Studies on the Construction of Mutant Diversity Pool (MDP) lines, and their Genomic Characterization in Soybean

  • Mutation breeding is useful for improving agronomic characteristics of various crops. In this study, we constructed soybean Mutant Diversity Pool (MDP) from 1,695 gamma-irradiated mutants through two selection phases over M1 to M12 generations; we selected 523 mutant lines exhibiting at least 30% superior agricultural characteristics, and, second, we eliminated redundant morphological phenotypes in the M12 generation. Finally, we constructed 208 MDP lines and investigated 11 agronomic traits. We then assessed the genetic diversity and inter-relationships of these MDP lines using target region amplification polymorphism (TRAP) markers. Among the different TRAP primer combinations, polymorphism levels and PIC values averaged 59.71% and 0.15, respectively. Dendrogram and population structure analyses divided the MDP lines into four major groups. According to an analysis of AMOVA, the percentage of inter-population variation among mutants was 11.320 (20.6%), whereas mutant inter-population variation ranged from 0.231 (0.4%) to 14.324 (26.1%). Overall, the genetic similarity of each cultivar and its mutants were higher than within other mutant populations. In an analysis of the genome-wide association study (GWAS) using based on the genotyping-by-sequencing (GBS), we detected 66 SNPs located on 13 different chromosomes were found to be highly associated with four agronomic traits: days of flowering (33 SNPs), flower color (16 SNPs), node number (6 SNPs), and seed coat color (11 SNPs). These results are consistent with those previously reported for other genetic resource populations, including natural accessions and recombinant inbred line. Our observations suggest that genomic changes in mutant individuals induced by gamma rays occurred at the same loci as those of natural soybean population. This study has demonstrated that the integration of GBS and GWAS can serve as a powerful complementary approach to gamma-ray mutation for the dissection of complex traits in soybean.

Feasibility of Gamma Knife Radiosurgery for Brain Arteriovenous Malformations According to Nidus Type

  • Objective : Gamma Knife radiosurgery (GKRS) is an effective and noninvasive treatment for high-risk arteriovenous malformations (AVMs). Since differences in GKRS outcomes by nidus type are unknown, this study evaluated GKRS feasibility and safety in patients with brain AVMs. Methods : This single-center retrospective study included patients with AVM who underwent GKRS between 2008 and 2021. Patients were divided into compact- and diffuse-type groups according to nidus characteristics. We excluded patients who performed GKRS and did not follow-up evaluation with magnetic resonance imaging or digital subtraction angiography within 36 months from the study. We used univariate and multivariate analyses to characterize associations of nidus type with obliteration rate and GKRS-related complications. Results : We enrolled 154 patients (mean age, 32.14±17.17 years; mean post-GKRS follow-up, 52.10±33.67 months) of whom 131 (85.1%) had compact- and 23 (14.9%) diffuse-type nidus AVMs. Of all AVMs, 89 (57.8%) were unruptured, and 65 (42.2%) had ruptured. The mean Spetzler-Martin AVM grades were 2.03±0.95 and 3.39±1.23 for the compact- and diffuse-type groups, respectively (p<0.001). During the follow-up period, AVM-related hemorrhages occurred in four individuals (2.6%), three of whom had compact nidi. Substantial radiation-induced changes and cyst formation were observed in 21 (13.6%) and one patient (0.6%), respectively. The AVM complete obliteration rate was 46.1% across both groups. Post-GKRS complication and complete obliteration rates were not significantly different between nidus types. For diffuse-type nidus AVMs, larger AVM size and volume (p<0.001), lower radiation dose (p<0.001), eloquent area location (p=0.015), and higher Spetzler-Martin grade (p<0.001) were observed. Conclusion : GKRS is a safe and feasible treatment for brain AVMs characterized by both diffuse- and compact-type nidi.

