The water quality of offshore waters around the Korean coast was evaluated by bioassay using gametes, embryoss and early development systems of a sea urchin species Hemicentrotus pulcherrimus. The results show that despite the inflows of several river systems, the west coast maintain the grade II water. This is thought to be due to the decrease in pollutant input resulting from the purification of inflow rivers (e.g., the control of waste water discharge and the construction of sewage treatment facilities), and the dilution of pollutants by a strong tidal mixing with a large difference between the ebb and flood tides. However, Asan, Hampyong and Chonsu bays, where circulations of water are relatively poor, and the Kunsan and Mokpo harbors which are influenced by pollutants from neighboring cities have shown the grade III water. The south coast has maintain the grade II or III because developments of sea urchins were moderately or strongly inhibited. Also, Kangjin, Duekryang, Kwangyang, Masan, and Jinhae bays where water circulations are relatively poor, show the grade III water, with strong inhibitions of the early development of sea urchins. The east coast has maintained the grade I and II due to monotonous coastlines and smooth circulation of sea water. However, Chongcho Lake shows the worst water quality due to the breakwater which is constructed to maintain port functions.
Ultrastructure of gametes in the three-spine stickleback, Gasterosteus aculeatus aculeatus was observed, utilizing light, scanning and transmission electron microscopes. The egg of three-spine stickleback is spherical and demersal type. The eggs are highly adhesived to each other but not to substrates. There are many oil droplets in vitelline membrane. The outer surface of egg envelope is arranged by mushroom-like structures and pore canals. The egg have a micropyle, sperm entry site, in the area of the animal pole. The egg envelope consists of three layers, an outer layer with high electron density, a middle layer consisting two layers and an inner layer consisting of 16 to 20 layers. In the fertilized egg envelope, the molecular weights of these components ranged from 14 kDa to 205 kDa. The molecular weights of nam protein bands are 19.4 kDa, 36.7 KDa, 39.4 kDa, 42.9 kDa, 46.1 kDa and 53.0 kDa. The head of spermatozoa is spherical shape and the acrosome is absent. The mitochondria in midpiece are arranged from one to three layers and separated from the axoneme by the cytoplasmic canal. The tail has two lateral fins and the axoneme is of the 9+2 structure.
This study was conducted in an attempt to quantify some traits that may be involved in the reproductive process of Pinus thunbergii. During the past 6 years (1995-2000), we surveyed flowering patterns of 60 P. thunbergii clones in a seed orchard which was established in Anmyon-do, 1979. From this survey, we estimated clonal contributions of male and female flowers and sexual asymmetry. Among 60 clones, 18 clones (30%) accounted for 0.37 (1999)~0.46 (1995) of clonal contribution in female flowers and 0.44 (1999)~0.57 (1995) of clonal contribution in male flowers. As compared with the previous reports in other pine species, more clones made contributions to reproductive process in P. thunbergii seed orchard. The relative effective population numbers for females (v♀) varied from 0.59 (1995) to 0.91 (1999) and those for males ( v♂) were between 0.56 (1995) and 0.83 (1998) at gamete level. This showed that the female gametes effectively contribute to the reproductive process more than did the male gametes. The relative effective population numbers at clonal level ($v_b$) were in the range of 0.72 (1995) and 0.93 (1999). More than 73% of the total clones showed values of 0.4~0.6 ($0.5{\pm}0.1$) in maleness index. The values averaged over all the clones were temporally variable with the range of 73% in 1996 and 100% in 2000. The degree of sexual asymmetry ($A_s$) ranged from 0.09 to 0.26. These values were relatively low when compared with those of other trees, suggesting that P. thunbergii seed orchard remains a high level of homogeneity in the number of male and female. All of the various types of effective population numbers were positively correlated with each other while they were negatively correlated with the degree of sexual asymmetry.

Primordial germ cell (PGC) is the progenitor cell of the germ cell lineage and eventually give rise to gametes that are responsible for creating individual organisms via a fertilization process. This means that PGC is a unique cell that can be converted into individual fish. This advantage of PGCs would make it possible to develop various applications in the field of fish bioengineering. First, PGCs may make it easier to preserve the genetic resources of fish. Cryopreservation of fish eggs or embryos has not been successfully achieved so far. Therefore, the only possible method to preserve genetic resources of fishes is to raise fish as live individuals. If PGCs isolated from various fishes could be cryopresewed, these cells could be converted into live fishes via germ-line chimera production. This is particularly useful for preserving genetic materials of endangered species. Even if the species of interest were to become extinct, it could be recovered by the transplantation of cryopreserved PGCs into the embryos of a closely related species. Another application of this technology is in what could be termed "surrogate broodstock technology". (중략)
Seasonal variation in reproductive condition of the Pacific oyster Crassostrea gigas was investigated from a suspended cultured oyster population in Gosung Bay, South Korea using histological techniques, Gametogenesis of oysters initiated in February when water temperature reached 11 to $13^\circ{C}$. Increase in oocyte size and the number resulting in follicle expansion was observed from March to May First spawning of oysters observed in mid Jun when the surface water temperature reached 22 to $25^\circ{C}$. Spawning activity of oysters extended from mid June to late September with two marked spawning peaks in June and August. Most oysters collected from October to December exhibited few residual eggs in packed follicles exhibiting a typical spent condition. No gametes were observed from December to February from oysters collected in the Bay. Gonadal development of oysters in the Bay seemed to follow a seasonal fluctuation in environmental conditions such as water temperature and food availability in the water column. Spawning of oysters in late June was in part associated with sudden drop in salinity due to vast amount of freshwater input in the Bay after the summer flooding. Sex ratio of oysters was 59.5% male and 39.8% female. Less than 1 percent (0.6%) of the oysters examined were hermaphrodite; few eggs were observed in testis.
This study is to select good diploid clones and develope them into FDR clones. Seventeen diploid clones were selected by tuber plants from the imported diploid potato seeds and from the progenies of the crosses between them. The characteristics of the selected clones were reported. Fifteen clones were crossed among them or open pollinated or bulk pollinated, and 18 selections were made from the seed progenies of these pollinations. Male and female unreduced gametes were searched by crossing 2x, 4x and by microtechnique. Only one clone, D6-21 was found to produce unreduced male gamete with the rate of 27-30% by FDR. Unreduced female gamete has not been found among these diploids.

