, the principal iridoid glucoside contained in gardenia fruit, was hydrolyzed to genipinic acid or genipin as a precursor for the pigment by enzymatic or chemical reaction. These red or blue pigments prepared with materials hydrolyzed of geniposide and amino acid and had properties governed by the electrostatic character of the amino acid. The pigments showed good stability to heat and pH but were gradually bleached by light while the other natural pigments are unstable in light, heat, acid, and base solution. The safety of the pigments was considered to be of little virulences in comparison to synthetic pigments.
Colonial morphology of the various yeasts grown on the yeast morphology agar medium containing orange-yellow pigments extracted from the fruits of Gardenia jasminoides (GJPM) was investigated in hopes of the differential identification of yeasts on primary cultures. Colonies of Candida lusitaniae and Ca. guilliermondii on GJPM turned to prussian blue within three days of incubation and Ca. tropicalis and Ca. viswanathii turned to bluish gray but the latter species turned to deep blue after 7 days. Ca. krusei, Saccharomyces cerevisiae, and Torulopsis glabrata showed neutral gray, grayish green, and baby blue respectively after one or two weeks. However, the colonies of Ca. albicans and parapsilosis remained unchanged even after 20 days. Colonial color of Cryptococcus neoformans showing brown to purple brown was distinguishable not only from buff color of Cr. laurentii after one or two weeks incubation but also from those of Candida spp. Growth of certain species was promoted on GJPM. The findings clearly showed that Ga. jasminoides pigments medium was useful to the morphological differentiation of medically important yeasts which were often encountered in sputum or other clinical specimens.

Anthracnose symptoms caused by Glomerella cingulata were observed on leaves and stems of gardenia in Sunchon, Jeonnam in Korea in 2000. Symptoms on infected plants typically appeared as irregularly circular, dark-brown ring spots and water-soaked brown lesions. Based on cultural and morphological characteristics, the fungus (G-00-03 isolate) from the diseased plants was identified as Colletotrichum gloeosporioides, and its teleomorph stage was Glomerella cingulata. Healthy gardenia artificially inoculated with fungal spores showed anthracnose symptom 7 days after inoculation. This is the first report of gardenia anthracnose caused by Glomerella cingulata in Korea.
Response surface methodology (RSM) was used for the optimization of antioxidant capacity in gardenia extracts. The antioxidant capacities of gardenia fruit extracts were investigated by ferric reducing ability (FRA) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity (RSA) assays. The optimum extraction parameters for the strongest antioxidant capacity were the ethanol concentration (EtOH) of 48.9%, extraction temperature of $72.9^{\circ}C$, and extraction time of 29.9 min. Analysis of variance (ANOVA) showed that the quadratics of EtOH and extraction temperature had highly significant effect on the antioxidant capacity (p<0.001). The antioxidant capacity was correlated with contents of bioactive components [crocin, geniposide, and total phenolic (TP) compounds] in gardenia extracts and mainly attributed to the content of the TP compounds.