Nutritional Quality of Restructured Pork Jerky with Electron Beam and Gamma Ray Irradiation (방사선 조사된 재구성 돈육포의 단백질 품질)

  • To evaluate the effect of irradiation on restructured pork jerky containing paprika and Japanese apricot extracts, the quality of protein was determined in vitro based on the formation of trypsin indigestible substrate inhibitor (TIS) and the computed protein efficiency ratio (C-PER) as determined based on the protein digestibility and amino acid analysis. In addition, we compared the effects of electron beam irradiation to those of gamma irradiation. Approximately 3% of the moisture content of pork jerky was reduced in response to irradiation with 3kGy administered using an electron beam however, no additional reduction was observed in samples that were subjected to higher doses of irradiation. In addition, there were no notable differences in the crude protein and fat content of pork jerky samples that were subjected to irradiation, regardless of dose. Furthermore, the total amino acids profiles did not change in response to electron beam irradiation. However, the in vitro protein digestibility increased by 7% in response to 3kGy of electron beam irradiation and 5kGy of gamma irradiation, but no significant changes in digestibility were not observed in response to treatment with higher doses. TIS quantified as trypsin inhibitors were formed in response to irradiation using the electron beam (3kGy) and gamma rays (5kGy), although there was a slight reduction in the production of TIS inhibitors in samples irradiated with higher doses. Moreover, only samples irradiated with 10kGy (electron beam and gamma ray) showed higher TBA values than those of the control samples. Finally, the C-PERs $(2.50{\sim}2.60)$ were greater in all of the irradiated pork jerky samples than in the control samples (2.22). Taken together, these results suggest that electron beam irradiation and the incorporation of extracts (paprika and Japanese apricot) may be useful methods of improving the nutritional quality of pork jerky.

Gamma-ray Irradiation on Radio Sensitivity in Yacon (Samallanthus sonchifolius (Poepp. & Endl.) H. Robinson) Breeding (돌연변이 육종을 위한 야콘의 최적 감마선 조사량)

  • Yacon [Samallanthus sonchifolius (Poepp. & Endl.) H. Robinson], a member of Compositae plants, has sweet taste and crisp texture. Unlike other Andean root crops such as potato and sweet potato, the cultivation area of yacon has increased recently, since it is known to have large content of fructooligosaccharides (FOS). Since there are no yacon varieties bred in Korea, we have been trying to create new genetic resources using gamma-ray. The optimal gamma-ray dosage for mutation breeding in yacon was investigated. Crown bud and green bud of yacon were exposed to doses of gamma rays from 20 Gy to 80 Gy, and subsequently planted in a greenhouse. After 50 days of sowing, the survival rates and growth decreased rapidly at doses above 40 Gy, while all of crown bud individuals died above 60 Gy. The median lethal dose (LD50) of crown bud and green bud was 22.4 and 36.6 Gy, and the median reduction doses (RD50) for plant height, fresh weights, and tuberous root weight were 20-40 Gy, respectively. A dose of 20-40 Gy was found to be optimal for mutation breeding in yacon. Considering the growth factors, the optimum doses were determined to be within the range of 20-40 Gy for the selection of useful mutant lines. M2-M3 mutant lines were obtained from 20-60 Gy gamma-ray-irradiated M1 plants through clonal propagation. These mutant lines will be used for the development of a new variety of yacon plant with high FOS and no crack tuberous root.

A New Composition of Nanosized Silica-Silver for Control of Various Plant Diseases

  • The present study addressed the efficacy of nanosized silica-silver for controlling plant pathogenic microorganisms. The nanosized silica-silver consisted of nano-silver combined with silica molecules and water soluble polymer, prepared by exposing a solution including silver salt, silicate and water soluble polymer to radioactive rays. The nanosized silica-silver showed antifungal activity against the tested phytopathogenic fungi at 3.0 ppm with varied degrees. In contrast, a number of beneficial bacteria or plant pathogenic bacteria were not significantly affected at 10 ppm level but completely inhibited by 100 ppm of nanosized silicasilver. Among the tested plant pathogenic fungi, the new product effectively controlled powdery mildews of pumpkin at 0.3 ppm in both field and greenhouse tests. The pathogens disappeared from the infected leaves 3 days after spray and the plants remained healthy thereafter. Our results suggested that the product developed in this study was effective in controlling various plant fungal diseases.

