α-Glucosidase inhibitory caged xanthones from the resin of Garcinia hanburyi

  • A yellow resin (gamboge) from Garcinia hanburyi has been widely used as folk medicine due to its antibacterial and antitumor activities. We isolated four ${\alpha}$-glucosidase inhibitory compounds from the methanol extract of gamboge. The compounds (1-4) were identified as gambogoic acid (1), moreollic acid (2), gambogic acid (3), and 10-methoxygambogenic acid (4), respectively through spectroscopic data including 2D-NMR and HREIMS. All compounds were examined in the enzyme inhibition assay against ${\alpha}$-glucosidase to identify their inhibitory potencies and kinetic behavior. All compounds (1-4) showed enzyme inhibition against ${\alpha}$-glucosidase, but the activity was significantly affected by the methoxy group on C-10 of ring A and pentenyl pyran moiety of ring D. For example, compound 1 ($IC_{50}=41.4{\mu}M$) bearing pyran ring eight times effective that 4 ($IC_{50}=350.6{\mu}M$) having geranyl group itself. Most active compound was found out to be gambogoic acid (1) which was analyzed most abundant metabolite in gamboge by LC-ESI-MS/MS. In kinetic study, compounds 1 and 2 were proved as noncompetitive inhibitors.

Effect of Garcinia kola seeds supplemented diet on growth performance and gonadal development of Oreochromis niloticus juveniles breed in ponds

  • Background: Despite the favorable geo-climatic potential of Cameroon, the national production of tilapia remains low due to poor tilapia growth reported by fish farmers. One of the underlying reasons is the early female maturation at a very small size and precocious breeding in earthen ponds, resulting in overpopulation which leads to stunted growth and therefore to the production of unmarketable fish size. Studies have shown that dietary supplementation of G. kola enhanced growth in young Clarias gariepinus and Oreochromis niloticus. It was also reported that G. kola inhibited spawning in Tilapia adult females. Therefore, this study sought to assess the effects of Garcinia kola as growth promoter and inhibitor of gonadal development in young Oreochromis niloticus. Methods: A total of 108 juveniles weighing $13.32{\pm}0.62g$ were randomly distributed in 9 hapas of 12 fishes each (9 females and 3 males) and fed for 70 days with three isonitrogenous diets, 40% crude protein with increasing Garcinia kola supplementation levels of 0 (normal diet), 6% and 10% (experimental diets). Physico-chemical parameters of the water (temperature, dissolved oxygen, pH, nitrate, nitrite, ammonia, and transparency) were measured twice a week. Every 14 days, fish were harvested, counted, and weighed. At the end of the experiment, three fish of each sex per replicate were sacrificed and their gonad and liver collected and weighed. Data were statistically analyzed using one-way analysis of variance repeated measure followed by Newman-Keuls multiple tests. Results: The results showed that all physico-chemical parameters of the water were within the recommended values for Tilapia culture. Tilapia fed 6% Garcinia kola supplemented diet displayed higher final body weight in males ($38.60{\pm}3.50g$) and females ($36.77{\pm}3.62g$) compared to those receiving normal diet ($36.23{\pm}1.36g$ and $25.87{\pm}3.32g$; respectively to the final body weight in males and females). The gonadosomatic index and hepatosomatic index indicated no significant variation in males while in females, these were significantly low in the experimental fish compared to control fish. Conclusion: The results of this study demonstrated that supplementation of G. kola seeds in diets of young Tilapia improved growth performance and impaired gonadal development in females.

Effect of Feeding Mixture of Soybean Peptides, L-Carnitine and Garcinia Cambogia Extract on Body Weight and Lipid Metabolism in Rats

  • This study was performed to investigate effects of the experimental mixture containing soybean peptides, L-carnitine and Garcinia Cambogia extract on body weight and lipid metabolism in rats. Male Sprague-Dawley rats (n=40) of eight weeks old were raised for four weeks with high fat diet (40% fat as calorie) to induce obesity. After induction of obesity, rats were feed control (C) diet, containing either 0.16% (+1D), 1.6% (+10D), 8% (+50D) of experimental mixture for eight weeks. Plasma aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activity and total protein and albumin concentration were not different among groups. The Body weight gain was significantly lower in experimental mixture diet group compared to control group. Weights of perirenal fat pad and epididymal fat pad in the +50D group were significantly lower than those in the +1D and +10D groups. Plasma total lipid and liver total cholesterol levels in the experimental groups were significantly lower than those in the control group. Fecal total lipid and total cholesterol excretions were highest in +50D group. These results suggest that the experimental mixture containing peptides, L-carnitine and Garsinia Canbogia extract is effective for reducing the body weight and adipose tissue weight which may be due to the modulation of lipid metabolism and the increased fecal excretion of lipid.