We previously pointed out that the polluted sediment elutriate manifestly affected the early events of reproductive outputs in the Pacific oysters, Crassostrea gigas. A serial dilution of priority xenobiotic sediment elutriates determined by gas chromatography/mass spectrometry (GC/MS) were exposed to gametes of the oyster with different stress burdens to detail the maternal stress transfer to its reproductive outputs. There was an apparent critical concentration over which survival and morphogenesis were significantly affected with more profound damage in morphogenesis. The critical concentration which drives mortality and abnormal morphogenesis of the larvae corresponded to a dilution between 10 and 20% of our elutriate. The adverse effects of the early lives by the xenobiotic exposure over the critical concentration were magnified by the maternal stress from the exposed benzo(a)pyrene (BaP), one of the priority polyaromatic hydrocarbons (PAHs) during the maturation condition. These results indicate that maternal transfer of the xenobiotic compounds from oysters living in the contaminated location might represent a significant adverse effect to their larval population of wild seeds.

Bioassays using gametes of sea urchins are widely used in ecotoxicological assessments of marine environments. Since most of sea urchin species in Korean coastal water spawn from spring to autumn, bioassay with them during the winter is impossible. In the course of developing standard methods for bioassays with Korean species, we found a winter-spawning starfish, Asterias amurensis, Since reproductive mode of asteroids is similar to echinoids, the bioassay protocol for sea urchins could be applied similarly to the starfish. Here, we tested and determined several conditions for the acceptability of bioassay with A. amurensis. The least required time for formation of fertilization membrane of fertilized eggs to be easily distinguished from unfertilized ones was 60 min. The threshold of sperm to egg ratio that could make acceptable fertilization rates in controls was 3000. The allowed time for manipulation of sperm after dilution in seawater was at most 3 hr. The optimal exposure time of sperms when the response against toxicant solution was relatively stable was in the range of 20-60 min. The tolerance range of sperms to the salinity of test solution was 26-38 psu. The sensitivity of A. amurensis sperm was intermediate among marine organisms commonly used in aquatic toxicity tests. The sperm bioassay with A. amurensis can be satisfactorily applied to toxicity assessments of marine environments.
During normal early embryonic development in mammals, the global pattern of genomic DNA methylation undergoes marked. changes. The level of methylation is high in male and female gametes. Thus, we cloned the cDNA of the porcine DNA methyltransferase 1 (Dnmt1) gene to promote the efficiency of the generation of porcine clones. In this study, porcine Dnmt1 cDNA was sequenced, and Dnmt1 mRNA expression was detected by reverse transcription-polymerase reaction (RT-PCR) in porcine tissues during embryonic development. The porcine Dnmt1 cDNA sequence showed more homology with that of bovine than human, mouse, and rat. The complete sequence of porcine Dnmt1 cDNA was 4,774-bp long and consisted of an open reading frame encoding a protein of 1611 amino acids. The amino acid sequence of porcine DNMT1 showed significant homology with those of bovine (91%), human (88%), rat (76%), and mouse (75%) Dnmt1. The expression of porcine Dnmt1 mRNA was detected during porcine embryogenesis. The mRNA was detected at stages of porcine preimplantation development (1-cell, 2-cell, 4-cell, 8-cell, morula, and blastocyst stages). It was also abundantly expressed in tissues (lung, ovary, kidney and somatic cells). Further investigations are necessary to understand the complex links between methyltransferase 1 and the transcriptional activity in cloned porcine tissues.
Initiation and activation of sperm motility are prerequisite processes for the contact and fusion of male and female gametes at fertilization. The phenomena are under the regulation of cAMP and $Ca^{2+}$ in vertebrates and invertebrates. Mammalian sperm requires $Ca^{2+}$ and cAMP for the activation of sperm motility. Cell signaling for the initiation and activation of sperm motility in the ascidians and salmonid fishes has drawn much attention. In the ascidians, the sperm-activating and attracting factors from unfertilized egg require extracellular $Ca^{2+}$ for activating sperm motility and eliciting chemotactic behavior toward the egg. On the other hand, the cAMP-dependent phosphorylation of protein is essential for the initiation of sperm motility in salmonid fishes. A decrease of the environmental $K^+$ concentration surrounding the spawned sperm causes $K^+$ efflux and $Ca^{2+}$ influx through the specific $K^+$ channel and dihydropyridine-sensitive L-/T-type $Ca^{2+}$ channel, respectively, thereby leading to the membrane hyperpolarization. The membrane hyperpolarization induces synthesis of cAMP, which triggers further cell signaling processes, such as cAMP-dependent protein phosphorylation, to initiate sperm motility in salmonid fishes. This article reviews the studies on the physiological mechanisms of sperm motility and its cell signaling in aquatic species.
