Effect of Cell-Wall Broken Spores of Ganoderma lucidum(Leyss. ex.Fr.) Karst on the Lipid Accumulation and Body Weight Reduction in C57BL/6J Mice fed High-Fat Diet (파벽(破壁) 영지포자가 고지방식이 섭취 마우스의 지방축적 및 체중감량에 미치는 영향)

  • This study was performed to investigate the effects of cell-wall broken spores of Ganoderma lucidum on the lipid accumulation and body weight reduction in C57BL/6J mice. Six-week-old C57BL/6J mice were randomly divided into 5 groups and assigned to one of these groups; normal chew diet(Nor) group, high-fat diet(HFD) group, HFD plus spores of Ganoderma lucidum 800 mg/kg/day (HFD + GS/B) group, HFD plus cell-wall broken spores of Ganoderma lucidum 400 mg/kg/day (HFD + BGS/A) group and finally HFD plus cell-wall broken spores of Ganoderma lucidum 800mg/kg/day (HFD + BGS/B). The experimental groups which were treated oral co-administration with cell-wall broken(or original) spores of Ganoderma lucidum and HFD significantly attenuated accumulative body weight gain, compared with HFD group. Administration of these experimental materials also resulted in significant reduction not only the serum levels of total cholesterol, homocysteine but also the lipid accumulation in liver tissue. But in the almost of results the cell-wall broken spores of Ganoderma lucidum were evaluated superior than the original one. These results indicate that cell-wall broken spores of Ganoderma lucidum may inhibit the lipid accumulation in blood as well as liver tissue. Therefore it may be a valuable candidate for the therapy preventing obese induced hyperlipidemic symptoms.

Purification and enzyme characteristics of laccase from Ganoderma lucidum (Ganoderma lucidum 균주에 의한 Laccase의 정제 및 효소적 특성)

  • The production media and enzymatic characteristics of laccase from Ganoderma lucidum was investigated. Potato dextrose yeast extract media was proved to be the best for laccase production. The enzyme has optimum pH of 6.45km value of 6.71 mM and appeared to be stable at wide pH range. The enzyme was inactivated partially by methanol and ethanol and totally by sodium azide but not at all by acetone. Also the enzyme purification was performed and the data is given.

  • A morphologically different form of Ganoderma lucidum was isolated from a cultivator's farm, and its optimum growth conditions were determined. A major difference in their morphology was color of fruit bodies. Fruit bodies of the mutant were white wherase those of normal Ganoderma lucidum were red. Spores of the mutant were global and mycelia were thin. Mycelial growth of this white mutant was favorable on potato sucrose agar medium, and optimum pH of the medium was 5.5.

A Study on the Hypoglycemic Effects and the Production Conditions of the Korean Organic Native Rice Cultured by Ganoderma lucidum (영지버섯으로 배양된 한국 유기농 토종 쌀의 생산 조건과 혈당강하 효과에 관한 연구)

  • This study was conducted to investigate the hypoglycemic effects and the production conditions of the Korean organic native rice cultured by Ganoderma lucidum. The broth culture of Ganoderma lucidum, the innoculation and culture of Ganoderma lucidum to the Korean organic native rice, and the oral administration of the Korean organic native rice cultured by Ganoderma lucidum to the streptozotocin-induced diabetic rats, were carried out. Then, the blood glucose level, the serum total cholesterol and triglyceride, and the activities of alanine aminotransferase and aspartate aminotransferase, were analyzed. The pH 4.5 at $30^{\circ}C$ was the optimal condition of the broth culture of Ganoderma lucidum. When the physical shape and the smell of the produced rice were considered, the optimal conditions to produce the Korean organic native rice cultured by Ganoderma lucidum, were the seeding of Ganoderma lucidum to Korean organic native rice in the rate of 7.5% weight, and the culture period of 9days at $28^{\circ}C$. In streptozotocin-induced diabetic rats, the blood glucose level of the dieted group by the Korean organic native rice cultured by Ganoderma lucidum for 2 weeks, were significantly decreased when compared with the control. And, the levels of the total cholesterol and triglyceride in serum were ranked less than those of the control. Also, in streptozotocin-induced diabetic rats, the activities of alanine aminotransferase and aspartate aminotransferase in the serum of the dieted group by the Korean organic native rice cultured by Ganoderma lucidum for 2 weeks, were lower than those of the control.