가시오갈피 잎 추출물이 흰쥐의 지질대사에 미치는 영향 (Effects of Ethanol Extract from Leaves of Eleutherococcus senticosu on Hyperlipidemia in Rats)

  • 본 연구는 가시오갈피 에탄올 추출물의 지질대사 개선 및 항비만 효과에 대해 구명하고자 3주령 된 흰쥐 40마리를 정상군, AIN-76 식이 대조군, 가시오갈피 추출물 처리군, 양성대조군(가르시니아)으로 나누어 6주간 실시하였다. AIN-76식이로 인하여 체중이 유의적으로 증가하였고, 가시오갈피 추출물은 AIN-76 식이군과 비교 시 10.55% 유의적으로 감소하였으며, 가르시니아 투여군도 13.60% 유의적으로 감소하였고 가시오갈피 잎 추출물 투여군과 유의적으로 같은 수준이었다. 식이섭취량은 정상군에 비하여 모든 군에서 전체적으로 감소하는 경향을 보였으며, 특히 가시오갈피 잎 추출물 투여군은 대조군 대비 16.87% 유의적으로(p<0.05) 감소함을 알 수 있었다. 혈청 중 총콜레스테롤 함량은 AIN-76식이 대조군이 정상군 대비 28.36% 유의적으로(p<0.05) 증가하였고, 가시오갈피 잎 추출물 투여군과 가르시니아 투여군은 AIN-76 식이군 대비 각각 25.47, 27.15% 유의적으로 감소하였다. 혈청 중 중성지방 함량은 AIN-76 식이군이 정상군 대비 35.04% 유의적으로 증가하였다. 가시오갈피 추출물 잎 투여군과 가르시니아 투여군은 AIN-76 식이군 대비 각각 37.54, 26.76% 유의적으로(p<0.05) 감소하였다. HDL-콜레스테롤 함량은 가시오갈피 잎 추출물 투여군과 가르시니아 투여군은 AIN-76 식이군 대비 각각 7.01, 5.00% 유의적으로 증가하였다. 간과 부고환 지방 조직의 무게는 가시오갈피 잎 추출물 투여로 유의하게 감소하였다. 간 조직 내 중성지방의 경우 AIN-76 식이군에서 정상군 대비 27.21% 유의적으로(p<0.05) 증가하였고, 가시오갈피 잎 추출물 투여군과 가르시니아 투여군에서는 AIN-76 식이군 대비 각각 6.77, 10.52% 유의적으로 감소하였다. 총 콜레스테롤은 AIN-76 식이군이 정상군과 비교하여 28.21% 증가하였고, 가시오갈피 잎 추출물 투여군과 가르시니아 투여군 에서는 각각 5.78, 6.91% 유의적으로 감소하였다. 위의 실험 결과 향후 가시오갈피 잎 추출물은 비만 예방 및 치료에 효과적인 소재로 활용될 수 있을 것으로 판단된다.

The Effects of Combination of Functional Beverage(Garcinia Cambogia, L-Carnitine, and Soy Peptide) and Exercise on the Improvement of Body Fat

  • Background: There is abundance of studies on the decrease in body fat through limited calorie intake or exercise; however, studies focusing simultaneously on functional foods and exercise are rare. This study was aimed to identify the effects of combination of a functional beverage and exercise on body composition and biochemical metabolic profiles. Method: Eighty-one healthy volunteers (69 females aged 19 to 50 years and 12 males aged 19 to 55 years), who maintained their body weight stable with 23 or higher in BMI (kg/$m^2$) and 25% or higher body fat for the last three months, were recruited in the study through written advertisement. All the subjects gave their informed consent, and the study was conducted in accordance with the Declaration of Helsinki. The study design was a randomized double-blind placebo-controlled parallel group design. All participants were given 12-week programed-exercise, which was performed 3 times a week. One bottle (100 ml) of test (Garcinia cambogia 300 mg, L-carnitine 20 mg, Soy peptide 1,000 mg) or placebo solution was given daily 30 min before each session of programed-exercise. BMI (body mass index), %fat, local fat amount (visceral and mid-thigh), waist circumference, skin fold thickness and some biochemical metabolic parameters like glucose, insulin and free fatty acids, etc in the blood were measured and compared before and after 12-week intervention within groups as well as between groups according to the protocol Results: Twenty six volunteers were dropped out and fifty five volunteers completed the study. At the end of 4, 8, and 12 weeks, approximately 1.98%, 3.00% and 3.50% losses of initial body weight were observed, respectively, in the test group (P<0.01), and 0.29, % ,0.74%, 1.60%, respectively, in the placebo group (P>0.05). BMI changed by 2.40%, 3.41% and 4.46%, respectively, in the test group, and 0.38%, 0.95% and 1.75%, respectively, in the placebo group, at each period of time. The reductions of body weight and BMI were significantly higher in the test group than in the placebo group at each period of time (P<0.05). Conclusions: It is thought that the combination of functional beverage, which contains mostly garcinia cambogia, L-carnitine, and soy peptide, and exercise have synergy effects on reducing body fat.

