Objective: This study was to investigate the effects of dietary cotton stalk on nitrogen and free gossypol in sheep. Methods: Treatments included 25% cotton stalk (Treat 1), 50% cotton stalk (Treat 2), and a control (no cotton stalk). Six Xinjiang daolang wethers were cannulated at the rumen and duodenum and fed one of these diets. The effects of these diets on nitrogen and free gossypol absorption and metabolism were determined. Fifteen healthy Xinjiang daolang wethers were assessed for daily gain, tissue lesions, and free gossypol accumulation. Results: Dry matter intake decreased with increasing dietary cotton stalk. Total tract dry matter digestibility did not significantly differ among treatments. Dietary cotton stalk significantly decreased volatile fatty acids and increased ammonium nitrogen in the rumen. Nitrogen intake was significantly higher in Treat 2 than in the control or Treat 1. Nitrogen retention and free gossypol intake increased with dietary cotton stalk. Duodenal free gossypol flow did not increase, and free gossypol almost disappeared from the rumen. The free gossypol content of plasma and tissue was increased with dietary cotton stalk with liver free gossypol>muscle free gossypol>kidney free gossypol. Elevated dietary free gossypol decreased platelets, hemoglobin, and serum iron. Aspartate aminotransferase and ${\gamma}$-glutamyltransferase increased in Treat 2. With high long-term dietary cotton stalk intake, liver cells were swollen, and their nuclei dissolved. Renal cells were necrotic and the interstitia were enlarged. Conclusion: With short-term cotton stalk administration, only a small amount of free gossypol is retained in the body. In response to long-term or high free gossypol cotton stalk feeding, however, free gossypol accumulates in, and damages the liver and kidneys.
The fertility of dairy cows has been declining worldwide. The number of services per conception has increased, and repeat breeder (RB) cows are considered important in the dairy industry. However, there has been little research on RB cows in Korea. The objective of this study was to investigate the relationship between serum biochemical profiles and RB Holstein cattle in Korea and compare the results with those of studies conducted abroad. In addition, we investigated hidden factors that are needed for RB cattle to become pregnant. Overall, 34 Korean Holstein Friesian cows were divided into three groups: pregnant with normal cycling (PNC), pregnant with repeat breeder (PRB), and non-pregnant with repeat breeder (NRB). Blood samples were collected from the jugular vein, and serum parameters (ALT, AST, ALP, GGT, LD, CK, TP, ALB, GLOB, TBIL, BUN, CRE, GLU, TC, TG, NEFA, CA, MG, and IP) were analyzed. GGT levels were significantly different among groups (P=0.01). The concentration of GGT was the highest in the NRB and the lowest in the PNC. In contrast to the findings of other studies, there were no differences in GLU, TP, TC, and BUN levels. This study is the first to investigate serum biochemistry in RB cattle and provides results that differ from those of previous studies; these findings would help establish a novel approach to improve fertility of RB cattle.
This study compared blood metabolites during peri- and postpartum periods among cows with clinical or subclinical endometritis and cows without endometritis. Blood samples from 207 Holstein dairy cows were collected at 4 weeks prepartum, just after calving, and at 1, 2, 4, and 6 weeks postpartum to measure serum concentrations of calcium, magnesium, non-esterified fatty acids (NEFAs), total cholesterol, albumin, urea nitrogen, ${\beta}$-hydroxybutyrate (BHBA), aspartate aminotransferase (AST), ${\gamma}$-glutamyltransferase, glucose, and phosphorus. Clinical endometritis was diagnosed by the observation of vaginal discharge (> 50% pus) and subclinical endometritis was diagnosed by the evaluation of uterine cytology (> 18% neutrophils) at 4 weeks postpartum. Cows were divided into three groups based on the presence or absence of clinical or subclinical endometritis: the control group (n = 104), the clinical endometritis group (n = 66), and the subclinical endometritis group (n = 37). Calcium and magnesium concentrations were lower in the clinical endometritis group than in the control and subclinical endometritis groups throughout the study period (p < 0.05 to 0.0001), whereas the NEFAs concentration was higher in the clinical endometritis group than in the control group throughout the study period (p < 0.01). The total cholesterol concentration was lower in the clinical endometritis group than in the control and subclinical endometritis groups throughout the pre- and postpartum periods (p < 0.05 to 0.001). The albumin concentration was lower in the clinical endometritis group than in the control and subclinical endometritis groups during the postpartum period (p < 0.05 to 0.001). The urea nitrogen concentration was lower in the clinical endometritis group than in the control and subclinical endometritis groups at 4 and 6 weeks postpartum (p < 0.01). At 1 week postpartum, the BHBA concentration was higher in the clinical endometritis group than in the control group (p < 0.05), whereas the AST concentration was higher in the clinical endometritis and subclinical endometritis groups than in the control group (p < 0.05). In conclusion, lower serum concentrations of calcium, magnesium, total cholesterol, albumin, and urea nitrogen, but higher concentrations of NEFAs, BHBA, and AST during the postpartum period were associated with the incidence of clinical endometritis, indicating the importance of balanced nutrition during the transition period.