In Situ Observation of Initial Rusting Process of Steel Containing Al Using Synchrotron Radiation X-Rays

  • We observed initial rusting process of steel containing Al under wet/dry cyclic condition with NaCl solution film using in situ X-ray diffraction spectroscopy at SPring-8 synchrotron radiation facility. It was found that mass fraction of iron oxides such as ${\alpha}-FeOOH$, ${\beta}-FeOOH$ and ${\gamma}-FeOOH$ varied with Al content. Some kinds of Al oxides were also found at the initial stage of corrosion. Those corrosion products might affect the corrosion process and corrosion rate of the steel.

  • CsI:Gd crystal was grown by the Bridgeman method and its scintillation properties were investigated. The wavelength peak of the luminescence spectrum for the crystal excited by X-ray was 419 nm. The range of the spectrum was from 300 nm to 800 nm. The spectrum well matched to the quantum efficiency of a typical bi-alkali photo-multiplier tube(PMT). An energy resolution of 48.2 % was obtained for 662 keV ${\gamma}$-rays of $^{137}Cs$. The three decay times were obtained as a fast(557.4 ns, 42.2 %), intermediate (1.78 ${\mu}s$, 29.7 %) and slow (5.43 ${\mu}s$, 28.1 %) components, respectively.

  • 본 연구는 사용후핵연료 수송 저장 용기인 저합금강(SA350 LF3)에 일정의 감마선을 조사하고 감마선 조사 전후 물성 및 내식 특성 변화와 표면처리에 의한 내식성 개선 효과에 관하여 연구하였다. 상온 항복강도 및 인장강도의 기계적 물성은 감마선 조사 여부에 따라 물성의 차이는 보이지 않았지만, 저온충격 특성은 감마선 조사를 하지 않은 충격 흡수에너지에 비교하여 조사후 시험편의 충격 흡수 에너지가 감소되었다. 양분극 곡선에 측정에 의해 관찰된 저합금강의 내식성은 감마선 조사된 시험편에서 감마선을 조사하지 않은 시험편 보다 낮은 부식전위를 나타내었다.

Radiation Sensitivity of Some Food Decay Fungi (몇몇 음식 부식 균류의 방사선 감수성에 대한 연구)

  • Five species of food decay fungl, Aspergillus flavus, Asp. uiger, Penicillum sp., Botrytis cinerea, and Rhizopus stolonifer, were examined for their radio sensitivity in several suspension media. Asp. flavus, Asp. niger and Penicillum sp. have almost the same sensitivity toward gamma rays. with D value in the range of 30 to 35 K rad, whereas Botrytis cinerea has a D value of approximately 55K rad and Rhizopus stolonifer, the most re4sistant fungus studied, has a crease in their radioresistnace when compared with spores irradiated in water. Asp. flavus and penicillium sp. spores irradiated in citrate buffer at pH3-7 showed almost no change in their radisensitiity with pH, but Botryis cinerea spores showed a distinct decrease in their radioresistnace at pH 6 and 7. Penicillum sp. spores irradiated in sucrose solutions showed no sinificant change in their radioresistance. Botrytis cinerea spores displayed a higher radioresistance when they were irradiated in sucrose solution than in water.

Design of a High Stable Measuring Circuit for Radioactive Pulses (방사선 펄스의 고안정 계측회로 설계)

  • The aim of this paper is to develop a high stable measuring device for radioactive pulses. The device consists of a high voltage supply unit using a fly-back converter principle, and a pulse detection unit for gamma-rays and neutrons. The high voltage supply unit designed can generate DC voltage up to 1,500v at 5V-input, and have a series voltage regulator to maintain the output voltage constantly, resulting in less than 1.63% of voltage regulation. The pulse detection parts consists of an active integrator, a pole-zero circuit, and a 3-stage amplifier of 60 dB, and its frequency bandwidth is from 37 Hz to 300 kHz. From the experimental results, it is confirmed that the measuring device can count at least 10,000 pulses in a second.