고지방식이를 섭취하는 흰쥐에서 가르시니아캄보지아 껍질추출물, 대두펩타이드 및 L-카르니틴 조성물 섭취와 규칙적인 트레드밀운동이 체중감량에 미치는 영향 (Effect of Treadmill Exercise Training and Dietary Intake of Garcinia Cambogia Extract, Soypeptide and L-Carnitine Mixture on Body Weight Reduction in Rats Fed High-Fat Diet)

  • This study was undertaken to examine effects of dietary intake of garcinia cambogia extract, soy peptide and L-carnitine mixture on body weight gain and obesity-related bio-markers in rats fed high-fat diet for 9 weeks with or without regular treadmill exercise. Forty 5-week-old male Sprague-Dawley rats were randomly divided into four groups; sedentary control group (SC), exercised control group (EC), sedentary formula-fed group (SF), and exercised formula-fed group (EF). The SC and EC rats were fed high-fat control diet (fat comprises$40\%$ of total caloris), and SF and EF rats were fed high-fat formula (composed of garcinia cambogia, soy peptide and L-carnitine) supplemented diet. Statistical analyses by two-way ANOVA indicated that the regular treadmill exercise significantly lowered cumulative body weight gain, total visceral fat mass, and epididymal, perirenal and retroperitoneal fat pad weights, and serum concentrations of total cholesterol and LDL + VLDL cholesterol, insulin, c-peptide and leptin. Feeding the formula also resulted in significant reductions in cumulative body weight gain and visceral fat pad weights, along with other related parameters including serum total and LDL + VLDL cholesterol levels, and hepatic enzyme activities involved in fatty acid synthesis. Statistical analyses by one-way ANOVA revealed that the formula consumption significantly improved body weight gain ($18\%$ reduction), total visceral fat weight ($20\%$ reductions), and serum total ($43\%$ reduction) and LDL + VLDL cholesterol ($54\%$ reduction) levels, as well as serum levels of insulin ($49\%$ reduction), and c-peptide ($41\%$ reduction) in sedentary rats, but failed to exhibit significant reductions in these indices in animals under treadmill exercise program. Taken together, these results suggest that the treadmill exercise per n exhibited significant improvements in body fat reduction and other related bio-markers, and so the formula consumption did not achieve a further significant reductions in these bio-markers in exercised rats. Nevertheless, animals fed the formula with regular exercise showed the most efficient weight reduction compared to other groups either fed formula without exercise or received regular exercise without dietary supplementation.

  • Various natural compounds act as antioxidants in protection against lipid peroxidation. Lipid peroxidation yields a variety of decomposition products which have been implicated in not only decreasing the nutritional value of food, but also in developing an off-flavor and toxic substances. As a source of safer and more effective natural antioxidants of natural origin have been widely investigated. Some Indonesian plants have evaluated for their antioxidative activity, and suggested the possible existence of various antioxygenic compounds in them. We attempted to study such antioxygenic compounds with simple method evaluation. As we are interested in the natural product compounds, we examined of several sample such as edible sea-weeds, and some edible fruits. Sea-weed, Eisenia bicyclis, one of the edible brown algae, exhibited the activity. As a traditional food additive consumed by Indonesian, Garcinia parvifolia is used as taste supplement in region West Sumatra, have been studied. Our current studies on the semi-polar fractions shows the activity by the thiocyanate method test. Another sample, Garcinia mangostana, a famous fruit with sweet taste, the part kernel have also evaluated. The acidic fraction of the extract showed antioxidative activity. Some other active components were found in the neutral and BuOH fractions.