Antioxidative and Antimutagenic Effects of Ganoderma lucidum Krast Extracts (영지버섯(Ganoderma lucidum Krast) 추출물의 항산화 및 항돌연변이 효과)

  • The antioxidative effect, antimutagenic capacity and inhibitory effect of cancer cell proliferation in ethanol extracts of Ganoderma lucidum were studied for suggestion of prevention and dietetic treatment of chronic diseases and development of antioxidative, antimutagenic and anticancer functional food by employing biological and biochemical assay. The IC/sub 50/ of MDA with BSA conjugation reaction, lipid peroxidation and scavenging effect on DPPH radical in ethanol extracts of Ganoderma lucidum showed 1026.21 mg/mL, 0.152 mg/mL and 0.412 mg/mL, respectively. So, the most effective antioxidative capacity in ethanol extracts of Ganoderma lucidum was the lipid peroxidation, among the method used this study. The indirect and direct antimutagenic effects of ethanol extracts of Ganoderma lucidum were examined by Ames test using Salmonella typimurium TA98 and TA100. The inhibition rates on indirect mutagenicity mediated by 2-Anthramine showed 100% in the Salmonella typimurium TA98 and 98.74% in the Salmonella typimurium TA100 and the direct mutagenicity mediated by sodium azide in Salmonella typimurium TA100 was 82.96%. But, the inhibitory effect on indirect mutagenicity mediated by 2-Nitrofluorene in Salmonella typimurium TA98 was low(7.81%). The inhibitory Effect of Ganoderma lucidum ethanol extracts on cell proliferation in Hela and MCF-7 by MTT test were 74.36% in HeLa and 73.90% in MCF-7 at the 0.50 mg/assay concentration and IC/sub 50/ were 0.163 mg/mL and 0.196 mg/mL respectively. From this result, it is suggested that Ganoderma lucidum is believed to have a possible antioxidative, antimutagenic and anticancer capacities.

Evaluation of Anti-Oxidant and Anti-Inflammatory Activities of Ganoderma lucidum Cultured on Hulled Barley (겉보리에서 배양한 영지버섯 추출물의 항산화 및 항염증 효능 평가)

  • Background: Ganoderma lucidum cultured on hulled barley was investigated as a potential natural source of antioxidants and anti-inflammatory agents. Methods and Results: The yields from Ganoderma lucidum cultured on hulled barley water and ethanol extract were 17.69% and 25.77%, respectively. The antioxidant activity of Ganoderma lucidum cultured on hulled barley extracts was confirmed by various methods including assayss of 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azino-bis-(3-ethylbenzo thiazoline-6-sulfonic acid) (ABTS), nitrite radical scavenging, and $Fe^{3+}$ to $Fe^{2+}$ reducing power activity. The ethanol extract of Ganoderma lucidum cultured on hulled barley showed improved DPPH, ABTS and nitrite radical scavenging activity compared with the water extract. After treatment of RAW264.7 cells with Ganoderma lucidum cultured on hulled barley ethanol extracts, the cell viability compared with the control was 92.82%, even at a concentration of $3,000{\mu}g/m{\ell}$. The ethanol extract inhibited reactive oxygen species (ROS) generation in RAW264.7 cells stimulated with $H_2O_2$, even at low concentrations. In addition, the ethanol extract showed an inhibitory effects on the production of lipopolysaccharide-induced nitric oxide (NO) in RAW264.7 cells. Conclusions: This study suggests that the extract of Ganoderma lucidum cultured on hulled barley is a potential source of natural antioxidants and anti-inflammatory agents.

