Optimization of Tannase Production by Aureobasidium pullulans DBS66

  • Tannase production by Aureobasidium pullulans DBS66 was optimized. The organism produced maximum tannase in the presence of 1% tannic acid after 36 h. Maximum gallic acid accumulation was observed within 36 h and tannic acid in the fermented broth was completely degraded after 42 h of growth. Glucose had a stimulatory effect on tannase synthesis at 0.1% (w/v) concentration. The organism showed maximum tannase production with $(NH_4)_2HPO_4$ as nitrogen source. Shaking speed of 120 rpm and 50-ml broth volume have been found to be suitable for maximum tannase production.

Expression and Localization of Inulinase in Recombinant Saccharomyces cerevisiae (재조합 Saccharomyces cerevisiae에서 Inulinase의 발현과 국재성)

  • Inulinase of Kluyveromyces marxianus origin was produced by recombinant yeast Saccharomyces cerevisiae under the control of GAL1 promoter, to examine the expression and localization of inulinase in a repressed(galactose-free) or derepressed(galactose-containinga) medium. The inulinase gene(INU1A) was constitutively expressed at 6.7 units/ml in a repressed medium. When the cell started to utilize galactose in a derepressed medium, the INU1A gene began to be expressed, and the final expression level reached about 45 units/ml. According to be the nondenaturingPAGE analysis, inulinase produced by S. cerevisiae was found to be less glycosylated than the bakers yeast invertase. In addition, its glycosylation pattern was less heterogeneous than the K. marxianus inulinase. The supplementation of inulin or raffinose into the derepressed medium increased the cell growth rate, while the expression of INU1A was repressed. Regardless of the carbon sources examined, most of inulinase activity (more than 98%) was found in the extracellular medium, indicating excellent secretion efficiency.

  • As a result of the transfer function analysis on the casual relationship between Andong Dam water level and fog time in Andong City from March of 1981 to December of 1992, it should be referring to monthly increases in the water level having an effect on 0.89 hour per m increases in the fog time with 4 months delay. It is evident, hence, the artificial lake of the dam is one of the direct causes of fog. Based upon this objective evidence, water resources development policy and dam managment policy must be formulated.

  • A new class of soluble PPV derivatives containing dimethyldodecylsilylphenyl unit as a pendant was synthesized by Gilch polymerization method. The resulting electroluminescent (EL) polymers showed good solubility, good film-forming ability onto the ITO substrate, and exhibited an amorphous morphology due to dimethyldodecylsilylphenyl branched group linked to the polymer backbone. The weight average molecular weights and polydispersities of the present EL polymers were in the range of 8.0-80.0 x $10^{4}$ and 2.67-7.80, respectively. The resulting EL polymers revealed a high thermal stability of up to $355-410^{\circ}C$. Their glass transition temperatures were in the range of $104-251^{\circ}C$. The emission colors could be tuned from green to orange-red colors by changing the MEH-PPV contents in copolymer systems. The turn-on voltages of the EL polymers were in the range of 1.8-4.0 V.

  • Dynamic response of ground due to train loads is analyzed. A numerical procedure based on finite element method is used to solve two-dimensional ground response. Dynamic train loads assumed in analysis is the point acceleration on train rail with magnitude of 2330 gal and thickness of surface of soil layer assumed is 60cm. In order to consider the effect of acceleration point, dynamic responses such as response acceleration and displacement are computed as a function of distance from acceleration point on rail. In addition, simple methods which reduce dynamic effects on ground are also proposed.

  • The present study was conducted to assess gene expression of bacterial lacZ driven by the SV40 promoter at early developmental stages of bovine embryos. The lacZ gene was linearized with BamHI digestion and introduced into the pronucleus by microinjection at 20 hrs after the commencement of in vitro fertilization. Intact bovine blastocysts were not stained with X-Gal, suggesting that there is no endogenous beta-galactosidase activity in these blastocysts. In contrast, the bovine blastocyst cells microinjected with the lacZ gene exerted a characteristic greenish-blue color originating from the bacterial beta-galactosidase activity, albeit at a low rate, i.e. 2.1% of the total fertilized oocytes injected. It was concluded, therefore, that the lacZ gene driven by the SV40 promoter could be used for an indirect screening method in which the presence of transgene is evaluated from the product of transgene expression.

  • To investigate antitumor components of Korean higher fungi, the carpophores of Ganoderma lucidum (Fr.)Karst. were collected at Gal-mae-ri in Gyeong-gi Province and extracted with 0.1N sodium hydroxide solution. The extract was purified by dialyzing through Visking tube and a dark brownish power was obtained. The antitumor activity of the fraction was tested against sarcoma 180 implanted in mice. The tumor inhibition ratio of the fraction against the tumor was 87.6% in doses of 50 mg/kg/day for the period of ten days. The tumor in five of 15 mice was completely regressed. The antitumor fraction was chemically analyzed and found to be a complex of polysaccharide and protein, from which four monosaccharides and eighteen amino acids were identified.

Identification of a Cellular Protein Interacting with Murine Retrovirus Gag Polyproteins

  • The retroviral Gag polyprotein directs the assembly of virion particles and plays an important role in some events after entry into a host cell. The Gag polyprotein of a virus mixture is responsible for inducing murine acquired immunodeficiency syndrome (MAIDS) when injected into susceptible strains of mice. In order to identify the host cellular proteins which interact with the MAIDS virus Gag proteins and possibly mediate the function of the Gag proteins, mouse T-cell leukemic cDNA expression library was screened using the yeast GAL4 two hybrid system. Of 11 individual positive clones, the clone Y1 was selected for the study of protein-protein interaction. Its DNA sequence revealed that it was an exact match to the murine SH3 domain-containing protein SH3P8. It is expressed as 2.4 kbp transcripts in testis at higher levels and in various tissues tested at lower levels. Glutathione S-transferase-Y1 fusion protein binds tightly to $Pr60^{def-gag}$ as well as $Pr65^{eco-gag}$.

  • Let $d$ be a positive integer with $d\not{\equiv}2$ mod 4, and let $K=\mathbb{Q}({\zeta}_{pd})$ for S an odd prime $p$ such that $p{\equiv}1$ mod $d$. Let $K_{\infty}={\bigcup}_{n{\geq}0}K_n$ be the cyclotomic $\mathbb{Z}_p$-extension of $K=K_0$. In this paper, explicit generators for the Tate cohomology group $\hat{H}^{-1}$($G_{m,n}$ are given when $d=qr$ is a product of two distinct primes, where $G_{m,n}$ is the Galois group Gal($K_m/K_n$) and $C_m$ is the group of cyclotomic units of $K_m$. This generalizes earlier results when $d=q$ is a prime.

  • A 4 year-old Doberman Pinscher was presented with periocular skin lesions which has developed after climbing a mountain with the client. Cytologic preparations of the periocular skin lesions revealed marked eosinophilic infiltration, and a few neutrophils, lymphocytes and epithelial cells. These findings are consistent with eosinophilic furunculosis of the face typically in medium breed dogs. The patient was administered orally with prednisolone 1mg/kg for 2 weeks. The skin lesions were regressed responsively. The patient has no recurrent signs.