Antimicrobial Effect of Ethanol Extract of Garcinia mangostana L. against Enterococcus faecalis Isolated from Human Oral Cavity

  • Enterococcus faecalis is a major causative agent of endodontic treatment failure. The purpose of this study was to investigate bactericidal effects of ethanol extract of Garcinia mangostana L. (mangosteen extract) on five strains of E. faecalis that were isolated from human oral cavities. The bactericidal effects of mangosteen extract were assessed by measurement of minimum bactericidal concentration (MBC) value. The cytotoxicity of mangosteen extract on immortalized human gingival fibroblasts, hTERT-hNOF, was determined based on cell counting method. The data revealed the MBC value of mangosteen extract against the E. faecalis strains was $4{\mu}g/ml$. Additionally, the cell viability of mangosteen extract on hTERT-hNOF was 83.7-89.1% at the 1 to $16{\mu}g/ml$. These findings indicated that mangosteen extract could be used as a root canal cleaner during management of endodontic treatment failure caused by E. faecalis.

Pattern of Sexual Dimorphism in Garcinia kola (Heckel) Plantation

  • A study was designed to investigate the pattern of sexual dimorphism in a plantation of Garcinia kola. Twenty trees were randomly selected for the study and have been observed to flower regularly. A total of 100 inflorescence were randomly collected from the crown of each tree and 500 flowers randomly assessed within the period of four (4) flowering seasons. Floral sex assessment was done visually and with a hand magnifying lens; floral morphometric measurements (i.e. pedicel and perianth length and breadth), inflorescence length, and breadth) was taken using a veneer caliper; number of flowers per inflorescence and inflorescence per twig was counted; while, data analysis was conducted on excel using analysis of variance and pairwise t-test comparison. Four floral sexes were identified in the G. kola plantation studied which were unisexual male flowers, unisexual female flowers, cosexual unisexual male flowers, and cosexual hermaphrodite flowers. Three tree sexes were identified viz: inconstant male, invariant female, and cosexual trees. The plantation was significantly sexually dimorphic in floral sex and phenotypic traits (i.e. pedicel and perianth size), and as well as sexually dimorphic in tree sex and reproductive phenotypic traits (i.e. inflorescence size, number of inflorescences per twig, and number of flower bud per inflorescence). The sexual system of the plantation was therefore trioecious with features suggestive of evolving dioecy through the gynodioecious pathway.

가르시니아 캄보지아 추출물함유 육류용 딥소스가 고지방식이 섭취 흰쥐의 지방축적 및 체중감량에 미치는 영향 (Effect of Garcinia cambogia Extract-containing Dip-sauce for Meat on Lipid Accumulation and Body Weight Reduction in Rats Fed High-fat Diet)

  • 본 연구의 목적은 가르시니아 캄보지아 추출물이 함유된 육류용 딥소스의 항비만 효과를 동물실험을 통하여 검증하기 위하여 고지방 식이로 유도한 비만 쥐의 체중 및 지방조직의 변화, 혈중 지질성분의 변화를 5주령 흰쥐 18마리를 정상식이에 식염수 투여군(NCD), 고지방 식이에 가르시니아 캄보지아 추출물-비함유 육류용 딥소스 투여군(HFD), 고지방 식이에 2% 가르시니아 캄보지아 추출물-함유 육류용 딥소스 투여군(HFD+GC)으로 나누어 90일 간 실시하였다. HFD군의 흰쥐에서의 체중 중가가 가장 두드러지게 나타났고, 이러한 체증증가는 HFD+GC군의 흰쥐에서 유의 있게 감소되는 것을 확인할 수 있었다. 또한 개복 후 부고환 및 신장주변 지방 조직을 적출하여 무게를 확인한 결과에서도 HFD+GC군의 흰쥐에서 지방의 무게가 유의하게 감소된 것을 확인할 수 있었다. 또한, 혈청 중의 지질성분인 total cholesterol, triglycerol, total lipid의 함량이 다른 식이 군에 비해 유의하게 감소한 결과도 HFD+GC군에서 확인할 수 있었다. 이상의 결과들은 가르시니아 캄보지아 추출물이 비만 유도 실험 모델 쥐에서 비만과 관련된 각종 지표들을 억제함으로써 항비만 효과를 나타내고 있음을 확인할 수 있었고, 이러한 효과는 가르시니아 캄보지아 추출물이 함유된 육류용 딥소스를 비만을 유발할 수 있는 음식과 동시에 섭취하여도 효과가 있음이 시사되었다.