Mitochondrial DNA Analysis in Fusants of Ganoderma lucidum and Lentinus edodes (영지버섯과 표고버섯 원형질 융합체의 미토콘드리아 DNA 검색)

  • It has been known that Ganoderma lucidum and Lentinus edodes have anticancer activity and immune enhancing activity. These two mushrooms were grown in liquid culture and harvested. From these mycelia, DNA was isolated and EtBr-CsCl density gradient ultracentrifugation was performed to purify it further. Then mitochondrial DNA was isolated by bisbenzimide-CsCl density ultracentrifugaton. Mitochondrial DNA of Ganoderma lucidum was digested by restriction enzymes, EcoR I, Hind Ⅲ, and Pst I, then electrophoresed. It showed 12, 22, 4 fragments. Mitochondrial DNA of Lentinus edodes was digested by EcoR I. Electric pattern showed 6 fragments. 4 fragments had appeared by Pst 1 digested mitochondrial DNA. Hind ill couldn't digest mitochondrial DNA of Lentinus edodes. Mitochondrial DNA of fusants was isolated to compare to those of parents. The results showed that fusant P₂S₄has new, recombined mitochondrial DNA. But P₂S₄had the same DNA that Ganoderma lucidum had.

The Taste Components Composition in Various Mushrooms-Added Korean Soybean Paste (Doenjang) (버섯첨가된장의 맛성분 조성)

  • The purpose of this study is to evaluate the sensory characteristics and the taste components of Korean soybean paste (doenjang) with mushrooms such as Lentinus edodes, Ganoderma lucidum and Phellinus Lentinus. edodes doenjang turned out to have a good taste, odor and color, but Ganoderma lucidum and Phellinus liteus doenjang were worse than control doenjang in the taste, odor and color as well as in sensory evaluation. The contents of amino type nitrogen were higher in Lentinus edodes and Phellinus liteus doenjang than in control doenjang. Of organic acids, succinic acid was the most abundant and Ganoderma lucidum doenjang had a little higher acetic, butyric and propionic acid. Fructose and glucose were detected as free sugar of mushroom doenjang, of which glucose was considerably contained in Ganoderma lucidum doenjang, Total contents of free amino acid were 2.247 ∼ 2.833mg/100g in doenjangs added mushrooms. Glutamic acid and alanine were dominant in mushroom doenjangs.

Molecular Taxonomy of Ganoderma cupreum from Southern India Inferred from ITS rDNA Sequences Analysis

  • Ganoderma is a cosmopolitan wood-rot basidiomycete that has been extensively studied for its pathogencity and medicinal properties. Identification of Ganoderma based on macro-microscopic features led to large number of synonyms which resulted in 250 taxonomic names. A Ganoderma species collected from Courtallam, Tamil Nadu was identified as G. cupreum. Phylogenetic analysis inferred from internal transcribed spacer rDNA region resolved the Indian isolate MYC1 as Ganoderma cupreum which clustered with Australian and Asian "cupreum" clade with 85% bootstrap support BS and shared 99% and 98% nucleotide similarity with Malaysian and Australian 'cupreum' respectively. This study represents the first molecular evidence of G. cupreum from Asian origin.

Study on Anti-Allergic Effecst of Ganoderma lucidum Herbal Acupuncture and Ganoderma lucidum Extract (영지(靈芝) 약침(藥鍼)과 영지(靈芝) 추출액의 항알레르기 효과에 대한 연구)

  • Objectives We studied on anti-allergic effects of Ganoderma lucidum herbal acupuncture(GHA) and Ganoderma lucidum extract(GE). Methods In vivo, Animals were herbal-acupunctured GHA at both B13s three times for 5 days. Then, we investigated compound 48/80-induced active systemic anaphylatic shock using ICR mice and anti-DNP IgE-induced passive cutaneous anaphylaxis using Sprague Dawley rat. In vitro, we measured cell viability, b-hexosaminidase release, IL-4 and TNF-a from RBL-2H3 cells, and nitric oxide from Raw264.7 cell after treatment of GE of various concentrations. Results In vivo, GHA pretreatments at both B13s inhibited compound 48/80-induced active systemic anaphylatic shock. Passive cutaneous anaphylaxis were inhibited by GHA10 and OP. In vitro, $0.1\;{\sim}\;2%$ GE treatments were not affect on cell viability and inhibited b-hexosaminidase release, IL-4, TNF-a and nitric oxide. Conclusions These results suggest that GHA and GE may be beneficial in the inhibition of allergic inflammatory response.