This study examined metabolic profiles of 1349 Holstein cows from 91 commercial herds. Thirteen parameters which are consisted of twelve blood components and body condition score were examined and their mean values. standard deviations and standard limits, which are 80% confidential limits, in each lactational stage were reported. The variations of each parameter affected by season, individual milk yield, adjusted corrected milk yield of herd. and lactation number were also reported. A model of metabolic profile test applicable to this country where the average number of cows in a herd is small as to be fifteen is designed. Metabolic profiles as reflected in each parameter were discussed in relation to adequacy of dietary intake for production, milk production, reproductive performance, and diseases, and the possible measure to improve productivity of dairy cows were proposed. Much of the variation in parameters was due to differences between herds, and less to differences between seasons, differences between individual milk yield, and differences between lactational stages. As the average herd size in this country is small, it is believed that all the cows in a herd must be sampled, and the individual result of each parameter was compared with the standard limit for each lactational stage, and the percentage of cows which are outside the standard limits in a herd was calculated to use as a criteria for evaluation of the herd. Data outside the 99% confidential limits were to be deleted at first, but when the trends of the data outside the 99% confidential limits are same as the trends of the data within 99% confidential limits, the deleted data must be reviewed again, otherwise some important informations would be missed. The mean concentration of blood urea nitrogen in this study was much higher than that was reported in England, U.S.A. and Japan, and it was similar to the upper limits reported in England, U.S.A. and Japan. So it was thought that the concentration of blood urea nitrogen is improper as a criteria for protein intake. The increase of serum total protein cocentration beyond standard limits was due to increase of serum globulin concentration in most of the cows. The correlation coefficient between serum and protein and serum globulin concentration was 0.83. Serum globulin concentration was negatively related to adjusted corrected milk of herd. Serum albumin, calcium and magnessium concentrations were negatively related to adjusted corrected milk of herd, which indicate that high-producing individual or high-producing herd have not taken sufficient protein/amino acids, calcium and magnessium. Packed cell volume was negatively related to adjusted corrected milk of the herd, and the trend was same In each lactational stage. The correlation coefficient between serum and packed cell volume was 0.16 and the correlation was very weak. Blood glucose concentration was lowest in early lactational stage, which indicates negative energy balance in early lactational stage. Blood glucose concentration was negatively related to adjusted corrected milk of herd from peak to late lactational stage, which indicates negative energy balance during the period in high-producing individuals or high-producing herds. Correlation coefficient between serum aspartate aminotransferase activity and serum ${\gamma}$-glutamyltransferase activity was 0.41, and this indicates that serum ${\gamma}$-glutamyltransferase should be included as a parameter of metabolic profile test to evaluate liver function. Body condition score of dairy cows in this country was lower than that of Japan in every lactational stages, and the magnitude of increase in body condition score during middle and late lactational stages was small. Metabolic profile can not be evaluated with solely nutritional intake. When an individual or large percentage of cows in a herd have adnormal values In parameters of metabolic profile test, veterinary clinician and nutritionist should cooperate so as to diagnose diseases and to calculate the e of no운ents simultaneously.
