Purification and Characterization of Storage Protein-1 from Galleria mellonella (꿀벌부채명나방(Galleria mellonella L.)의 저장단백질-1(storage protein-1)의 정제 및 물리화학적 연구)

  • Storage protein-1 (SP-1) of Gallerio mellonella was identified in hemolvmph and fat body by electrophoresis. SP-1 was purified from hemolvmph by KBr density gradient ultracentrifugation , DEAE-cellulose (DE52) ion-exchange chromatography, and gel permeation chromatography (Sephadex G-200). Purity of SP-1 was confirmed by Non-SDS PAGE and electron microscope. SP-1 is 9.4 nm in diameter and regular octahedron in shape. SP-1 has isoelectric point of 5.7 and native molecular weight of 365 K dalton and is composed of one type of subunit with molecular weight of 82 K dalton. Ttiacylslvcerol and phospholipid were found to be maior lipid components in SP-1.

  • Male specific protein (MSP) was identified and purified from the hemolymph of Galleria melloneffa L. by electrophoresis and anion exchange chromatography. MSP has a native molecular weight of 55 kDa as determined by gel filtration chromatography and consists of a single unit with the apparent molecular weight of 27 kDa and has the pl of approximately 5.8. MSP is present in the hemolymph from day 8 pupae throughout the male adult. MSP was also found in pupal fat body, adult fat body, and adult testis.

  • Uptake and accumulation of vitellogenin into ovary were traced by radiolabeled vitellogenin produced in the fat body. Vitellogenin reached the oocytes through the follicle cells and the intercellular space and penetrated the oocyte membrane via the endocytotic vesicles and then accumulated to the growing yolk granules.

Development of Economical Artificial Diets for Greater Wax Moth, Galleria mellonella (L.) (꿀벌부채명나방[Galleria mellonella (L.)] 사육을 위한 경제적 인공사료 개발)

  • This research was carried out to develop economical artificial diets of the greater wax moth, Galleria mellonella (L.). Diets were mixed with malt or sugar instead of whole honey for cheaper ones. fecundity, developmental period, pupation rate, emergence rate and adult longevity of G. mellonella depending on diet were examined. The head width and length of larvae were not significantly different among diets tested. However, the weight of larva was the heaviest, 255.5 mg, at 1/2 honey + 1/2 malt diet and lightest, 144.3 mg, at sugar diet. The weight of pupa was also the heaviest at 1/2 honey + 1/2 malt representing 196.7 mg. Larval period was the shortest at honey diet as 30.9 days but the longest at sugar diet as 36.5 days. Pupation and emergence rates were over 79.3% at all diets. The longevity of mated females was ranged from 6.3 to 8.0 days and those of mated males ranged from 7.9 to 11.2 days. The highest number of eggs, 1269, was obtained at sugar diet. Artificial diets replaced by rice bran, malt, and sucrose reduced costs compared with wheat bran plus honey diet. The cost of rice bran was only 10% of wheat bran. Honey was much more expensive than malt and sugar, that is 6 and 13 times, respectively.

Purification and Characterization of Apolipophorin-III in the Hemolymph of the War Moth, Galleria mellonella L.

  • Two molecular species of apolipophorin-III (spoLp-III) were purified from the last instar larval hemolymph of Galleria mellonella by gel permeation chromatography (Sephadex G-100), ion exchange chromatography (DE-52), heat treatment (90C for 30 min) and Mono S FPLC, and were named apoLp-III-a and apoLp-lll-b, respectively. They were indistinguishable by SDS-PAGE but could be separated by native PAGE. The molecular mass of apoLp-III determined by SDS-PAGE was approximately 18 kDa. The N-terminal amino acid sequence of apoLp-III-b revealed high similarities with the apoLp-III from Manduca sexta.