Taurine has been reported to prevent cardiovascular disease and improve liver function, diabetes, and platelet function. However, there are few studies on the effects of taurine in Koreans. The purpose of this study was to investigate the effect of basal doses of taurine on blood glucose, liver disease, and lipid diseases among Korean college students. The study included a taurine intake group and a control group; each group consisted of 15 students. Taurine was administered at a standard dose of 1,000 mg for 2 weeks postprandial. All subjects were excluded from medication or other food besides meals provided by the dormitory. The liver test gamma-glutamyl transferase (GGT) in the taurine group decreased to $23.53{\pm}25.73IU/L$ before intake and to $15.15{\pm}4.91IU/L$ after intake (P=0.186). Lipid metabolite triglyceride (TG) was $100.42{\pm}28.33mg/dL$ before intake and $80.22{\pm}17.08mg/dL$ after intake (P<0.05). Total cholesterol (T-Cho), low density cholesterol (LDL-C), and high density lipoprotein cholesterol (HDL-C). Consequently, taurine improved liver function and lipid metabolism. Hematologic tests showed a decrease in segmented neutrophil percentage and an increase in lymphocyte percentage. Thus, taurine also seems to be related to immunological function.

This study was conducted to investigate the protective effects of water extract from Crassostrea gigas (CGW) against ethanol-induced hepatic toxicity in rats. Seventy-two male Wistar rats (6-week-old) were divided into six groups of 12 animals each: control group (1 mL saline/d), ethanol-treated group, positive control group (ethanol+Hovenia dulcis Thunb extract), CGWL group (ethanol+low dosage of CGW), CGWM group (ethanol+medium dosage of CGW), and CGWH group (ethanol+high dosage of CGW). All groups except the control group received ethanol (40% ethanol 5 g/kg) orally. CGW administration with ethanol resulted in prevention of ethanol-induced hepatotoxicity by increasing levels of serum alanine aminotransferase and ${\gamma}-glutamyltransferase$. CGW supplementation significantly reduced formation of malonaldehyde and inhibited reduction of hepatic glutathione and peroxidase levels, as compared with the ethanol-administration group. Further, CGW suppressed expression of CYP2E1, which was elevated by ethanol administration. Consequently, our results indicate that Crassostrea gigas may exert hepatoprotective effects against alcohol-induced hepatocyte injury by intensifying the anti-oxidative defense system.

Purpose: This study aimed to investigate the association of total dietary antioxidant capacity (TAC) with oxidative stress and metabolic markers among patients with metabolic syndrome according to gender. Methods: A total of 346 subjects aged 30~59 years with two or more risk factors of metabolic syndrome were recruited from a general hospital near Seoul in South Korea between 2010 and 2012 based on data from the medical checkup. Biochemical indices for oxidative stress and metabolic markers were measured. Food consumption data from 3-day food records were linked with the antioxidant capacity database for commonly consumed Korean foods to estimate individual's TAC. Results: Average dietary TAC of the study subjects was 132.0 mg VCE/d/1,000 kcal in men and 196.4 mg VCE/d/1,000 kcal in women. Levels of ${\gamma}$-glutamyltransferase (GGT), systolic blood pressure, diastolic blood pressure, and blood triglycerides were reduced significantly according to increasing TAC in men, but there was no significant trend in women. Intakes of total flavonoids and carotenoids were significantly negatively correlated with GGT (p < 0.05) and d-ROMs (p < 0.01) in men, whereas those of ${\alpha}$-tocopherol (p < 0.05) and ${\gamma}$-tocopherol (p < 0.05) were positively correlated with biological antioxidant potential (BAP) in women. The odds ratio of high oxidative stress indices and abnormal metabolic markers according to TAC level were not significant in either men or women. Conclusion: The results show that dietary TAC was partially associated with oxidative stress and metabolic markers among patients with metabolic syndrome. Further research is required for elucidating the association between dietary TAC and incidence of metabolic syndrome and chronic diseases within a large population in prospective studies.
In order to understand the pathogenetic mechanism of adult respiratory distress syndrome (ARDS), the role of phospholipase A2 (PLA2) in association with oxidative stress was investigated in rats. $Interleukin-1{\alpha}\;(IL-1,\;50\;{\mu}g/rat)$ was used to induce acute lung injury by neutrophilic respiratory burst. Five hours after IL-1 insufflation into trachea, microvascular integrity was disrupted, and protein leakage into the alveolar lumen was followed. An infiltration of neutrophils was clearly observed after IL-1 treatment. It was the origin of the generation of oxygen radicals causing oxidative stress in the lung. IL-1 increased tumor necrosis factor (TNF) and cytokine-induced neutrophil chemoattractant (CINC) in the bronchoalveolar lavage fluid, but mepacrine, a PLA2 inhibitor, did not change the levels of these cytokines. Although IL-1 increased PLA2 activity time-dependently, mepacrine inhibited the activity almost completely. Activation of PLA2 elevated leukotriene C4 and B4 (LTC4 and LTB4), and 6-keto-prostaglandin $F2{\alpha}\;(6-keto-PGF2{\alpha})$ was consumed completely by respiratory burst induced by IL-1. Mepacrine did not alter these changes in the contents of lipid mediators. To estimate the functional changes of alveolar barrier during the oxidative stress, quantitative changes of pulmonary surfactant, activity of gamma glutamyltransferase (GGT), and ultrastructural changes were examined. IL-1 increased the level of phospholipid in the bronchoalveolar lavage (BAL) fluid, which seemed to be caused by abnormal, pathological release of lamellar bodies into the alveolar lumen. Mepacrine recovered the amount of surfactant up to control level. IL-1 decreased GGT activity, while mepacrine restored it. In ultrastructural study, when treated with IL-1, marked necroses of endothelial cells and type II pneumocytes were observed, while mepacrine inhibited these pathological changes. In histochemical electron microscopy, increased generation of oxidants was identified around neutrophils and in the cytoplasm of type II pneumocytes. Mepacrine reduced the generation of oxidants in the tissue produced by neutrophilic respiratory burst. In immunoelectron microscopic study, PLA2 was identified in the cytoplasm of the type II pneumocytes after IL-1 treatment, but mepacrine diminished PLA2 particles in the cytoplasm of the type II pneumocyte. Based on these experimental results, it is suggested that PLA2 plays a pivotal role in inducing acute lung injury mediated by IL-1 through the oxidative stress by neutrophils. By causing endothelial damage, functional changes of pulmonary surfactant and alveolar type I pneumocyte, oxidative stress disrupts microvascular integrity and alveolar barrier.

Objectives: This study describes the results of a mass screening and secondary questionnaire conducted among sampled residents around Camp Carroll. Methods: The subjects were sampled based on Waegwan groundwater ingestion history via a primary health questionnaire survey. However, the study population included voluntary participants and there were no grounds for disqualification. Among the 1,033 residents, excluding people living outside Waegwan, 844 subjects age 30 and over were analyzed. History of physician-diagnosed disease (including detailed history of cancer), eating habits, drinking and smoking histories were queried through questionnaires. Health screening consisted of a blood pressure check and blood test (complete blood cell, liver enzyme, lipid, blood sugar test, etc.). Results: The proportion of abnormal gamma-glutamyltransferase levels was higher in the groundwater ingesting female group than the non-ingesting female group. The odds ratios of the ingested '1 to 9 years' and '10 years and over' groups were 3.09 and 0.87, respectively. Proportions of hypertension in males, abnormal serum triglyceride levels in all and in females, and abnormal serum high density lipoprotein cholesterol levels in males were higher in the '10 to 29-year' resident group than in the '1 to 9 year' group. However, there were no significant trends according to length of residence. Conclusions: Physician-diagnosed prevalence and laboratory test results are not different by histories of Waegwan groundwater ingestion and by length of residence. Even if there are partially significant differences, they do not tend to follow increases of exposure amount and trends.
Objectives: This study aimed to report a case that showed improvements in the symptoms of patients with alcoholic hepatitis without any indication of deterioration of the disease. Methods: Western medicine with Urusa tablets and Godex capsules and Korean medicine therapeutic approaches, including Shihogayonggolmoryo-tang, acupuncture, and moxibustion, were administered to a patient during the period of treatment. Blood tests were used to determine levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), AST/ALT ratio, alkaline phosphatase (ALP), gamma glutamyltransferase (GGT), total bilirubin, direct bilirubin, and total cholesterol. Fatigue was measured using the numeric rating scale (NRS), and the patient's total sleeping time was checked, daily. Results: After the combined treatment, the AST/ALT ratio and the AST, ALT, ALP, GGT, total cholesterol, and direct bilirubin levels were decreased. Through Oriental medicine for the purpose of improving symptoms, NRS of fatigue decreased from 10 to 5, and the amount of sleeping time increased from 2 to 5 hours. Conclusions: The herbal medicine had no effect on the hepatoprotective drugs such as Urusa tablets and Godex capsules used to treat alcoholic hepatitis, and no adverse reaction from the combined administration was observed. To reduce fatigue and insomnia in patients with alcoholic hepatitis, it might be helpful to combine Western medications with Korean medicine treatments, including acupuncture, moxibustion, and Shihogayonggolmoryo-tang.
